Expression of Twist,Snail,Slug and its significance in oral squamous cell carcinoma progression

Abstract:PURPOSE: To investigate the expression of Twist,Snail and Slug in oral squamous cell carcinoma(OSCC) patients as well as its clinical and pathologic significance.METHOD: Twist,Snail and Slug expressions were analyzed in 60 patients with OSCC by immunohistochemistry(IHC).The correlation between the expression of Twist,Snail and Slug and patients' clinicopathologic characteristics was evaluated,including prognosis.The data was analyzed with SPSS10.0 software package.RESULTS: Twist expression was detected in 70% of 60 OSCC patients.Twist protein expression was detected in 35.7% cases of primary cancer tissues without lymph node metastasis,64.3% cases of primary cancer tissues with lymph node metastasis,55.6% cases of early stage primary cancer tissues and 81.8% cases of advanced stage primary cancer tissues.The presence of lymph node metastasis and tumor stage had association with Twist expression(P=0.012,P =0.046).Snail and Slug expression were detected in 63.3%,58.3% of 60 OSCC patients.There was no significance correlation between Snail,Slug expressions and the clinicopathological parameters.During the follow-up period(6-59 months),18 patients(30%,18/60) suffered tumor recurrence.Recurrences were found in 2 of 16 patients with Twist negative expression and in 16 of 42 patients Twist positive expression.A correlation between Twist protein expression and recurrence was detected(P=0.025).In Cox regression analysis,Twist expression remained the only independent variate(P=0.042) and the other factors were not significant.But no significant correlations between Snail,Slug expressions and recurrence were found(P>0.05).CONCLUSIONS: Twist,Snail and Slug protein was overexpressed in OSCC.Twist expression was involved in OSCC progression.Twist may serve as a predictor for lymph node metastasis and poor prognosis.But,there was no significant difference between the expression of Snail,Slug and the clinicopathological parameters.

Abstract:Objective To detect the gene expression difference between gastric cancer tissue and lymph node metastasis,screen different express genes,and study its mechanism of metastasis and the relationship with biological behavior.Methods Using U133plus 2.0 gene chip technology,we detected the gene expression difference between gastric cancer tissue and epithelial cells of lymph node metastasis in five patients,and screened out the differentially expressed gene Twist-l.In vitro,the cell proliferation and apoptosis level were measured by using gene over-expression and gene knockout.Metastasis ability of carcinoma cells was detected by cell scratch assay.The expression changes of epithelial-mesenchymal transition (EMT)-associated protein (E-cadherin,Vimentin) in Twist-1 overexpression and gene knockout cells were determined by Western blot.Finally,we detected the expression of Twist-1 in gastric cancer tissue,and its correlation with TNM stage was analyzed.Results The expression of Twist-1 was higher in epithelial cells than in gastric cancer tissue (12.12±3.21 vs.2.07±0.71,P＜0.01).There was no correlation of the expression of Twist-1 with cell proliferation(absorbance of cell proliferation:negtive control 0.84±0.16,null vector control 0.74 ±0.06 and Twist-1 expression cell 0.71 ± 0.07) and apoptosis [cell apoptosis rate:negtive control (2.05±0.08)％,null vector control (4.31±0.07)％ and Twist-1 expression (3.95±0.09)％],but cell migration ability enhanced.In Twist-1 over-expression group,the level of E-cadherin was decreased,while vimentin increased.Conclusions Twist-1 gene changes might be correlated with the metastasis of gastric cancer by the way of EMT.
Wnt1 Correlates with Lymph Node Metastasis in OSCC

