Abstract:Portable, low-cost and quantitative detection of a broad range of targets at home and in the field has the potential to revolutionize medical diagnostics and environmental monitoring. Despite many years of research, very few such devices are commercially available. Taking advantage of the wide availability and low cost of the pocket-sized personal glucose meter—used worldwide by diabetes sufferers—we demonstrate a method to use such meters to quantify non-glucose targets, ranging from a recreational drug (cocaine, 3.4 µM detection limit) to an important biological cofactor (adenosine, 18 µM detection limit), to a disease marker (interferon-gamma of tuberculosis, 2.6 nM detection limit) and a toxic metal ion (uranium, 9.1 nM detection limit). The method is based on the target-induced release of invertase from a functional-DNA–invertase conjugate. The released invertase converts sucrose into glucose, which is detectable using the meter. The approach should be easily applicable to the detection of many other targets through the use of suitable functional-DNA partners (aptamers, DNAzymes or aptazymes).

Abstract:Heavy metal pollution of water can pose a serious threat to public health. Because pollution can occur at multiple stages of water distribution, portable on-site detection methods are highly desirable for routine monitoring of water contamination. Here, we developed a method that enables the detection of heavy metal ions using a personal glucose meter (PGM), a readily accessible analytical device. The method utilizes a cell-free genetic circuit that generates glucose in response to heavy metal ions. The cell-free protein synthesis reaction mixture includes template DNA encoding the invertase enzyme under the control of operators for heavy metal-responsive transcription factors. When supplied with heavy metal ions cognate to the transcription factors, the reaction mixture successfully generated functional invertase, which subsequently hydrolyzed sucrose to glucose. The resulting glucose levels measured using a PGM correlated well with the concentrations of the heavy metal ions, supporting the feasibility of this method to detect water pollution without sophisticated equipment. Our results demonstrate a successful example of harnessing cell-free protein synthesis to transform analytes into surrogate molecules that can be readily measured with generic analytical devices. We expect that the method described here could be adapted for widespread application in numerous fields in which routine monitoring of analytes is required.

Fully Integrated Battery-Free and Flexible Electrochemical Tag for On-Demand Wireless in Situ Monitoring of Heavy Metals

Abstract:Migration of heavy metals from food contact materials to contained foods and drinks will cause severe health problems. The in situ analysis of the heavy metal ions inside these containers is in high demand. In this study, a fully integrated battery-free and flexible electrochemical tag is presented for wireless in situ detections of heavy metal ions in various containers. The circuit part of the tag is integrated with square wave anodic stripping voltammetry (SWASV) technique and near field communication (NFC) module, enabling on-board electrochemical sensing, wireless data transmission, and power harvesting. The circuits showed high consistency with commercial electrochemical workstation in SWASV analysis. A screen-printed flexible electrode array is connected with the circuit board. The working electrodes were modified with gold and bismuth nanoparticles respectively, which showed high sensitivity, selectivity, linearity, and reproducibility in the detections of lead and cadmium. The tag achieved quantitative detections of heavy metal ions in various containers, including enamelled cup, ceramic teapot, and dark-red enameled pottery. With proper encapsulation, the miniaturized passive and flexible tag can be attached to the inside surface of various containers for a long time. The wireless detections of heavy metal ions can be achieved just with an NFC-enabled smartphone outside the container. The platform provides a simple and effective solution for in situ analysis of toxic and harmful substances, which can be widely used in the areas of food safety and water quality monitoring.

Abstract:A smart voltammetric procedure in an original automatic trace metal monitoring system is presented in this study. Its aim is to expand the measurement range of a gold-band microelectrode array (MEA) for monitoring heavy metal ions such as Zn2+, Pb2+ and Cu2+. A voltammetric controller to execute a smart voltammetric procedure and a homemade mercury-free gold-band MEA are introduced. Three particular features are included in the smart procedure, which are self-calibration, multi-standard solution fluidics control and automatic adjustment of deposition time. The measurement range of a gold-band MEA is investigated by a traditional voltammetric set-up. Experiments show good linearity from 10 to 50 mu g L-1 for Zn2+, Pb2+ and Cu2+; the MEA detection limit for Zn2+, Pb2+ and Cu2+ is about 0.4, 1 and 0.4 mu g L-1, respectively. Moreover, self-calibration, multi-standard solution fluidics control and automatic adjustment of deposition time are introduced in smart voltammetric measurement with the method of standard additions. The upper concentration limit can be increased to twice that of conventional measurements; the detection limit for Zn2+, Pb2+ and Cu2+ can reach 0.1, 0.2 and 0.1 mu g L-1, respectively. In addition, this design has been validated by monitoring natural concentrations of Zn2+, Pb2+ and Cu2+ (West Lake, China) and comparing with atomic absorption spectrometry.
On Site Determination of Heavy Metal Ions with Portable Electrochemical Instrument Based on Smartphone

