Abstract:Enantioselective functionalization of inert C(sp3)–H bonds of prochiral alkyl methylene groups is particularly challenging. Direct functionalization of aliphatic C–H bonds is becoming a more and more popular and powerful tool in organic synthesis because of its advantages over traditional transformations, including the ability to avoid tedious, costly, and somewhat hazardous processes. Accompanied by difficulties in activating C–H bonds and achieving specific transformations from the activated C–H bonds, so-called reactivity and selectivity are major concerns in direct C–H functionalization. The renaissance of transition-metal chemistry since the late 1950s has provided powerful solutions for these problems. Many transition-metal complexes were synthesized and found to be capable of selectively reacting with C–H bonds to form organometallic complexes. With select transition-metal complexes, site-selective and direct C–H functionalization controlled by electronic, steric, and/or stereoelectronic factors has been showcased in many elegant examples. In particular, a recently developed directing strategy has allowed for the control of regioselectivity by relying on the distance between targeted C–H bonds and directing groups.1Rouquet G. Soc. 2015; 137: 13776-13779Crossref Scopus (81) Google Scholar One of the biggest challenges in direct C–H functionalization is the control of both diastereo- and enantioselectivity. In past decades, gratifying achievements have been made through enantioselective carbene and nitrene insertion mediated by transition-metal catalysts supported by rationally designed chiral ligands.4Liao K. Nature. 2016; 533: 230-234Crossref Scopus (237) Google Scholar In comparison, hampered by harsh reaction conditions and limited efficiency of chiral ligand sets, direct transition-metal insertion of C–H bonds to induce asymmetric C(sp3)–H bond conversion is far behind other types of asymmetric catalysis. In general, the cleavage of C–H bonds through transition-metal insertion requires high energy, which means that elevated reaction temperatures are essential, complicating proper differentiation of C(sp3)–H bonds with highly similar chemical environments. Therefore, suitable transition-metal catalysts and chiral ligands are highly sought after to milden the reaction conditions and control the stereoselectivity. Although challenges remain, efforts to achieve highly stereoselective C(sp3)–H functionalization are still ongoing. In 1993, Schröder and Schwarz reported the gas-phase dehydrogenation of racemic 3-methylpentan-2-one by bare Fe(I) via remote C–H bond activation.5Schröder D. Schwarz H. J. Am. Chem. Soc. 1993; 115: 8818-8820Crossref Scopus (26) Google Scholar They found that this process exhibited stereoselectivity in discriminating C(sp3)–H bonds at the C4 position. This is the first elegant report of stereoselectivity in a C–H activation process not involving carbene insertion and exemplifying its great mechanistic importance. In terms of generating chiral centers from prochiral substrates, there are two categories in asymmetric C–H functionalization. (1) If the cleavage of C–H bonds does not occur at the prochiral carbon, the outcome of stereoselectivity is discrimination of the two identical moieties of symmetric substrates, thus called desymmetrization.6Giri R. Rev. 2009; 38: 3242-3272Crossref Scopus (1329) Google Scholar (2) On the other hand, there is the distinction between two C–H bonds of prochiral methylene groups with almost the same chemical environment. This kind of reaction is both much more challenging and important because of its broader potential substrate scope and application in synthetic organic chemistry. In the past, palladium-catalyzed C(sp3)–H insertion of methylene groups by a directing strategy proved to be sufficient. For discriminating similar C–H bonds at prochiral methylene groups, chiral groups were introduced to substrates to control the diastereoselectivity by remote chirality relay.7Chen K. Li Z.-W. J. 2015; 21: 7389-7393Crossref Scopus (43) Google Scholar However, given that stoichiometric amounts of the chiral source are required and insufficient suitable chiral auxiliaries have been reported to date, this is not an ideal solution. Obviously, asymmetric catalysis utilizing only a catalytic amount of chiral ligands is more appealing, because it could highly decrease costs and enhance practicability. In contrast to the aforementioned asymmetric carbene and nitrene insertions, examples of asymmetric C(sp3)–H functionalization from methylene groups via transition-metal insertion have rarely been reported. Applying a bidentate directing strategy, the Yu group recently used catalytic amounts of L-tert-leucine as a transient chiral directing group to achieve enantioselective arylation of benzylic C–H bonds of ortho-alkyl-substituted benzaldehydes with excellent enantiomeric ratios.8Zhang F.