Abstract:Phenylethylamine and phenoxylalkylamine fragments were introduced into pyridazinone derivatives (as alpha(1)-adrenoceptor antagonist) for the first time. 30 new compounds of pyridazinone were designed and synthesized. The structures of the new compounds were identified by H-1 NMR, IR and HRMS spectra. Target compounds have been assayed with function analysis in anococcyeus muscle. The results indicate that 28 target compounds have good blocking activity to alpha(1)-adrenoceptor (pA(2)>6.00), The pA(2) of compounds 6o, 6p, 6q, 6v, 6x, 6y, 10c and 10d were beyond 7.00.

Abstract:Aim To study the blocking activity of carbamoyl substituted phenoxylalkylamine derivatives as α1-adrenocepter(α1-AR) antagonist.Methods A series of carbamoyl substituted phenoxylalkylamine derivatives were synthesized through the condensation of 4a or 4b with various substituted phenylethanamine and reduction.Results Twelve new compounds were synthesized and their structures were confirmed by MS,1H-NMR,IR spectra.Their bioactivities had been tested.Conclusion The test result indicated that most the target compounds showed some blocking activity to α1-adrenoceptor.

Abstract:In order to search for new α1-adrenoceptor antagonists bearing 1,4-benzodioxane moiety, six new target compounds (7a-7f) were synthesized from catechol via ring-closing, hydrolysis, amidation, substitution and salt formation. Their structures were confirmed by ESI-MS, 1H NMR, IR and HR-MS. Preliminary biological evaluation result showed that most of the target compounds had modest blocking activity against α1- adrenoceptor and the pA2 value of 7e was greater than 7.00. 3D-QSAR study of the title compounds was performed via Sybyl software. Based on our previous work, CoMFA model was constructed from eighteen 1-(1,4-benzodioxan-2- ylcaronyl)-4-aryloxy-alkyl-piperazine analogues (q2=0.753, r 2=0.986), and these results would be helpful for further structural optimization of 1,4-benzodioxane α1-adrenoceptor antagonists.

Abstract:Aim To find new α_1-adrenoceptor antagonists bearing 1,4-benzodioxane moiety.Methods The target compounds were synthesized from catechol via ring-closing,hydrolysis,amidation,substitution and salt formation.Preliminary biological activities were evaluated.Result and conclusion Thirteen new target compounds were synthesized and their structures were confirmed by ESI-MS,IR,()~1H-NMR,and HRMS.Biological assay showed the pA_2 values of eight target compounds were greater than 6.00.Preliminary bioassay suggested that most of the target compounds showed potent blocking activity to α_1-AR and 1-(1,4-benzodioxan-2-yl-carbonyl)-4-aryl-oxyalkylpiperazine should be a kind of valuable α_1-adrenoceptor antagonists to be investigated.

Abstract:Aim To study the antihypertensive effect and quantitative structure-activity relationship(QSAR)of halophenoxylalkylamines as α 1-adrenocepter(α 1-AR)antagonist.Methods Based on the biological activity of DDPH metabolites,12 compounds of halophenoxylalkylamines were designed and prepared.The target compounds were synthesized from 2,6-dimethylphenol and their structures were confirmed by MS, 1H-NMR,IR spectra.Their antihypertensive effects were tested and the QSAR was subjected to weighted holistic invariant molecular(WHIM)descriptors.Results The test indicates that all the target compounds exhibited antihypertensive activities.Compounds 3,10 have potent antihypertensive effect but their duration time is shorter than that of DDPH.Conclusion The QSAR indicates that the antihypertensive duration time of the compounds could be prolonged by enhancement in molecular symmetry or simplification of the molecular substituents.

Abstract:AIM: To study the synthesis and anti-α_ 1 -adrenoceptor activity of 5-methoxytryptamine phenylpiperazine compounds. METHOD: Cyclization,substitution and amidation reactions were used in the synthesis of the title compounds.The inhibition activity of the target compounds were tested. RESULT: Fourteen new compounds were synthesized. Structure of the title compounds were determined by IR, 1 HNMR and ESI-MS. Compounds WBⅠ-2,WBⅠ-3,WBⅠ-4,WBⅠ-8,WBⅠ-9 and WBⅠ-14 showed inhibition activity. CONCLUSION: The potency (pA_ 2 ) of the title compounds was lower than those of control Prazosin.