Abstract:Introduction Most heavy metal ions are harmful to human body and may cause a variety of diseases, among which chromium(VI), cadmium, arsenic and their compounds are classified as Group 1 (human carcinogen) by IARC [1]. Heavy metal ions can enter the human body through direct drinking and the enrichment of food chain. Therefore, the allowed concentration of heavy metal ions are recommend by WHO, which are 50 μg/L for chromium(VI), 3 μg/L for cadmium, 10 μg/L for arsenic, respectively [2]. Compared with other methods, electrochemical method is gaining wide recognition and has the advantages of fast speed, simple operation and possibility of use in portable detection on site [3]. Therefore, a portable electrochemical instrument based on smartphone was designed, in order to detect the concentration of heavy metal ions in the field. Method A block diagram of the portable electrochemical instrument is shown in Fig. (a). The instruction of controlling electrochemical process is sent out by MCU and executed by potentiostat. The electrochemical signal obtained is simply processed and returned to MCU. The communication between MCU and smartphone software based on Android system is realized through Bluetooth. The completed printed circuit board (PCB) of the instrument is shown in Fig. (b). The determination of chromium(VI) is realized by gold nanoparticles modified gold electrode, in order to obtain a larger electrochemical response. The gold nanoparticle modification was achieved by applying a potential of -0.3 V for 120 seconds with stirring in 0.2 mmol/L chloroauric acid with 0.5 mol/L sulphuric acid, which is simple, stable and repeatable for on site determination. As shown in Fig. (c), the increase of electrode surface area can be observed by Atomic Force Microscope (AFM). After parameters optimization, the detection of Chromium(VI) was carried out in 0.3 mol/L nitric acid by linear sweep voltammetry (LSV) with a potential range of 0.8 V to 0.2 V. There is no accumulation process during detection, so it takes less than 10 seconds. Results and Conclusions The linear sweep voltammograms in chromium(VI) concentration range of 10 μg/L to 100 μg/L with a step of 10 μg/L are shown in Fig. (d). The standard curve of Cr(VI) is shown in Fig. (e), where it shows a linear relationship with the correlation coefficient of 0.98. A detection limit of 6.4 μg/L can be calculated, suggesting the sensitivity of the method for Cr(VI) detection. These results indicate that the portable electrochemical instrument based on smartphone can be a promising alternative for heavy metal ions detection on site. Abstract:A portable, quantitative and selective DNA detection biosensor requiring only one step for target recognition and signal reporter generation using a loop-based DNA competitive hybridization assay and personal glucose meter (PGM) was developed. In the presence of target DNA, due to its competitive binding, the invertase-DNA conjugate was released which could be collected with the help of a magnet subsequently. The released invertase-DNA was used to catalyze the hydrolysis of sucrose into glucose with millions of turnovers which was target concentration dependent. We found that there was a linear relationship between the PGM signals and the target DNA concentration in the range of 100 pM to 100 nM with a detection limit of 100 pM. In addition, the sensor exhibits excellent sequence selectivity, being able to differentiate a single mismatch in the target DNA, and excellent anti-interference ability, having almost no effect in serum on the detection performance. The biosensor reported here is easier to operate, owning great potential in point of care testing in environments with limited resources and skilled personnel for rapid and sensitive detection of specific DNA sequence in real biological samples.

Abstract:DNA detection is commonly used in molecular biology, pathogen analysis, genetic disorder diagnosis, and forensic tests. While traditional methods for DNA detection such as polymerase chain reaction (PCR) and DNA microarrays have been well developed, they require sophisticated equipment and operations, and thus it is still challenging to develop a portable and quantitative DNA detection method for the public use at home or in the field. Although many other techniques and devices have been reported to make the DNA detection simple and portable, very few of them are currently accessible to the public for quantitative DNA detection because of either the requirement of laboratory-based instrument or lack of quantitative detection. Herein we report application of personal glucose meters (PGMs), which are widely available, low cost, and simple to use, for quantitative detection of DNA, including a hepatitis B virus DNA fragment. The quantification is based on target-dependent binding of cDNA-invertase conjugate with the analyte DNA, thereby transforming the concentration of DNA in the sample into glucose through invertase-catalyzed hydrolysis of sucrose. Instead of amplifying DNA strands through PCR, which is vulnerable to contaminations commonly encountered for home and field usage, we demonstrate here signal amplifications based on enzymatic turnovers, making it possible to detect 40 pM DNA using PGM that can detect glucose only at the mM level. The method also shows excellent selectivity toward single nucleotide mismatches.
Portable Detection of Copper Ion Using Personal Glucose Meter

