Abstract:The aqueous extract of lignum sappan can kill cell lines of HL-60, K562, L929 and Yac-1 at the concentration of 2 microliters/ml. The survival time of mice bearing EAC is increased by 185% (P less than 0.01) by ip 0.2 ml/mouse x 7d. The three-stage sequential test standard can be passed smoothly.

Abstract:Lysimachia capillipes Hemsl (Primulaceae), a folk medicinal plant in China, showed significant anti-tumor activities in vivo and in vitro. Capilliposide B (LC-B) and capilliposide C (LC-C) are the main bioactive components in this plant. To explore their tissue distribution, a reliable bioanalytical method for the quantification of LC-B, LC-C and their bioactive metabolite, capilliposide A (LC-A), in mouse tissues was developed and validated. In this study, the tissue distribution profiles of the three compounds were examined after intravenous administration of pure LC-B and oral administration of total saponins of L. capillipes Hemsl extract (LCE) for 10days. Method validation was conducted over the curve range 10.0-5000ng/mL for all three analytes in various tissue homogenates. The relative standard deviation of intra-day and inter-day precision of the QC samples was <14.7%, and the accuracy ranged from 85.9 to 114.0%. The results indicated that LC-B was rapidly and widely distributed throughout the whole body except for muscle following intravenous administration of LC-B. In addition, LC-A was only in liver, intestine, lung and stomach. After oral administration of LCE, LC-B and LC-C were distributed into various tissues. The highest levels were observed in stomach and intestine.

Abstract:The antitumor properties in vitro and in vivo of ethyl acetate acidic extract from Solidago canadensis L.flowers were evaluated.MTT was used to identify cytotoxic activity in vitro.The in vivo antitumor activity was evaluated by the antitumor bioassay against transplanted mouse solid tumor S180 and EAC.Cytotoxicity of acidic ingredient from ethyl acetate extract was distinct(IC5050 μg·mL-1) in vitro was assayed by four cell lines.Also,it has the strong antitumor activity against transplanted mouse solid tumor S180 and EAC in vivo,showing a positive correlation with the concentration.The inhibitory rates at the dose of 0.01 g·kg-1 ip against S180 and EAC were 52.06% and 32.90%,respectively.Solidago canadensis L.(tribe Astereae,family Compositae) is a promising herb which has antitumor activity in vitro and in vivo.

Abstract:Lignin, the most abundant renewable aromatic polymer, has been shown to suppress the growth of mammalian tumor cells. Despite being a textbook example, there is little information on the biological activity of lignin in relation to its molecular structure or the molecular mechanisms by which lignin suppresses tumor cells in mammalian species. Here, we prepared Microwave-assisted Acid-catalyzed Solvolysis Lignin (MASL) and assessed its effects on human and mouse tumor cells. Our data showed MASL significantly reduced viability of tumor cells by modulating apoptotic pathways. MASL treatment upregulated TNF-α, Fas, and FasL expression levels, while suppressing anti-apoptotic NF-κB and mTOR pathways in tumor cells. In-vivo experiments were also performed using tumor-bearing mice to confirm the anti-tumor effects of MASL. An administration of MASL significantly suppressed tumor growth in mice in association with elevation of caspase 3 expression. These findings strongly suggest the potential usefulness of low molecular weight lignin as an effective therapeutic against malignancies.

Abstract:Bamboo shavings are a kind of Chinese traditional medicine, which have been certificated as a material of functional food by the Ministry of Health in China. The anti-tumor activities of a triterpenoid-rich extract of bamboo shavings (EBS) and its main component, friedelin were evaluated in the present study. It was proved that EBS could inhibit the growth of P388 and A549 cancer cell lines effectively by SRB and MTT assay. Meanwhile, EBS had notable inhibitory effect on the sarcoma-loaded mice S180 model, which led to a depressed trend of tumor weight, compared to cyclophosphamide. Furthermore, the anti-tumor activity of friedelin monomer, a main triterpenoid separated from EBS, was tested by MTT assay and results showed that friedelin displayed rather strong anti-tumor activities on the proliferation of four cancer lines, A375, L929, Hela and THP-1, with a time-dose relationship compared to de-methyl-cantharidin, respectively. Therefore, it is suggested that EBS has a great potential to be applied in functional food for its anti-tumor activity, in which friedelin was one of the most important active factors.

