Abstract:A one-pot reaction for the transformation of common secondary amides into amines with C-C bond formation is described. This method consists of in situ amide activation with Tf2O-partial reduction-addition of C-nucleophiles. The method is general in scope, which allows employing both hard nucleophiles (RMgX, RLi) and soft nucleophiles, as well as enolates. With the use of soft nucleophiles, the reaction proceeded with high chemoselectivity at a secondary amide in the presence of ester, cyano, nitro, and tertiary amide groups.

Abstract:The direct partial reduction of highly stable secondary amides to more reactive aldimines and aldehydes is a challenging yet highly demanding transformation. In this context, only three methods have been reported. We report herein an improved version of the Charette’s method. Our protocol consists of activation of secondary amides with triflic anhydride/2-fluoropyridine,and partial reduction of the resulting intermediates with 1,1,3,3-tetramethyldisiloxane(TMDS), which delivered aldimines or aldehydes upon acidic hydrolysis. Aromatic amides were reduced to the corresponding aldimines in 85%–100% NMR yields,and yields(NMR) from aliphatic amides were 72%–86%. Acidic hydrolysis of the aldimine intermediates afforded, in one-pot,the corresponding aldehydes in 80%–96% yields. A simple protocol was established to isolate labile aldimines in pure form in92%–96% yields. The improved method gave generally higher yields as compared to the known ones, and features the use of cheaper and more atom-economical TMDS as a chemoselective reducing agent. In addition, a convenient extraction protocol has been established to allow the isolation of amines, which constitutes a mild method for the N-deacylation of amides, another highly desirable transformation. The extended method retains the advantages of the original method of Charette in terms of mild conditions, good functional group tolerance, and excellent chemoselectivity.
General Direct Transformation of Secondary Amides to Ketones Via Amide Activation

Abstract:Carbon-carbon bond formation and functional group transformation are two cornerstones of organic synthesis.Generally,most of the established methods involve the transformation of a more reactive molecule to a more stable one,while the inverse transformation from a more stable molecule to a more reactive one is challenging.Secondary amides are a class of quite stable compounds,and ketones are a class of extremely versatile compounds enabling a number of fundamental transformations.The direct transformation of secondary amides into ketones is of high relevance in organic synthesis.Nonetheless,no such method has been reported when this work was disclosed.Here,we report in detail the general direct transformation of secondary amides into ketones by Tf2O-mediated deaminative alkylation with organocerium reagents.The influence of the base additive and the organometallic reagent on this transformation was investigated.It was found that 2-fluoropyridine gave the best results of the bases screened,and both organocerium reagents（RCeCl2） generated in situ from RLi and CeCl3and cerium complexes generated in situ from RMgX and CeCl3are superior to organomagnesium,organolithium and organozinc reagents.The optimum reaction condition was thus determined as successive treatment of a dichloromethane（0.25 mol/L） solution of secondary amide（1.0 equiv.） and 2-fluoropyridine（1.2 equiv.） with 1.1 molar equivalents of Tf2O（-78 ℃,then 0 ℃）,and 3.0 molar equivalents of RM/CeCl3（-78 ℃）,followed by hydrolysis with 2 mol/L aqueous HCl solution.This protocol shows wide substrate applicability.Using the developed method,a variety of ketones including alkyl-alkyl ketones,alkyl-aryl ketones,aryl-aryl ketones,α,β-unsaturated ketones,and β-chloroenones have been synthesized in 65%～90% yields.It should be noted that the reaction of α,β-unsaturated amides led to a highly selective 1,2-addition and the use of alkynyl cerium reagents generated from lithium acetylides yielded β-chloroenones.Moreover,weakly basic alkynylborane reagents and allyltrimethyl silane,and even weakly nucleophilic styrene can also be applied as nucleophiles for the deaminative alkylation of secondary amides.On the basis of the experimental results,a plausible mechanism involving nitrilium ion intermediate and ketimine is proposed.As one significance of these transformations,secondary amides successfully serve as effective acylating reagents in the intermolecular reactions with organometallic reagents,allyltrimethyl silane and styrene（reductive acylation）.

Abstract:Amides are widely used as stable synthetic intermediates in organic synthesis and medicinal chemistry. Versatile and chemoselective C-C bond forming methods for the direct transformation of amides are highly demanding. In this paper, we report the reductive cycloaddition of common secondary amides with the Danishefsky diene to produce 2-substituted 2,3-dihydro-4-pyridones. This one-pot procedure involves amide activation with triflic anhydride, partial reduction, and [4+2] cycloaddition. The synthetic utility of this step-econimical method was demonstrated by the short and protecting-group-free total syntheses of alkaloids (+/-)-lasubine I and (+/-)-myrtine.

Abstract:We report the first one-pot reductive homocoupling reaction of secondary amides and cross-coupling reaction of secondary amides with ketones to give secondary vicinal diamines and amino alcohols. This method relies on the direct generation of α-amino carbon radicals from secondary amides by activation with trifluoromethanesulfonic anhydride, partial reduction with triethylsilane and samarium diiodide-mediated single-electron transfer. The reactions were run under mild conditions and tolerated several functional groups.

