Abstract:Summary Diabetes was generally recognized as a critical issue in global public health , and has become the third noninfectious disease after angiocardiopathy and cancer . So far , the combination of applying insulin and controlling diet , as well as changing the way of living were the major therapeutic method for treating diabetes . Recently , the adjuvant therapy of natural extracts has been the research highlight in this field . To observe the intervention effect of Haematococcus pluvialis extract on blood glucose tolerance , insulin tolerance , lipid adjustment and the possible functional mechanism of improving diabetic patients condition , the diabetic mouse models were induced by alloxan through intraperitoneal injection , and given by gavage with different doses of H . pluvialis extract for 28 days . The contents of fasting blood glucose and their body masses were detected at 7‐day intervals . The capability of blood glucose tolerance and insulin tolerance was measured during later period of administration . Meanwhile , the contents of triacylglycerol ( TG) , serum total cholesterol ( TC) , high density lipoprotein cholesterol ( HDL‐C ) and low density lipoprotein cholesterol ( LDL‐C ) were detected . After modeling , the mice had dark and messy hair , and the typical diabetic symptoms including polydipsia , polyphagia and polyuria along with significant mass loss . Compared with the model group , by giving different doses of H . pluvialis extract in 28 d for experimental groups , the high , middle and low dose groups had statistically significant differences between the body masses and blood glucose contents , respectively ( P<0 .01 , P<0 .05 , P<0 .05) . The difference between high dose group and model group was most evident . During the experiment of blood glucose tolerance , the blood glucose level of each group reached the peak in 30 min of glucose loading . The glucose levels appeared downtrend after 30 min ,of which the high dose group and middle dose group were much lower than the model group in 120 min ( P<0 .01) . In the insulin experiment , after injecting 0 .8 U/kg insulin for 15 min , the blood glucose level of high dose group reduced by 20 .9% compared with the basic level , while the low dose group only reduced by 0 .6% . Three dose groups had the lowest blood glucose levels which reduced by 53 .6% , 23 .9% and 17 .9% respectively in 90 min , while the model group reduced by 5 .3% . The blood glucose increased in 120 min;however , the level in the high dose group of H . pluvialis extract was still much lower than that of model group ( P<0 .01) . The TC , TG and LDL‐C contents of the high dose group of H . pluvialis extract were much lower than the model group , which were statistically significant differences ( P<0.05,P<0.01,P<0.05) . Haematococcus pluvialis extract has remarkable effect of decreasing blood glucose level , relieving the symptoms of polydipsia , polyphagia , polyuria and mass loss , improving the glucose tolerance and insulin tolerance , increasing the utilization of glucose and regulating blood lipid level on diabetic mice . However , it has no effect on the blood glucose and lipids of normal mice . Among the three dose groups , the high dose group ( 8 g/kg) has the best effect .
Comparative Study on Koali Vine Polysaccharides Components in Regulating Glucose and Lipid Metabolism

Abstract:The hyperlipidemia mice models which induced by feeding high fat diet or by intraperitoneal injected with egg yolk were used to observe the lipid-decreasing effect of STDT components,and the diabetic mice induced by alloxan were used to observe the hypoglycemic effect of STDT components.The results showed that at the dose level of 1000 mg/kg,each component of STDT can reduce in varying degrees the total cholesterol(TC) and triglyceride(TG) levels in hyperlipidemia mice which induced by feeding high fat diet,and show a positive correlation with molecular weight,but in hyperlipidemia mice which induced by intraperitoneal injected with egg yolk,the lipid-decreasing effect of STDT components show no correlation with molecular weight;Though STDT components have no significant hypoglycemic effect on diabetic mice induced by alloxan,but can improve the living condition significantly.STDT components can significantly improve the Starch tolerance on diabetic mice induced by alloxan,and show a positive correlation with molecular weight.We concluded that the lipid-decreasing or hypoglycemic effects that each component of STDT has is correlated with its molecular weight,and its mechanisms may be related to preventing absorption of lipid material as well as sugar in the intestinal.
Effect of Echinacoside on Lipid Metabolism in Type 2 Diabetic Mice