Abstract:In the present study, we investigated the in vitro and in vivo antitumor effects of a sulfated polysaccharide fraction from the brown alga Laminaria japonica (LJSP) on cervical carcinoma. In vitro, the results showed that LJSP exhibited the highest cell growth inhibitory effect on cervical carcinoma U14 cells among five tumor cell lines. In vivo, the results showed that LJSP could not only inhibit the growth of the tumor but also enhance the spleen and thymus indexes, as well as the body weight of U14 tumor-bearing mice. Moreover, the white blood cell count of H22 tumor-bearing mice showed no change in the LJSP-treated groups and little toxicological effects were observed on hepatic function and renal function in LJSP-treated mice bearing U14 tumor cells. Besides, LJSP induced apoptosis of transplanted tumor tissues by increasing the ratio of Bax/Bcl-2. These data showed that LJSP exhibited prominent antitumor activities and low toxic effects; thus, it could be developed to a safe and effective anticancer agent.

Abstract:Actinidia macrosperma (AM) is a medicinal plant in China and has been well known for its activities against cancers, especially of lung, liver and digestive system. The immunomodulatory effects of AM aqueous extract were examined using S180-bearing mice. Young adult (20 +/- 2 g) ICR mice inoculated with S180 cells were divided randomly into six groups: S180-bearing control group, normal control group, positive control group (Ginseng Radix Rubra 2000 mg/kg) and 50, 100, 250 mg/kg AM treatment groups. Each group consisted of ten mice. Body and tumor weights were obtained after 12 consecutive days, and their humoral, cellular and nonspecific immune functions were also determined by relative assays. The results showed that the aqueous extract of AM was lack of significant inhibit on transplantable sarcoma S180, with a inhibit rate of 1.5%-14.8% (dose at 250 mg/kg was the best), but significantly increased the overall immune functions (especially at 100 and 250 mg/kg). The immunomodulatory effect was dose-dependent in a nonlinear fashion with the optimal dose of 100 mg/kg. The AM-induced antitumor effects were at least partially indirect and were associated with the modulation of immune functions.

Abstract:Objective To evaluate the antitumor effect of Radix Acanthopanacls Senticosi extract with different extracting methods,which will provide pharmacological evidence of its preparation process.MethodsS_(180)cell and tumor-bearing mice were treated in three types of extract of Radix Acanthopanacls Senticosi(extract Ⅰ,Ⅱ and Ⅲ) to evaluate the effect of anti-tumor and immune adjustment by inhibiting rate and NK cell activity.ResultsThe inhibiting rate of extract Ⅱ in different dosage groups range from 19.74% to 35.67%,and the date of extract Ⅲ is from 32.27% to 48.17%.There is a significant difference(P(0.05))between the various dosage of extract Ⅲ and the black group.The inhibiting rate was in direct proportion to the extract Ⅱ and Ⅲ dosage.ConclusionThe extract Ⅲof Radix Acanthopanacis Senticosi is clear to reflect the effect of anti-tumor.

Abstract:Two saponins, methyl protodioscin and dioscin, were extracted from the root of Polygonatum Zanlanscianense Pamp. One of them, dioscin exerted significant inhibitory effects on the growth of the human leukemia cell HL-60, inducing differentiation and apoptosis. HL60 cells were induced mainly along the granulocytic lineage. In addition, we have found that dioscin affects many cancer cells. These studies may have important significance in treating related cancers.

Abstract:Lignin, as an abundant aromatic biopolymer in plants, has great potential for medical applications due to its active sites, antioxidant activity, low biotoxicity, and good biocompatibility. In this work, a simple and ecofriendly approach for lignin fractionation and modification was developed to improve the antitumor activity of lignin. The lignin fraction KL-3 obtained by the lignin gradient acid precipitation at pH = 9-13 showed good cytotoxicity. Furthermore, the cell-feeding lignin after additional structural modifications such as demethylation (DKL-3), sulfonation (SL-3), and demethylsulfonation (DSKL-3) could exhibit higher glutathione responsiveness in the tumor microenvironment, resulting in reactive oxygen species accumulation and mitochondrial damage and eventually leading to apoptosis in HepG2 cells with minimal damage to normal cells. The IC50 values for KL-3, SL-3, and DSKL-3 were 0.71, 0.57, and 0.41 mg/mL, respectively, which were superior to those of other biomass extractives or unmodified lignin. Importantly, in vivo experiments conducted in nude mouse models demonstrated good biosafety and effective tumor destruction. This work provides a promising example of constructing carrier-free functionalized lignin antitumor materials with different structures for inhibiting the growth of human hepatocellular carcinoma (HepG2) cells, which is expected to improve cancer therapy outcomes.
Antitumour Activity of Lycium Chinensis Polysaccharides in Liver Cancer Rats.

