Abstract:An enantioselective synthesis of platensimycin, a novel antibiotic natural product that inhibits bacterial beta-ketoacyl-(acyl-carrier-protein) synthase (FabF), is described. Our synthetic strategy for the construction of the oxatetracyclic core involved an intramolecular Diels-Alder reaction. Our preliminary studies provided a complex tetracyclic product by first undergoing an interesting 1,5-hydride shift followed by a Diels-Alder reaction. Further optimization of the diene's electronic properties, by incorporation of a methoxy group, led to the oxatetracyclic core of platensimycin. The three required chiral centers, including two all-carbon quaternary chiral centers, were built in the intramolecular Diels-Alder step. The synthesis utilized natural (+)-carvone as the key chiral starting material, which determined the stereochemistry of the final product. The synthesis also featured an efficient Petasis olefination, a hydroboration sequence, a Gais's asymmetric Horner-Wadsworth-Emmons reaction, and a mercury salt catalyzed enol ether isomerization.

Abstract:A mild and efficient dual-mode Lewis acid induced Diels-Alder (DA)/carbocyclization cascade cyclization reaction has been developed for construction of the tricyclic core of ent-kaurenoids in one pot with the aid of a theoretical study on the π,σ-Lewis acidities of a variety of Lewis acids. With ZnBr2 as the dual-mode Lewis acid, a series of substituted enones and dienes underwent DA/carbocyclization cascade cyclization reaction smoothly at room temperature and provided the tricyclic cyclized products in one pot with good yields and high diastereoselectivity. The tricyclic cyclized product has been successfully utilized as a common intermediate for formal syntheses of (±)-platensimycin and (±)-platencin.

Abstract:Platencin (1) was isolated from Streptomyces platensis MA 7339 in 2007 which showed highly potent and broad-spectrum activity against many multidrug-resistant pathogens. [1] In this formal synthesis, we have developed two different procedures to synthesize tricyclic core 3 from o-vanillin (2) by intermolecular and intramolecular Diels-Alder strategies, respectively. In the intermolecular Diels-Alder strategy, Diels-Alder reaction of masked o-benzoquinone 4 and an aldol condensation were the key steps in the construction of the tricyclic core of platencin. In the intramolecular Diels-Alder strategy, Diels-Alder reaction of masked o-benzoquinone 6, an aldol condensation, and stereoselective hydrogenation were the key steps. The detail will be discussed.

Abstract:A bioinspired photo-Fries/imine capture cascade reaction was developed in continuous-flow mode, which facilitated the rapid construction of a series of diversely functionalized 2,7-heterocycle-fused tetrahydrocarbazoles, the ubiquitous core structures embedded in strychnos and akuammiline-type monoterpene indole alkaloids. The synthetic utility of this novel method has been preliminarily explored by the first total synthesis of (+)-alsmaphorazine C and formal synthesis of (+)-strictamine in a concise and efficient manner.

Abstract:An enantioselective route to the oxatetracyclic core of (-)-platensimycin (1) has been investigated by using an intramolecular Diels-Alder reaction as the key step. The thermal reaction of E/Z mixture (1:1) provided oxatetracyclic core 2 from the E-diene and the Z-diene was recovered unchanged. The Diels-Alder substrate was conveniently assembled in optically active form with use of (S)-carvone as the starting material.

Abstract:A stereoselective formal synthesis of platencin was accomplished from bromophenol 8 in eleven synthetic steps with 6 % overall yield. The intramolecular Diels‐Alder reaction of masked o‐benzoquinone, intramolecular aldol condensation, and stereoselective hydrogenation were the key steps in the construction of the tricyclic core of platencin. A stereoselective construction of the tricyclic core of platencin was accomplished in eleven synthetic steps from bromophenol 8 via the intramolecular Diels‐Alder reaction of masked o‐benzoquinone, intramolecular aldol condensation, and stereoselective hydrogenation.

Abstract:In the work reported herein, the concise and enantioselective total synthesis of the Schizozygine alkaloid (-)-strempeliopine was developed. This synthetic strategy featured the palladium-catalyzed decarboxylative asymmetric allylic alkylation of N-benzoyl lactam to set up the absolute configuration at the C20 position, a highly diastereoselective one-pot Bischler-Napieralski/lactamization and iminium reduction sequence for the construction of the pentacyclic core structure, and the late-stage dearomative addition of indole, leading to the otherwise difficult-to-achieve hexacyclic indoline framework with complete control of four neighbouring stereocenters.

Abstract:The first enantioselective total synthesis of (+)-steenkrotinA has been achieved in 18 steps and 4.2% overall yield. The key features of the strategy entail a Rh-catalyzed O-H bond insertion followed by an intramolecular carbonyl-ene reaction, two sequential SmI2-mediated Ueno-Stork and ketyl-olefin cyclizations, and a cascade intramolecular aldol condensation/vinylogous retro-aldol/aldol process with inversion of the relative configuration at the C7 position. The absolute configuration of (+)-steenkrotin A was determined based on the stepwise construction of the stereocenters during the total synthesis.