Abstract:Objective: To study whether the expression of Wnt1 in patients with oral squamous cell carcinoma (OSCC) were correlated to lymph node metastasis. Methods: Immunohistochemistry was used to detect Wnt1 expression in OSCC tissues of 60 patients. Western blot was used to detect Wnt1 expression in 5 pairs of OSCC tissues and adjacent normal mucosa tissues . Real-time PCR and Western blot were used to detect Wnt1 expression on mRNA and protein levels re-spectively in HN-12 cells, which was highly metastatic compared with HN-4 cells. Growth curve, scratch test and tran-swell assay were used to detect the growth, migration and invasion ability of HN-12 and HN-4 cells which were treated with or without Wnt1 recombinant protein. Results: Wnt1 protein was up-regulated in 24 cases of OSCC with lymph node metastasis compared to 16 cases without lymph node metastasis (P<0.01). Moreover, the expression of Wnt1 on mRNA and protein levels was both much higher in HN-12 cells than HN-4 cells. Lastly, Wnt1 could notably enhanced cell growth, migration and invasion. Conclusion: The high expression of the key member Wnt1 of Wnt signal pathway was re-lated to lymph node metastasis of OSCC.
Expression of Twist Protein in Laryngeal Squamous Cell Carcinoma and Its Relationship with Prognosis

Abstract:Objective To investigate the expression of Twist protein in laryngeal squamous cell carcinoma,and to explore its roles in the progression of the disease.Methods Immunohistochemical S-P methods were used to detect Twist protein in 80 specimens of laryngeal squamous cell carcinoma selected from the Pathology Department of our hospital during 2005 and 2010,and 30 specimen of side cancer tissues were taken as controls.Results ①Expression of Twist protein in laryngeal squamous cell carcinoma was higher than that in side cancer tissue,and the differences were statistically significant(χ2=11.657,P0.01).②The expression of Twist protein in patients in Ⅲ-Ⅳ stages,T3-T4,or with lymph node,distant metastasis and poorly differential degrees was significantly higher than that in the patients in Ⅰ-Ⅱ stages,T1-T2 stages or with great and moderate differentiation,non-lymph node and non-distant metastasis(P0.05,P0.01),but it was not related to gender and age(P0.05).③The total 5-year survival rate in the 75 patients with laryngeal squamous cell carcinoma was 42.7%,in the Twist protein-positive patients it was 32.1%,and in Twist protein-negative patients it was 55.6%.Kaplan-Meier survival analysis showed that the survival rate in Twist protein-positive patients with laryngeal squamous cell carcinoma was significantly lower than that in wist protein-negative patients,and the differences were statistically significant(χ2=6.219,P0.05).Conclusion The overexpression of Twist protein might be an important biological marker for malignant transformation and invasion and matastasis of laryngeal squamous cell carcinoma,and the up-regulation of the expression of Twist predicts more invasion and poor prognosis.

Abstract:Objective To investigate the expression of Twist in colorectal carcinomas and identify the clinical and pathological use of it.Methods The expressions of Twist were detected by Immunohistochemical S-P method in 112 cases of colorectal carcinomas and 40 cases of normal colorectal tissues from surgical resections.Results The positive rate of Twist in colorectal carcinomas was 69.6% while the positive rate in normal gastric tissues was 17.5%,a significant difference was found between them(χ2=12.11,p=0.001); Twist overexpression was related with the tumor differentiation(χ2=9.575,p=0.008),the depth of invasion(χ2=8.183,p=0.017),lymph node metastasis(χ2=4.05,p=0.044) and Duke's staging(χ2=11.114,P=0.011),while not related with age(p=0.286),sex(p=0.887) difference.Conclusion Twist may play an important role in the formation and development of colorectal carcinomas,The expression of Twist is a useful marker in the clinical and pathological diagnosis of colorectal carcinomas.
Hyperthermia Inhibited the Migration of Tongue Squamous Cell Carcinoma Through TWIST2.