Abstract:A simple and sensitive method for Cu2+ detection was developed using the Cu+-catalyzed alkyne–azide cycloaddition reaction, Fe3O4 magnetic nanoparticles (MNPs) as the reaction platform, and a portable blood glucose meter (PGM) as the detection method. Gold nanoparticles (AuNPs) were labeled with glucose oxidase (GOx) and alkyne-functionalized, terminally thiolated ssDNA (C2). In the presence of Cu2+ and ascorbate, the functionalized AuNPs were captured onto MNPs modified with azide-functionalized ssDNA (C1) via the Cu+-catalyzed alkyne–azide cycloaddition reaction. The GOx on the AuNPs' surface oxidized glucose (Glu) into gluconic acid and H2O2, reducing the Glu content in the reaction solution, which was quantitatively detected by the PGM. Under optimal conditions, the PGM response of the system showed a good linear relationship with the logarithm of Cu2+ concentration in the range of 0.05 to 10.00 μmol/L, with a detection limit of 0.03 μmol/L (3σ). In actual tap water samples, the spiked recovery rate of Cu2+ was between 92.30% and 113.33%, and the relative standard deviation was between 0.14% and 0.34%, meeting the detection requirements for Cu2+ in real water samples.

Abstract:Personal glucose meter (PGM) is one of the most commercially available POC (point-of-care) devices for monitoring the level of glucose reliably, yet its non-glucose quantification ability is limited since such strategy needs ingenious interface design and tedious enzyme conjugation. Herein, we constructed a portable and sensitive platform that can detect non-glucose target by combining enzyme-encapsulated zeolitic imidazole framework-90 (ZIF-90) with personal glucose meter. ZIF-90 is an ideal carrier and susceptor due to the extraordinary capability of packaging enzyme and stimuli-responsiveness. We selected adenosine-5'-triphosphate (ATP) as the target model of non-glucose analytes. Upon ATP-induced decomposition of MOF, the released enzyme (glucose oxidase or invertase) catalyzed substrate and gave rise to the change of the glucose concentration for PGM assay. This method determined ATP with a remarkably sensitivity of 233 nM and effective recovery in real serum samples. Our strategy provides a facile and practical approach for measuring the non-glucose target using PGM, and could potentially be applied in bimolecular detection in point-of-care diagnosis.

Abstract:Developing portable and low-cost methods for quantitative detection of large protein biomarkers and small molecular toxins can play a significant role in controlling and preventing diseases or toxins outbreaks. Despite years of research, most current methods still require laboratory-based or customized devices that are not widely available to the general public for quantitative analysis. We have previously demonstrated the use of personal glucose meters (PGMs) and functional DNAs for the detection of many nonglucose targets. However, the range of targets detectable by functional DNAs is limited at the current stage. To expand the range of targets that can be detected by PGMs, we report here the use of antibodies in combination with sandwich and competitive assays for quantitative detection of protein biomarkers (PSA, with a detection limit of 0.4 ng/mL) and small molecular toxins (Ochratoxin A, with a detection limit of 6.8 ng/mL), respectively. In both assay methods, with invertase conjugates as the link, quantitative detection is achieved via the dependence between the concentrations of the targets in the sample and the glucose measured by PGMs. Given the wide availability of antibodies for numerous targets, the methods demonstrated here can expand the range of target detection by PGMs significantly.
Using a Glucose Meter to Quantitatively Detect Disease Biomarkers Through a Universal Nanozyme Integrated Lateral Fluidic Sensing Platform.

