Abstract:A novel and practical chiral ruthenium NHC-diamine system is disclosed for the enantioselective hydrogenation of isocoumarins, which provides a new concept to apply (chiral) NHC ligands in asymmetric catalysis. A variety of optically active 3-substituted 3,4-dihydroisocoumarins were obtained in excellent enantio-selectivities (up to 99% ee). Moreover, this methodology was utilized in the synthesis of O-methylmellein, mellein, and ochratoxin A.

Abstract:An efficient synthesis of optically active 4-substituted 2-oxazolidinones by the ruthenium(II)-NHC-catalysed asymmetric hydrogenation of 2-oxazolones was developed. Excellent enantioselectivities (up to 96% ee) and yields (up to 99%) were obtained for a variety of substrates, bearing a range of functional groups and useful motifs. The hydrogenation reaction was successfully scaled up to gram scale using low catalyst loading. Moreover, the utility of this methodology was demonstrated by the transformation of the enantioenriched product into the corresponding chiral beta-amino alcohol, a bisoxazoline ligand, and the formal synthesis of (-)-aurantioclavine.

Abstract:Highly effective asymmetric hydrogenation of various ferrocenyl ketones, including aliphatic ferrocenyl ketones as well as the more challenging aryl ferrocenyl ketones, was realized in the presence of a Ru/diphosphine/diamine bifunctional catalytic system. Excellent enantioselectivities (up to 99.8% cc) and activities (S/C = 5000) could be obtained. These asymmetric hydrogenations provided a convenient and efficient synthetic method for chiral ferrocenyl alcohols, which are key intermediates for a variety of chiral ferrocenyl ligands and resolving reagents.

Abstract:Comprehensive Summary Chiral ruthenium‐catalyzed enantioselective hydrogenation of tetrapyridine‐type N‐heteroarenes was developed. The partially reduction of adjacent tetraheteroaromatic substrates proceeded smoothly in the presence of phosphine‐free chiral cationic ruthenium diamine complexes, affording unprecedented high reactivity, enantioselecitivity and diastereoselectivity (up to 93% yield, >99% ee and 92:8 dr). The potential application of chiral tetradentate pyridine‐amine products as chiral ligands has been demonstrated in the Cu‐catalyzed asymmetric Friedel–Crafts alkylation reaction between indoles and nitroalkenes. Abstract:Highly efficient asymmetric hydrogenation of dibenzo[b,f][1,4]oxazepine, dibenzo[b,f][1,4]thiazepine and dibenzo[b,e]azepine derivatives with chiral cationic ruthenium diamine catalysts has been developed, giving diverse chiral seven-membered N-heterocycles with excellent results (up to 99% yield and 99% ee). Moreover, it was found that the catalyst counteranion regulated the enantioselectivity obviously in the hydrogenation of dibenzo[b,f][1,4]thiazepine and dibenzo[b,e]azepine derivatives.

Abstract:A highly efficient P-stereogenic diphosphine-rhodium complex was applied to the chemo- and enantioselective hydrogenation of allylic hydrazones for the synthesis of chiral allylic hydrazines in 89-96 % yields and with 82-99 % ee values. This methodology was successfully applied to the preparation of versatile chiral allylic amine derivatives.

Abstract:A soluble polymer (MeO-PEG) supported biphenylbisphosphine (BIPHEP)-Ru/chiral diamine (1,2-diphenylethylenediamine) complex, in which the polymer is attached to the two phenyl rings of BIPHEP ligand, has been prepared, and shown to be highly active with good enantioselectivity for the catalyzed asymmetric hydrogenation of unfunctionalized aromatic ketones. The derived chiral ruthenium complex 5 proved to be stable in air allowing facile catalyst recycling. Especially for 4′-tert-butyl-acetophenone and 1-acetonaphthone, excellent ee values up to 96.5% and 95.9% have been obtained which are comparable to or even higher than the enantioselectivity achieved with 2,2′-bis(diphenylphosphino)-1,1′-binaphthyl-Ru-DPEN catalyst under similar conditions.

