Abstract:Abstract Background Hydrogen-rich saline has been reported to have antioxidant and anti-inflammatory effects and effectively protect against organ damage. Oxidative stress and inflammation contribute to the pathogenesis and/or development of pulmonary hypertension. In this study, we investigated the effects of hydrogen-rich saline on the prevention of pulmonary hypertension induced by monocrotaline in a rat model. Methods In male Sprague-Dawley rats, pulmonary hypertension was induced by subcutaneous administration of monocrotaline at a concentration of 6 mg/100 g body weight. Hydrogen-rich saline (5 ml/kg) or saline was administred intraperitoneally once daily for 2 or 3 weeks. Severity of pulmonary hypertension was assessed by hemodynamic index and histologic analysis. Malondialdehyde and 8-hydroxy-desoxyguanosine level, and superoxide dismutase activity were measured in the lung tissue and serum. Levels of pro-inflammatory cytokines (tumor necrosis factor-α, interleukin-6) in serum were determined with enzyme-linked immunosorbent assay. Results Hydrogen-rich saline treatment improved hemodynamics and reversed right ventricular hypertrophy. It also decreased malondialdehyde and 8-hydroxy-desoxyguanosine levels, and increased superoxide dismutase activity in the lung tissue and serum, accompanied by a decrease in pro-inflammatory cytokines. Conclusions These results suggest that hydrogen-rich saline ameliorates the progression of pulmonary hypertension induced by monocrotaline in rats, which may be associated with its antioxidant and anti-inflammatory effects.
Effects of Ang-(1-7) on Pulmonary Hypertension and Right Ventricular Remodeling in Monocrotaline-Treated Rats

Abstract:Objective To investigate the therapeutic effects of Ang-(1-7) on pulmonary hypertension and right ventricular remodeling in monocrotaline-treated rats.Methods Thirty SD rats were randomly divided into three groups: control group,MCT group and Ang-(1-7) group.Rats in MCT group and Ang-(1-7) group received 60mg/kg MCT injection subcutaneously and after 24 hours received either saline or 24ug/kg/h of Ang-(1-7) injection via osmotic minipumps for four weeks.The rats in control group were firstly injected saline subcutaneously and then received saline injection via osmotic minipumps for four weeks.Right ventricular free wall thickness(RVFWT),pulmonary arterial acceleration time(PAAT) and tricuspid regurgitation were measured by echocardiography.Right ventricular systolic pressure(RVSP),mean arterial pressure(MAP) and heart rate(HR) were measured.The animals' hearts were measured to calculate the ratio of right ventricle to left ventricle plus septum(RVHI=RV/LV+S) and right ventricle mass to body weight(RVMI=RV/BW).Histopathological changs of lung and right ventricle were observed by hematoxylin and eosin staining.Percentage of wall thickness(WT%) and percentage of wall area(WA%) of pulmonary arterioles were evaluated.Restults Four weeks after monocrotaline injection,compared with control group,RVSP,RVHI,RVMI,WT%,WA % and RVFWT were significantly increased in MCT group,PAAT were significantly decreased in MCT group.Some rats occured tricuspid regurgitation and right ventricle occured cardiomyocyte hypertrophy in MCT group.However,no significant difference was found between Ang-(1-7) group and control group among the above parameters.Conclusion Ang-(1-7) may inhibit pulmonary arterial hypertension and right ventricular remodeling in monocrotaline-treated rats.

Abstract:The preventive effect of Radix Salciae Miltiorrhizae (RSM) on pulmonary hypertension (PHT) and right ventricular hypertrophy (RVH) induced by monocrotaline (MCT) in rats was observed with the methods of measuring the right ventricular systolic pressure (PVSP), the ratio of the right ventricle to the left ventricle plus interventricular septum RV/(LV+ S) and the pulmonary small artery morphological analysis. The results show that RSM can reduce the PHT, and prevent the RVH and the increase of the medial thickness of pulmonary small arteries. It can also prevent the endothelial cell injury produced by MCT. The pharmacological effects of RSM on pulmonary circulation were also discussed.