Abstract:Along with the advance in medical research, more biomarkers emerge as useful indicators for both disease and health index. However, majority of them have no practical or economic testing methods available yet, or rely on high-costing methods such as Enzyme-Linked Immuno-Sorbent Assay (ELISA), High-Performance Liquid Chromatography (HPLC), Mass Spectrum, and immunohistochemistry (IHC). In this article, we develop a universal nanozyme integrated testing platform for biological molecules that incorporates the electrochemical measurement of glucose with lateral flow immunostrip (LFS) for target analytes. This design involves the quantitative conversion of analytes into invertase and then glucose, which can be measured by an extremely affordable meter. The feasibility of this design was validated using 8-hydroxy-2'-deoxyguanosine (8-OHdG) and prostate specific antigen (PSA) as representatives for small molecules and moderate to large proteins respectively. Our approach yields results comparable to commercial diagnostic ELISA kits at a substantially reduced cost and reaction time. Specifically, the design has a detection limit of 0.23 ng mL(-1) for 8-OHdG and 1.26 ng mL(-1) for PSA, and a detection range of 0.1-100 ng mL(-1) for 8.0HdG and 1-100 ng mL(-1) for PSA. By combining the accessibility of well-established glucose testing and LFS, our design can serve as a point of care testing method that can be fully integrated into the personal lifestyle without requiring professional assistance.

Abstract:Self-powered electrochemical biosensors based on enzyme biofuel cells (EBFC) are uniquely valuable in developing real-time and on-site detection of heavy metal ions. This work combines graphdiyne/graphene (GDY/GR) heterojunction with Deoxyribozyme (DNAzyme) trigger catalytic hairpin self-assembly (CHA) circuit. In addition, the smartphone is introduced to obtain the detection signals. The results show that the designed sensor platform can be used for single-tube ultrasensitive and visual lead ion detection with a wide linear range of 0.003–5000 nM and a low detection limit (LOD) of 0.005 nM. This design enriches the routine screening methods for heavy metal ions in food and environmental samples and introduces new idea for the development and application of self-powered electrochemical biosensors.

Abstract:Pervasive detection of blood glucose is rather critical for the real-time disease diagnosis which would provide valuable guidance for treatment planning. Here, we established a health care platform for this purpose through incorporating the glucose detection with liquid metal printed sensor and the smart phone monitoring system together. The liquid metal ink composed of bismuth indium stannic (BIS) alloy was identified as an appropriate sensor material to be quickly written or printed on polyvinyl chloride (PVC) substrate at around 59 °C to form desired electrodes. It thus eliminated the complicated procedures as usually required in conventional sensor fabrication strategies. The alloy electrodes were characterized via cyclic voltammetry to demonstrate their practical functionality. Further, unlike using the commonly adopted glucometer, a smart phone was developed as the data acquisition and display center to help improve the portability and ubiquitous virtue of the detection system. Glucose solution in different concentrations was assayed via this platform. It was shown that there is a good linear relationship between the concentration and the integral value of the curve recorded by the mobile phone, which confirms the feasibility of the present method. This quantitative point-of-care system has pervasive feature and is expected to be very useful for future low-cost electrochemical detection.

Abstract:We herein developed a label-free and washing-free method to detect biological thiols (biothiols) on a personal glucose meter (PGM) utilizing the intrinsic glucose oxidase (GOx)-mimicking activity of gold nanoparticles (AuNPs). By focusing on the fact that this activity could be diminished by target biothiols through their binding onto the AuNP surface, we correlated the concentration of biothiols with that of glucose readily measurable on a PGM and successfully determined cysteine (Cys), homocysteine (Hcy), and glutathione (GSH) down to 0.116, 0.059, and 0.133 μM, respectively, with high specificity against non-target biomolecules. We further demonstrated its practical applicability by reliably detecting target biothiol in heterogeneous human serum. Due to the meritorious features of PGM such as simplicity, portability, and cost-effectiveness, we believe that this work could serve as a powerful platform for biothiol detection in point-of-care settings.

Abstract:Real-time, on-site detection and quantification of different trace analytes is a challenge that requires both searching a general class of molecules to recognize a broad range of contaminants and translating this recognition to easily detectable signals. Functional nucleic acids, which include DNAzymes (DNA with catalytic activity) and aptamers (nucleic acids that bind an analyte), are ideal candidates for the target recognition. These nucleic acids can be selected by a combinatorial biology method called in vitro selection to interact with a particular analyte with high specificity and sensitivity. Furthermore, they can be incorporated into sensors by attaching signaling molecules. Due to the high extinction coefficients and distance-dependent optical properties, metallic nanoparticles such as the commonly used gold nanoparticles have been shown to be very attractive in converting analyte-specific functional DNA into colorimetric sensors. DNAzyme directed assembly of gold nanoparticles has been used to make colorimetric sensors for metal ions such as lead, uranium, and copper. To make the operation even easier and less vulnerable to operator's errors, dipstick tests have been constructed. Here, we describe protocols for the preparation of DNAzyme-linked gold nanoparticles (AuNP) that are then immobilized on to lateral flow devices to make easy-to-use dipstick tests for metal ions.

