Abstract:OBJECTIVE To establish a method to determine the contents of indigo,indirubin,tryptanthrin and isorhamnetin in roots,stems and leaves from Baphicacanthus cusia by high performance liquid chromatography (HPLC) and to evaluate the effects of three different drying methods (oven drying,shade drying and freeze drying).METHODS This experiment was conducted by using Dikma Diamonsil C18 column and Agilent 1200 HPLC-VWD system.Gradient elution was applied by using 0.2％ phosphoric acid methanol solution and 0.2％ phosphoric acid aqueous solution as the mobile phase.The flow rate was kept at 1.0 mL·min-1.The detection wave length was set at 289 nrn.The column temperature was 25 ℃ and the injection volume was 10μL.RESULTS Totally 36 samples were determined in four batches of roots,stems and leaves from B.cusia by oven drying,shade drying and freeze drying treatments,respectively.Only tryptanthrin could be detected in roots from B.cusia.Indigo,indirubin and tryptanthrin could be detected simultaneously in stems from B.cusia.Indigo,indirubin,tryptanthrin and isorhamnetin could be detected simultaneously in leaves from B.cusia.The content of indigo in stems from B.cusia by freeze drying treatment was the highest among three different drying methods,as well as in leaves from B.cusia.The contents of indirubin in stems from B.cusia by three different drying treatments were basically the same,as well as in leaves from B.cusia.The content of tryptanthrin in roots from B.cusia by shade dryingtreatment was the highest among three different drying methods,as well as in stems from B.cusia,as well as in leaves from B.cusia.The contents of isorhamnetin in leaves from B.cusia by oven drying and freeze drying treatments were basically the same,but isorhamnetin could not be detected by shade drying treatment.CONCLUSION This established HPLC method is simple and quick,precise and accurate,with high sensitivity and good reproducibility,and suitable for quality control of medicinal materials in different parts from B.cusia.The drying method of roots,stems and leaves from B.cusia should be appropriately chosen according to its medicinal parts and the active ingredients.