Abstract:This study investigated the effects of hydroxycitric acid tripotassium hydrate on right ventricular function, myocardial and pulmonary vascular remodeling in rats with pulmonary hypertension, and possible mechanisms.METHODS:Pulmonary hypertension was induced in male Sprague-Dawley rats by a single subcutaneous injection of monocrotaline or hypoxic chamber. In vivo, inflammatory cytokine (including TNF-α, IL-1β, IL-6, and TGF-β, the level of SOD) expression, superoxide dismutase and hydrogen peroxide levels, and p-IκBα and p65 expressions were detected. In vitro, pulmonary artery smooth muscle cell proliferation and migration, ROS production, and hypoxia-inducible factor-1 expression were also studied.RESULTS:Hydroxycitric acid tripotassium hydrate decreased right ventricular systolic pressure and reduced right ventricular fibrosis and pulmonary vascular remodeling in rats with two kinds of pulmonary hypertension. Moreover, the expression of both inflammatory and oxidative stress factors was effectively reduced, and the p65 signaling pathway was found to be inhibited in this study. Additionally, hydroxycitric acid tripotassium hydrate inhibited human pulmonary artery smooth cell proliferation and migration in vitro.CONCLUSIONS:This study shows that hydroxycitric acid tripotassium hydrate can alleviate pulmonary hypertension caused by hypoxia and monocycloline in rats, improve remodeling of the right ventricle and pulmonary artery, and inhibit pulmonary artery smooth muscle cell proliferation and migration. The protective effects may be achieved by regulating inflammation and oxidative stress through the p65 signaling pathway.
[Effect of Elastase Inhibitor on Pulmonary Hypertension Induced by Monocrotaline].

Abstract:OBJECTIVE:Pulmonary hypertension is a proliferative vascular disease characterized by pulmonary vascular structural remodeling. Until now, the pathogenesis of pulmonary hypertension is still not fully understood. Although considerable progress has been made, there is, to date, no cure for advanced pulmonary vascular disease. Recently, a number of studies suggest that endogenous vascular elastase (EVE) play a role in the vascular changes associated with pulmonary hypertension. The purpose of the study was to determine whether an elastase inhibitor might reverse advanced pulmonary vascular disease produced in rats by injection of monocrotaline.METHODS:One hundred and twenty male Sprague-Dawley rats were used in this study. The rats were divided into three groups: control, model and ZD-0892 groups. In the model and ZD-0892 groups, the rats were subjected to a single subcutaneous injection of monocrotaline (60 mg/kg) in the hind flank, while the rats in control group received an equivalent volume of 0.9% saline. From day 21, the rats in the ZD-0892 and model groups received twice-daily gavage tube feedings of either ZD-0892 at a dose of 240 mg/kg per day or its administration vehicle, while the rats in control group were subjected to an equivalent volume of 0.9% saline. On days 21, 28 and 35 post-injection, the elastolytic activity was measured with a fluorescence microplate reader and pulmonary artery pressure was detected via catheterization. Meanwhile, the lungs were evaluated morphologically, using the barium-gelatin perfusion technique.RESULTS:The injection of monocrotaline led to severe pulmonary hypertension in rats 21 days later and pulmonary artery elastolytic activity increased remarkably. A 1-week treatment with ZD-0892 resulted in declines in elastase activity. This was associated with significant declines in pulmonary artery pressure, decreases in muscularization of peripheral arteries and reductions in medial hypertrophy. After 2 weeks, elastase activity returned to normal level. Pulmonary artery pressure and structure were normalized.CONCLUSION:Increased elastase activity is important in the development of vascular changes and progressive pulmonary hypertension. ZD-0892 can suppress the elastase activity and completely reverse the fatal pulmonary hypertension induced by monocrotaline in rats.
Genistein, Bosentan and Tadalafil in Treatment of Monocrotaline-Induced Pulmonary Arterial Hypertension in Rats: a Comparison of Efficacy