Abstract:Objective To explore the characteristics of the effect of echinacoside on lipid metabolism in type 2 diabetic mice,and to investigate the content of growth factor 21 in liver tissue of diabetic mice,providing further insight into the possible way of regulating blood lipid metabolism in type 2 diabetic mice with Echinacea.Methods Twenty db/db mice (6-week-old) were randomly divided into two groups:experimental group and model group;while six db/m mice (SPF) (6-week-old) were control group.We detected the fasting blood glucose after 2 weeks that was higher than 16.7mmol/L in experiment.Each group was given sterilized distilled water,echinaco side,sterilized distilled water.The serum total,cholesterol,triglyceride,high density lipoprotein,low density lipoprotein,alanine aminotransferase level,aspartate,aminotransferase level,and the liver tissue measurement of fibroblast growth factor 21 levels were detected after eight weeks.Results The model group serum TG,TC and LDL were significantly higher than the control group (P ＜ 0.01).The content of serum HDL was significantly lower than the control group (P ＜ 0.01).Experimental group of serum TG,TC and LDL levels were significantly lower than the model group (P ＜ 0.01).The content of serum HDL was significantly higher than that in the model group (P ＜ 0.01).The expression of FGF21 in model group in liver were significantly lower than that of control group (P ＜ 0.01).The experimental group the expression of FGF21 in mouse liver was significantly higher than that in the model group (P ＜ 0.01).The content of ALT and AST in serum of model group was significantly higher than the control group (P ＜ 0.01).Experimental group serum ALT,AST were significantly lower than the experimental group (P ＜ 0.01).Conclusion After 8 weeks of administration,the serum lipid metabolism disorder of db/db diabetic mice was significantly improved,and the liver function damage was not increased.The mechanism of FGF21 may be related to the improvement of the expression level of liver tissue.

Abstract:Uraria crinita (UC) is a traditional edible plant and has been used as herbal medicine for a long history in China. There is no report on its anti-diabetic activity. In the present study we evaluated anti-hyperglycemic and anti-hyperlipidemic effect of U. crinita water extract (UCWE) in diabetic mice. Type II diabetes was induced in mice by injection of streptozotocin (STZ) in combination with high fat and protein food. After 3 weeks, oral administration of UCWE significantly decreased the fasting blood glucose in oral glucose tolerance test (OGTT), as well as significantly suppressed the increase of blood glucose after glucose challenge. Plasma concentration of triglyceride (TG) and free fatty acids (FFAs) were also decreased. Furthermore, UCWE increased the concentration of plasma insulin. Taken together, UCWE exhibited hypoglycemic and hypolipidemic activity, indicating a beneficial effect in diabetes treatment. 　 Key words: Uraria crinita water extract (UCWE), diabetes, oral glucose tolerance test (OGTT), TG, FFAs, insulin.

Abstract:Using KM mouse as test animals,the effect of mulberry leaves extract(MLE) on blood glucose level was studied and the quantities of four components,named 1-deoxynojimycin(DNJ),flavone,polyphenol and polysaccharide were mensurated.The result showed that the MLE was able to inhibit the increase of blood glucose in diabetic model mice.When preparing MLE,there were remarkable differences among the mulberry varieties on the content of active components and the function of MLE in reducing the blood glucose level.Among the five tested mulberry varieties,the Shansang305 was the best one.Moreover,the optimum leaf order was the upper half of shoot from mulberry trees.There was a close relationship between the effect of MLE in reducing blood glucose and the content of DNJ.

