Abstract:MicroRNAs (miRNAs) play an important role in several major events during life, such as embryonic development, cellular differentiation, proliferation, apoptosis, and metabolism. The mis-regulation of miRNAs is linked to the development of cancer. In breast cancer, the expression of miRNAs is frequently deregulated, and specific miRNAs have been identified as oncogenes or tumor suppressors that modulate all of the stages of breast cancer progression. The unique properties of miRNAs suggest that they may be useful for the early clinical diagnosis, classification, and grading of breast cancer. Additionally, miRNAs may aid in determining prognosis and patient response to treatment. A comparative analysis of miRNA expression between physiologic and pathophysiologic states and a clarification of the role of miRNAs in breast cancer development are essential for determining the mechanism underlying miRNA-mediated cancer development and progression. Such information may uncover new objectives and methodologies for the treatment of breast cancer. Recently, miRNA-related breast cancer studies have shifted from the examination of single molecules to the exploration of entire pathways or even entire networks. Studies have also shifted from examining the single cell phenotype to examining tissues as a whole throughout the multiple stages of breast cancer development. Moreover, extracellular miRNAs have been identified and are being thoroughly studied. In the tumor microenvironment, we found that miRNAs secreted by macrophages can promote breast cancer cell invasion. This finding reveals a new mechanism by which exogenous miRNAs can regulate the progression of breast cancer. In this chapter, we will discuss the emerging role of miRNAs in breast cancer with respect to the following: (1) miRNAs as oncogenes and tumor suppressors in breast cancer; (2) miRNAs in breast cancer metastasis, including the function of miRNAs in breast cancer stem cells; and (3) exogenous and secretory miRNAs. Specific examples from each category will be described.
