Abstract:Seventeen compounds were isolated from the leaves of PANAX GINSENG C. A. Meyer. Among them, a new minor saponin was established as 3beta,6alpha,12beta-trihydroxy-dammar-20(22), 24-diene-6- O-alpha- L-rhamno-pyranosyl-(1-->2)-beta- D-glucopyranoside ( 2). Abstract:From the tap root of Panax notoginseng,ten compounds were isolated by repeated column chromatography with silica gel and C18.Their structures were identified by spectroscopic methods as ginsenoside Rh4(1),notoginsenoside R2(2),ginsenoside Rh1(3),ginsenoside F1(4),20(S)-ginsenoside Rg2(5),3-O-β-D-glucopyranosyl-(12)-β-D-glucopyranoside-12β,25-dihydroxydammar-(E)20(22)-ene(6),vinaginsenoside R15(7),vinaginsenside R3(8),pseudoginsenoside RT3(9),and 6-O-(β-D-glucopyranosyl)-20-O-(β-D-xylopyranosyl)-3β,6α,12β,20(S)-tetrahydroxydammar-24-ene(10),respectively.Compounds 6-10 were first isolated from this plant,and compounds 6 and 10 were identified from the nature for the first time.

Abstract:The minor bioactive saponins of the total saponin from the leaves of Panax notoginseng (Burk.) F. H. Chen. were separated and purified by column chromatographies on silica gel and thin layer gel. Four compounds were identified as: ginsenoside C-K (I) [20 (S) -protopanaxadiol 20-O-beta-D-glucopyranoside], ginsenoside-Rh1 (II) [20 (S) -protopanaxatriol-6-O-beta-D-glucopyranoside], ginsenoside-Mc (III) [20 (S)-protopanaxadiol 20-O-alpha-L-arabinofuranosyl (1 --> 6) -D-glucopyranoside] and notoginsenoside-Fe (IV) [20(S) -protopanaxadiol-3-O-beta-D-glucopyranosido-20-O-alpha-L-arabinofuranosyl(1 --> 6)-beta-D-glucopyranoside]. Abstract:Three new dammarane-type triterpene ginsenosides, together with six known ginsenosides, were isolated from the leaves of Panax ginseng C.A. Meyer. The new saponins were named as ginsenoside Rh11, ginsenoside Rh12, and ginsenoside Rh13. Their structures were elucidated as (20S)-3,6,12,20-tetrahydroxydammara-25-ene-24-one 20-O--d-glucopyranoside (1), (20S)-3,12,20,24,25-pentahydroxydammarane 20-O--d-glucopyranoside (2), and (20S,23E)-3,12,20,25-tetrahydroxydammara-23-ene 20-O--d-glucopyranoside (3) on the basis of 1D and 2D NMR experiments and mass spectra. The known ginsenosides were identified as ginsenoside M7cd, ginsenoside Rg6, ginsenoside Rb3, gypenoside XVII, gypenoside IX, and 20-(E)-ginsenoside F4.

Abstract:Four new dammarane-type triterpenoid saponins, namely notoginsenosides SFt 1 -SFt 4 ( 1–4 ) were isolated from the steamed leaves of Panax notoginseng (Burk.) F. H. Chen (Araliaceae), together with 17 known saponins. Their structures were established on the basis of detailed spectroscopic analyses and acidic hydrolysis. The known ginsenosides Rk2 and Rh3 were obtained from P. notoginseng for the first time. All of these new saponins showed no in vitro cytotoxicity against five human cancer cell lines (HL-60, SMMC-7712, A-549, MCF-7, and SW480).

Abstract:Leaves of Panax notoginseng,P.ginseng and P.quinquefolius have similar saponin components but with different exclusive components.In this work,the contents of saponins in the leaves of Panax notoginseng,P. ginseng and P.quinquefolius were determined with reversed-phase high-performance liquid chromatography.The chromatographic separation was achieved by using a Kinetex-C18 column(100×4.6 mm,2.6 μm,phenomenex,at 30 ℃).Eleven compounds were separated with a gradient elution and detected at 203 nm.The results showed that the order of total content of saponins,protopanaxadiol saponins and protopanaxatriol saponins in three kinds of leaves was P.quinquefolius P.ginseng P.notoginseng,P.quinquefolius P.notoginseng P.ginseng,and P. ginseng P.quinquefolius P.notoginseng,respectively.The content of protopanaxadiol saponins in the leaves of P.quinquefolius and the content of protopanaxatriol saponins in the leaves of P.ginseng obviously were higher than those in others.And the total content of Ginsenosides Rb3 and Rd was accounted for over 60% of the total 11 ginsenosides in P.quinquefolius leaves.Conclusively,the leaves of Panax notoginseng,P.ginseng and P. quinquefolius should be well used as a wonderful source for saponins due to their high contents.In addition,all the three kinds of saponins should be exploited on the basis of their contents and compositions.
Integrated metabolomic and transcriptomic analyses revealed the distribution of saponins in Panax notoginseng