Abstract:The present research is carried out to study the hypoglycemic and hypolipidemic effects of the Dendrobium huoshanense C. Z. Tang et S. J. Cheng extract on type 2 diabetic mice. D. huoshanense extract was prepared using the condensation reflux method. The chemical components were separated by UPLC Triple-TOF MS/MS to further identify the components that mainly exerted hypoglycemic and hypolipidemic effects. Type 2 diabetic C57BL/6 mice were induced by a high-fat diet (HFD) followed by intraperitoneal streptozotocin (STZ) and were daily gavaged with different doses of D. huoshanense extract for 5 weeks. Various methods such as HE staining, ELISA, and Western blot were used to evaluate the hypoglycemic and hypolipidemic effects of D. huoshanense extract on mice with type 2 diabetes mellitus model. The experimental results revealed that the main compounds of D. huoshanense extract were polysaccharides, flavonoids, alkaloids, amino acids, and many other chemical components. Oral administration of 200 mg/kg of the extract of D. huoshanense significantly reduced blood glucose and insulin levels of high-dose group mice and lowered total cholesterol (TC), triglyceride (TG), and low-density lipoprotein cholesterol (LDL-C) levels, but increased high-density lipoprotein cholesterol (HDL-C) levels and liver glycogen content. In addition, the protein expression levels of insulin receptor (Ins R), phosphatidylinositol-3 kinase (PI3K) and p-protein kinase B (Akt) were upregulated. These results imply that the D. huoshanense aqueous extract can improve insulin resistance and lipid metabolism in type 2 diabetic mice, and it may regulate the function through the PI3K/AKT pathway.

Effects of Taohe Chengqi Decoction on Glycose and Lipid Metabolism in Diabetic Rats Induced by STZ

Abstract:[Objective] To study the effect of Taohe Chengqi Decoction on glucose,insulin and lipid metabolism in diabetic rats induced by STZ. [Method] Successful rat with diabetes model was established by STZ injection through caudal vein. Selected SD rats were randomly divided into two groups as diabetes group and treatment group. Control group was set up. Treatment group were orally administered by Taohe Chengqi Decoction 10 ml / kg·d( with 1 g raw medicine per millilitre) for 56 days. At the 14th,28th,42th and 56th day,Glu,TG,TC, LDL-C and HDL-C was measured in serum. At the 56th day,insulin was measured. [Result]Taohe Chengqi Decoction reduced the contents of blood glucose,TG,TC,LDL-C,the HDL-C level of treatment group showed significantly increased than diabetes group,the insulin level showed no significant differences in each group. [Conclusion] Taohe Chengqi Decoction can lower blood glucose and improve lipid metabolism,which may play therapeutic role in diabetic rats.

Abstract:Aims: To investigate the therapeutic effects of Tangningtongluo Tablet on diabetic mice and its mechanism. This study was established the scientific basis for the clinical application of Tangningtongluo Tablet in the treatment of diabetes mellitus and provided data supporting the transformation of Tangningtongluo Tablet from an in-hospital preparation to a new Chinese medicine. Methods: In this study, a diabetic mouse model was established by high-glucose and high-fat diet feeding in combination with STZ injection for 4 weeks. Glucose metabolism, lipid metabolism, liver histomorphological changes and liver function related indexes were detected, pancreatic histomorphological changes and insulin resistance related indexes were observed, and the expression of pathway related proteins and inflammatory factors were examined. Results: Glycemia and glycated hemoglobin were reduced in diabetic mice after the treatment of Tangningtongluo Tablet, and glucose tolerance and lipid results were modified. The insulin resistance status of the mice was diminished and tissue damage to the pancreas and liver was repaired. Expression of ERS/NF-κB related pathway proteins was reduced in liver tissues, and inflammatory factors such as TNF-α, IL-6 and IL-1β were reduced in serum. Conclusions: Tangningtongluo Tablet could reduce blood glucose in diabetic mice, regulate the disorder of lipid metabolism, enhance insulin sensitivity, improve insulin resistance, repair pancreatic tissue damage and protect mouse liver in diabetic mice. The mechanism of action might be related to the regulation of ERS/NF-κB signaling pathway and the reduction of TNF-α, IL-6 and IL-1β production.
Hypoglycemic Effect of Triterpenoid-Rich Extracts from Euryale Ferox Shell on Normal and Streptozotocin-Diabetic Mice.