-L. Hong K. Li T.-J. Park H. Yu J.-Q. Science. 2016; 351: 252-256Crossref Scopus (457) Google Scholar However, for more inert alkyl methylene C–H bonds, similar reactions have not been approached. Conceptually, Duan and colleagues reported the first example of asymmetric benzylic C(sp3)–H arylation with the assistance of a chiral ligand. On the basis of the well-developed β-arylation reactions of aliphatic carboxylic amides bearing an 8-aminoquinoline, they used monodentate chiral phosphoric amides as ligands to control the enantioselectivity. It is possible, though, that this arylation might also proceed without involvement of the ligand, thus giving rise to the racemic product and lowering the enantioselectivity. Moreover, such a transformation proved to be unsuccessful with an alkyl methylene group.9Yan S.-B. Zhang S. Duan W.-L. Org. Lett. 2015; 17: 2458-2461Crossref Scopus (131) Google Scholar Thus, bidentate directing groups might not be ideal for asymmetric functionalization of methylene C(sp3)–H bonds with external chiral ligands, probably because they occupy an additional coordinating site on the metal center and compete with ligand coordination. To solve this problem, applying a monodentate directing strategy seems reasonable because it allows for another free coordinating site on the catalytic Pd center and the possibility of using chiral bidentate chelating ligands, which has proved to be very successful in asymmetric catalysis in the past. Ideally, the proper bidentate ligand can both accelerate the activation rate and promote the enantioselectivity. Following this intelligent analysis and design, Yu and coworkers10Chen G. Gong W. Science. 2016; 353: 1023-1027Crossref Scopus (225) Google Scholar now present in Science the first example of an enantioselective arylation of β-methylene C–H bonds of aliphatic amides with newly designed chiral ligands (Scheme 1). They elaborately designed bidentate and chiral acetylprotected aminoethyl quinoline (APAQ) ligands and applied them successfully in β-arylation reactions of aliphatic amides with aryl iodides. As a key issue, the electron-deficient amide moity acted as a weakly coordinating directing group. Installing an aminoethyl or aminopropyl group in the ligand to form a six- or seven-membered chelate with Pd proved crucial for catalyst activity in this direct arylation of methylene C–H bonds. Ligands bearing a sterically hindered 3,5-di-tert-butylphenyl group gave rise to the best yields and enantiomeric ratios. Further optimization of the ligands led to the introduction of another stereocenter in the homobenzylic position, such that a syn-arrangement resulted in higher er values and the absolute configuration in the α position controlled enantioselection. This method proved to have a wide substrate scope for applied amides, and a variety of both electron-rich and electron-poor aryl iodides (meta- and para-substituted) were applicable. It also showed portability toward carboxylic acids acting as the directing group instead of the amide group, as demonstrated in the cis-selective β-arylation of a cyclopropane and cyclobutane amino acid. Finally, density functional theory (DFT) calculations of the transition states showed the difference in their free energies, further proving the concept of the rational design. Indeed, combining weakly coordinating monodentate substrates and ligand acceleration provides an intriguing approach to solving challenges in the enantioselective C–H functionalization of methylene groups. With a deep understanding of the intrinsic properties of the efficient ligand sets, Yu and colleagues found the sweet spot of enhancing both reactivity and selectivity in the reported reaction. Their approaches to developing these chiral ligands will inspire new ideas to further push this field forward. At this stage, the scope is still limited by the essentiality of the specific directing groups, and new catalytic systems should be developed for other kinds of substrates. Thus, in order to find more general solutions, chemists devoted to this field still have a long way to go.