Abstract:Objective To compare the effects of genistein,bosentan,and tadalfil in treatment of monocrotaline-induced pulmonary arterial hypertension (PAH) in rats.Methods Monocrotaline-induced PAH SD-rat model was established by injecting monocrotaline (50 mg/kg) intraperitoneally (n=48),while the rats in control group (n=8)were given normal saline injection.Then all model rats were randomly divided into 6 groups (n=8) according to different drugs given by gavage after 3 weeks:PAH group (PAH rats were treated with normal saline),genistein G30,G60,G120 groups (PAH rats were treated with genistein 30,60,120 mg· kg-1 · d-1,respectively),bosentan group (PAH rats were treated with bosentan 5 mg · kg-1 · d-1),and tadalafil group (PAH rats were treated with tadalafil 0.5 mg · kg-1 · d-1).After 2 weeks of treatment,survival rate,tricupid regurgitation (TR),pulmonary artery diameter (PAD),mean pulmonary arterial pressure (PAP),acetylcholine (Ach)-induced endothelium-dependent relaxation (EDdR),sodium nitroprusside (SNP)-induced endothelium-independent relaxation (EDiR),right ventricular hypertrophy index (RVHI),and lung phathological of rats in each group were compared.Results Compared with the control group,the TR,PAD,mean PAP and RVHI of rats were significantly increased in PAH group (P＜0.05).Compared with the PAH group,the TR and RVHI of rats were significantly decreased in G60 and G-120 groups,PAD was significantly decreased in G120 group,mean PAP was significantly decreased in G30,G-60 and G120 groups,survival rate was significantly increased in G120 group,and endothelium-dependent relaxation was significantly improved in G30,G60 and G120 groups,while pulmonary vascular luminal stenosis,wall thickening and vascular smooth muscle cell proliferation were significantly inhibited in G30,G60 and G120 groups (P＜0.05).However,the above effects of genistein were less than bosentan and tadalafil (P＜0.05).Conclusion Genistein has a certain effect on monocrotaline-induced PAH in rat;although its efficacy is less than that of bosentan and tadalafil,genistein is cheaper,easy to obtain and has less side effects,and is also expected to be a new auxiliary drug for PAH.
Betaine Attenuates Monocrotaline-Induced Pulmonary Arterial Hypertension in Rats via Inhibiting Inflammatory Response

Abstract:BACKGROUND: Pulmonary arterial hypertension (PAH) is characterized by increased pulmonary vascular resistance, leading to right ventricular failure and death. Recent studies have suggested that chronic inflammatory processes are involved in the pathogenesis of PAH. Several studies have demonstrated that betaine possesses outstanding anti-inflammatory effects. However, whether betaine exerts protective effects on PAH by inhibiting inflammatory responses in the lungs needs to be explored. To test our hypothesis, we aimed to investigate the effects of betaine on monocrotaline-induced PAH in rats and attempted to further clarify the possible mechanisms.METHODS: PAH was induced by monocrotaline (50 mg/kg) and oral administration of betaine (100, 200, and 400 mg/kg/day). The mean pulmonary arterial pressure, right ventricular systolic pressure, and right ventricle hypertrophy index were used to evaluate the development of PAH. Hematoxylin and eosin staining and Masson staining were performed to measure the extents of vascular remodeling and proliferation in fibrous tissue. Monocyte chemoattractant protein-1 (MCP-1) and endothelin-1 (ET-1) were also detected by immunohistochemical staining. Nuclear factor-κB (NF-κB), tumor necrosis factor alpha (TNF-α), and interleukin-1β (IL-1β) were assessed by Western blot.RESULTS: This study showed that betaine improved the abnormalities in right ventricular systolic pressure, mean pulmonary arterial pressure, right ventricle hypertrophy index, and pulmonary arterial remodeling induced by monocrotaline compared with the PAH group. The levels of MCP-1 and ET-1 also decreased. Western blot indicated that the protein expression levels of NF-κB, TNF-α, and IL-1β significantly decreased (<i>p</i> < 0.01).CONCLUSION: Our study demonstrated that betaine attenuated PAH through its anti-inflammatory effects. Hence, the present data may offer novel targets and promising pharmacological perspectives for treating monocrotaline-induced PAH.