Abstract:Angiotensin II (A II) receptor antagonists are clinically useful for the management of cardiovascular disease. Based on the analysis of the quantitative structure-activity relationships of forty-two new compounds reported in literatures as A II receptor antagonists, a series of novel 5-amido-1H-imidazo[5,4-b]pyridine derivatives were designed and synthesized by application of the methods of computer aided drug design and principles of bioisosterism and hybridization. All of them were first reported and their structures were confirmed by IR, H-1 NMR and MS spectra. The results of preliminary pharmacological tests in vitro show that some target compounds have certain antagonistic activity of A I receptor. Moreover, the analysis of SAR suggests that N-phenylpyrrole-2-tetrazole can be used as a substitution for future designing of novel A I receptor antagonists.

Abstract:Aim To study the synthesis and anti-hypertensive activity of NO-donating benzopyran compounds and search for novel anti-hypertensive agents with high activity and less side-effects.Method Ten novel compounds Ⅲ_1～Ⅲ_(10) were synthesized by coupling of organic nitrate and furoxan with trans-3,4-benzopyran on the C-4 position through succinic acid.Their structures were confirmed by spectral data and elemental analysis.The inhibition of the target compounds on KCl-induced contraction of the aortic strips and the effects on systolic aortic pressure(SAP)and diastolic aortic pressure(DAP) of the spontaneously hypertensive rats(SHR)were detected.The amount of NO released in vitro of the compounds was also determined by Griess method.Result and conclusion The preliminary pharmacological tests showed that most of the target compounds inhibited the KCl-induced contraction to a certain extented.Among them Ⅲ_9 decreased SAP(22.1%)and DAP(23.8%) of the SHR with the same duration as control pinacidil.The amount of NO released by Ⅲ_9 is 0.9 mg·L~(-1).

Abstract:AM: In a search for new cardiovascular drug candidates, a series of novel oxime ethers derived from a natural isochroman-4-one were synthesized. METHOD: Compounds 3 and 6, derived from the natural antihypertensive compound 7, 8-dihydroxy-3-methyl-isochroman-4-one (XJP), were designed and synthesized. Subsequently, a series of novel isochroman-4-one codme ether hybrids were prepared by hybridizing various N-substituted isopropanolamine functionalities to isochroman-4-one codme. Furthermore, beta(1)-adrenergic blocking activities of the synthesized compounds were assayed using the isolated rat left atria. RESULTS: Twenty target compounds were obtained, and the preliminary structure-activity relationships were deduced. The most promising compound Ic exhibited beta(1)-adrenoceptor blocking activity (inhibition: 52.2%) at 10(-7) mol.L-1, which was superior to that of propranolol (inhibition: 49.7%). CONCLUSION: The results suggested that natural product XJP/isopropanolamine moiety hybrids may provide a promising approach for the discovery of novel cardiovascular drug candidates.

Abstract:AIM:To design and synthesize novel AchE inhibitors.METHODS:The condensation of 2-bromo-5, 6-dimethoxy-indan-1-one with various aminoalkyl phenols in the presence of K2CO3 and acetonitrile gave the corresponding title compounds, and the in vitro AchE and BchE inhibitory activities were evaluated by the modified Ellman method.RESULTS:Sixteen novel target compounds 8a - p were synthesized, their structures were confirmed by 1H NMR, MS, IR and elemental analysis. Preliminary pharmacological test demonstrated that most of these compounds displayed high AchE inhibitory activities, the IC50 of the most potent inhibitor 8h was 50.0 nmol x L(-1), similar to that of Huperzine A (IC50 = 53.0 nmol x L(-1)), while all the compounds were almost inactive against BchE.CONCLUSION:2-Phenoxy-indan-1-one derivatives exhibit high activities of AchE inhibition and are worthy of further investigation.

Abstract:A series of novel cyclosulfamido substituted phenoxylalkylamine derivatives were synthesized through the condensation and reduction of various substituted phenoxypropan-2-one with 7-(2-aminoethyl-3,4-dihydro-1,1-dioxo-2H-benzo[e][1,2]thiazine)hydrocholoride (5). Their structures were identified by H-1 NMR, HRMS, and IR data. Abstract:A series of phenoxyisoquinolines, N-phenoxyethyl-1-(2-nitrophenyl)-1,2,3,4-THIQs 3a–3d, N-phenoxyethyl-1-benzyl-1,2,3,4-THIQ 3e, N-phenoxyethyl-1-(2-aminophenyl)-1,2,3,4-THIQs 5f–5i, N-phenoxyethyl-1-(2-phenoxyethylaminophenyl)-1,2,3,4-THIQs 5f′–5i′, have been synthesized and tested in isolated rat vas deferens α-adrenoreceptors. Comparison of pA2 values for these compounds in the presence of phenylephrine confirms that α1-adrenoceptor blocking activity of 3a–3d (–NO2 series) is more active than 6a–6c (–NH2 series) in the aortic rings isolated from SD rats. On the other hand, the electron-donating group at the 6-position of isoquinoline ring either increases or decreases the α1-adrenoceptor blocking activity.