Abstract:A versatile, selective, solvent (methanol vs ethanol)- and base (potassium vs lithium carbonate)-assisted switchable synthesis of saturated ketone and α-methyl saturated ketone from α,β-unsaturated ketone is developed. Mechanistic aspects, evaluated from spectroscopic studies, in situ monitoring of the reaction progress, control studies, and labeling studies, further indicate the involvement of a tandem dehydrogenation-condensation-hydrogenation sequence in the reaction, in which the interconvertible coordination mode (imino N → Ru and amido N-Ru) of coordinated imidazole with Ru(II)-para-cymene is crucial, without which the efficiency and selectivity of the catalyst are completely lost. The catalyst demonstrates good efficiency, selectivity, and functional group tolerance and displays a broad scope (69 examples) for monomethylation and hydrogenation of unsaturated chalcones, double methylation of ketones, and N-methylation of amines.

Abstract:Asymmetric C−H activation has surfaced as a transformative toolbox for the efficient assembly of chiral molecules. However, despite of major advances in rhodium and palladium catalysis, ruthenium(II)‐catalyzed asymmetric C−H activation has thus far largely proven elusive. In contrast, we herein report on the unprecedented ruthenium(II)‐catalyzed highly regio‐, diastereo‐ and enantioselective C–H alkylation. The key to success was proved by the identification of C2‐symmetric chiral imidazolidine carboxylic acids (CICAs), which can be easily prepared in a one‐pot fashion. This ruthenium/CICA system enabled the efficient installation of central and axial chirality, and featured excellent branched to linear ratios with generally >20:1 dr and up to 98:2 er. Mechanistic studies by experiment and computation were carried out to unravel the catalyst mode of action.

Abstract:Heating tert-butyl-tetraline with [(p-cymene)RuCl2]2 produces the racemic complex [(arene)RuCl2]2, which can be separated into enantiomers by chromatography of its diastereomeric adducts with chiral phosphine ligand. The resolved chiral complex catalyzes C-H activation of N-methoxy-benzamides and their annulation with N-vinyl-pivaloyl amide giving dihydroisoquinolones in 50-90% yields and 70:30-90:10 enantiomeric ratio.

Abstract:Five ruthenium(ii) complexes were synthesized, including (HO-C5H3N-CH2-C5H3N-C5H4N)Ru(PPh3)Cl2 (3), [(HO-C5H3N-CH2-C5H3N-C5H4N)Ru(PPh3)2Cl][PF6] (4) and [(HO-C5H3N-CH2-C5H3N-C5H4N)Ru(PPh3)2OH][PF6] (5) bearing an unsymmetrical pincer NNN ligand with a 2-hydroxypyridylmethylene fragment, and [(CH3O-C5H3N-CH2-C5H3N-C5H4N)2Ru][Cl]2 (6) and [(CH3O-C5H3N-CH2-C5H3N-C5H4N)2Ru][PF6]2 (7) containing 2-methoxypyridylmethylene moieties. 4 reacts with H2O at room temperature to give 5 whose crystal structure reveals the existence of intramolecular hydrogen-bonding between its two -OH groups. 3 exhibits high catalytic activity for transfer hydrogenation of ketones.

Abstract:Highly enantioselective tandem hydrogenation/lactonization of various 2-acylarylcarboxylates including 2-aroylarylcarboxylates were realized by using [RuCl(benzene)(S)-SunPhos]Cl as the catalyst under mild reaction conditions. Excellent enantioselectivities (up to 99.6% ee) and activities (S/C=1000) were obtained. This convenient and practical method enables a direct access to a series of highly optically pure 3-substituted phthalides that are very important molecules as valuable pharmacological compounds and diversified synthons for medicinal chemistry. Moreover, a gram-scale reaction was performed to further demonstrate the practicality of this approach.