Abstract:The Plakortin polyketides represent a structurally and biologically fascinating class of marine natural products. Herein, we report a unified strategy that enables the divergent syntheses of various Plakortin polyketides with high step-economy and overall efficiency. As proof-of-concept cases, the enantioselective total syntheses of (+)-hippolachnin A, (+)-gracilioether A, (-)-gracilioether E, and (-)-gracilioether F have been accomplished based on a series of bioinspired, rationally designed, or serendipitously discovered transformations, which include (1) an organocatalytic asymmetric 1,4 conjugate additidn to assemble the common chiral gamma-butenolide intermediate enroute to all of the aforementioned targets, (2) a challenging biomimetic [2+2] photocycloaddition to forge the oxacyclobutapentalene core of (+)-hippolachnin A, (3) a [2+2] photocycloaddition followed by one-pot oxidative cleavage of methyl ether/Baeyer-Villiger rearrangement to access (-)-gracilioether F, and (4) an unprecedented hydrogen-atom-transfer-triggered oxygenation of vinylcyclobutane to afford (+)-gracilioether A and (-)-gracilioether E in one pot.

Abstract:Amphidinolide Y, a 17-membered cytotoxic macrolide isolated from marine dinoflagellates, has been synthesized via ring-closing metathesis to assemble the congested trisubstituted (E)-double bond. The seco precursor was prepared from readily available chiral synthons with the tetrahydrofuran ring formed via 5-endo epoxide ring-opening cyclization. It was found that the C6-keto seco substrate showed higher reactivity toward Grubbs' second generation catalyst while Schrock's Mo catalyst was completely inactive for formation of the macrocycle.

Abstract:The first and enantioselective total synthesis of (+)-plumisclerinA, a novel unique complex cytotoxic marine diterpenoid, has been accomplished. Around the central cyclopentane anchorage, a sequential ring-formation protocol was adopted to generate the characteristic tricycle[4.3.1.0(1,5)]decane and trans-fused dihyrdopyran moiety. Scalable enantioselective La-III-catalyzed Michael reaction, palladium(0)-catalyzed carbonylation and SmI2-mediated radical conjugate addition were successfully applied in the synthesis, affording multiple grams of the complex and rigid B/C/D-ring system having six continuous stereogenic centers and two all-carbon quaternary centers. The trans-fused dihyrdopyran moiety with an exo side-chain was furnished in final stage through sequential redox transformations from a lactone precursor, which overcome the largish steric strain of the dense multiring system. The reported total synthesis also confirms the absolute chemistries of natural (+)-plumisclerinA.

Abstract:The molecular design, chemical synthesis, and biological evaluation of two distinct series of platensimycin analogues with varying degrees of complexity are described. The first series of compounds probes the biological importance of the benzoic acid subunit of the molecule, while the second series explores the tetracyclic cage domain. The biological data obtained reveal that, while the substituted benzoic acid domain of platensimycin is a highly conserved structural motif within the active compounds with strict functional group requirements, the cage domain of the molecule can tolerate considerable structural modifications without losing biological action. These findings refine our present understanding of the platensimycin pharmacophore and establish certain structure-activity relationships from which the next generation of designed analogues of this new antibiotic may emerge.

Abstract:Over the past decade, Gold complexes have emerged as efficient and mild catalysts for the transformation of substrates possessing alkyne functionality into a range of useful scaffolds. These powerful methods have enabled the development of novel approaches for the total synthesis of biologically active natural products by gold catalysis. In this case, we found that the intramolecular nucleophilic addition of a hydroxyl group to a carbon-carbon triple bond, which activated by a gold catalyst, followed by further useful transformation has proven to be an excellent method for rapid construction of structural diversity of molecular scaffolds. The cortistatins are a family of 11 steroidal alkaloids which exhibit significant biological activities. The intriguing biological properties and their low natural abundance have elevated cortistatins to be a typical target for both partial and total synthesis. Up to now, more than a dozen research groups have published approaches directed toward the synthesis of cortistatins, including one semi-synthesis, five total syntheses and five formal syntheses, as well as a number of synthetic studies about the pentacyclic core and some illuminating model studies. One of the biggest challenges for the synthesis of cortistatins is how to construct the unprecedented oxabicyclo[3.2.1] octane ring system which lies within a complex tetracarbocyclic skeleton. In our previous work, we have developed a gold-catalyzed semi-pinacol rearrangement strategy to diastereoselective synthesis of the oxabicyclo[3.2.1] octane ring system. The wide substrate scope as well as the high diastereoselectivity have made us to apply this method into the asymmetric formal synthesis of Cortistatins. Herein, full details about our efforts towards the formal synthesis of cortistatins were described by employing our developed gold-catalyzed cascade reaction to oxabicyclo[3.2.1] octane ring systems. This route is featured with a novel gold-catalyzed cascade reaction involving intramolecular nucleophilic addition of hydroxyl group to the carbon-carbon triple bond, followed by an oxonium ion initiated semi-pinacol-type 1,2-migration to construct the key oxabicyclo[3.2.1] octane skeleton.
