Abstract:Objective: To explore the features of microsatellite alterations of circulating DNA in the plasma of patients with hepatocellular carcinoma (HCC) and whether they are in concordance with those in the carcinoma tissues. Methods: Peripheral blood samples were collected from 62 HCC patients and the corresponding tumor tissues were obtained during operation. Three high-polymorphic microsatellite markers located at chromosome 8p, D8S277, D8S298, and D8S1771, were selected to be used to detect the loss heterozygosity (LOH) and microsatellite instability (MSI) by PCR and sequencing. Results: Joint detection showed that 39 out of the 62 tissue samples showed LOH at all the 3 loci, and the same alterations at the same loci were seen in 33 matched plasma samples with a concordance rate of 84.6%. Nineteen tissue samples showed MSI at all 3 loci, and the same alterations were shown in 14 matched plasma samples with a concordance rate of 73.3%. The rate of LOH for at least one locus in the plasma DNA was 58.1% (36/62), significantly higher than the rate of MSI at at least one locus in the plasma samples (29.0%, 18/62, P<0.01). The MSI positive rate in the loci D8S277 of the plasma DNA was 22.6%, significantly higher than those of the other 2 loci (4.8% and 4.8% respectively, both P <0.05). The MSI positive rate at the loci D8S277 of the cancer tissue was 46.8%, significantly higher than those of the other 2 loci (38.7% for D8S298 and 37.1% for D8S1771, both P<0.05). Conclusion: Microsatellite alterations show a high concordant pattern between the tissue and plasma DNA in HCC, which indicates that the microsatellite alterations of tumor tissue are reflected by plasma DNA, LOH may play an important role in hepatocarcinogenesis whereas MSI may also contribute to this progress in a less significant way, and D8S277 is a sensitive locus to MSI in HCC.
Loss of Heterozygosity at D8S298 is a Predictor for Long-term Survival of Patients with Tumor-Node-Metastasis Stage I of Hepatocellular Carcinoma

Abstract:PURPOSE:Our previous studies have shown that chromosome 8p deletion correlates with metastasis of hepatocellular carcinoma (HCC). This study was to determine whether 8p deletion could be used in predicting the prognosis of patients with HCC, particularly in those with early stage of HCC.EXPERIMENTAL DESIGN:A total of 131 patients with tumor-node-metastasis (TNM) stage I of HCC who underwent curative liver resection were enrolled. Loss of heterozygosity (LOH) was examined using 10 microsatellite markers at chromosome 8p, as well as 14 microsatellites at chromosome 1p, 17p, 4q, 13q, and 16q, and their association with 5-year overall survival (OS) and disease-free survival (DFS) of patients was analyzed.RESULTS:In the entire cohort of patients, the mean LOH frequency at these 24 loci was 43.2%; LOH frequencies at D8S298 and D1S199 were 31.5% and 33.7%, respectively. LOH at D8S298 was associated with a worse 5-year OS (P = 0.008) and DFS (P = 0.038) in patients with TNM stage I of HCC. Likewise, the patients with LOH at D1S199 had a worse 5-year OS (P < 0.001) and DFS (P = 0.014) compared with those without LOH at D1S199. In multivariate analyses, LOH at D8S298 was an independent predictor of decreased DFS (hazard ratio, 0.372; 95% 95% confidence interval, 0.146-0.948; P = 0.038), whereas LOH at D1S199 was an independent predictor of decreased OS (hazard ratio, 0.281; 95% confidence interval, 0.123-0.643; P = 0.003).CONCLUSIONS:LOH at D8S298 and D1S199 is independently associated with a worse survival in patients with TNM stage I of HCC after curative resection and could serve as novel prognostic predictors for this subgroup of patients.

