Differential Expression and Regulation of Angiopoietin-2 in the Mouse Ovary During Sexual Maturation and Luteal Development

Abstract:Angiopoietins (Ang), a family of major angiogenic growth factors, are glycosylated and secreted proteins, which have characteristic protein structures that contain coiled-coil domains in the N-terminal portion and fibrinogen-like domains in the C-terminal portion of the molecule. So far, four members of the angiopoietin family have been discovered. Ang-2 is the endogenous antagonist of Ang-1 and plays an important role in the regulation of vascular remodeling together with vascular endothelial growth factor (VEGF). Ang-2 plays an important role in female reproduction. The aim of this study was to examine the expression and regulation of Ang-2 in the mouse ovary during sexual maturation, gonadotropin treatment and luteal development by in situ hybridization and and reverse transcription polymerase chain reaction (RT-PCR). The results showed that Ang-2 mRNA was mainly localized in the granulosa cells, thecal cells and corpus luteum, but not in the oocytes by in situ hybridization. Moreover, Ang-2 was highly expressed in the corpus luteum and granulosa cells of atretic follicles. By RT-PCR, Ang-2 mRNA was lowly expressed on day 10 after birth, then the expression gradually increased and reached the highest level on day 25 after birth. In the superovulated model of immature mice, the levels of Ang-2 mRNA expression were high at 48 h after eCG treatment and 3 h post-hCG injection. Ang-2 mRNA was highly expressed in the corpus luteum from days 2 to 9 post-hCG injection by in situ hybridization, then the expression gradually declined on days 11 and 13 after hCG treatment. By RT-PCR, the levels of Ang-2 mRNA expression showed a decline after hCG injection, with a nadir on day 3 followed by an increase reaching the highest level on day 9 post-hCG injection. Then, Ang-2 expression gradually declined from days 11 to 15 after hCG injection. These results suggest that Ang-2 may be involved in follicular development, atresia, ovulation, and corpus luteum formation and regression.