Abstract:In the present study, polysaccharides were extracted from the Lycium chinensis (LCP). Rats were divided into four groups. Two groups (Groups A) were maintained on the basal diet, whereas the remaining three groups (Groups B, C and D) had free access to the basal diet and were orally fed with LCP at 200mg/kg b.w. for Group B, 400mg/kg b.w. for Group C and 600mg/kg b.w. for Group D, respectively. Following 4 weeks of this dietary regimen, hepatocarcinogenesis was initiated in all animals by a single intraperitoneal DENA (Sigma–Aldrich, St. Louis, MO, USA) injection at a dose of 200mg/kg body weight (mixed with peanut oil). Results still showed that L. chinensis polysaccharides (LCP) increased spleen, thymus indexs, antioxidant enzymes activities and decreased oxidative injury. In addition, LCP still significantly affect VEGF and Cyclin D1 proteins expression in liver cancer rats. It can be concluded that LCP exhibited remarkable protective effects against diethylnitrosamine (DEN)-induced oxidative hepatic injury in liver cancer rats.
Possible Reduction of Hepatoma Formation by Smmu 7721 Cells in Scid Mice and Metastasis Formation by B16f10 Melanoma Cells in C57bl/6 Mice by Agaricus Blazei Murill Extract

Abstract:Agaricus blazei Murill extract (ABM) has been reported to possess antitumor effects. In this study, the role of ABM in tumor growth and metastasis in vivo was evaluated in experimental Smmu 7721 hepatoma cells in severe combined immunodeficiency (SCID) mice and B16F10 melanoma cells lung metastasis in C57BL/6 mice. For the tumor growth model, the size of the liver tumor mass was about 10 mm to 20 mm in the control group. In comparison with the control group, the tumor mass seem to grow slowly with ABM treatment, especially at the high dose. For the tumor metastasis model, after a six-week treatment, the survival rates of B6 mice were 0%, 30%, 10% and 50% for control group, low, median and high concentration ABM treatment groups, respectively. The survival rate showed that pretreatment of C57BL/6 (B6) mice with ABM lengthened their lifespan after tumor cell inoculation, which supports the notion that ABM successfully reduced lung metastasis formation by B16F10 melanoma cells. The treatment effect was dependent on the concentration of ABM for tumor growth and metastasis in these models.
Antitumor activity of a polysaccharide from longan seed on lung cancer cell line A549 in vitro and in vivo

Abstract:In this study, a water-soluble longan seed polysaccharide (WLSP), with a molecular weight of 57 kDa, was isolated from longan seed. Gas chromatography (GC) analysis showed that WLSP was composed mainly of rhamnose (Rha), mannose (Man), arabinose (Ara), galactose (Gal), and glucose (Glc), with molar ratios of 2.4:1.5:2.3:5.6:6.5. The result in 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay showed that WLSP showed a dose-dependent antiproliferative effect on the proliferation of A549 human lung cancer cells, which is consistent with the amount of lactate dehydrogenase (LDH) release from A549 cells. Prompted by this antiproliferative effect, we further examined its antiproliferative mechanism and in vivo anticancer effect. Our results showed that WLSP had the ability to cause cell cycle arrest in G1 phase, activation of caspases 3 and 9, and cleavage of poly[ADP (ribose)] polymerase (PARP) in A549 cells. The result of this in vivo study showed that WLSP could suppress the growth of xenograft A549 tumors and induce apoptosis. Taken together, these results indicate that WLSP exert an anticancer effect in vitro and in vivo and may be useful for the prevention of lung tumorigenesis.

