Abstract:Objective To explore the immunomodulatory effects on intestinal tract by polysaocharide from Dendranthema mrifolium and chlorogenic acid. Methods Polysaccharide was extracted from Dendranthema mrifolium with water and ethanol;the enteric mucosa lymphocytes were isolated from rats,and co-cultured with polysaccharide and chlorogenic acid respectively.The level of IFN-γ and TNF-α in supernatants were assayed by ELISA.The immune modulatory effects on intestinal lymphocytes were compared between these two componnets. Results Polysaccharide and chlorogenic acid both increased the level of TNF-α and IFN-γ;moreover,chlorogenic acid shown more effective on IEL than polysaccharide. Conclusion Polysaccharide and chlorogenic acid both exert immune modulatory effect on lymphocytes of enteric mucosal immune system.

Abstract:The effects of L.deliciosus Polysaccharides on the immune function of mice were investigated.The methods of weighing the immune organs of mice,of carbon clearance,swimming and enduring high temperature were used.The result shows that high concentration group could significantly increase the thymic and splenic indexes of mice and the swallowing coefficient compared with the control((p<0.05)) and that it also significantly increased the splenic index of mice compared with low concentration group((p<0.05)) and enhanced the swallowing coefficient compared with the control and low concentration group(p<0.05).Furthermore,the time of swimming was significantly prolonged in high concentration group compared with the control(p<0.05).However,the different groups had no obvious effect on the livability of enduring high temperature.It indicates that polysaccharides of L.deliciosus can improve the immune function of mice.

Abstract:Effect of Tricholoma mongolicum and its polysaccharide on immune function in mice were studied.The immune organ indexes were determined using the weight method.The serum antibody levels were measured by spectrophotometry.The monocyte-macrophage phagocytosis was measured by carbon clearance test and the lysozyme content in serum of mice was measured by immunization turbidimetry.The results showed that the powder and polysaccharide extract of Tricholoma mongolicum fruit bodies had no significant effect on the thymus weight;The high dose powder of Tricholoma mongolicum fruit bodies increased spleen weight significantly;The low dose powder of Tricholoma mongolicum mycelium increased thymus weight significantly;The low dose polysaccharide extract of Tricholoma mongolicum mycelium increased spleen weight significantly;The powder and polysaccharide extract of Tricholoma mongolicum fruit bodies and mycelium could enhance the average content of hemolytin and lysozyme in serum and could enhance the monocyte-macrophage phagocytosis.The powder and polysaccharide extract of Tricholoma mongolicum fruit bodies and mycelium could significantly improve immune function in mice.
Physical and Chemical Properties of Corn Silk Polysaccharide and Its Immunity Activity

Abstract:In order to study the chemical composition and immune activity of corn silk,we applied a systematic extraction and separation method to obtain crude polysaccharide from corn silk(CSCP for short).HLPC and GC used to analysis the molecular weight and monosaccharide composition.Mouse splenocyte proliferation was used to determine the immune activity of CSCP.The results showed that 75% of the CSCP molecular weight was between the 1.0×105 and 5.0×105.CSCP contained rhamnose,xylose,arabinose,mannose,glucose and galactose and other monosaccharides.The molar ratios were 1.00∶1.36∶1.30∶0.66∶2.52∶1.60.The largest proportion was glucose.CSCP significantly stimulated the proliferation of mouse spleen cells and had a definite dose-effect relationship.

Abstract:Objective To study the effect of polysaccharides from Nostoc commune vauch on immunological function in mice.Methods Polysaccharides with different dosages were given to mice by intragastric administration.A series of tests,such as the serum hemolysin level (HC50),the phagocytosis of phagocytes,the activities of NK cells and the proliferation of splenic lymphocytes were examined.Results Polysaccharides from Nostoc commune vauch enhanced the phagocytotic ability of macrophages and the activities of NK cells,which indicated polysaccharides from Nostoc commune vauch can improve the non-specific immune function of normal mice.The serum HC50 in mice and spleen lymphocyte proliferation were increased when administrated with polysaccharides from Nostoc commune vauch.There were no significant differences in viscera index among all group mice.Conclusion Polysaccharides from Nostoc commune vauch may significantly enhance the immunological function of mice.

