Effect of Hypenhemia on Caspase-3 Expression in Caski Cell

Abstract:Objective To investigate the influence of hyperthermia on the expression of Caspase-3 in Caski cell. Methods Caski cells were heated at 43 ℃,45 ℃,47 ℃ and 37 ℃ in temperature-controlled water baths.The expression of Caspase-3 was evaluated by real time PCR and Western Blot. Results The expressed multiple of Caspase-3 mRNA after 24 h in 37 ℃,43 ℃,45 ℃,and 47 ℃ was 1,2.738±0.156,1.35±0.071,and 0.78±0.093,respectively.The gray scale of Caspase-3 protein expression in 37 ℃,43 ℃,45 ℃,and 47 ℃ was 0.894±0.064, 1.237±0.182,1.196±0.104,and 0.401±0.051,respectively.The Caspase-3 protein expression in 43 and 45 ℃ was higher than that in 37 ℃(P0.05),while that in 47 ℃ was lower than that in 37 ℃(P0.05). Conclusions The hyperthermia at 43 ℃ or 45 ℃ could increase the expression of Caspase-3 mRNA and protein in Caski cell.