Abstract:This paper aims to study the anti-tumor activity and mechanism of Dendrobium denneanum in vitro and in vivo. In vitro, the inhibitory effects of D. denneanum on proliferation of human hepatoma SMMC-7721 cell and human natural liver HL-7702 cell were measured by MTT colorimetric assay. The transplantable models of S180 in mice were established to investigate the inhibitory effects of the three extracts (polysaccharide, ethanol extraction and water extraction ) against tumor. The drug treatment lasted for 10 days, and the MDA and SOD contents of mouse blood serum were measured on d 11. In vitro, 160 μg/mL polysaccharides were shown to have a prominent inhibition on SMMC-7721 for 48 hours, but an unapparent inhibition on HL-7702. In vivo, the SOD content of mouse blood serum in polysaccharide group was significantly higher than that of physiological salt solution group (P < 0.01). The MDA content of mouse blood serum in polysaccharide group was significandy lower than that of physiological salt solution group (P < 0.01). This study indicates that D. denneanum polysaccharide is a potential anti-tumor drug.
Inhibitory Effect of Ethyl Acetate Part of Ferula sinkiangensis on S180 Solid Tumor in Mice

Abstract:OBJECTIVE:To study the inhibitory effect of ethyl acetate part from Ferula sinkiangensis on S180 solid tumor in mice.METHODS:60 mice were randomly divided into normal control group,tumor model group,ethyl acetate part of F. sinkian-gensis low-dose,middle-dose and high-dose groups(1.0,2.0,3.0 g/kg,ig),cisplatin control group(positive drug,5 mg/kg,iv), with 10 mice in each group. Except for normal control group,other groups were given mice ascites infected by S180 solid tumor to induce S180 solid tumor model. The next day after modeling,those groups were given relevant medicine,and normal control group and tumor model group were given constant volume of normal saline,once a day,for consecutive 10 days. Body weight,organ (spleen,thymus)index,the change of tumor volume,volume and weight of solid tumor,inhibitory rate of tumor and pathological change were observed in each group. ELISA method was used to determined the serum levels of IL-2,IFN-γ and TNF-α. RE-SULTS:Compared with normal control group,spleen index and TNF-α of tumor model group were increased significantly,while IL-2 and IFN-γ were decreased significantly (P<0.05);dysplastic cells of tumor tissue appeared lumpy,accompanied by a num-ber of ischemic necrosis of dysplastic cells. Compared with tumor model group,body weight,spleen index,thymus index and vol-ume and weight of solid tumor in cisplatin control group were decreased significantly,while IL-2 and TNF-α levels were increased significantly. The spleen index and TNF-α level of ethyl acetate part of F. sinkiangensis groups were increased significantly,while the volume and weight of solid tumor were decreased significantly(P<0.05). The tumor tissue was inhibited to different extents, especially in cisplatin control group;the number of tumor cells was decreased,accompanied by small number of cellular necrosis and infiltration. Inhibitory rates of cisplatin control group,ethyl acetate part of F. sinkiangensis groups were (73.26 ± 4.55)%, (40.34±3.45)%,(50.86±9.34)%,(58.20±3.70)%,respectively. CONCLUSIONS:ethyl acetate part of F. sinkiangensis can in-hibit S180 solid tumor in mice.

Abstract:Treatment options are limited in patients with diffuse large B-cell lymphoma (DLBCL). Salvianolic acid A (SAA) is a water-soluble phenolic acid extracted from Salvia miltiorrhiza (Danshen) with anti-tumor properties. The anti-leukemic activity of SAA that has been shown in our recent finding prompts us to investigate the therapeutic effect and mechanism of action of SAA in DLBCL. Here, we find that SAA inhibits the viability of DLBCL cells by inducing cellular apoptosis, which is accompanied by upregulation of Bax and cleavage of PARP. Pre-incubation of SAA increases the phosphorylation of JNK, while decreases the phosphorylation of p38 and ERK in DLBCL cells. Importantly, pharmacological JNK inhibition partially mitigates the anti-survival effect of SAA, and inhibitions of p38 and ERK synergize with SAA. Furthermore, SAA suppresses DLBCL tumor growth in a xenograft mouse model in vivo. Therefore, our data suggests the therapeutic utility of SAA in the management of DLBCL.