Abstract:Background/Aims: Recurrence and metastasis in hepatocellular carcinoma remains a major challenge to further improve, survival. High frequency of loss of heterozygosity at D14S62, and,D14S51 in tumor tissue has been shown to be closely related to metastasis and recurrence in breast cancer. But, loss of heterozygosity on 14q in plasma and tumor tissue DNA of hepatocellular carcinoma patients ha's not been investigated. To establish a way to Predict metastasis and recurrence after curative hepatic resection, we analyzed loss of heterozygosity on 14q in plasma and tumor tissue DNA of hepatocellular carcinoma patients with curative resection.Methodology: We used a simple, rapid and non-radioactive method to analyze loss,of heterozygosity at D14S62 and D14S51 in paired plasma, lymphocyte and tumor tissue DNA of 85 hepatocellular carcinoma patients with curative resection.Results: From 79 cases informative for D14S62 and 78 cases informative,for,D14S51 of 85 hepatocellular carcinoma tissue DNA, logs. of heterozygosity at D14S62 and D14S51 was present in 45 (57.0%) and 41 (52.6%) cases respectively. And in 96.0% of the tissues which showed loss of heterozygosity we were able to detect loss of heterozygosity in their matched plasma. in matched 85 cases of hepatocellular carcinoma plasma DNA, we detected loss of heterozygosity at D14S62 in 55.7% and at D14S51 in 50.0% of the respective informative DNA samples. The loss of heterozygosity patterns of plasma DNA were almost identical to their corresponding tumor tissues. A comparison of these, genetic changes with clinicopathological data of these checked hepatocellular carcinoma patients showed that loss of heterozygosity at D14S62 and D14S51 was adversely correlated significantly with the presence of tumor thrombosis as well as intrahepatic metastases and tumor size, with 35.4% at both the D14S62 and D14S51 locus in the HTMR (high-tendency to metastasis and recurrence) group compared with 72.9% and 59.4% in the LTMR (low-tendency to metastasis and recurrence) group at D14S62 and D14S51, respectively (P=0.001 and P=0.027, respectively).Conclusions: Our results suggest that loss' of heterozygosity at D14S62 and D14S51 plays an important role in the metastasis and recurrence of hepatocellular carcinoma patients, following curative resection. Loss of heterozygosity at D14S62 and D14S51 in the plasma DNA of hepatocellular carcinoma patients detected by a simple and non-radioactive method has great potentials to be clinically used to predicate metastasis and recurrence after curative hepatic resection.

Abstract:To explore the features of loss of heterozygosity (LOH) on 8p and their association with clinicopathological characteristics of hepatocellular carcinoma (HCC). Methods Five high-polymorphic microsatellite markers located at 8p were selected to detect the LOH in 62 HCC tissues by laser capture microdissection. Results LOH at one or more loci was observed in 35 of 62 HCC patients (56.5%). The three most frequently altered loci were D8S298 (51.1%, 24/47), D8S1771 (48.8%,21/43) and D8S264 (43.5%,20/46), respectively. LOH at D8S298 was more frequently detected in tumors with intrahepatic metastasis than those without metastasis (P 0.05). LOH at D8S1771 was more frequent in tumors larger than 3 cm in size (P0.05).Conclusion LOH at specific loci on 8p was obvious in HCC, and one or more putative tumor suppressor genes located at these regions may confer a tumor growth advantage and contribute to the progression of HCC. LOH at some specific loci on 8p is associated with certain clinicopathological parameters of HCC.

Abstract:B54 Purpose: Our previous studies have demonstrated that chromosome 8p deletion, particularly 8p11-12 and 8p21-23, correlates with metastasis of human hepatocellular carcinoma (HCC). The purpose of this study was to determine whether allelic losses on these regions including 8p could be used in predicting the prognosis of patients with HCC. Experimental Design: A total of 208 patients with TNM stage I or II of HCC who underwent curative liver resection from January 1999 to March 2000 were enrolled. Genomic DNA was extracted from microdissected HCC tissues. LOH was examined using 10 microsatellite markers located at the regions of chromosome 8p mentioned above, and its association with 5-year overall survival (OS) and disease-free survival (DFS) of patients was analyzed. Results: The mean heterozygosity of these loci was 69.8%. The LOH frequencies assessed from informative cases at the loci tested were also high, with a range from 32.6% (D8S1721) to 48.8% (D8S261), and the three most frequently altered loci were D8S261(8p22). In univariate analyses, LOH on D8S29857 (8p22) (39.6%) was associated with a worse 5-year OS ( P =0.037) and DFS ( P P =0.008) and DFS ( P =0.038). In a multivariate model, D8S298 LOH was one of the independent predictors for decreased DFS (P=0.019) and decreased OS (P=0.014). Conclusion: LOH at D8S298 is independently associated with a worse survival in stage I and II patients with curative resection for HCC.v:textbox>v:textbox>

