Abstract:The first phosphine-catalyzed enantioselective γ-addition with prochiral nucleophiles and 2,3-butadienoates as the reaction partners has been developed. Both 3-alkyl- and 3-aryl-substituted oxindoles could be employed in this process, which is catalyzed by a chiral phosphine that is derived from an amino acid, thus affording oxindoles that bear an all-carbon quaternary center at the 3-position in high yields and excellent enantioselectivity. The synthetic value of these γ-addition products was demonstrated by the formal total synthesis of two natural products and by the preparation of biologically relevant molecules and structural scaffolds.

Abstract:The enantioselective 1,4-addition of diarylphosphine oxides to α,β-unsaturated N -acylindoles and N -acylpyrroles catalyzed by a commercial chiral oxazaborolidine is reported. This methodology tolerates a variety of substrates and diarylphosphine oxides, leading to high yields and enantioselectivities. Importantly, the resulting chiral products exhibit excellent transformation abilities to form corresponding acids, amides, and esters with high potential applications in the synthesis of chiral phosphine ligands.

Abstract:The first asymmetric Michael addition of 3-substituted phthalides to nitroolefins promoted by amino acid-incorporating multifunctional catalysts has been developed. The reported method led to the synthesis of 3,3-disubstituted phthalide derivatives in high yields, and in a highly diastereoselective and enantioselective manner. Facile synthesis of a chiral bicyclic lactam has also been demonstrated.

Abstract:A chiral phosphine-catalyzed tandem Michael addition of tryptamine-derived oxindoles to ynones has been developed, which provides facile access to a series of optically enriched spiro[pyrrolidine-3,3 '-oxindole] compounds in good yields with good to excellent enantio- (up to 97% ee) and diastereoselectivities (up to 20 : 1 dr). Mechanistic studies indicate that this tandem reaction proceeds via an intermolecular aza-Michael/intramolecular Michael addition process.

Abstract:An efficient bifunctional cinchona alkaloid derived thiourea-promoted enantioselective conjugate addition of nitroalkanes to indolylidenecyanoacetates has been developed under neat conditions. The process leads to synthetically interesting densely functionalized 3,3'-disubstituted oxindoles with creation of up to three stereogenic centers.

Abstract:Over the past decades, asymmetric catalysis has been intensely investigated as a powerful tool for the preparation of numerous chiral biologically active compounds. However, developing general and practical strategies for preparation of both enantiomers of a chiral molecule via asymmetric catalysis is still a challenge, particularly when the two enantiomers of a chiral catalyst are not easily prepared from natural chiral sources. Inspired by the biologic system, we report herein an unprecedented catalytic enantiodivergent Michael addition of pyridazinones to enones by subtle adjustment of achiral amino moiety of dipeptide phosphine catalysts. These two dipeptide phosphine catalysts, P5 and P8, could deliver both enantiomers of a series of N2-alkylpyridazinones in good yields (up to 99%) with high enantioselectivities (up to 99% ee) via the catalyst-controlled enantiodivergent addition of pyridazinones to enones.

Abstract:Here we introduce a metal‐free, catalytic and enantioselective strategy from α,β‐unsaturated 2‐acyl imidazoles to the chiral phosphorous 2‐acyl imidazoles. Interestingly, this methodology was catalyzed by the classical and commercial oxazaborolidine under mild conditions. This strategy features a wide range of substrates scope with good yields and excellent enantioselectivities. The possible mechanism further suggests the key of this reaction through the cleavage of diarylphosphine oxides using Frustrated Lewis Pairs theory.

Abstract:Chiral diamine catalysts 11a-e derived from α,α-diphenyl prolinol were prepared and successfully applied to the Michael addition of aromatic oximes to α,β-unsaturated aldehydes in mediocre to good yields (up to 78%) and good to high enantioselectivities (up to 93% ee).

Abstract:A highly efficient synthesis of chiral 3,3-disubstituted oxindoles was developed using a chiral spirocyclic phosphoric acid catalyzed 1,6-conjugate addition reaction of para-quinone methides derived from N-unprotected isatins with indoles. The reaction proceeds under mild reaction conditions to provide indole-containing N-unprotected oxindoles bearing quaternary stereocenters in good yields and with moderate to excellent enantioselectivities (up to 97% ee).

Abstract:Enantioselective hydroxylation of N-protected 3-substituted oxindoles has been developed via chiral iminophosphorane catalysis with oxaziridines as oxidants. As such, a variety of optically active 3-substituted-3-hydroxy-2-oxindoles were obtained in excellent yields (91-99%) and moderate to excellent level of enantiomeric excess (up to 94% ee). Abstract:A highly efficient organocatalyzed Michael addition of 2-oxindole-3-carboxylate esters to nitroolefins using a Cinchona alkaloid-thiourea and an achiral Bronsted acid as cooperative organocatalysts is reproted that affords significantly improved enantioselectivity and diastereoselectivity. It also provides an efficient approach to the synthesis of spirooxindole derivatives with high enantioselectivity.

Abstract:The enantioselective organocatalytic phospha-Michael reaction of alpha,beta-unsaturated aldehydes with diaryl phosphine oxides is explored for the first time using (S)-2-(diphenyl(trimethylsilyloxy) methyl) pyrrolidine as a catalyst. Abstract:The rhodium-catalyzed diastereo- and enantioselective Michael addition of arylboronic acids to 3-alkylenyloxindoles has been developed with (R)-binap as a ligand. A wide variety of the desired functionalized oxindoles are smoothly obtained in high yields (up to 99%) with high enantioselectivities (up to 92% ee) and good diastereoselectivities (up to 82:18).

Abstract:The first example of a photoexcitated amine-catalyzed process for asymmetric Michael addition of o-quinodimethanes to enones is described. In the presence of simple chiral amino acid esters, a variety of Michael adducts were generally obtained in good yields and excellent stereoselectivities. This strategy can be successfully applied to 3-substituted-2-cyclohexenones and provides an asymmetric access to all-carbon quaternary centers. Furthermore, the high stereocontrol was explained by means of density-functional theory (DFT) calculations.

Abstract:Chiral nitrogen-containing molecules such as chiral amines, azetidines, and 2-substituted tetrahydroquinolines are important privileged scaffolds in medicinal chemistry. In this paper, an efficient and highly enantioselective chiral phosphoric acid catalyzed asymmetric addition of 2-(vinyloxy)ethanol to imines has been developed for the first time, providing the corresponding chiral amines containing dioxolane acetals that can transform into useful chiral N-heterocycles including azetidines and 2-substituted tetrahydroquinolines with excellent optical purity.
