Protective effect of Bilberry anthocyanidin on microwave radiation damage of vascular endothelial cells

Abstract:Objective To discuss the protective effect of different concentrations of Bilberry anthocyanidin on microwave radiation damage of vascular endothelial cells and to clarify the application value of anthocyanidin in radiation injury.Methods The anthocyanidin was extracted by the method of microwave-assisted.The EVC 304cells were cultivated in vitro.The cells were divided into control group,20mW·cm-2 microwave radiation group and 25,50,and 100mg·L-1 anthocyanidin group.The changes of the cell morphology were observed by inverted microscope,the inhibitory rate of proliferation was detected by MTT method,the cell cycle and apoptotic rate were detected by flow cytometry,and the activity of caspase-3was assayed.Results The cell morphology in microwave radiation group showed contraction,rounded,smaller size,cell gap widening,and most of the cells showed breaking phenomenon.Compared with microwave radiation group,the morphology of cells in anthocyanidin group was significantly improved.The MTT experiment result showed the inhibitory rate of proliferation of EVC 304cells reached 58.6%after 20mW·cm-2 microwave radiation for 24hand the inhibitory rate of proliferation of cells in anthocyanidin group was 30.5%,which was lower than that in microwave radiation group(P0.05).The flow cytometry results showed that the apoptotic rate of the cells in control group was 2.9%;25,50,and100mg·L-1 anthocyanidin could reduce the apoptotic rate of the cells in microwave group from 14.9%to 6.1%,4.2%,and 3.5%.Compared with microwave radiation group,the activity of caspase-3of the cells in microwave radiation group was higher than that in control group(P0.05).25,50,and 100mg·L-1 anthocyanidin could reduce the activity of caspaase-3,which was lower than that microwave radiation group(P0.05).Conclusion Bilberry anthocyanidin has protective effect on the radiation damage of vascular endothelial cells.