One-Pot Reductive 1,3-Dipolar Cycloaddition of Secondary Amides: A Two-Step Transformation of Primary Amides

Abstract:The one-pot reductive 1,3-dipolar cycloaddition of secondary aromatic N-(trimethylsilylmethyl)amides with reactive dipolarophiles is reported. The method relies on the in situ generation of nonstabilized NH azomethine ylide dipoles via amide activation with triflic anhydride, partial reduction with 1,1,3,3-tetramethyldisiloxane (TMDS), and desilylation with cesium fluoride (CsF). Running under mild conditions, the reaction tolerated several sensitive functional groups and provided cycloadducts in 71-93% yields. The use of less reactive dipolarophile methyl acrylate led to the cycloadduct in only 40% yield. A (Z) geometric intermediate of NH-azomethine 1,3-dipole was postulated to account for the observed higher yields and higher cis diastereoselectivity for the substrates bearing an electron-withdrawing group. This model features an unconventional cyclic transition state via carbanion-aryl ring interaction. Because the starting secondary amides can be prepared from common primary amides, the current method also constitutes a two-step transformation of primary amides.

Abstract:Amides are a class of easily available compounds, and widely serve as versatile intermediates in organic synthesis medicinal chemistry. Amide-based transformations could lead to many useful compounds and intermediates including various amines, ketones and enaminones. Though direct transformation of amides is of high demand, many current chemoselective transformations are only achieved in multistep approaches. In recent years, direct transformation of amides is emerging as an exciting area. A number of recent progresses on nucleophilic addition to amide carbonyl group that led to new C-C bond formation are highlighted in this review, including (1) in situ amide activation with trifluoromethanesulfonic anhydride (Tf2O) followed by addition of pi- and s-nucleophiles or reactive organometallic reagents; (2) direct transformation of N-alkoxyamides; (3) direct transformation of amides using Schwartz reagent; and (4) catalytic reductive C-C bond forming reactions of amides, and metal catalyzed coupling of amides.

Abstract:A direct transformation from methyl ketones to secondary or tertiary amides has been developed through a novel electrochemical approach and a wide scope of formamides could be utilized as the amide sources. This transformation was promoted by in situ generated iodine through electrolysis of sodium iodide under mild, metal-free conditions. This electrochemical procedure could avoid the use of stoichiometric iodine and afforded the target products in good to excellent yields.

Abstract:The secondary amine participating asymmetric reductive amination remains an unsolved problem in organic synthesis. Here we show for the first time that secondary amines are capable of effectively serving as N-sources in direct asymmetric reductive amination to afford corresponding tertiary chiral amines with the help of a selected additive set under mild conditions (0-25 degrees C). The applied chiral phosphoramidite ligands are readily prepared from BINOL and easily modified. Compared with common tertiary chiral amine synthetic methods, this procedure is much more concise and scalable, as exemplified by the facile synthesis of rivastigmine and N-methyl-1-phenylethanamine.

Abstract:The catalytic asymmetric geminal bis-nucleophilic addition to nonreactive functional groups is a type of highly desirable yet challenging transformation in organic chemistry. Here, we report the first catalytic asymmetric reductive/deoxygenative alkynylation of secondary amides. The method is based on a multicatalysis strategy that merges iridium/copper relay catalysis with organocatalysis. A further combination with the palladium-catalyzed alkyne hydrogenation allows the one-pot enantioselective reductive alkylation of secondary amides. This versatile protocol allows the efficient synthesis of four types of α-branched chiral amines, which are prevalent structural motifs of active pharmaceutical ingredients. The protocol also features excellent enantioselectivity, chemoselectivity, and functional group tolerance to be compatible with more reactive functional groups such as ketone and aldehyde. The synthetic utility of the method was further demonstrated by the late-stage functionalization of two drug derivatives and the concise, first catalytic asymmetric approach to the κ-opioid antagonist aticaprant.

Abstract:Triflic anhydride in combination with 2-fluoropyridine effectively dehydrates secondary amides to afford nitriles under mild reaction conditions. The reaction is general in scope and compatible with the use of aliphatic, α,β-unsaturated, aromatic, and heteroaromatic amides bearing either secondary, tertiary, or benzylic N-alkyl groups. The reaction also shows good to excellent chemoselectivity for the secondary amide and tolerates several labile functional groups.

Abstract:Summary of main observation and conclusion In recent years, exciting progress has been made in the field of direct transformation of amides, nevertheless, the condensation between two amides remains rare and restricted to homo‐coupling reactions. Herein, we report the cross‐coupling of secondary amides with tertiary amides, which provides a synthesis of ketones under mild conditions, and features the use of tertiary amides as surrogates of alkyl carbanions. The method relies on the coupling of enamines, generated from tertiary amides by catalytic partial reduction of tertiary amides with Vaska's catalyst, with nitrilium ions, formed in situ from secondary amides via activation with trifluoromethanesulfonic anhydride, and on the subsequent deformylation.