Abstract:The antioxidant effects of the triterpenoid-rich extracts from Euryale ferox shell (ES) have been confirmed in vitro. This study examined whether the triterpenoid-rich extract from ES eases human hyperglycemia and diabetes caused by metabolic disorders. Normal and streptozocin (STZ)-induced diabetic mice were used as controls for the four groups that received the triterpenoid-rich extracts of ES suspended in distilled water orally at doses of 200, 300, 400, 500±2 mg/L. Body weight, blood glucose and pancreatic tissue morphology were observed after 4 weeks. The expression of protein tyrosine phosphatase-1B (PTP1B) and insulin receptor substrate (IRS-1) proteins, which are related to the regulation of glucose metabolism in vivo, were also investigated. Compared with the model group (LD50 900±2 mg/L), it was found that the triterpenoid-rich extracts of ES could regulate glucose metabolism (P<0.01) and cause body weight to return to normal levels (P<0.05). Islet morphology recovered well, the expression of the negative regulation protein PTP1B gene was reduced and insulin receptor IRS-1 protein expression was increased. These data prove that the triterpenoid-rich extracts from ES have a therapeutic effect on diabetes by insulin resistance.

Abstract:OBJECTIVE:To study the effect of Supplemented Taoren Chengqi decoction (STCD) on the secretion of insulin and proliferation of NIT-1.METHOD:The effect of STCD and the serum of rat after orally administrating of STCD on the secretion of insulin and proliferation of NIT-1 were studied. The proliferation of NIT-1 was measured by 3H-TdR incorporation and cell counting methods, while the secretion of insulin was measured from the cultured medium by the ultra sensitive rat insulin ELISIA kit.RESULT:Both the STCD and the serum of rat after orally administrating of STCD significantly could increased the secretion of insulin and proliferation of NIT-1.CONCLUSION:The treatment of the diabetic patients by STCD might be through with its improvement of secretion of insulin and proliferation on pancreatic beta-cell.

Abstract:The effect of the water extract of Panax Ginseng C.A.on the metabolism of blood glucose and blood lipid in normal mice and the hypoglycemic effect in ALX-induced diabetic mice were investigated.The normal mice were fed with water extract of ginseng at different dose of 100,200,400 mg/kg every day for 20 days by oral administration.The extract at the doses 200 and 400 mg/(kg·d) reduced the fasting blood glucose significantly,and 100 mg/(kg·d) dose reduced the blood lipid(P<0.05).The oral glucose tolerance test(OGTT) in normal mice indicated that both the water extract at the doses 200 and 400 mg/(kg·d) significantly reduced the blood glucose at 30 min after orally administrating water extract for 20 days.In ALX-induced diabetic mice,the doses of 100,300 mg/(kg·d)water extract were chose to test the effect on the fasting blood glucose.At both doses the extract reduced the fasting blood glucose and at the dose of 300 mg/(kg·d)the P value was less than 0.01.In summary,the water extract of ginseng can improves the glucose tolerance of normal mice,and reduces the blood glucose of diabetic mice.The machnisms might be that the water extract of ginseng promots the transformation from glucose to lipid of mice.
Effect of 1-Deoxynojirimycin Isolated from Mulberry Leaves on Glucose Metabolism and Gut Microbiota in a Streptozotocin-Induced Diabetic Mouse Model