Abstract:The novel bioactive polysaccharide, Sacha inchi shell polysaccharide‐3 (SISP‐3), was isolated from the Sacha inchi (Plukenetia volubilis L.) shell using anion exchange and gel filtration chromatography. This neutral polysaccharide fraction, characterized by a triple‐helix structure and a molecular weight of 17,684 Da, was successfully isolated. The composition analysis of SISP‐3 revealed the presence of Rha, Ara, Gal, Glc, and Fru in molar ratios of 7.2%:41.7%:40.3%:4.1%:6.7%. The dominant glycosidic linkage in SISP‐3 was identified as ⟶4)‐Galp‐(1⟶, accompanied by a complex branching pattern. The results from RAW264.7 cell experiments showed that SISP‐3 significantly enhanced cell viability, phagocytosis, and NO production. Additionally, it significantly stimulated the production of cytokines, including TNF‐α, IL‐1β, and IL‐6, in RAW264.7 cells. These findings suggest that SISP‐3 serves as an immune active agent with immunomodulatory properties, indicating its potential as a natural immunomodulatory agent for various applications.

Abstract:Sijunzi decoction (SJZD) is a classic recipe in Traditional Chinese Medicine (TCM) with strong immune-enhancement activity. To further understand the characterization and immunomodulatory effect of polysaccharides from SJZD, the monosaccharide compositions of crude polysaccharide (SJZDP), polysaccharide fraction (S-3), and homogeneous polysaccharide (S-3-AG) from SJZD were compared by GC analysis, as well as their immunomodulatory effects on Peyer’s patch cells, splenocytes, and macrophages which are related to intestinal immunity, specific immunity, and nonspecific immunity. The results showed that S-3-AG mainly contained Ara with a proportion of 38.9%, while Glc accounted for the largest proportion in S-3 (55.6%) and SJZDP (87.6%). The SJZDP, S-3, and S-3-AG all showed strong capability to stimulate Peyer’s patch cells to proliferate and produce IgA and promoted the proliferation and IFN-γ production of splenocytes and increased the NO production and TNF-α production of macrophages. However, S-3 and S-3-AG were able to stimulate splenocytes to secret IL-4, SJZDP had no effect on IL-4 production of splenocytes in the tested concentrations. In addition, S-3 could stimulate the phagocytic activity of macrophages, and S-3-AG restrained the proliferation of macrophages at the concentration of 50–200 µg/mL. These results suggested that SJZDP, S-3, and S-3-AG might have different immunomodulatory effects on intestinal immunity, specific immunity, and nonspecific immunity due to their different monosaccharide compositions. It will provide references for the material basis and mechanism of SJZD immunomodulation activity.

Abstract:Objective To investigate the effect of Sipunculus nudus Polysaccharides(SNP)on immunological functions in mice.Methods By lymphocyte transformation test and NK cells activity test,the effects of SNP on cellular immunity were evaluated.By the hemolysin level of serum and plaque-forming cell(PFC) test of mice sensitized by sheep red blood cell(SRBC),the effect of SNP on humoral immunity was measured.Carbon clearance test was used to detect the promotion of phagocytosis function caused by SNP.Results Compared with the normal group,SNP could promote the cellular immunity and humoral immunity,and promote eyewinker's phagocytosis function of liver and spleen in mice.Conclusion SNP can stimulate the immunological function in mice.
Immune Function and Antioxidant Activity of Intracellular Polysaccharides from Phellinus baumii

Abstract:An immune-deficient mouse model was established by intraperitoneal injection of cyclophosphamide(CTX) to explore the immune enhancement and antioxidant capability of crude polysaccharides(SH) from Phellinus banmii and purified polysaccharide fractions(P-47000 and P-8700).Meanwhile,the bioactivity differences among SH,P-47000 and P-8700 were investigated.The results showed that SH,P-47000,and P-8700 could enhance immune function of CTX-induced immune-deficient mice to some extent.The order of immune enhancement function from strong to weak was P-8700,SH and P-47000.P-47000 exhibited a lower reducing power than SH and P-8700.However,no significant difference in reducing power between SH and P-8700 was observed.In addition,P-47000 revealed lower ability to scavenge superoxide anion free radicals than SH and P-8700.Moreover,P-8700 revealed stronger hydroxyl free radical scavenging capacity with a scavenging rate of 53.421%,which was obviously higher than that of SH and P-47000.All the polysaccharides had high scavenging capability against DPPH free radicals.SH and P-8700 revealed the highest and lowest scavenging capability on DPPH free radicals with a scavenging rate of respectively 93.221% and 66.435%.