Abstract:BackgroundHyperthermia has been shown promising in the treatment of head and neck squamous cell carcinoma (HNSCC); however, the mechanism underlying hyperthermia reducing tumor metastasis is poorly elucidated. TWIST2, an important transcription factor of epithelial-mesenchymal transition (EMT), plays a critical role in the tumor progression and metastasis. The role of TWIST2 in tongue squamous cell carcinoma (TSCC) and its association with hyperthermia still have not been reported.MethodThe correlations between TWIST2 expression and the clinical-pathologic characteristics of 89 patients with TSCC were evaluated by immunohistochemical staining. TSCC cell lines transfected with siRNA against TWIST2 were heated for 40min at 42.5 degrees C, and the migration capability of cells was examined by migration assay. Xenograft tumors in nude mice were treated by hyperthermia, and TWIST2 expression was measured.ResultsOur data showed that TWIST2 expression was associated with the metastasis of human TSCC. In Tca8113 and Cal-27 cells, TWIST2-siRNA treatment can reduce cell migration ability and has no effect on the cell proliferation and apoptosis. Hyperthermia can decrease the level of TWIST2 in TSCC and inhibit the migration of cells.ConclusionsThis demonstrated that hyperthermia might decrease the migration of Tca8113 and Cal-27 cells by reducing TWIST2 expression. Altogether, these findings suggest an as yet undescribed link between TWIST2 and hyperthermia in TSCC.
High Vimentin Expression Associated with Lymph Node Metastasis and Predicated a Poor Prognosis in Oral Squamous Cell Carcinoma

Abstract:Oral squamous cell carcinoma (OSCC) is a common public health problem worldwide with poor prognosis, which is largely due to lymph node metastasis and recurrence. Identification of specific molecular markers of OSCC with lymph node metastasis would be very important for early and specific diagnosis. In this study, we screened for the potential prognosis markers via unbiased transcriptomic microarray analysis in paired two OSCC cell lines, a lymph node metastatic HN12 cell line and a low metastatic parental HN4 cell line. The results showed that vimentin, with 87-fold increase of expression, was on the top of all upregulated genes in metastatic HN12 cells compared to non-metastatic HN4 cells. Treatment of non-metastatic HN4 cells with TGF-β1 induced epithelial to mesenchymal transition (EMT), with increased vimentin expression as well as enhanced migration activity. Consistently, knockdown of vimentin via siRNA resulted in suppressed invasion and migration activities of HN12 cells, suggesting an essential role of vimentin in EMT-related functions of OSCC cells. Finally, immunohistochemical (IHC) staining analysis showed that high vimentin expression was strongly associated with high lymph node metastases (p < 0.05), and poor overall survival (p < 0.05) in OSCC patients. Thus, high vimentin expression is strongly associated with increased metastatic potential, and may serve as a prediction marker for poor prognosis in OSCC patients.
Prognostic Value of Twist in Lung Cancer: Systematic Review and Meta-Analysis.

Abstract:BACKGROUND Twist is identified as an epithelial-mesenchymal transition (EMT) regulator which has been considered to induce metastasis. However, the prognostic value of Twist in patients with lung cancer remains controversial. METHODS A search of database including EMBASE, Medline, ISI Web of knowledge and PubMed was performed. Eligible articles studying on the prognostic significance of Twist were included in this meta-analysis. Pooled hazard ratios (HRs) and their corresponding 95% confidence intervals (CIs) were assessed to quantify the prognostic role. RESULTS A total of five studies were included. The pooled HR for Twist was 1.949 (95% CI: 1.408-2.698, I(2)=42.7%, P<0.001), suggesting high Twist expression is associated with poor prognosis in lung cancer patients. Our sensitivity analyses suggested the robustness of the results. Neither Begg's test nor Egger's test found publication bias in any analysis. CONCLUSIONS Overexpressed Twist in lung cancer tissue indicated poor prognosis.
Expression of Twist in Cervical Cancer and Its Relationship with Clinical Biological Behaviors

Abstract:Objective To investigate the expression of transcription factor Twist in human cervical cancer(squamous cell carcinoma),cervical intraepithelial neoplasia(CIN) and normal cervical tissues,and explore its clinicopathological significance. Methods Samples of cervical cancer(cervical cancer group,n=61),CINⅠ(CINⅠgroup,n=22),CINⅡ-Ⅲ(CINⅡ-Ⅲ group,n=44) and normal cervical tissues(normal control group,n=22) were collected,the expression of Twist protein in cervical tissues was detected by immunohistochemistry,and its relationship with clinical biological behaviors of cervical cancer was analysed. Results The expression rates of Twist protein in normal control group,CIN Ⅰ group,CINⅡ-Ⅲ group and cervical cancer group were 0,40.90%,68.18% and 70.49%,respectively.The expression rates of Twist protein in cervical cancer group,CINⅠ group and CINⅡ-Ⅲ group were significantly higher than that in normal control group(P0.01).However,there was no significant difference in the expression rates of Twist protein among cervical cancer group,CIN Ⅰ group and CINⅡ-Ⅲ group(P0.05).There was no significant difference in the expression rates of Twist protein among patients with different ages,FIGO clinical stages and lymphovascular space invasion status in cervical cancer group(P0.05).The expression rate of Twist protein in poorly-differentiated and undifferentiated tumors(94.44%) was significantly higher than that in well-differentiated and medium-differentiated tumors(60.47%)(P0.05),and the expression rate of Twist protein in patients with lymph node metastasis(93.75%) was significantly higher than that in those without lymph node metastasis(62.22%)(P0.05). Conclusion There is high expression of Twist protein in cervical cancer,which may be helpful in the evaluation of differentiation and metastasis of cancer.