Abstract:Objective To explore the prognostic value of chromosomal loss of heterozygosity (LOH) in molecular staging for tumor-node-metastasis (TNM) stage Ⅰ of hepatocellular carcinoma (HCC) using survival analysis.Methods 131 patients with TNM stage Ⅰ of HCC who underwent curative liver resection from January 1999 to March 2000 were enrolled.Genomic DNA was extracted from the microdissected HCC paraffin-embedded tissue samples for LOH detection using 24 high-polymorphic microsatellite markers located at chromosomes 1p,8p,17p,4q,13q,and 16q,and its associations with the 5-year overall survival (OS) and the disease-free survival (DFS) of these patients were analyzed.Results LOH was frequent at the chromosomal loci analyzed.The LOH frequencies at D8S298 and D1S199 were 31.5％ and 33.7％,respectively.On univariate analysis,D8S298 LOH in the entire cohort was associated with a significantly worse 5-year OS (46 ± 5 vs.62 ± 2,P ＜ 0.05) and DFS (44 ± 5 vs.57 ± 3,P ＜ 0.05).Likewise,patients with D1 S199 LOH had significantly worse 5-year OS (42 ± 4 vs.61 ± 2,P ＜ 0.05) and DFS (41 ± 5 vs.57 ±3,P ＜ 0.05) than those without.On multivariate analyses,LOH at D8S298 was an independent predictor of decreased DFS (HR =0.372;95％ CI=0.146 ～0.948;P ＜0.05),whereas LOH at D1S199 was an independent predictor of decreased OS (HR =0.281;95％ CI =0.123 ～ 0.643;P ＜ 0.05).Conclusion LOH at D8S298 and D1S199 could serve as novel biomarkers of prognostic prediction for patients with TNM stage Ⅰ of HCC after curative liver resection,and it would be of great value in molecular staging for TNM stage Ⅰ of HCC.
The Association of Frequent Allelic Loss on 17p13.1 with Early Metastastic Recurrence of Hepatocellular Carcinoma after Liver Transplantation