Abstract:Objective To explore the effects of aspirin on moncrotaline-induced pulmonary arterial hypertension in rats.Methods 120 Sprague-Dawley rats were randomly divided into six groups with each for 20:normal control group (Ctrl group),untreated pulmonary arterial hypertension group (PAH group) and various aspirin-dosage treated pulmonary arterial hypertension group (ASA 0.5,1.0,2.0 and 4.0 mg·kg~(-1)·d~(-1) by intragastrically after monocrotaline injection from day 1 to day 30).The puhnonary arterial hypertension was induced by intraperitoneal injection of 50 mg/kg monocrotaline on day 0.On day 31,results were measured as following:(1)Survival rates and weights were measured; (2) Right heart catheter was performed to measure systolic Pulmonary Arterial Pressure (sPAP); (3)Heart specimens were separated and weighed to determine the Right Ventricular Hypertrophy Index (RVHI); (4)With pulmonary arterioles staining,Pulmonary Arterioles Thickness Index (PATI) were evaluated by IPP 6.0 image analysis software.Results In comparison with PAH group,weights in ASA 2 group and ASA 4 group were increased (P<0.05 respectively).Survival rates were significantly increased in overall ASA treatment group compared with PAH group (P<0.05 respectively). sPAPs and RVHIs in various ASA treatment group were significantly decreased than that in PAH group (P<0.05 respectively); PATIs in ASA treatment group,except ASA 0.5 group,were significantly decreased than that in PAH group (P<0.05 respectively).Conclusions Aspirin (ASA 1,ASA 2 and ASA 4) could reduce puhnonary arterial pressure, attenuates right ventricular hypertrophy and pulmonary arterioles proliferation in monocrotaline-induced pulmonary arterial hypertension model.

Abstract:This study was designed to investigate whether rosuvastatin could attenuate monocrotaline-induced pulmonary hypertension via regulation of Akt/eNOS signaling pathway and asymmetric dimethylarginine (ADMA) metabolism in rats. After a single-dose injection of monocrotaline (60mg/kg), oral administration of rosuvastatin (5mg/kg) was started from day 1 to day 28 (preventive administration) or from day 15 to day 28 (therapeutic administration), or with vehicle as corresponding controls. 28days after monocrotaline, significant pulmonary hypertension characterized by pulmonary arterial medial wall thickening, right ventricular hypertrophy and right heart failure was observed. Rosuvastatin (5mg/kg, for 14days and 28days) treatment significantly attenuated monocrotaline-induced pulmonary vascular remodeling, right ventricular hypertrophy and dysfunction, and normalized the down-regulated pulmonary Akt/p-Akt and eNOS/p-eNOS expressions, while increased DDAH2 expression accompanied by decreased serum level of ADMA. However expression of PRMT1 and GSK3β/p-GSK3β did not differ among all groups (all P>0.05). We concluded that rosuvastatin inhibits monocrotaline-induced pulmonary hypertension through normalization of Akt, eNOS and DDAH2 expressions, and decreasing the level of ADMA.
Effect and mechanism of nitrendipine on pulmonary hypertension induced by monocrotaline

Abstract:Objective: To explore the effect and mechanism of nitrendipine (NIT) on pulmonary hypertension (PH), adult male Sprague-Dawley rats were given a single subcutaneous injection of monocrotaline (MCT, 60 mg �� kg-1) to induce the mold of PH. Methods: Rats were given injection of NIT (10 mg �� kg-1) by ip 30 min before and once daily after MCT injection for 4 weeks. The parameters of pulmonary hemodynamics were monitored. Concentration of endothelin-like immunoreactivity (ir-ET), nitric oxide (NO), malondialdehyde (MDA) and activity of superoxide dismutase (SOD) in plasma and pulmonary homogenate measured by radioimmunoassay and colorimetric analysis. Results: Continuous injection of NIT significantly inhibited the progression of pulmonary artery pressure (3.6��0.5 versus 4.5��0.9 kPa) and pulmonary vascular resistance (79��18 versus 118��17 kPa �� min �� L-1). Histological examination revealed that NIT also effectively prevented pulmonary arterial medial thickening. NIT significantly raised both concentration of NO(16��7 versus 0.6��0.4 ��ol �� L-1) in pulmonary homogenate and activity of SOD (144��18 versus 106��45 NU �� ml-1) in plasma and depressed the increase of MDA concentration. Conclusions: Continuously using NIT can effectively prevent pulmonary hypertension induced by MCT of which mechanism may be partly related to the increase in activity of SOD and concentration of NO besides its inhibition of inflow of Ca2+.
All-TRANS RETINOIC ACID INTERFERES DEVELOPMENT OF PULMONARY HYPERTENSION INDUCED BY MONOCROTALINE IN RATS

