Abstract:A palladium-catalyzed regioselective decarboxylative direct C-H arylation of boron dipyrromethenes (BODIPYs) at the 2,6-positions has been developed as a late-stage approach to rapidly assemble a diversity-oriented BODIPY library. With the complement of this protocol, the direct C-H arylation of BODIPYs becomes regiocontrollable at alpha- and beta-positions. A new type of indole-fused BODIPY exhibiting bright red/NIR fluorescence with a large molar extinction coefficient (145 500 M-1 cm(-1)) and a high quantum yield (71%) has been synthesized for the first time.

Novel Meso-Polyarylamine-bodipy Hybrids: Synthesis and Study of Their Optical Properties

Abstract:A series of 14 meso-polyarylamine-BODIPY (borondipyrromethene) hybrids of the general structure A were synthesized. Two methods were used to prepare them. The first protocol involved a direct Liebeskind-Srogl cross-coupling of thiomethylbodipys 1-2 with arylaminoboronic acids (4 examples, 75-98%). The second method involves a two-step sequence: a Liebeskind-Srogl reaction to prepare 6 meso-bromoarylbodipys (58-83%) followed by a Suzuki coupling of these Br-containing BODIPYs with arylaminoboronic acids (10 examples, 44-84%). Seven of these derivatives displayed emission in the near-infrared region. The optical properties of compound 18 were rationalized in terms of its crystal structure.

Abstract:Boron dipyrromethene (BODIPY) derivatives 1 and 2 consisting of donor and acceptor units with dual photoresponses to solvent polarity and luminogen aggregation are developed through taking advantage of twisted intramolecular charge transfer (TICT) and aggregation-induced emission (ATE) processes. In nonpolar solvents, the locally excited (LE) states of the BODIPY luminogens emit intense green lights. Increasing solvent polarity brings the luminogens from the LE state to the TICT state, causing a large bathochromic shift in the emission color but a dramatic decrease in the emission efficiency. The red emission is greatly boosted by aggregate formation or ATE effect: addition of large amounts of water into the solutions of 1 and 2 in the polar solvents causes the luminogens to aggregate supramolecularly and to emit efficiently. The emission can be enhanced by increasing solvent viscosity and decreasing solution temperature, indicating that the ATE effect is caused by the restriction of the intramolecular rotations in the aggregates of the luminogens.

Abstract:A mild and efficient 1,4-diazabicyclo[2.2.2]octane (DABCO)-catalyzed three-component domino reaction was developed for the synthesis of highly functionalized NH-pyrroles from arylglyoxal monohydrates, enamino esters, and cyclic 1,3-dicarbonyl compounds in 1,4-dioxane at room temperature for 0.5 hours. Abstract:The molecular engineering of boron dipyrromethenes (BODIPYs) has garnered widespread attention due to their structural diversity enabling tailored physicochemical properties for optimal applications. However, catalytic enantioselective synthesis of structurally diverse boron-stereogenic BODIPYs through intermolecular desymmetrization and BODIPYs with atroposelectivity remains elusive. Here, we showcase rhodium(II)-catalyzed site-specific C-H (hetero)arylations of prochiral BODIPYs and polysubstituted BODIPYs with diazonaphthoquinonesand diazoindenines, providing efficient pathways for the rapid assembly of versatile (hetero)arylated boron-stereogenic and axially chiral BODIPYs through long-range desymmetrization and axial rotational restriction modes. The synthetic application of the procedures has been emphasized by the efficient synthesis of BODIPY derivatives with various functions. Photophysical properties, bioimaging, and lipid droplet-specific targeting capability of tailored BODIPYs are also demonstrated, indicating their promising applications in biomedical research, medicinal chemistry, and material science.

Abstract:The high tunabilities of the optical properties of BODIPYs have been demonstrated by various peripheral modifications over years. However, chemical modifications at the 1-and 7-positions of 8-substituted BODIPYs have been scarcely explored, since these sites are electronically and sterically least reactive. Herein, we report an effective rhodium(Rh)-catalyzed C-H activated alkynylation at the 1-and 7-positions of 8-pyrrole-appended BODIPYs, where the appended pyrrole moiety serves as a directing group to promote the alkynylations. In addition, some double annulation products of these alkynylated compounds were obtained by photo-cyclization with green light.

Abstract:Boron dipyrromethenes (BODIPYs) are some of the most popular and indispensable tetracoordinate boron compounds and have found widespread applications owing to their excellent spectroscopic and photophysical properties. BODIPYs possessing boron-stereogenic centres are scarce, and strategies for the synthesis of enantioenriched boron-stereogenic BODIPYs with structural diversity remain underdeveloped. In theory, the BODIPY core skeleton has several sites that could be decorated with different substituents. However, due to the lack of general and efficient asymmetric synthetic methods, this potential diversity of chiral BODIPYs has not been exploited. Here we demonstrate a modular enantioselective assembly of multi-substituted boron-stereogenic BODIPYs in high efficiency with excellent enantioselectivities. Key to the success is the Pd-catalysed desymmetric Suzuki cross-coupling, enabling the precise discrimination of the two α C–Cl bonds of the designed prochiral BODIPY scaffold, giving access to a wide range of highly functionalized boron-stereogenic BODIPYs. Derivatizations, photophysical properties and applications in chiral recognition of the obtained optical BODIPYs are further explored.