Abstract:Extracellular polysaccharides (EPS1-1) was extracted from fermentation liquor of Rhizopus nigricans and evaluated its immune-enhancing activities in vitro and in vivo. Results suggested that the proliferation of lymphocyte was stimulated after treated with EPS1-1. Moreover, the activities of macrophages were enhanced by increasing the activities of phagocytosis and acid phosphatase, the production of NO and the mRNA levels of IL-2, TNF-α and iNOS. Furthermore, EPS1-1 could significantly boost the immunity of normal and immunosuppressed mice, which included the increase of loaded swimming time, footpad swelling, organ index and the secretion of IL-2 and TNF-α in serum, thus suggesting that EPS1-1 could improve the body immunity through cellular immunity and humoral immunity. These findings provided further insights into the potential use of EPS1-1 as immunopotentiator or new function food.

Abstract:Polysaccharides coded as CP were extracted from Athyrium Multidentatum (Doll.) Ching and then fractionated into five fractions (FP-1, FP-2, FP-3, FP-4 and FP-5). A purified polysaccharide designated as FP-3-4 was prepared from FP-3 by Sephadex G-100 column chromatography. Chemical analysis disclosed that CP and these fractions were heteropolysaccharides and mainly composed of glucose, galactose, arabinose, mannose, rhamnose, xylose, fucose, ribose and uronic acid with different molar ratios. They presented different images of SEM. FP-3-4 was highly branched polymers with sixteen types of linkages. The in vitro immunomodulatory results stated that CP and these fractions could promote macrophage proliferation, enhance macrophage phagocytosis and increase the production of NO, TNF-α, IFN-γ, IL-1β, IL-6, IL-10 and IL-2, indicating remarkable immune enhancement activities. RNA sequencing analysis revealed that CP and FP-3 induced macrophage activation mainly through MAPK and alternative NF-κΒ signaling pathways via CD14/TLR4 and Dectin-2 receptors, which were verified by RT-qPCR and western blot.

Abstract:In this study, nanoparticles were prepared by using positively charged quaternized chitosan and negatively charged mucopolysaccharide such as chondroitin sulfate, heparin and hyaluronic acid. The nanoparticles have a stable nanostructure with particle size in 336.2-424.5 nm, potential in 18.5-31.1 mV and polydispersity index PDI of 0.172-0.335. Moreover, their encapsulation efficiency was 68.77 % and 64.89 %, and they have low endotoxin and good stability. It can significantly promote the expression of IL-6, TNF-α, and IL-1β of DCS cells. Moreover, the in vivo immune activity of heparin-quaternized chitosan-OVA nanoparticles against BALB/C mice was showed that, the nanoparticles could significantly promote the secretion of immunoglobulins in mice including IgG1 and IgG2. And nanoparticle also can promote the production of immune factors. Meanwhile, the expression of immune factor genes was also elevated. Furthermore, the results of tissue section experiments showed that the nanoparticles are safety of the body.

Abstract:In this study, a typical Chinese medicine injection (Mai-luo-ning, MLN) was chosen to study the transformation possibility of its by-products (polysaccharides). What’s more, the basic properties of polysaccharides were also detected and its immunoprotect and anti-inflammatory activity were both proved in vivo and in vitro. The experiment determined that MLN-1 significantly increased the level of anti-inflammatory-related factors and the number of lymphocytes. Acute oral toxicity study established that MLN-1 has a non-toxic effect. The present work successfully showed that MLN-1 has the potential to be developed as a food additive.