Abstract:Ethnopharmacological relevance: India is a rich source of medicinal plants and a number of plant extracts are used against diseases in various systems of medicine such as Ayurveda, Unani and Siddha. Only a few of them have been scientifically explored. One of such plants is Ammannia baccifera L., traditionally used as an antitumor agent. Aim of the study: The objective of the present study was to explore the anticancer activity of the methanol extract of A. baccifera against Dalton's ascites lymphoma (DAL)-bearing Swiss albino mice and in vitro cytotoxicity against human cervical cancer cell line (HeLa). Materials and methods: In vitro cytotoxic effects of A. baccifera was evaluated against HeLa and NIH 3T3 cells using MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] method. Anticancer activity of the extract was evaluated in Swiss albino mice against DAL cell line at the doses of 100 and 200mg/kg p.o. The extract was administered to animals for 14 consecutive days. After 12h fasting from last dose, the mice were sacrificed and the anticancer effect of A. baccifera was assessed by evaluating tumor volume, viable and nonviable tumor cell count, tumor weight, hematological parameters and certain antioxidant biochemical parameters against DAL bearing control group. Results: The extract of A. baccifera was cytotoxic to the HeLa cancer cell line but relatively non-toxic to the normal cell line NIH 3T3. A. baccifera extract treatment resulted in significant decreases in tumor volume, viable cell count and tumor weight and enhanced the life span of DAL bearing mice. Hematological parameters such as RBC, hemoglobin and lymphocyte count reverted to normal level in A. baccifera treated mice. The extract also significantly (p<0.05) decreased the levels of lipid peroxidation and increased the activities of GSH, GPx, SOD and CAT. The anticancer effects of the A. baccifera extract is comparable with that of the standard drug 5-Fluorouracil. Conclusion: The results showed that the methanol extract of A. baccifera is effective in inhibiting the tumor growth in ascetic models that is comparable to 5-fluorouracil. This plant has potential in the development of anticancer therapy and this study scientifically validated the folklore use of this plant.

Abstract:A lectin (AMML) from the roots of Astragalus mongholicus was extracted and purified by affinity chromatographic technique. Human cervical carcinoma cell line (HeLa), human osteoblast-like cell line (MG63) and human leukemia cell line (K562) were used to check the effects of AMML on cell proliferation, apoptosis and cell cycle. Maximum growth inhibition (92%) was observed with HeLa cells, followed by K562 cells (84%) and MG63 (48%) cells. Morphological observation showed that AMML-treated HeLa cells displayed outstanding apoptosis characteristics, such as nuclear fragmentation and appearance of membrane-enclosed apoptotic bodies. The apoptosis of HeLa cells was confirmed by flow cytometry using Annexin V/FITC and propidium iodide (PI) staining technique. For the first time we also report a significant cell cycle arrest at S phase of HeLa cells by AMML. Therefore, the present investigation may lead to the possible therapeutic use of Astragalus mongholicus lectin.
Mechanisms of action of Sappan lignum for prostate cancer treatment: network pharmacology, molecular docking and experimental validation

Abstract:Background: Prostate cancer (PCa) is the most common non-cutaneous malignancy in men globally. Sappan lignum, which exists in the heartwood of Caesalpinia sappan L., has antitumor effects; however, its exact mechanism of action remains unclear. This study elucidated the underlying mechanisms of Sappan lignum in PCa through network pharmacology approaches and molecular docking techniques. Moreover, the therapeutic effects of Sappan lignum on PCa were verified through in vitro experiments. Methods: The constituent ingredients of Sappan lignum were retrieved from the HERB database. Active plant-derived compounds of Sappan lignum were screened based on gastrointestinal absorption and gastric drug properties. Disease targets for PCa were screened using unpaired and paired case datasets from the Gene Expression Omnibus. Intersection targets were used for gene ontology and Kyoto encyclopedia of genes and genomes (KEGG) pathway enrichment analysis. Core targets were identified through topological analysis parameters and their clinical relevance was validated through The Cancer Genome Atlas database. The affinity between the phytochemicals of Sappan lignum and core proteins was verified using the molecular docking technique. Validation experiments confirmed the significant potential of Sappan lignum in treating PCa. Results: Twenty-one plant-derived compounds of Sappan lignum and 821 differentially expressed genes associated with PCa were collected. Among 32 intersection targets, 8 were screened according to topological parameters. KEGG analysis indicated that the antitumor effects of Sappan lignum on PCa were primarily associated with the p53 pathway. The molecular docking technique demonstrated a strong affinity between 3-deoxysappanchalcone (3-DSC) and core proteins, particularly cyclin B1 (CCNB1). CCNB1 expression correlated with clinicopathological features in patients with PCa. Experimental results revealed that 3-DSC exhibited anti-proliferative, anti-migratory, and pro-apoptotic effects on 22RV1 and DU145 cells while also causing G2/M phase cell cycle arrest, potentially through modulating the p53/p21/CDC2/CCNB1 pathway. Conclusion: This research highlights the promising therapeutic potential of Sappan lignum in treating PCa, with a particular focus on targeting the p53 pathway.
Potential anticancer activity of litchi fruit pericarp extract against hepatocellular carcinoma in vitro and in vivo.