Abstract:Background and Objectives: Identification and characterization of loss of heterozygosity (LOH) can determine putative tumor suppressor genes (TSGs) and provide a variety of molecular markers for hepatocellular carcinoma (HCC). This study aimed to investigate LOH status on chromosomes 4q, 6q, 8p, 9p, and 17p, and to explore their clinical significances in HCC post-liver transplantation.Methods: A total of 37 patients with HCC who underwent liver transplantation were enrolled. LOH was examined using 34 microsatellite markers located on 4q13-3q5, 6q27, 8p22-p23, 9p21-p22, and 17p12-p13.Results: The frequency of LOH at each microsatellite locus ranged from 23% to 75%, with a mean value of 53.1%. Frequencies of LOH on 4q, 6q, 8p, 9p, and 17p were 62% (23 of 37), 30% (11 of 37), 49% (18 of 37), 46% (16 of 35), and 68% (25 of 37), respectively. LOHs on certain chromosomal regions were significantly associated with age, AFP level, tumor size, tumor multiplicity, histological grade, and metastatic recurrence.Conclusions: LOH on 17p13.1 correlated to metastatic HCC recurrence, while LOH on 4q and 8p was found to be associated with progression of HCC. Abstract:Objective To explore the value of chromosomal loss of heterozygosity(LOH)in prognosis prediction of hepatocellular carcinoma(HCC)patients with curative resection by survival analyses.Methods A total of 225 patieats who underwent curative resection for HCC from January 1999 to May 2000 were enrolled.Genomic DNA was extracted from microdissected HCC paraffin-embedded tissue samples for LOH detection using 24 high-polymorphic microsatellite markers located at chromosomes 1 p,8p,17p,4q,13q and 16q,and its association with 5-year overall survival(OS)and disease-free survival (DFS) of patients was analyzed.Results LOH was frequent at chromosomal loci analyzed.In univariate analyses,D8S298 LOH in the entire cohort was associated with a statistically significant worse 5-year OS [(39±6)vs(54±5),P＜0.05]and DFS[(36±7)vs(60±6),P＜0.01].Likewise,cases with D1S199 LOH had a statistically significant worse 5-year OS[(28±7)vs(55±5),P＜0.01]and DFS [(29±8)vs(53±5),P＜0.05]than cases without D1S199 LOH.No statistically significant associationa with OS or DFS were observed for any othar marker we anlyzed in this study.In a multivariate model,D8S298 LOH and D1S199 LOH were independently associated with decreased DFS(P＜0.05)and decreased OS(P＜0.05)in the entire cohort,respectively.Conclusion D8S298 LOH and D1S199 LOH may serve as biomarkers of metastatic recurrence and OS prediction in HCC patients after curative resection,respectively.
The Prognostic Value of Circulating Plasma DNA Level and Its Allelic Imbalance on Chromosome 8p in Patients with Hepatocellular Carcinoma

Abstract:Purpose: We demonstrated that chromosome 8p deletion is associated with metastasis of human hepatocellular carcinoma (HCC). This study assesses the value of circulating plasma DNA level and its allelic imbalance (AI) on chromosome 8p in the prediction of HCC prognosis. Methods: Blood samples were collected from 79 patients with HCC before operation, 20 patients with liver cirrhosis, and 20 healthy volunteers. The HCC and adjacent non-tumor liver tissues were obtained from surgical specimens. Plasma DNA was extracted and quantified. Two microsatellite markers on chromosome 8p, D8S258 and D8S264, were selected and used in the AI analysis. Results: The circulating plasma DNA level was found to closely associate with tumor size (P=0.008) and TNM stage (P=0.040), negatively associate with the 3-year disease-free survival (DFS) (P=0.017) and overall survival (OS) (P=0.001). AI at D8S258 in plasma DNA was significantly correlated with tumor differentiation (P=0.050), TNM stage (P=0.010), and vascular invasion (P=0.023), negatively correlated with the 3-year DFS (P=0.005) and OS (P=0.036). However, AI at D8S264 was only closely associated with 3-year DFS (P=0.014). Combined detection of AI at D8S258 and circulating plasma DNA level was independently associated with DFS (P=0.018) and OS (P=0.002) of patients with HCC. For patients with both AI at D8S258 and a higher level of plasma DNA, the 3-year DFS and 3-year OS rates were decreased remarkably (P=0.014 and 0.044). Conclusion: Combination of circulating plasma DNA level and AI at D8S258 might be an independent predictor for prognosis of HCC patients.

Abstract:Objective To study the loss of heterozygosity(LOH)on chromosome 3p in breast cancers and precancerous lesion.Methods LOH at 11 microsatellite loci was detected in 41 cases of breast cancers and 12 cases of precancerous lesion by polymerase chain reaction and silver stain.The expressions of ER,PR,FHIT and hMLH1 were detected in breast cancer by immunohistochemistry.Results LOH on 3p was detected in 97% of breast cancers.D3S1295,D3S1029 and D3S1038 located at 3p14,3p21-p22 and 3p25 were identified as the loci with most frequent LOH(53.1%,43.6% and 52.5%).LOH of D3S1038 and expression of hMLH1 protein correlated with several clinicopathological features.LOH of D3S1295 had significant negative correlation with the expression of FHIT.In breast precancerous lesions,LOH on 3p was detected in 41.7% lesions.D3S1295 and D3S1029 were also identified as the most frequent LOH locus (27.3% and 16.7%).The smallest common LOH region seems likely lie between 3p14 and 3p25. Conclusions The smallest LOH region indicates the existence of breast tumor related genes and some of them affect gene expression.
Clinical Significance of Heterozygosity Loss at Mfn2 Gene in Hepatocellular Carcinoma

