Abstract:The copper-catalyzed sulfenylation of indoles with aryl iodide and sulfur powder has been investigated both experimentally and theoretically. This protocol provides a direct and facile approach to prepare 3-sulfenylindoles with moderate to excellent yields and good functional-group tolerance. The in-situ IR analysis provided evidence for that NaOAc could promote the synthesis of diphenyl disulfide by the coupling of aryl iodide and sulfur powder. According to DFT calculations, the coupling pathway involving the intermediate N-methyl-3-iodoindole is more favored than the direct coupling by the CH activation of indole. The N-methyl-3-iodoindole was identified as a crucial intermediate in the catalytic cycle.

Abstract:Catalytic asymmetric sulfenylation of 3-substituted oxindoles has been developed through efficient catalysis by tartaric acid derived chiral iminophosphoranes. With N-(phenylthio)phthalimide as the sulfur source, a wide range of optically active 3-phenylthiooxindoles were obtained in excellent yields (90-99%) and good enantiomeric excess (up to 90% ee). Interestingly, 3-aryl and 3-methyl substituted oxindoles afforded sulfenylated products in S-configuration, whereas substituted oxindoles with 3-arylidene or 3-isobutyl substituents gave the corresponding R-configured sulfenylated products.

Abstract:Asymmetric synthesis of 3-sulfonylated 3-substituted oxindoles through the addition of sodium sulfinate salts to 3-bromo-3-substituted oxindoles has been achieved using chiral nickel complexes of N,N'-dioxides. This method facilitates the creation of diverse chiral sulfonyl oxindoles, several of which display promising anticancer properties. Notably, the catalyst demonstrates remarkable tolerance to water, crucial for maintaining enantioselectivity. Furthermore, the utilization of topographic steric maps of the catalysts offers valuable insights into the mechanism underlying enantioselection reversal.

Abstract:An enantioselective thiocyanation of oxindoles has been developed for the first time using a bifunctional cinchona-derived organo-catalyst and N-thiocyanatophthalimide as the electrophilic thiocyanation source in the presence of 2-naphthol as the additive. Various enantioenriched 3,3'-disubstituted oxindoles with SCN-containing quaternary carbon stereocenters were synthesized under mild conditions in high yields (up to 99%) and good enantioselectivities (up to 6:94 er).

Abstract:A C6' bulky substituted quinine-catalyzed S(N)V reaction between 3-substituted oxindole and (E)-3-(nitromethylene)-oxindole was developed. This enantioselective C(sp3)-C(sp2) coupling furnished bisoxindole scaffolds featuring a vinyl-substituted all-carbon quaternary stereocenter with high stereoselectivities. In addition, the gram-scale synthesis and synthetic post-transformations were conducted to demonstrate the potential synthetic usefulness.

Abstract:Background: While C-S bonds coupling is playing a crucial role in the syntheses of many biologically active substances, the well concerned C-H functionalization reactions have been barely employed yet in construction of such sulfur compounds. Objective: The primary objective of this study was to expand the application of copper, a cheap metal, to catalyze C-S bond construction reactions of indoles. Methods: We chose copper compounds of various oxidant states as catalysts. Substituted indoles were picked as substrates, and disulfides as sulfur source. The variants of catalyst, solvent and reaction temperature were screened to optimize the reaction condition. After that, we explored the scope of diversely substituted indoles and disulfides substrates of the reaction. Results: The reactions were all smoothly performed to provide corresponding products in good yields. The main product is tunable between C3-sulfenylindoles and C2,3-sulfenylindoles according to the amount of disulfides. C3-sulfenylindoles were synthesized in 85 to 99% yields. C2,3-sulfenylindoles were also fruitfully obtained in from 66 to 98% yields. Conclusion: A facile and efficient method for sulfenylation of indole, which employed CuI as catalyst, to give C3- and C2,3-sulfenylindoles, was presented. Our methodology features easy operation, excellent yields and cheapness of catalyst.

Abstract:β-Chiral sulfones are structural motifs widely found in natural products and bioactive molecules, while also serving as important intermediates for the synthesis of valuable chiral scaffolds. In contrast to the rapid growth of sulfur dioxide insertion chemistry over the last decade, enantioselective catalytic variants for accessing β-chiral sulfones, especially those bearing a quaternary carbon stereocenter, remain rare. Herein, we report an enantioselective copper/bisoxazoline catalyzed oxysulfonylation of alkenes to yield isoxazolinyl-containing β-chiral sulfones bearing quaternary carbon stereocenters with moderate to excellent yields and up to 96:4 er. The advantage and irreplaceability of this sulfur dioxide insertion catalytic system have been demonstrated by achieving transformations clearly unavailable with the previously reported sulfonyl chloride system. Additionally, the recoverability of the new-developed bisoxazoline ligand L9 and its reusability without any erosion of the catalytic activity and enantioselectivity further demonstrate the usefulness of the protocol.

Abstract:A highly efficient synthesis of chiral 3,3-disubstituted oxindoles was developed using a chiral spirocyclic phosphoric acid catalyzed 1,6-conjugate addition reaction of para-quinone methides derived from N-unprotected isatins with indoles. The reaction proceeds under mild reaction conditions to provide indole-containing N-unprotected oxindoles bearing quaternary stereocenters in good yields and with moderate to excellent enantioselectivities (up to 97% ee).

The synthetically useful 2,2-disubstituted indolin-3-one derivatives with vicinal chiral quaternary-tertiary stereocenters were obtained in high yields with excellent stereoselectivities. The adduct can be readily transformed into a structurally interesting heterocyclic architecture by means of further synthetic elaboration.

Abstract:A pentanidium-catalyzed highly enantioselective conjugate addition of 3-alkyloxindoles to phenyl vinyl sulfone has been demonstrated. This approach allows the construction of 3,3-dialkyl-substituted oxindole frameworks with high yield and excellent enantioselectivity (up to 99%) under simple phase-transfer conditions. A variety of oxindoles bearing all-carbon quaternary stereogenic centers were obtained in the presence of 0.25mol% pentanidium. Meanwhile, practicality was illustrated by a gram-scale asymmetric synthesis of two 3,3-dialkyl-substituted oxindoles. The resulting adduct can be smoothly transformed to the natural product analogue in a short synthetic route.