Abstract:Tumor cell plasticity, including epithelial to mesenchymal transition (EMT) and its reverse program, mesenchymal to epithe-lial transition (MET), regulates circulating tumor cells and carcinoma metastasis. Twist is overexpressed in rhabdomyosarcoma, breast cancer, gastric cancer, and other tumors. Twist, as a transcriptional factor, cross-talks with multiple signaling pathways, forming a com-plex network to participate in the regulation of EMT/MET in circulating tumor cells, which in turn promotes metastasis of tumor cells. Therefore, monitoring the level of Twist and epithelial–mesenchymal phenotypic molecules is important as it may be beneficial for in-creasing the detection ratio of circulating tumor cells as tumor biomarkers and for evaluating the effects of anticancer drugs.
Prognostic Significance of P21, P27 and Survivin Protein Expression in Patients with Oral Squamous Cell Carcinoma

Abstract:Oral squamous cell carcinoma (OSCC) accounts for >80% of head and neck malignancies. p21, p27 and survivin proteins are abnormally expressed in OSCC and have been previously reported to correlate with cell proliferation and apoptosis. However, the prognostic significance of p21, p27 and survivin remains controversial. The aim of the present study was to investigate the association of clinical parameters and prognosis with the levels of p21, p27 and survivin expression in patients with OSCC. The levels of the three biomarkers were evaluated by immunohistochemical staining in specimens from 110 patients with OSCC and each section was scored according to the percentage of positive tumor cells and staining intensity. Log-rank test and Cox proportional hazards regression were performed to assess the correlation between biomarkers and clinical events. The association between the immunoexpression of p21, p27 and survivin and clinical pathological variables were analyzed by the χ2 test and a non-parametric analysis. The expression of p21 in patients with OSCC was found to correlate with the expression of p27 and survivin. The results of the current study revealed that the five-year survival rate was significantly lower in patients with high p21 expression. In addition, the expression of p27 also showed a negative correlation with the five-year survival rate of OSCC, but to a lesser extent. By contrast, the expression of survivin was not a prognostic factor for OSCC. A Kaplan-Meier analysis and Cox proportional hazards model showed that lymph node metastasis and p21 expression were independent prognostic factors of OSCC.
Expressions of SOX2, TWIST, and SNAIL in Breast Cancer and Their Clinical Implications

Abstract:Objective As important transcription factors, SOX2, TWIST and SNAIL are closely related to breast cancer me?tastasis. The aim of this study was to explore the expressions of SOX2, TWIST and SNAIL in breast cancer and the relationship between the three factors. Methods We collected 74 specimens of breast cancer and another 74 from paired normal tissue from January 2012 to January 2013. Using immunohistochemistry, we determined and compared the expression of SOX2 in the breast cancer and normal tissues. We also detected the expressions of TWIST and SNAIL proteins and analyzed the relationship of SOX2, TWIST and SNAIL ex?pressions with the clinicopathological characteristics of breast cancer as well as the correlation between the three proteins. Results The expression of SOX2 was significantly higher in the breast cancer than in the normal tissues (25.7% vs 8.1%, P=0.004) and corre?lated with the grade, lymph node metastasis, and TNM stage of breast cancer ( P<0.05) , while the expressions of TWIST and SNAIL were significantly correlated with lymph node metastasis and clinical TNM stages of the malignancy ( P<0.05) . The protein expression of TWIST was correlated positively with those of SOX2 ( Cc=0. 325, P<0.005) and SNAIL (Cc=0.308, P<0.008), and so was that of SNAIL with that of SOX2 (Cc=0.275, P<0.018). Conclusion The SOX2 protein is highly expressed in breast cancer. The expres?sions of SOX2, TWIST and SNAIL are closely related to lymph node metastasis of the malignancy and the three factors are interacted coor?dinatively in the metastasis of breast cancer.
Relationship between TWIST expression and epithelial-mesenchymal transition of oesophageal squamous cell carcinoma.