Abstract:1-Deoxynojirimycin (DNJ) exerts hypoglycemic effects. However, the traditional method for DNJ extraction is inefficient, and the hypoglycemic mechanism of DNJ remains unclear. In this study, the mixed fermentation by Lactobacillus fermentum and Saccharomyces cerevisiae was used to enhance DNJ extraction efficiency. It was found that this strategy was more efficient than the traditional method as the yield improved from the original 3.24 mg/g to 5.97 mg/g. The purified DNJ significantly decreased serum glucose (P < 0.01) and insulin levels (P < 0.05), improved serum lipid levels (P < 0.05), and reversed insulin resistance (P < 0.05) in diabetic mice. These changes were caused by up-regulating the protein expression of insulin receptor and glycolysis enzymes (GK, PK, and PFK) (P < 0.05) and down-regulating the protein expression of insulin receptor substrate-1 and gluconeogenesis enzymes (PCB, PEPCK, FBPase, and G-6-Pase) (P < 0.05), thus alleviating glucose tolerance. Additionally, DNJ treatment relieved gut dysbiosis in diabetic mice by promoting the growth of Lactobacillus, Lachnospiraceae NK4A136 group, Oscillibacter, norank Lachnospiraceae, Alistipes, and Bifidobacterium (P < 0.05) and suppressing the growth of Ruminococcaceae UCG-014, Weissella, Ruminococcus, Prevotellaceae Ga6A1 group, Anaerostipes, Klebsiella, Prevotellaceae UCG-001, and Bacteroidales S24-7 group (P < 0.05).
Tetradecyl 2,3-Dihydroxybenzoate Improves the Symptoms of Diabetic Mice by Modulation of Insulin and Adiponectin Signaling Pathways

Abstract:Background: Tetradecyl 2,3-dihydroxybenzoate (ABG-001) derived from Chinese medicine, gentiana regescens Franch is a leading compound with NGF mimic effect, it can induce neurite outgrowth of PC12 cells via the IGF-1/PI3K/ERK signaling pathway. Thus, we inferred that this compound had anti-diabetic effect and used streptozocin (STZ)-induced diabetic mice to indicate it. Methods: ABG-001 was synthesized with 2,3-dihydroxybenzoic acid and tetradecyl alcohol under certain reaction conditions. STZ-induced diabetic mice were used to investigate anti-diabetic effect. Oral glucose tolerance test, insulin tolerance test, RT-PCR, Western blot, ELISA assays and histological section were performed to do the analysis of action mechanism. Results: ABG-001 showed anti-diabetic effect in STZ-induced diabetic mice. In diabetic mice, the anti-diabetic effect of ABG-001 at a dose of 20 mg/kg was equal with metformin at a dose of 140 mg/kg. Moreover, glucose tolerance and insulin sensitivity were significantly improved on diabetic mice. The plasma insulin, adiponectin and leptin were notably increased, whereas glucagon remarkably decreased. The gene expressions of adiponectin and leptin in adipose tissue, glucose transporter 4 and adiponectin receptor 1 in liver and gastrocnemius, ADR2 in hypothalamus and pancreas were obviously increased. Conclusion: ABG-001 exerts antidiabetic effects via modulation of insulin and adiponectin signaling pathways. This new type of molecule could be a promising drug candidate for treatment of diabetes.
[Effect of Sijunzi Decoction on intestinal barrier of type 2 diabetic mice]

Abstract:This study aims to explore the improvement effect of Sijunzi Decoction on intestinal barrier in diabetic mice. A type 2 diabetes mellitus(T2DM) model was established in C57BL/6J mice by feeding them with high-sugar and high-fat diet combined with streptozotocin(STZ). The T2DM mice were randomly divided into a control group, a T2DM group, a donepezil(DON) group, a rosiglitazone(RGZ) group, and Sijunzi Decoction groups(7. 5, 15, and 30 g·kg~(-1)), and orally administered for six weeks. The body weight and fasting plasma glucose(FBG) of mice were recorded. Fasting plasma insulin(FINS) and insulin resistance index(HOMA-IR) were observed to assess insulin resistance(IR). Intestinal flora and levels of serotonin(5-HT), lipopolysaccharide(LPS), and short-chain fatty acids(SCFAs) in serum were analyzed. Changes in colonic structure and tight junction proteins occludin, claudin-1,and ZO-1 were observed through HE staining and immunohistochemistry. Spontaneous alternation test was conducted to observe the effect on spatial memory ability. Compared with the results in the control group, FBG and HOMA-IR in the T2DM group were significantly increased(P< 0. 01); species richness index(Sobs index), Shannon diversity index(Shannon index), and species abundance estimate index(Chao index) were decreased; LPS was significantly increased(P< 0. 001), while the levels of 5-HT,SCFAs, occludin, claudin-1, and ZO-1 were significantly decreased(P< 0. 01), indicating impaired colonic barrier function;spontaneous alternation accuracy was significantly decreased(P<0. 05). After 6 weeks of Sijunzi Decoction treatment, compared with the results in the T2DM group, FBG and HOMA-IR in the Sijunzi Decoction 15 g·kg~(-1) group were significantly decreased(P<0. 01);Sobs index, Shannon index, and Chao index were increased; LPS was significantly decreased(P<0. 01), while the levels of 5-HT,SCFAs, occludin, claudin-1, and ZO-1 were significantly increased(P< 0. 05), indicating improved colonic barrier function;spontaneous alternation accuracy was increased(P<0. 001). In conclusion, Sijunzi Decoction has the effect of improving intestinal barrier in diabetic mice.