Abstract:Finding effective chemotherapeutic agents for clinical use is a long-lasting goal in medicinal chemistry. In this study, we report a new class of alpha(1)-adrenoceptor (alpha(1)-AR) antagonists. Specifically, we describe the synthesis and the blocking activities toward alpha(1)-AR of 7-(2-hydroxypropoxy)-3,4-dihydroisoquinolin-1(2H)-one 1 and its structurally perturbed analogs 2-11 that were designed according to the principle of bioisosterism. Their structures were identified with IR, 1H NMR, MS, HRMS and elemental analysis. The blocking activities of compounds 1-11 were evaluated on isolated rat anococcygeus muscles. The results indicated that these compounds showed moderate to good activities. Among them, compound 1 exhibited the highest activity that was comparable to those of known alpha(1)-AR antagonists tamsulosin and DDPH (1-(2,6-dimethylphenoxy)-2-(3,4-di- methoxyphenylethylamino)propane hydrochloride) and thus may be exploitable as a lead compound for the discovery of promising alpha(1)-AR antagonists.
Study on NO-donating antihypertensive agents I. Synthesis and antihypertensive activity of C-3 nitrate or furoxan substituted benzopyrans

Abstract:A series of nitric oxide-donating derivatives of [1,2,4]triazol-5(4H)-one (9a-f and 15a-f) as a novel class of angiotensin II receptor AT1 antagonists have been designed and synthesized by coupling furoxan and nitric oxide with lead compound 1. The synthesized compounds were evaluated for their antagonism of AT1 receptor with induced contraction in the rabbit thoracic aortic ring and the results showed that compounds 9b, 15b and 15d exhibited potent antagonistic activity of AT1 receptor. Moreover 9b, 15b, 15d, 15e had good maximum NO release amount of this series.

Abstract:Two series of novel NO-releasing benzimidazole derivatives (8a-e, 9a-g) were designed and synthesized by coupling nitro ester and furoxan NO-donor moieties with benzimidazole biphenyl skeleton. The NO-releasing assay indicated that all the target compounds had different level of NO-releasing ability. Furthermore, the isolated organ assay (rat aortic strips) was used to evaluate the antagonism of Ang II-induced vasoconstriction ability. It was observed that the pA(2) values of compounds 8e and 9e were better than that of lead compound 6. Moreover, the pharmacological investigation showed that the antagonism of Ang II-induced pressure response by oral administration of compound 8e was obviously superior to that of lead compound 6, and comparable to that of the positive control losartan. These results suggested that NO-releasing hybrids may provide a promising approach for the discovery of novel antihypertensive agents.

Abstract:7,8-Dihydroxy-3-methyl-isochromanone-4 (XJP), is a polyphenolic natural product with moderate antihypertensive activity. To obtain new agents with stronger potency and safer profile, we employed XJP and naftopidil as the lead compounds to design and synthesize a novel class of hybrids as antihypertensive agent candidates. In the present study, a series of hybrids (6a–r) of XJP bearing arylpiperazine moiety, which is identified as the pharmacophore of naftopidil, were designed and synthesized as novel α1-adrenergic receptor antagonists. The biological evaluation showed that target compounds 6c, 6e, 6f, 6g, 6h, 6m and 6q possessed potent in vitro vasodilation potency and α1-adrenergic receptor antagonistic activity. Furthermore, the most potent compound 6e significantly reduced the systolic and diastolic blood pressure in spontaneously hypertensive rats (SHRs), which was comparable to that of naftopidil, and it had no observable effects on the basal heart rate, suggesting that 6e deserves to be further investigated as a potential clinical candidate for the treatment of hypertension.

Abstract:A series of 2-alkylbenzimidazoles bearing a N-phenylpyrrole moiety were synthesized and evaluated as a novel class of AT(1) receptor antagonists. Among them, compounds 10a and 10g inhibited [(125)I] AngII-binding affinity to AT(1) receptor at nanomolar level and potently inhibited the Ang II-induced pressor response by oral administration. Moreover, evaluation in spontaneously hypertensive rats showed that 10a is an orally active AT(1) receptor antagonist.