Abstract:We have investigated mRNA and protein expression of TWIST, Vimentin and E-cadherin in ESCC (oesophageal squamous cell carcinoma) and explored their relationship with tumour's infiltration and metastasis. RT-PCR (reverse transcriptase-PCR) was used to evaluate mRNA expression of TWIST, E-cadherin and Vimentin in 40 cases of ESCC. The protein expression of the genes was examined by immunohistochemical staining in each specimen. Expression of TWIST, E-cadherin and Vimentin mRNA and protein with clinicopathologic parameters were analysed. mRNAs of TWIST, Vimentin and E-cadherin were expressed in 75, 55 and 35% respectively of ESCC, i.e. significantly different from that in normal oesophageal mucosa (15, 0 and 85% respectively; P<0.01). In ESCC with LN (lymph node) metastasis, expression of TWIST and Vimentin mRNA, but not E-cadherin mRNA was significantly higher (100 and 83%) than in ESCC without LN metastasis (64 and 43%, P=0.018) respectively. Levels of mRNA expression of the 3 genes followed similar patterns to their above-mentioned frequencies. Protein expression of TWIST, E-cadherin and Vimentin were observed in 70, 35 and 50% respectively of ESCC, which were significantly different from normal mucosa (15, 80 and 0%; P<0.001). In ESCC with LN metastasis, protein expression of TWIST and Vimentin, but not E-cadherin, were significantly higher (100 and 75%) than in ESCC without LN metastasis (61 and 39%). Protein expression of TWIST was positively correlated with Vimentin (r=0.327, P=0.039), but negatively correlated with E-cadherin (r=-0.633, P=0.000). Thus, both mRNAs and proteins of TWIST and Vimentin were significantly overexpressed in ESCC, especially ESCC with LN metastasis. The mRNA and protein of E-cadherin were down-regulated in ESCC. These results suggest potential roles of TWIST as the promoter of tumour invasion and metastasis associated with down-regulation of E-cadherin.
Risk factors and impact of occult and skip metastasis in early-stage oral tongue squamous cell carcinoma

Abstract:Objectives: To investigate risk factors associated with occult lymph node metastases (ONM) and skip metastasis in early-stage oral tongue squamous cell carcinoma (OTSCC) patients. Meanwhile, to analyze the contribution of metastatic nodes to survival outcomes. Materials and methods: 544 OTSCC patients who were clinically staged T1-T2N0 with pathologic results from May 2018 to January 2024 were enrolled. Those with ONM were divided into subgroups with or without skip metastasis. Clinical, laboratorial, radiological and pathological factors between groups were analyzed by using univariate analysis and multivariate logistic analysis. The association of tumor growth behavior with the metastatic pattern of lymph nodes was summarized. Additionally, disease free survival (DFS) among different groups were compared using Kaplan-Meier analysis. Results: Tumor growth behavior was associated with ONM. Tumor thickness with a threshold of 6.4 mm was not inferior to histological depth of invasion in predicting ONM. Only 1.3% of patients had nodal involvement of neck level IV or V. The DFS of patients with ONM were significantly reduced than those without ONM (P < 0.001). The DFS between patients with and without skip metastasis exhibited no statistical significance(P = 0.246). The 1-year, 2-year recurrence rates of patients with or without ONM were 31.9%, 37.5%, 10.1% and 14.0%, correspondingly. Conclusions: Tumor thickness with a threshold of 6.4 mm could be used as a preoperative predictor for ONM. Elective neck dissection of level I - III might be sufficient for early stage OTSCC patients. OTSCC patients with ONM should be closely observed during the first 2 years after surgery. Clinical relevance: The risk of ONM in early stage OTSCC patients might be predicted by tumor thickness calculated on MR imaging. Elective neck dissection of level I - III could remove micrometastases timely and effectively.

