Long non-coding RNA ST8SIA6-AS1 facilitates proliferation and invasion in hepatocellular carcinoma by regulating microRNA-142-3p

Abstract:Objective:To explore the effect of ST8SIA6-AS1 targeted binding of microRNA (miRNA, miR)-142-3p on the proliferation and invasion of hepatocellular carcinoma (HCC).Methods:Gene expression profiling interactive analysis (GEPIA) was used to verify the differential expression of ST8SIA6-AS1 in hepatic cell carcinoma (HCC) and adjacent normal tissues. Real-time quantitative reverse transcriptase-polymerase chain reaction (RT-qPCR) was used to detect the tissue level of ST8SIA6-AS1 and miR-142-3p and cell level (human liver cancer cell lines HepG2, SSMC-7721, HCCLM3 and Hep3B and normal liver cell line L02). Cell counting kit-8 (CCK-8) and colony formation assays were used to detect the proliferation of Hep3B cells. Transwell method was used to detect the migration and invasion of Hep3B cells. Bioinformatics and double luciferase reporting experiments verified the targeting relationship between ST8SIA6-AS1 and miR-142-3p. The data were expressed by mean±standard deviation ( Mean± SD). T-test was used between the two groups. Univariate analysis of variance was used to compare the two groups. Spearman rank test was used to analyze the correlation. Results:The results of GEPIA showed that the expression of ST8SIA6-AS1 in HCC tissues was higher than that in normal tissues. The results of RT-qPCR showed that the expression of ST8SIA6-AS1 in HCC was significantly higher than that in adjacent tissues (0.778±0.363 vs. 1.951±0.575, t=11.370, P<0.05). The miR-142-3P in HCC was significantly lower than that in paracancerous tissues (0.881±0.374 vs. 0.371±0.164, t=9.395, P<0.05). The expression of ST8SIA6-AS1 in HepG2, SMMC-7721, HCCLM3 and Hep3B cells (3.84±0.27, 5.81±0.60, 4.47±0.55, 5.94±0.69) was significantly higher than that in L02 cells (0.26±0.04 vs. 1.00±0.01, t=9.423, P<0.05). The relative expression of ST8SIA6-AS1 in Hep3B cells with ST8SIA6-AS1 knock-down was significantly lower than that in control cells (0.26±0.04 vs. 1.00±0.013, t=9.423, P<0.05). The results of colony formation experiment showed that the colony number of Hep3B cells with ST8SIA6-AS1 knock-down was lower than that of the control group [(32.60±6.70) vs. (100.00±10.00), t=10.040, P<0.05], and CCK-8 results showed that the 72-h survival rate of Hep3B cells with ST8SIA6-AS1 knock-down was lower than that of the control group [(0.71±0.06)% vs. (1.14±0.09)%, t=6.886, P<0.05]. The results of Transwell assay showed that the vertical migration rate of Hep3B cells was lower than that in the control group [(53.38±6.49)% vs. (100.00±13.00)%, t=5.641, P<0.05]. The invasion rate of Hep3B cells knocked down by ST8SIA6-AS1 was lower than that of the control group [(44.98±8.42)% vs. (100.00±10.00)%, t=7.485, P<0.05]. Double luciferase report showed that the overexpression of miR-142-3P decreased the fluorescence activity of ST8SIA6-AS1-wild-type (WT) cells, and ST8SIA6-AS1 negatively regulated the expression of miR-142-3p. Interfering with the expression of ST8SIA6-AS1 can inhibit the proliferation, migration and invasion of HCC cells, which can be reversed by silencing miR-142-3p. Conclusion:ST8SIA6-AS1 exhibited oncogenic function in HCC by competitively sponging miR-142-3p.