Abstract:OBJECTIVE:To investigate the loss of heterozygosity (LOH) at mitofusin-2 (Mfn2) gene in hepatocellular carcinoma (HCC) and its clinicopathological significance. METHODS:Four high polymorphic microsatellite markers flanking Mfn2 were selected for LOH analysis in 29 cases of HCC. RESULT:The frequencies of LOH on D1S2667, D1S2740, D1S434 and D1S228 were 21%, 23%, 21% and 22%, respectively. LOH at Mfn2 was closely correlated with tumor size, age, capsule, differentiation and t HBV infection (P<0.05), not with gender, thrombosis, cirrhosis and serum AFP levels (P>0.05). CONCLUSION:LOH at Mfn2 gene in HCC is associated with the clinicopathological features of patients.

Abstract:The carcinogenesis of primary liver cancer is a complicated multi-factor, multi-step, multi-gene process.Hepatocellular carcinoma (HCC) is the most important type of primary liver cancer. Allele deletions and inactivation of tumor suppressor genes (TSGs) play critical roles during the carcinogenesis, progress and recurrence of HCC. Loss of heterozygosity (LOH) associated with HCC occurs at most autosomes and the Y chromosome. Losses at chromosomes 1p, 1q, 4q, 8p, 10q, 16q, and 17p have been documented frequently. In this article, we previewed the investigations of clinical significance of LOH, and looked forward to the application of LOH in early detection, therapy, and prediction of prognosis and susceptibility in HCC patients.

Abstract:Objective The loss of heterozygosity (LOH) of chromosome 8p microsatellite was used as the genetic marker to identify the clonal disparity among different cell groups in the same hepato- cellular carcinoma (HCC) nodule.Methods In 10 patients with HCC,the parts with different colors in the same nodule were taken as different cell groups;Relative pure tumor cells were obtained by hand-as- siatant microdissection;LOH of 5 microsatellites in chromosome 8p was detected.Results LOH was de- tected in 7 patients,and 6 of them had different LOH alterations among different cell groups in the same tumor nodule.One had 3 different cancer cell groups with same LOH pattern.Conclusion In the same HCC nodule,different cell groups have dissimilar LOH,indicating that there are clonal differences among these cell groups and maybe the different cell groups have different clonal origins.

Abstract:AIM: To analyze loss of heterozygosity (LOH) and homozygous deletion on p53 gene (exon2-3, 4 and 11), chromosome 10q22-10q23 and 22q11.2 -22q12.1 in human hepatocellular carcinoma (HCC).METHODS: PCR and PCR-based microsatellite polymorphism analysis techniques were used.RESULTS: LOH was observed at D10S579 (10q22-10q23) in 4 of 20 tumors (20%), at D22S421 (22q11.2-22q12.1) in 3 of 20(15%), at TP53.A (p53gene exon 2-3) in 4 of 20 (20%), at TP53.B (p53gene exon 4) in 6 of 20(30%), and at TP53.G (p53gene exon 11)in 0 of 20(0%). Homozygous deletion was detected at 10q22-10q23(8/20; 40%), 22q11.2-22q12.1(8/20; 40%), p53 gene exon 2-3(0/20;0%), p53gene exon 4(6/20; 30%), and p53gene exon 11(2/20; 10%).CONCLUSION: There might be unidentified tumor suppressor genes on chromosome 10q22-10q23 and 22q11.2-22q12.1 that contribute to the pathogenesis and development of HCC.