Abstract:Objective: Herein, we aimed to identify biomarkers that affect lymphatic metastasis of oral squamous cell carcinoma (OSCC) through bioinformatic analysis, and clinicopathological and in vitro verifications. Design: The OSCC-related gene expression dataset was retrieved from The Cancer Genome Atlas (TCGA) and analyzed to identify differentially expressed genes (DEGs), which were subjected to pathway analysis. Weighted gene co-expression network analysis (WGCNA) and protein-protein interaction (PPI) network analysis were performed to identify hub genes. Expression of potential biomarkers was examined using quantitative real-time polymerase chain reaction, immunohistochemistry, and western blotting. Statistical analyses were performed to determine the association between biomarker expression and clinicopathological characteristics of patients with OSCC. Effects of selected biomarkers on proliferation, migration, and invasion were evaluated using in vitro assays. Results: For DEGs, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis revealed potential lymphatic metastasis-related biological processes and signaling pathways. Eight hub genes - ALOXE3, CSTA, PLA2G4E, PPL, SPRR1A, SPRR2A, SPRR2D, and SPRR2E, were identified via WGCNA and PPI analyses. CSTA expression was markedly downregulated in primary OSCC tissues, and low CSTA expression significantly correlated with high tumor grade (P = 0.001), nodal metastasis (P = 0.028), and poor overall survival (P < 0.001). CTSA overexpression inhibited OSCC cell migration and invasion in vitro, with little effect on OSCC cell proliferation. Conclusions: Our study revealed that CSTA is a promising biomarker and therapeutic target with prognostic implications in patients with OSCC. CSTA may play an essential role in OSCC lymphatic metastasis and tumor differentiation.

Abstract:Recently, TWIST, a basic helix-loop-helix transcription factor, has been reported to play a key role in the metastatic progression of several types of human cancer. The aim of this study was to investigate the significance of TWIST expression in bladder cancer using tissue microassays generated from 226 bladder tissue specimens. Using immunohistochemical staining, we studied TWIST expression levels in nonmalignant bladder tissues (n = 37), primary bladder cancer tissues (n = 164), and 25 cases of matched lymph node metastatic lesions. The association between TWIST expression levels and tumor staging and grading, as well as metastatic potential, was analyzed by statistical analysis. Our results showed that TWIST protein expression was significantly higher in bladder cancer specimens compared with nonmalignant tissues (P < .001), indicating its positive role in the development of bladder cancer. In addition, increased TWIST expression levels were associated with advanced-stage and high-grade tumors, suggesting its involvement in the progression of this cancer. Furthermore, TWIST expression was much higher in the metastatic lesion compared with its primary site (P < .05). More importantly, the increased TWIST expression in bladder cancer specimens was correlated with decreased membranous expression of E-cadherin, a cell adhesion molecule that plays a key role in the metastatic progression of human cancer. Our results demonstrate TWIST as a novel positive factor in the development and progression of bladder cancer and suggest a marker for advanced bladder cancer.
The significant clinical correlation of the intratumor oral microbiome in oral squamous cell carcinoma based on tissue-derived sequencing