Abstract:Boron dipyrromethenes (BODIPYs) are a class of tetracoordinate boron compounds that have attracted considerable interest in recent decades due to their excellent spectroscopic properties and structural versatility, leading to their widespread applications in chemical, biological, pharmaceutical and materials science. However, BODIPYs possessing boron-stereogenic centres in enantioenriched forms are rare and catalytic methodology to enantioselectively prepare a boron-stereogenic BODIPY remains elusive. Here we report a palladium-catalysed protocol for the enantioselective synthesis of boron-stereogenic BODIPYs via a desymmetric intramolecular C–H arylation reaction. This method gives access to a wide range of highly functionalized boron-stereogenic BODIPYs, including six- to nine-membered boron heterocycles, with good to excellent enantioselectivities. The discrimination of the two α C–H bonds of the BODIPY core is enabled by the co-action of steric hindrance and attractive interaction in the catalytic chiral pocket. Photophysical properties, derivatizations and applications in chiral recognition of the obtained chiroptical BODIPYs are investigated. This work enriches the chemical diversity of chiroptical BODIPY dyes.

Abstract:The synthesis of new B-O BODIPY derivatives functionalized with different alkoxy or diarylalkoxy derivatives is described. These compounds were synthesized from the reaction of different B-F BODIPY precursors with various alcohols and phenols, in the presence of AlCl3. Water-soluble dyes could be synthesized as well with this method, specifically by the introduction of polyethyleneglycol (PEG) groups. A photophysical study of the different compounds was performed, and showed that the B-O BODIPY derivatives exhibit rich fluorescence properties. Finally, the conjugation of the BODIPY core has been extended using two distyryl groups, hence providing NIR emitting BODIPY derivatives, in which one or two PEG groups have been anchored, making these systems very promising for future medical imaging applications.

Abstract:Fluorophores based on the 4-bora-3a,4a-diaza-s-indacene (aka boron dipyrromethene, boron dipyrrin, or BODIPY) platform have found diverse applications in different fields of modern science, medicine and (bio)technology. In this review we describe the numerous postfunctionalization methodologies of the boron dipyrromethene core designed and realized by research groups around the globe. In the postfunctionalization approach, boron dipyrromethenes with reactive functionalities attached directly to the core (halogen or hydrogen atoms, methyl, formyl, or alkylthio groups) are used as starting materials for further derivatization. The various synthetic methods towards these starting compounds and their postmodification are reviewed. We discuss the different strategies devised for postderivatization of the BODIPY nucleus at all possible positions (the pyrrole carbons, the meso-carbon, and the boron atom) and compare them concisely with the standard prefunctionalization methodology. Important properties and applications of a number of substituted BODIPYs made by the methods described in this review are also presented.

Abstract:Direct structural modification of small‐molecule fluorophores represents a straightforward and appealing strategy for accessing new fluorescent dyes with desired functionalities. We report herein a general and efficient visible‐light‐mediated method for the direct C‐H functionalization of BODIPY, an important fluorescent chromophore, using readily accessible and bench‐stable aryl and alkenylthianthrenium salts. This practical approach operates at room temperature with extraordinary site‐selectivity, providing a step‐economical means to construct various valuable aryl‐ and alkenyl‐substituted BODIPY dyes. Remarkably, this protocol encompasses a broad substrate scope and excellent functional‐group tolerance, and allows for the modular synthesis of sophisticated symmetrical and asymmetrical disubstituted BODIPYs by simply employing different combinations of thianthrenium salts. Moreover, the late‐stage BODIPY modification of complex drug molecules further highlights the potential of this novel methodology in the synthesis of fluorophore‐drug conjugates.

Abstract:Aromatic annulation on the periphery of BODIPY has proven to be particularly effective in expanding its pi-conjugated system, thereby enabling near-infrared (NIR) absorption and emission. However, achieving this shift in BODIPY's absorption and emission to the NIR region without compromising fluorescence quantum yield remains a significant challenge. Herein, we report a new family of symmetrical dibenzothieno and dibenzothieno[2,3-d]thieno[a]-fused BODIPYs (5a-c and 8a-c) synthesized via a simple four-step route involving regioselective 1,7-bromination to selectively enhance the reactivity specifically at the 1,7 positions. These aromatic heterocyclic [a]-fused BODIPYs dyes exhibit "butterfly-shaped" crystal structures, with a tunable dihedral angle between the indacene plane and its "wings" ranging from 2.47 degrees to 11.49 degrees. The symmetrical [a]-fusion of dibenzothieno and dibenzothieno[2,3-d]thieno rings results in NIR absorption and emission with high fluorescence quantum yields (Phi(F) > 0.8), and effectively narrows the HOMO-LUMO gap by elevating the energy level of the HOMO. The high fluorescence quantum yields and tunable HOMO levels of these aromatic heterocyclic [a]-fused BODIPY dyes make them highly promising as functional chromophores, particularly in the field of bioimaging. This work offers valuable insights for developing NIR fluorescent dyes with high fluorescence quantum yields for use as photosensitizers.