Abstract:OBJECTIVE:To detect putative suppressor loci involved in tumor progressing or metastases.METHODS:Thirty microsatellite marker primers were employed to amply the corresponding loci of the genome DNA from 83 patients with sporadic colorectal cancer. The PCR products were electrophoresed on a 377 PRISM sequencer and the fluorescent signals were analyzed by Genotyper and Genescan software.RESULTS:The data were obtained from 24 loci, with an average LOH frequency of 15.16%. The LOH at D2S206 and D2S364 was more frequent than 30%, and was less than 20% at the rest loci. Significant difference was observed between the percentage of LOH and tumor staging or differentiation at D2S142 (2q24.1), D2S126 (2q35), D2S2211 (2p24.2), D2S305 (2p23.3). Occarrence of deletion at the later two loci was correlative.CONCLUSIONS:Frequent LOH was not observed at the loci around known mismatch repair genes on chr. 2. The region between D2S305 (2p23.3) and D2S2211 (2p24.2) deleted holistically, and was correlated to the stage and differentiation of tumor attended by D2S142 (2q24.1) and D2S126 (2q35) on 2q. It is suggested that unknown genes associated with tumor progressing or metastases reside in the two loci on 2q or the region on 2p.
Detailed Deletion Mapping of Loss of Heterozygosity on 22q13 in Sporadic Colorectal Cancer

Abstract:AIMBoth development and progression of malignancies occur as a multistep process, requiring the activation of oncogenes and the inactivation of several tumor suppressor genes. The loss of heterozygosity (LOH) of tumor suppressor genes is believed to play a key role in carcinogenesis of colorectal cancer (CRC). In this study, we analyzed the LOH of seven loci on chromosome 22q13 in an effort to identify candidate tumor suppressor genes involved in colorectal carcinogenesis.METHODSMatched tumor and normal tissue DNA were analyzed by PCR using fluorescence-labeled polymorphic microsatellite markers in 83 CRC patients. PCR products were eletrophoresed and LOH was determined by calculating the peak height acquired through computer software. Comparisons between LOH frequency and clinicopathological features were performed by chi2 test. P<0.05 was considered as statistical significance.RESULTSThe average LOH frequency of chromosome 22q13 was 28.38%. The highest LOH frequency was 64.71% on D22S1160 locus, and the lowest was 21.43% on D22S1141 locus. We detected two obvious minimal deletion regions: one between markers D22S1171 and D22S274, the other flanked by markers D22S1160 and D22S1149, each about 2.7 and 1.8 cm, respectively. None had lost in all informative loci. LOH frequency on D22S1171 is 50% on distal colon, which was higher than that on proximal one (P = 0.020); on D22S114 locus, none LOH event occurred in patients with liver metastasis, whilst 46.94% occurred in patients without liver metastasis (P = 0.008); on D22S1160 locus, LOH frequency in lymph nodes metastasis patients was 83.33%, which was much higher than 43.75% without lymph nodes metastasis ones (P = 0.016). There was no statistical significance between clinicopathological features and other loci.CONCLUSIONThis study provides evidence of two minimal deletion regions, which may harbor putative tumor suppressor genes related to progression and metastasis in sporadic colorectal carcinoma on chromosome 22q13.

Abstract:Objective The loss of heterozygosity (LOH) on tumor suppressor genes is believed to play a key role in carcinogenesis of colorectal cancer. The aim of this study is to identify additional loci involved in colorectal tumorigenesis. Methods D22S274,a polymorphic microsatellite marker, was analyzed in 83 cases of colorectal and normal DNA by PCR. 83 sporadic colorectal cancer patients include 40 males and 43 females, which ages ranged from 31 to 84 years. PCR products were eletrophoresed on an ABI 377 DNA sequencer; Genescan 3.1 and Genotype 2.1 software were used for LOH scanning and analysis. Comparison between LOH frequency and clinicopathological data was performed by χ 2 test. Results The LOH frequency of D22S274 (22 q 13.32-13.33) was 34.04%. On this locus, LOH frequency of rectal cancer was 50% (9/18), which was higher than that of proximal colon cancer (12%, 2/17) (P=0.018). The frequency of distal colon cancer was 42% (5/12), also higher than that of proximal colon cancer. Putting both the tumors, distal colon and rectum, together into consideration, the frequency, 47% (14/30), was higher than that of proximal colon cancer (P=0.015),suggesting the mechanism of carcinogenesis was different in both groups. Conclusion This study seems to show the evidence for the putative tumor suppressor genes related to the sporadic colorectal carcinoma on 22q13.32-13.33.

