Abstract:Transition metal-catalysed C-H bond functionalisations have been extensively developed in organic and medicinal chemistry. Among these catalytic approaches, the selective activation of C(sp(3))-H and C(sp(2))-H bonds is particularly appealing for its remarkable synthetic versatility, yet it remains highly challenging. Herein, we demonstrate the first example of temperature-dependent selective C-H functionalisation of unactivated C(sp(3))-H or C(sp(2))-H bonds at remote positions through palladium catalysis using 7-pyridyl-pyrazolo[1,5-a]pyrimidine as a new directing group. At 120 degrees C, C(sp(3))-H arylation was triggered by the chelation of a rare [6,5]-fused palladacycle, whereas at 140 degrees C, C(sp(2))-H arylation proceeded instead through the formation of a 16-membered tetramer containing four 7-pyridyl-pyrazolo[1,5-a]pyrimidine-palladium chelation units. The subsequent mechanistic study revealed that both C-H activations shared a common 6-membered palladacycle intermediate, which was then directly transformed to either the [6,5]-fused palladacycle for C(sp(3))-H activation at 120 degrees C or the tetramer for C(sp(2))-H arylation at 140 degrees C with catalytic amounts of Pd(OAc)(2) and AcOH. Raising the temperature from 120 degrees C to 140 degrees C can also convert the [6,5]-fused palladacycle to the tetramer with the above-mentioned catalysts, hence completing the C(sp(2))-H arylation ultimately.

Abstract:A C-H arylation of thiopyran derivatives with aryl halides has been developed. Under the catalysis of Pd(OAc)2/Ag2CO3, the C-H arylation takes place at the α-position of the thiopyran ring. When dibromo-substituted compounds are used as reactants, double C-H arylations may occur on the same thiopyran ring at its α- and β-positions.

Abstract:Pd-catalyzed decarboxylative cross-couplings of 2-(2-azaaryl)acetates with aryl halides and triflates have been discovered. This reaction is potentially useful for the synthesis of some functionalized pyridines, quinolines, pyrazines, benzoxazoles, and benzothiazoles. Theoretical analysis shows that the nitrogen atom at the 2-position of the heteroaromatics directly coordinates to Pd(II) in the decarboxylation transition state.

Abstract:C2-arylation of N-acyl pyrroles with aryl halides is developed for the first time using Pd(PPh3)4 as a catalyst in combination with Ag2CO3 under air, which allowed the application of a good compatibility catalytic system. This protocol provides a straightforward method for the preparation of valuable arylated pyrroles in moderate to good yields under the standard conditions with good substrate tolerance. Interestingly, while N-benzoyl pyrroles reacted well, the use of substrates with a thiophene or furan ring indicated that the thiophene and furan rings are more reactive than pyrrole for the present catalytic system.

Abstract:Palladium-catalyzed C-H functionalization of acyldiazomethanes with aryl iodides has been developed. This reaction is featured by the retention of the diazo functionality in the transformation, thus constituting a novel method for the introduction of diazo functionality to organic molecules. Consistent with the experimental results, the density functional theory (DFT) calculation indicates that the formation of Pd-carbene species in the catalytic cycle through dinitrogen extrusion from the palladium ethyl diazoacetate (Pd-EDA) complex is less favorable. The reaction instead proceeds through Ag2CO3 assisted deprotonation and subsequently reductive elimination to afford the products with diazo functionality remained. This C-H functionalization transformation can be further combined with the recently evolved palladium-catalyzed cross-coupling reaction of diazo compounds with aryl iodides to develop a tandem coupling process for the synthesis of α,α-diaryl esters. DFT calculation supports the involvement of Pd-carbene as reactive intermediate in the catalytic cycle, which goes through facile carbene migratory insertion with a low energy barrier (3.8 kcal/mol).