Abstract:Litchi fruit pericarp (LFP) extract contains significant amounts of polyphenolic compounds, and exhibits powerful antioxidative activity against fat oxidation in vitro. The purpose of this study is to confirm the anticancer activity of LFP extract against hepatocellular carcinoma in vitro and in vivo, and to elucidate the mechanism of its activity. Human hepatocellular carcinoma cell line was tested in vitro for cytotoxicity, colony formation inhibition, and cell cycle distribution through flow cytometry after treatment with water-soluble crude ethanolic extract (CEE) from LFP. Murine hepatoma bearing-mice were fed doses of 0.15, 0.3, and 0.6g/kg/day of water-soluble CEE in DH2O p.o. for 10 days, respectively, to test the anticancer activity and BrdU incorporation of cancer cells in vivo. LFP extract demonstrated a dose- and time-dependent inhibitory effect on cancer cell growth; IC50 was 80μg/ml, and significantly inhibited colony formation in vitro, tumor growth and BrdU incorporation into cancer cells in vivo. The tumor inhibitory rates at doses of 0.15, 0.3, and 0.6g/kg/day were 17.31% (P>0.05), 30.77% (P<0.05), and 44.23% (P<0.01), respectively. BrdU labeled tumor cells of treated animals were 11.80±2.79%, and were significantly lower than that in untreated controls (23.00±5.42%, P<0.05). Our findings showed that LFP extract exhibited potential anticancer activity against hepatocellular carcinoma in vitro and in vivo through proliferating inhibition and apoptosis induction of cancer cells.
Antitumor and Immune Regulation Activities of the Extracts of Some Chinese Marine Invertebrates

Abstract:Extracts of 21 marine invertebrates belonging to Coelenterata, Mollusca, Annelida, Bryozoa, Echiura, Arthropoda, Echinodermata and Urochordata were screened for the studies on their antitumor and immune regulation activities. Antitumor activity was determined by MTT method and immune regulation activity was studied using T- and B-lymphocytes in mice spleenin vitro. It was found that the n-butanol part ofAsterina pectinifera, the acetic ether part ofTubuaria marina, 95% ethanol extract ofAcanthochiton rubrolineatus have a high inhibition rate of 96.7%, 63.9% and 50.5% respectively on tumor cell line HL-60 at the concentration of 0.063 mg/ml. The inhibition rate of the acetic ether part ofTubuaria marina on the tumor cell line A-549 is 65.4% at concentration of 0.063 mg/mL. The 95% ethanol extract ofMeretrix meretrix has so outstanding promoting effect on T-lymphocytes that their multiplication increases 25% when the sample concentration is only 1 μg/ml. On B-lymphocytes, the 95% extract ofRapana venosa, at concentration of 100 μg/ml, has a promotion percentage of 60%. On the other hand, under the condition of no cytotoxic effect, the 95% ethanol extracts ofAcanthochiton rubrolineatus andCellana toreum can reach 92% inhibition rate on T lymphocyte at concentration of 100 μg/ml, while the inhibition rate on B lymphocyte of the 95% extract ofAcanthochiton rubrolineatus reaches 92% at the same concentration.