Abstract:Background/Objectives: Type 2 diabetes mellitus (T2DM) is a disorder characterized by insulin resistance, hyperglycemia, and dyslipidemia. Myricetin, a flavonoid found in various plants, has shown potential anti-diabetic effects in murine studies. This meta-analysis aimed to evaluate the impact of myricetin supplementation on glucose metabolism and lipid profiles in mouse models of metabolic diseases. Methods: A systematic review and meta-analysis were conducted in accordance with PRISMA guidelines (PROSPERO: CRD42084518999). Studies involving mice with metabolic disease models and exclusively using myricetin supplementation were checked across four databases (Embase, Scopus, PubMed, and WoS) until 23rd September 2024. The primary outcomes assessed were blood glucose (BG), insulin levels, triacylglycerol (TAG), total cholesterol (TC), HDL, and LDL. A random-effects model was applied to estimate standardized mean differences (SMD), and SYRCLE's risk-of-bias tool for animal studies was used. Results: Twenty-one studies with 514 mice met the inclusion criteria. Myricetin supplementation significantly reduced BG (SMD = −1.45, CI: −1.91 to −0.99, p < 0.00001, I2 = 74%), insulin (SMD = −1.78, CI: −2.89 to −0.68, p = 0.002, I2 = 86%), TAG (SMD = −2.60, CI: −3.24 to −1.96, p < 0.00001, I2 = 81%), TC (SMD = −1.86, CI: −2.29 to −1.44, p < 0.00001, I2 = 62%), and LDL (SMD = −2.95, CI: −3.75 to −2.14, p < 0.00001, I2 = 74%). However, the effect on HDL was not statistically significant (SMD = 0.71, CI: −0.01 to 1.43, p = 0.05, I2 = 83%). Conclusions: Myricetin supplementation improved glucose metabolism and lipid profiles in mouse models, suggesting its potential as a therapeutic agent for managing T2DM. However, further research is needed to confirm these findings in human studies.

Abstract:Diabetes mellitus is a clinically complex disease characterized by hyperglycaemia with disturbances in carbohydrate, fat and protein metabolism. The aim of this study was to determine the therapeutic effect of ethanolic extract (EE) from the green cocoon sericin layer of silkworm Bombyx mori on mice with type 2 diabetes mellitus (T2DM) and its hypoglycaemic mechanisms. The results showed that oral EE for 7 weeks had significant ameliorative effects on all the biochemical parameters studied in vivo . The levels of oral glucose tolerance and insulin tolerance were significantly improved. The hypoglycaemic rate in the 350 mg kg −1 high dosage group was 39.38%. The levels of nuclear factor kappa B (NFκB), interleukin 6 (IL-6) and tumour necrosis factor alpha (TNF-α) in the high dosage EE-treated group were significantly reduced, while activities of antioxidant enzymes superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were obviously increased. The islet area and the number of insulin-positive beta cells increased significantly in the high dose group. Furthermore, expression levels of insulin receptor (IR), insulin receptor substrate (IRS), phosphatidylinositide 3-kinase (PI3K), p-Akt and phospho-glycogen synthase kinase-3β (p-GSK3β) involved in insulin signalling were increased. Adenosine 5′-monophosphate-activated protein kinase (AMPK) and glucose transporter 4 (GLUT4) also were activated to regulate glucose metabolism in EE-treated groups. The levels of glucose 6-phosphatase (G6pase) and phosphoenolpyruvate carboxykinase (PEPCK) decreased, while the glucokinase (GK) level increased to promote glycolysis. The results clearly indicated that oral EE, especially at a high dose, could improve the glucose metabolism of T2DM by reducing inflammatory reactions, enhancing the antioxidant capacity and insulin sensitivity, and regulating the balance between glycolysis and gluconeogenesis, which means that EE has potential ameliorative effects on T2DM mice.