Abstract:A mild and selective C6 arylation strategy for pyrrolo[2,3-d]pyrimidine derivatives with arylboronic acids at room temperature is described. This unified protocol has been achieved by the synergistic combination of Pd(II)/TEMPO catalysis and CF3CO2H promotion under silver-, base-, and additive-free conditions. The broad substrate scope, good functional group tolerance, excellent regioselectivity, and air and moisture tolerant conditions make this process attractive for the effective synthesis and modification of targeted small molecule drugs.

Abstract:In this study, we demonstrate that phosphinoacridines are efficient bidentate ligands for palladium‐catalyzed carboxylative C‐N coupling reactions under blue LED irradiation. This method facilitates the direct synthesis of arylcarbamates using a range of non‐activated aryl halides, such as iodides and bromides, with various amines under atmospheric pressure of CO2. The optimized conditions exhibit high tolerance to sensitive functional groups, resulting in very good to excellent yields of the desired products. This approach expands the scope of palladium‐catalyzed carboxylation reactions and offers an environmentally friendly route to arylcarbamates.

Abstract:Palladium-catalyzed arylation of unactivated beta-C(sp(3))-H bonds in carboxylic acid derivatives with aryl iodides is described for the first time using 2-amino-5,6-difluorophenyl-1H-pyrazole as an efficient and readily removable directing group. Two fluoro groups are installed at the 5- and 6-position of the anilino moiety in 2-aminophenyl-1H-pyrazole, clearly enhancing the directing ability of the auxiliary. In addition, the protocol employs Cu(OAc)(2)/Ag3PO4 (1.2/0.3) as additives, evidently reducing the stoichiometric amount of expensive silver salts. Furthermore, this process exhibits high beta-site selectivity, compatibility with diverse substrates containing alpha-hydrogen atoms, and excellent functional group tolerance.

Abstract:Methods that can directly arylate pyridines have significant potential in synthetic chemistry. Herein, we present palladium catalyzed C-H arylation of pyridines with aryl triflates that occurs regioselectively at the C3 positions of the pyridine core. A variety of aryl triflates and pyridines were applicable to the reaction to provide C3-arylated pyridines regardless of sterics or electronics of the pyridine cores.

Abstract:The apparent and the real: What looks like a Friedel-Crafts alkylation reaction of electron-deficient pyrroles is actually a Pd(II)-catalyzed, norbornene-mediated C-H activation reaction, in which the alkylation of the pyrrole core occurs by reductive elimination. As well as ethyl-1H-pyrrole-2-carboxylate, several other 2,3-disubstituted pyrroles underwent the selective C5 alkylation in good yield.

Abstract:The first example of catalytic thiocarbonylation of aryl iodides using CO2 has been achieved employing a combination of PdCl2 and carbazole-derived phosphine ligands. Under mild conditions, a broad scope of aryl iodides were converted to the desired thioester products in the presence of aryl or alkyl thiols (33 examples, up to 96% yields). The choice of metal, ligands, and reductant were crucial for high efficiency and chemoselectivity. Moreover, this strategy provided an effective method for the late-stage functionalization of biorelevant molecules.

In this new transformation, the aromatic C(sp2)H bond was successfully acylated to give diverse aromatic ketones regioselectively in moderate to good yields. Abstract:The C-H arylation of (benzo)oxazoles or (benzo)thiazoles with aryltrimethylammonium triflates was carried out via Pd-catalyzed C-H/C-N cleavage. Oxazoles, thiazoles, benzoxazole, and benzothiazole were arylated using activated and deactivated aryltrimethylammonium triflates to give 2-aryl(benzo)oxazoles or 2-aryl(benzo)thiazoles in reasonable to excellent yields.

Abstract:A novel palladium catalyzed direct ortho-arylation of N-phenacylpyridinium bromide was developed. The amazing N-phenacyl group regioselectively activates the C-H bond of pyridine and automatically departs from the arylated products. A kinetic isotope effect study proved that the reaction went through a C-H-bond activation pathway and 2,6-diphenylpyridine was produced stepwise from 2-phenylpyridine.