Abstract:OBJECTIVE:Investigate the frequency of loss of heterozygosity (LOH) at chromosome arm the short arm chromosome 3p14, 25 in the serum DNA from ovarian cancer and its clinical application.METHODS:Thirty-eight ovarian cancer serum samples with 18 corresponding tumor tissues and 8 benign ovarian tumors were obtained, and DNA samples extracted from serum and tissue were examined for 3p14, 25 LOH by using of polymerase chain reaction with four polymorphic microsatellite markers (D3S1029, D3S1228, D3S1300, D3S1481).RESULTS:Matched serum and tissue DNAs from 18 ovarian cancer patients showed significant concordance of 3p14, 25 LOH (P < 0.05). 3p14, 25 LOH in at least one of four loci occurred in 29 out of 38 (76%) serum samples, while 17 serum samples (45%) exhibited LOH at more than one locus. According to International Federation of Gynecology and Obstetrics (FIGO) staging, there was a trend that the rate of LOH was higher in advanced stages, and the frequency of loss in stage II, III, IV was 2/4, 78%, 8/9 respectively. It was also found that the number of microsatellite markers with 3p14, 25 LOH was increased with tumor progressed. No significant association of pathological types with LOH in serum DNA could be demonstrated except at locus D3S1029.CONCLUSIONS:Since the strong correlation of 3p14, 25 LOH between serum DNA and tumor tissue DNA, as well as the frequency of 3p14, 25 LOH associated with malignancy of ovarian cancer, it is suggested that detection of serum DNA 3p14, 25 LOH may be used as a molecular marker for staging and monitoring human ovarian cancer.

Abstract:OBJECTIVE:Detecting the loss of heterozygosity in paraffin-embedded pancreatic cancer tissues.METHODS:Analysing loss of heterzygosity (LOH) of microsatellite DNA on chromosome loci 3,5,7,9 and 18 with PCR-SSLP-silver stain method in pancreatic cancer.RESULTS:In 45 sporadic pancreatic cancer samples and their paired control tissue, LOH was detected on site for D18S46( 18q21.1,31.0%), D18S474 (18q21.1, 20.0%), D9S176 (9q22-31,20.0%), D3S1234 (3p14.2,17.5%),D 3S1289 (3p21.1, 15.7%), D3S1481 (3p14.2,4.9%), D7S486 (7q22,3.6%), D5S365 (5q32, 3.2%) and D3S587 (3p24-26, 2.6%).CONCLUSION:Different percentages of loss of heterzygosity on specific chromosomal regions were found, and the meaning of the results was discussed. Some key genes may play a role in the pathogenesis of pancreatic cancer.

RESULTS: The average LOH frequency on chromosome 22q was 28.38 %. The region between markers D22S280 and D22S274 (22q12.2-q13.33) exhibited relatively high LOH frequency. The two highest LOH loci with frequencies of 35.09 % and 34.04 % was identified on D22S280 (22q12.2-12.3) and D22S274 (22q13.32-13.33). 8 cases showed LOH at all informative loci, suggesting that one chromosome 22q had been completely lost. On D22S274 locus, LOH frequency of rectal cancer was 50 % (9/18), which was higher than that of proximal colon cancer (12 %, 2/17) (P=0.018). The frequency of distal colon cancer was 42 % (5/12), also higher than that of proximal colon cancer. But there was no statistical significance. The tumor-suppressor-gene(s) might locate on the 22q12.2-12.3 and/or 22q13.32-13.33.