Abstract:Panax notoginseng is famous for its important therapeutic effects. Saponins are bioactive compounds found in different parts and developmental stages of P. notoginseng plants. Thus, it is urgently to study saponins distribution in different parts and growth ages of P. notoginseng plants. In this study, potential biomarkers were found, and their chemical characteristic differences were revealed through metabolomic analysis. High-performance liquid chromatography data indicated the higher content of saponins (i.e., Rg1, Re, Rd, and Rb1) in the underground parts than that in the aerial parts. 20(S)-Protopanaxadiol saponins were mainly distributed in the aerial parts. Additionally, the total saponin content in the 3-year-old P. notoginseng plant (188.0 mg/g) was 1.4-fold higher than that in 2-year-old plant (130.5 mg/g). The transcriptomic analysis indicated the tissue-specific transcription expression of genes, namely, PnFPS, PnSS, PnSE1, PnSE2, and PnDS, which encoded critical synthases in saponin biosyntheses. These genes showed similar expression patterns among the parts of P. notoginseng plants. The expression levels of these genes in the flowers and leaves were 5.2fold higher than that in the roots and fibrils. These results suggested that saponins might be actively synthesized in the aerial parts and transformed to the underground parts. This study provides insights into the chemical and genetic characteristics of P. notoginseng to facilitate the synthesis of its secondary metabolites and a scientific basis for appropriate collection and rational use of this plant.

Abstract:Nine new minor dehydrogenated and cleavaged dammarane-type triterpenoid saponins, namely notoginsenosides ST6-ST14 (1-9) were isolated from the steamed roots of Panax notoginseng, together with 14 known ones. Among them, 5-7 and 21-22 were protopanaxadiol type and the left 18 compounds, including 1-4, 8-20, and 23 were protopanaxatriol type saponins. Their structures were identified by extensive analysis of MS, 1D and 2D NMR spectra, and acidic hydrolysis. Resulted from the side chain cleavage, the new saponins 1 and 2 featured in a ketone group at C-25, and 3-5 had an aldehyde unit at C-23. The known saponins 12, 16 and 18 displayed the enhancing potential of neurite outgrowth of NGF-mediated PC12 cells at a concentration of 10 μM, while 20 exhibited acetyl cholinesterase inhibitory activity, with IC50 value of 13.97 μM.

Abstract:Seven new protopanaxatriol type saponins, 20S-sanchirhinosides A1 (1), A2 (2), A3 (3), A4 (4), A5 (5), and A6 (6), and sanchirhinoside B (7) were obtained as minor constituents from the root extract of Panax notoginseng (Burkill, F. Abstract:Background: Panax quinquefolius and Panax notoginseng are widely used and well known for their pharmacological effects. As main pharmacological components, saponins have different distribution patterns in the root tissues of Panax plants. Methods: In this study, the representative ginsenosides were detected and quantified by desorption electrospray ionization mass spectrometry and high-performance liquid chromatography analysis to demonstrate saponin distribution in the root tissues of P. quinquefolius and P. notoginseng, and saponin metabolite profiles were analyzed by metabolomes to obtain the biomarkers of different root tissues. Finally, the transcriptome analysis was performed to demonstrate the molecular mechanisms of saponin distribution by gene profiles. Results: There was saponin distribution in the root tissues differed between P. quinquefolius and P. notoginseng. Eight-eight and 24 potential biomarkers were detected by metabolome analysis, and a total of 340 and 122 transcripts involved in saponin synthesis that were positively correlated with the saponin contents (R > 0.6, P < 0.05) in the root tissues of P. quinquefolius and P. notoginseng, respectively. Among them, GDPS1, CYP51, CYP64, and UGT11 were significantly correlated with the contents of Rg1, Re, Rc, Rb2, and Rd in P. quinquefolius. UGT255 was markedly related to the content of R1; CYP74, CYP89, CYP100, CYP103, CYP109, and UGT190 were markedly correlated with the Rd content in P. notoginseng. Conclusions: These results provided the visual and quantitative profiles of and confirmed the pivotal transcripts of CYPs and UGTs regulating the saponin distribution in the root tissues of P. quinquefolius and P. notoginseng.
