Abstract:von Willebrand disease (vWD), which is caused by the quantitative and qualitative defects of von Willebrand factor (vWF), is one of the most common inherited human bleeding disorders. The analysis of vWF gene was performed in some patients with type 2 and type 3 vWD by denature gradient gel electrophoresis and sequencing. Six point mutations of vWF gene were found in these patients with vWD. They were ArgdllHis,Ala737-Glu,Arg834-Trp, Gly22-Glu, Met37-Val and Ser71-stop code. Among them, Ala737-Glu, Gly22-Glu, Met37-Val and Ser71-stop substitutions were the first reported mutations according to international database. We constructed an expression plasmid pSVA737EvWF containing the full length of cDNA of vWF which included the Ala737-Glu substitution by site-direct mutagenesis. Recombinant vWF containing the candidate mutation was transitory expressed in COS-7 cells. The mutation belonged to Group II. In addition, a study of 142 chromosomes in 71 Chinese people in Suzhou and Kunming areas showed that there was ATCT tandem repeated sequence in vWF gene of Chinese people and there were 8 alleles in the area of the 31/1880-1980 in vWF gene. The total theoretical heterozygous rate was 79.4%. The vWF gene from 52 unrelated individuals of Han ethnic, 44 of Yi tribe, 42 of Dai tribe were amplified by PCR and subsequently analysed restriction enzyme digestion of Sma I, Hha I, Msp I and Rsa I. The allele frequencies of Sma I, Hha I of vWF gene are very different between Chinese and Caucasians, though the theoretical heterozygous rates in the two populations are almost the same. But in the three races of Chinese, the allele frequencies and theoretical rates only have a little difference. The allele frequencies and theoretical rates of Mspl of vWF gene in Chinese are mild lower than that of Caucasians. As for Ras I RELP of vWF gene, the theoretical rate is much lower in Chinese Hans than that in Caucasians. So it has a lower diagnosis value in the family analysis of vWD in Chinese population.