Herein, taking advantage of the pocket size, reliable quantitative results and simple operation of the pocket-sized personal glucose meter (PGM), we report an approach of circulating microRNA-21 (miR-21) detection with high precision and low cost. Via target-induced release of invertase from the DNA-invertase conjugate, which could convert sucrose into glucose, the detection of miR-21 in serum was linked to PGM readings. Combining the DNAzyme feedback amplification (DFA) program and highly efficient enzymatic turnover, an ultralow detection limit of 7 × 10-16 M for miR-21 was achieved using a PGM as the reporter. The high sensitivity and selectivity of the proposed method meets the requirement of quantifying cell free nucleic acids in serum. In addition, this approach fills the shortage of quantitative RT-PCR and next-generation sequencing in quantifying miRNAs with a short length and greatly reduces the cost of detection. We believe that widely used personal diagnosis devices could hold an important place in the booming area of liquid biopsy.

Abstract:Nanomaterials have shown great promise in advancing biomedical and environmental analysis because of the unique properties originated from their ultrafine dimensions. In general, nanomaterials are separately applied to either enhance detection by producing strong signals upon target recognition or to specifically extract analytes taking advantage of their high specific surface area. Herein, we report a dual-functional nanomaterial-based platform that can simultaneously enrich and enable sensitive detection of multiple metal ions. The macroporous graphene foam (GF) we prepared displays abundant phosphate groups on the surface and can extract divalent metal ions via metal-phosphate coordination. The enriched metal ions then activate the metal-responsive DNAzymes and produce the fluorescently labeled single-stranded DNAs that are adsorbed and quenched by the GF. The resultant fluorescence reduction can be used for metal quantitation. The present work demonstrated duplexed detection of Pb2+ and Cu2+ using the Pb- and Cu-responsive DNAzymes, achieving a low detection limit of 50 pM and 0.6 nM, respectively. Successful quantification of Pb2+ and Cu2+ in human serum and river water were achieved with high metal recovery. Since the phosphate-decorated GF can enrich diverse types of divalent metal cations, this dual-functional GF-DNAzyme platform can serve as a simple and cost-effective tool for rapid and accurate metal quantification in determination of human metal exposure and inspection of environmental contamination.
An efficient biosensor using a functionalized microneedle of Cu2O-based CoCu-LDH for glucose detection

Abstract:Glucose detection with small and micro volume sampling has recently received increasing attention in monitoring personal health. Herein, a cauliflower-type cluster of Cu2O nanoparticles (NPs) was directly deposited on the tip surface of a stainless steel acupuncture needle electrode (ANE) by electrochemical deposition, and then this pre-formed cuprous basis was used to further prepare the neatly arranged CoCu-layered double hydroxide (CoCu-LDH) nanosheets that interconnected to form nano-sized pores in the range from 100 to 500 nm. The microstructure and spectral characteristics of the surface modification materials were comprehensively characterized by FE-SEM, EDS, XRD, FT-IR and TEM. Cu2O-based CoCu-LDH composites with special morphology had been proven to accelerate the rate of electron transport and provide more available active centers, and moreover, the mixed valence of Cu/Co induced an excellent synergism for the electrocatalytic oxidation of glucose. As a result, CoCu-LDH/Cu2O/ANE as a sensitive glucose probe exhibited two wider linear ranges of 0.03-0.40 mM and 0.40-6.00 mM, with sensitivities of 116.13 μA mM-1 and 52.08 μA mM-1, respectively, and the detection limit as low as 0.46 μM (S/N = 3). The response time only took 3 s and it kept working stably in the interference of ascorbic acid (AA), dopamine (DA), uric acid (UA), and Cl-. In the stability test, the CoCu-LDH/Cu2O/ANE sensor exhibited a stable monitoring sensitivity after 15 days. Finally, the CoCu-LDH/Cu2O/ANE sensor had been successfully applied to glucose analysis in human serum, proving that our design was an attractive strategy for developing a portable, minimally invasive, and low-cost non-enzymatic electrochemical glucose sensing platform.