Abstract:Background: The microbiota is a critical component of the complex human microenvironment, impacting various physiological processes and disease development via the microbe–host interaction. In particular, the oral microbiota profoundly affects tumor development and progression. There is increasing evidence that oral microbiota is associated with the development of oral cancer, especially oral squamous cell carcinoma (OSCC). Methods: We comprehensively analyzed the oral microbiota in 133 OSCC samples worldwide. Subsequently, we evaluated the microbial compositions between OSCC patients and healthy people and their correlation with clinical parameters. The value of the oral microbiota as a diagnostic and prognostic biomarker was also determined. Results: This study found differences in critical oral microbiota between OSCC and normal controls. The most notable differences are present in p_Firmicutes , p_Actinobacteria , c_Fusobacteriia , o_Fusobacteriales , f_Fusobacteriaceae , and g_Fusobacterium . All six-level oral microorganisms were also associated with the clinical characteristics of OSCC, particularly with the clinical outcomes (survival time and status). We developed a predictive model based on this. We found that five different oral microorganisms have high confidence and can be used for clinical diagnosis and prognostic prediction, except for p_Actinobacteria . Conclusion: This study revealed that the intratumor oral microbiota of OSCC patients worldwide and the microbial signatures of OSCC patients possess similar properties in different regions, further refining the shortcomings of the current research field. We revealed that the oral microbiota could be used as a biomarker to reflect human health and disease progression status. This will provide new directions for tumor microbiome research. This means we can develop strategies through diet, probiotics, and antibiotics for cancer prevention and treatment.

Abstract:Introduction: CX3CL1 exhibits chemoattraction for T-cells, monocytes, and CD57+ natural killer cells mediating antitumor immunity. The role of CX3CL1 has been studied in tumors of the breast, lung, colon, pancreas, prostate, etc. The current study was undertaken to understand the importance of CX3CL1 and its correlation with CD57+ cells in oral squamous cell carcinoma (OSCC). Material and methods: Seventy-five primary OSCC were staged and histopathologically graded, followed by immunohistochemistry for CX3CL1 and CD57. Mann-Whitney U-test, Kruskal-Wallis test, Post hoc Bonferroni test, and Pearson's correlation coefficient were applied. Results: CX3CL1 assessment within the tumor cells was high in 62.66% of cases, and the CD57 Labeling Index (LI) varied over a wide range of 8.2-111.6. A statistically significant reduction in expression of both CX3CL1 and CD57 was observed with an increase in histologic grade (p = 0.021 and 0.038, respectively). Discussion: It is concluded that CX3CL1 and CD57 may be important players in the immune surveillance of OSCC. Further studies with detailed follow-up for the overall survival of patients will help in studying the diagnostic, prognostic, and therapeutic roles of CX3CL1 in OSCC.
Expression of Dickkopf-1 and Twist2 in Cervical Squamous Cell Carcinoma and Their Correlation with Vasculogenic Mimicry

Abstract:Wnt/β-catenin signaling, epithelial-mesenchymal transition (EMT), and vasculogenic mimicry (VM) all exert important effects in tumors. Dickkopf-1 (DKK1) is an antagonist of the Wnt/β-catenin, Twist homolog 2 (Twist2) is a key EMT transcription factor involved in cancer cell migration and invasion, and VM participates in the progression and metastasis of a variety of cancers. However, the correlation of DKK1, Twist2, and VM in cervical squamous cell carcinoma(CSC) is still unclear. This study focuses on correlations among these factors as well as their correlation with clinicopathologic data and survival in CSC. DKK1, Twist2, and VM expressions were immunohistochemically examined in 116 CSC tissues and 37 normal cervical tissues. Furthermore, clinical data were processed. The expression levels of these three factors differed between CSC and normal tissues. VM was observed in CSC, but not in normal cervical tissues. Twist2 expression was high in CSC but low in normal cervical tissues, whereas DKK1 expression had the opposite pattern. Tumor cells with VM had a high expression of Twist2 and low expression of DKK1. In addition, DKK1 expression was negatively correlated with Twist2 expression. Analyzing the relationships of DKK1, Twist2, and VM with the data of patients with CSC revealed that DKK1 expression was negatively correlated with the clinical stage, degree of differentiation, depth of infiltration, and lymph node metastasis of tumors. VM and Twist2 expression were positively correlated with the degree of differentiation, the depth of infiltration, and lymph node metastasis. The positive rate of VM was greater in stage II than in stage I. The patients who expressed VM and Twist2 had a reduced overall survival (OS) when compared with patients not expressing these proteins. However, the patients who expressed DKK1 had an increased OS when compared with patients who did not show any DKK1 expression. Multivariate analysis indicated that the expressions of DKK1, Twist2, and VM were prognostic factors for CSC. VM and the expression of DKK1 and Twist2 can be the potential prognostic biomarkers and therapeutic targets for CSC.