Abstract:OBJECTIVE:To study the immune activity of polysaccharide extracted from root of Salvia miltiorrhiza.METHODS:Investigated the effects of polysaccharide extracted from root of Salvia miltiorrhiza on lymphocyte proliferation response of mouse induced by LPS (the lipopolysaccharide LPS), phagocytosis of the peritoneal macrophage of mice to chick erythrocytes and the mouse models of delayed type hypersensitivity (DTH) response induced by DNFB.RESULTS:Lymphocyte proliferation and phagocytosis of the peritoneal macrophage of mice could be promoted by the polysaccharide, which could inhibit ear edema and capillary permeability increase induced by DNFB and enlarged the thymus and splenic index in mice. The expression of iNOS, IFN-alpha and IL-1beta was inhibited significantly in the treatment group.CONCLUSION:The polysaccharide extracted from Salvia miltiorrhiza can improve immune function.

Abstract:Objective:To investigate the immune regulating effect of polysaccharide from Scutellaria baicalensis Georgi.Method:The contents of immunoglobulin (IgG,IgA,IgM),contents of complement (C3,C4),activity of lysozyme in the serum,thymus index and spleen index of mice were determined by animal testing.Results:Polysaccharide from Scutellaria baicalensis Georgi can increased the weight of thymus of mice,significantly increased the weight of spleen of mice.It increased the content of IgG in serum,significantly increased the content of IgA and IgM in serum.It significantly increased the content of complement and the activity of lysozyme in serum.Conclusion:Polysaccharides from Scutellaria baicalensis Georgi were shown to have the function of improving the nonspecific and specific immune ability of mice.And humoral immunity of mice has been enhanced to a certain extent.

Abstract:The immune-enhancing activities of four sulfated polysaccharides, sOPS(t), sOPS(80), sJPS(t) and sJPS(50) picked out in our previous researches, were compared taking four corresponding unmodified polysaccharides as control. In vitro experiment, the effects of eight polysaccharides on chicken peripheral lymphocyte proliferation were determined by MTT assay. The result displayed that four sulfated polysaccharides could significantly stimulate lymphocyte proliferation, their actions were significantly or numerically stronger than those of corresponding unmodified polysaccharides, sOPS(80) presented better efficacy. In vivo experiment, 300 14-day-old chickens were averagely divided into ten groups. The chickens except blank control (BC) group were vaccinated with Newcastle disease vaccine, repeated vaccination at 28 days old. At the same time of the first vaccination, the chickens in eight polysaccharides groups were injected respectively with 0.5 ml (1mg) of eight polysaccharides, in vaccination control (VC) and BC group, with 0.5 mL of physiological saline, once a day for three successive days. On days 7, 14, 21 and 28 after the first vaccination, the peripheral lymphocyte proliferation and serum ND antibody titer were determined. The result showed that four sulfated polysaccharides could significantly promote lymphocyte proliferation and enhance serum antibody titer. Their actions were significantly or numerically stronger than those of corresponding unmodified polysaccharides, sOPS(80) possessed the best efficacy. These results indicated that sulfation modification could enhance the immune-enhancing activity of OPS and JPS, sOPS(80) possessed the best efficacy and would be expected as a component drug of new-type immunopotentiator.

In Vivo Immunomodulatory Activity of Polysaccharides Derived from Chlorella Pyrenoidosa

Abstract:The effects of polysaccharides from Chlorella pyrenoidosa on immunomodulatory activity were investigated in this paper. Phagocytosis of peritoneal macrophages was detected through ingesting chicken red blood cells (CRBC) test to evaluate nonspecific immune response. The cellular immune response was measured using the delayed-type hypersensitivity response to sheep red blood cells (SRBC) in terms of antibody titration level. Oral administration of C. pyrenoidosa polysaccharides, 1 g and 2 g kg−1 body weight significantly enhanced phagocytic rate (23.00 and 26.25%, respectively) and phagocytic index (0.30 and 0.36, respectively) compared with saline-treated control (P<0.05). The value of ear thickness rate was greatly elevated from 10.09% in saline-treated control to 16.22 and 19.48%, respectively in group treated with 1 g and 2 g kg−1 body weight polysaccharides (P<0.05). There was a significant correlation between the dosage of C. pyrenoidosa polysaccharides and antibody titre (r=0.99, P=0.019) and the antibody titre was significantly increased by 11.80, 21.90 as well as 39.80 in 2 g kg−1 bw compared to 10.30 in the control. Dietary supplementation with C. pyrenoidosa polysaccharides did not affect the growth of the mice during the experiment. These results indicated that immunomodulatory activities in mice were enhanced by administration of polysaccharides derived from C. pyrenoidosa.
Investigating immune-modulatory function of α-glucopyranose-rich compound polysaccharides by MC38-N4/OT-I co-culture system