Abstract:Background. Type 2 diabetes mellitus (T2DM) is a kind of disorder of glucose and lipid metabolism with the main clinical manifestation of long‐term higher blood glucose level than the normal value. Farnesol X receptor (FXR)/ceramide signaling pathway plays an important role in regulating cholesterol metabolism, lipid homeostasis, and the absorption of fat and vitamins in diet. Gegen Qinlian Decoction (GQD) is a classical herbal formula, which has a good clinical therapeutic effect on diabetes-related metabolic syndrome. Objective. To investigate the effect of Gegen Qinlian Decoction (GQD) on hepatic gluconeogenesis in obese T2DM rats based on the FXR/ceramide signaling pathway regulating mitochondrial metabolism and endoplasmic reticulum stress (ERS). Methods. ZDF (fa/fa) rats were fed with high-fat diet to establish the T2DM model; GQD was given to T2DM model rats by gavage; changes of the general state and body weight of rats were recorded; fasting blood glucose was detected; blood insulin, blood ceramide, glycosylated hemoglobin in blood, acetyl CoA in liver mitochondria, and bile salt lyase in intestinal tissue were detected by ELISA. The content of T-β-MCA in blood was detected by LC-MS; the content of glycogen in liver tissue was detected by PAS staining; the expression of FXR, Sptlc2, and Smpd3 in ileum tissue, P-PERK, ATF6α, GRP78 BIP, and P-IRE1 in the liver, and CS and PC protein in liver mitochondria was detected by immunohistochemistry and western blot assay. The mRNA expression levels of FXR, Sptlc2, and Smpd3 in the ileum, PERK, ATF6α, GRP78 BIP, and IRE1 in the liver, and CS and PC in liver mitochondria were detected by qRT-PCR. Results. GQD can improve the general state of T2DM rats, slow down their weight gain, reduce the levels of fasting blood glucose, fasting insulin, glycosylated hemoglobin, blood ceramide, bile salt hydrolase in intestinal tissue, and acetyl CoA in liver mitochondria of T2DM rats, and increase the contents of liver glycogen and T-β-MCA in blood of T2DM rats. At the molecular level, GQD can inhibit the expression levels of FXR, Sptlc2, and Smpd3 in the ileum of T2DM rats and the protein and mRNA expression levels of oxidative stress-related factors in the liver. At the same time, GQD can increase the expression of CS and reduce the expression of PC in liver mitochondria of T2DM rats. Conclusion. GQD can inhibit the FXR/ceramide signaling pathway, regulate endoplasmic reticulum stress, enhance the CS activity of liver mitochondria, reduce the acetyl CoA level and PC activity of liver mitochondria, inhibit hepatic gluconeogenesis, protect islet β-cells, and control blood glucose.