Abstract:The potential antitumor function of polysaccharides is well accepted, it is unclear whether polysaccharides have immunoregulatory effect on CD8+ T lymphocyte cells to attack tumor cells. To evaluate the CD8+ T function enhancing role of polysaccharide compounds, the MC38-N4/OT-I co-culture system was established. The synergistic and complementary immune effect of α-glucopyranose-rich compound polysaccharides can be achieved by manipulating the antigen-specific T-cell expansion capacity and efficacy. This study was designed to investigate the antitumor-enhancement activity of a α-glucopyranose-rich compound polysaccharides by determining the activation of CD8+ T cells in a co-culture system. Compared to the control group (42.5 % ± 0.72 %), the specific α-glucopyranose-rich compound polysaccharides, comprising Agaricus blazei Murill, Grifola frondosa and Pericarpium Citri Reticulatae, demonstrated a significant decrease (20.4 % ± 1.23 %, p < 0.05) in the survival rate of MC38-N4 cells in the co-culture system. Additionally, the α-glucopyranose-rich compound polysaccharides resulted in a substantial increase (p < 0.01) in the proportion of CD8+ T cells and CD62L+ central memory T cells, which is a less differentiated T cell subset with high immune activity. Collectively, we reported that specific polysaccharide combination, which remodel the function of cytotoxic T cells and provided a basis for improving immune functions by using the specific types of polysaccharides.
Effects of Crude Polysaccharides from Polygonati Rhizoma on Immune Function in Normal Mice

Abstract:OBJECTIVE To study the effects of crude polysaccharides from Polygonati Rhizoma(CPPR) on immune function in normal mice.METHODS The cell immune function was assessed by delayed type hypersensitivity(DTH) induced by dinitrofluorobenzene(DNFB) in normal mice.The humoral immune function was assessed by determining HC50 in normal mice.The mononuclear phagocyte immune function was assessed by determining the capacity of carbon particle clearance in normal mice.RESULTS Compared with blank control group,0.72 and 0.36 g-kg-1-d-1 CPPR could significantly increase ear edema of normal mice induced by DNFB in a dose-dependent manner.And 0.72,0.36 and 0.18 g-kg-1-d-1 CPPR could significantly enhance haemolysin level in vivo in a dose-dependent manner,and could significantly enhance the capacity of carbon particle clearance in normal mice.CPPR could promote development of thymus and spleen in normal mice.CONCLUSION CPPR could enhance cell,humoral and mononuclear phagocyte immune functions in normal mice.

Abstract:The goal of the present study was to investigate the immune-enhancing activity of polysaccharides from Cyrtomium macrophyllum (CMP). Two experiments were carried out. In immunosuppression experiment, the immune-enhancing effect of CMP in immunosuppressive mice was performed. The results showed that CMP at high and medium doses was able to overcome the CY-induced immunosuppression, significantly increases the thymus and spleen indices, enhances lymphocyte proliferation activity and macrophage function, improves immunoglobulin and cytokines levels compared with negative control group. In macrophage immunomodulatory experiment, the immune-enhancing effect of CMP in RAW264.7 cells was measured. The results showed that CMP induced the elevation of NO production, TNF-α secretion and iNOS protein of RAW264.7 cells. CMP can also strongly increase NF-κB levels in nuclear, which is an important transcription factor that can modulate expressions of iNOS, NO and TNF-α. Therefore, the CMP could be an effective immunomodulatory agent.