Difference in Bone Mineral Quality Between Caucasians and African Americans

Abstract:Introduction: African-Americans have a lower fracture risk than Caucasians [1] but the reason for this difference remains unclear. Arguments have been offered that fracture risk is governed by extrinsic factors [2, 3], e.g., differences in bone size, or intrinsic factors [4-6], e.g. differences in bone mineral density (BMD) [7, 8]. The latter finding, which was primarily obtained from dual energy X-ray absorptiometry (DXA), generally concluded that bone from African-Americans had a greater BMD than bone from Caucasians. Other mineral density assessment techniques, notably quantitative backscatter electron imaging (qBEI) and computerized tomography (CT), provided data that suggested the opposite [9, 10]. To gain new insight regarding the race-related intrinsic material properties of bone, the present study sought to test the null hypothesis that there are no differences in the relative quantity or quality of trabecular bone mineral obtained from African-American and Caucasian subjects.
Study on Renal Protection of Gui-Zhi Decoction in Hyperuricemic Mice

Abstract:This study was aimed to investigate renal protective effects and mechanism of Gui-Zhi(GZ) decoction in hyperuricemic mice.Potassium oxonate was used to induce hyperuricemia mouse model.Mice were randomly divided into 6 groups,which were the blank control group,model group,allopurinol group (5 mg·kg-1) and GZ decoction group (900,1 799 and 3 598 mg·kg-1).Hematoxylin eosin staining was used to observe the histopathological changes of renal tissues in mice.Commercial assay kits were used to measure levels of uric acid (UA),creatinine (Cr) and blood urea nitrogen (BUN) in serum and urine,as well as the xanthine oxidase (XOD) activity in liver.Renal protein levels of urate transporter 1 (URAT1),glucose transporter 9 (GLUT9),ATP-binding cassette G member 2 (ABCG2),organic cation transporter 1 (OCT1),OCT2,organic cation/carnitine transporter 1 (OCTN1) and OCTN2 were detected by western blot.The results showed that compared with the model group,GZ decoction can obviously decrease serum levels of UA,Cr and BUN,increase urine levels of UA and Cr,resulting in the elevation of fractional excretion of UA in hyperuricemic mice.Additionally,GZ decoction obviously inhibited hepatic XOD activity in hyperuricemic mice.Furthermore,GZ decoction downregulated renal URAT1 and GLUT9 protein levels,upregulated renal ABCG2,as well as OCT1,OCT2,OCTN1 and OCTN2 protein levels in hyperuricemic mice.It was concluded that GZ decoction had hypouricemic and renal protective effects in hyperuricemic mice,which might be associated with the reduction of UA production via inhibiting hepatic XOD activity,promoting UA and other organic ion excretion via regulating renal organic ion transporter protein levels.
Genistein modulation of streptozotocin diabetes in male B6C3F1 mice can be induced by diet

Abstract:Diet and phytoestrogens affect the development and progression of diabetes. The objective of the present study was to determine if oral exposure to phytoestrogen genistein (GE) by gavage changed blood glucose levels (BGL) through immunomodulation in streptozotocin (STZ)-induced diabetic male B6C3F1 mice fed with three different diets. These three diets were: NTP-2000 diet (NTP), soy- and alfalfa-free 5K96 diet (SOF) and high fat diet (HFD) with 60% of kcal from fat, primarily rendered fat of swine. The dosing regimen for STZ consisted of three 100mg/kg doses (i.p.): the first dose was administered at approximately 2weeks following the initiation of daily GE (20mg/kg) gavage, and the second dose was on day 19 following the first dose, and the third dose was on day 57 following the first dose. In mice on the NTP diet, GE treatment decreased BGL with statistical significances observed on days 33 and 82 following the first STZ injection. In mice fed the HFD diet, GE treatment produced a significant decrease and a significant increase in BGL on days 15 and 89 following the first STZ injection, respectively. In mice fed the SOF diet, GE treatment had no significant effects on BGL. Although GE treatment affected phenotypic distributions of both splenocytes (T cells, B cells, natural killer cells and neutrophils) and thymocytes (CD4/CD8 and CD44/CD25), and their mitochondrial transmembrane potential and generation of reactive oxygen species, indicators of cell death (possibly apoptosis), GE modulation of neutrophils was more consistent with its diabetogenic or anti-diabetic potentials. The differential effects of GE on BGL in male B6C3F1 mice fed with three different diets with varied phytoestrogen contents suggest that the estrogenic properties of this compound may contribute to its modulation of diabetes.