Abstract:Objective To determine whether all-trans retinoic acid (atRA) affects the metabolism of collagen in main pulmonary artery and exerts an inhibitory effect in rats with pulmonary hypertension induced by monocrotaline . Methods All rats (n=72) were divided into 3 groups as control, model, and atRA . In model and atRA groups, rats (n=48) were assigned at random to be given a single subcutaneous injection of monocrotaline (60mgg/kg) and administrated with either atRA (30rng·kg-1·d-1) for atRA group or saline through oral-gastro intubation for model group. In control group, rats (n=24) received a single subcutaneous injection of an equal volume of 0. 9% saline. On day 7, 14,21 and 28 after monocrotaline or saline injection, cardiovascular catheters were inserted into the pulmonary artery of rats in each group to examine their mean pulmonary artery pressure, in addition with their hydroxyproline content determined by chromometry. Results In comparison with the control rats, the mean pulmonary artery pressure of rats in model group increased significantly on day 21 and up to the peak on day 28 (P<0.01), while their hydroxyproline contents decreased significantly on day 14 ( P < 0.05) and increased significantly on day 21 and 28. The atRA group when compared with the model group show reduction in the content of hydroxyproline and the mean pulmonary artery pressure ( P < 0.01 ). Conclusion The atRA inhibits the accumulation of collagen in main pulmonary artery and interferes the development of pulmonary hypertension which might elicit favorable geometric remodeling of rat pulmonary hypertension induced by monocrotaline.

Abstract:BACKGROUND:Ethyl pyruvate (EP) is an anti-inflammatory and anti-oxidant agent associated with many diseases. In this study, we evaluated whether EP could attenuate monocrotaline-induced pulmonary arterial hypertension (PAH). METHODS:A PAH model was established by subcutaneously injecting a single dose of monocrotaline (60 mg/kg). And then a daily intraperitoneal injection of EP (50 mg/kg) was administered on day 1 to day 28 (preventive EP treatment) or day 15 to day 28 (therapeutic EP treatment). Hemodynamic changes were measured by catheterization, and the right ventricle hypertrophy index, the medial wall thickness, and the medial wall areas were also calculated. Enzyme-linked immunosorbent assay and immunohistochemical analysis were used to determine the serum levels and expression of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and endothelin-1 (ET-1) in the lung tissue. RESULTS:Both preventive and therapeutic EP treatment significantly ameliorated hemodynamic changes and vascular remodeling indicators (all P < 0.05). The serum levels and expression of TNF-α, IL-6, and ET-1 in the lung tissue were also significantly decreased (all P < 0.05). CONCLUSIONS:EP ameliorates monocrotaline-induced PAH and reverses pulmonary vascular remolding in rats by inhibiting the release of TNF-α and IL-6 and reducing the expression of ET-1.

Abstract:BACKGROUND:Ginsenosides have been shown to exert beneficial pharmacological effects on the central nervous, cardiovascular, and endocrine systems. We sought to determine whether total ginsenosides (TG) inhibit monocrotaline (MCT)-induced pulmonary hypertension and to elucidate the underlying mechanism. METHODS:MCT-intoxicated rats were treated with gradient doses of TG, with or without N (G)-nitro-l-arginine methyl ester. The levels of molecules involving the regulation of nitric oxide and mitogen-activated protein kinase pathways were determined. RESULTS:TG ameliorated MCT-induced pulmonary hypertension in a dose-dependent manner, as assessed by the right ventricular systolic pressure, the right ventricular hypertrophy index, and pulmonary arterial remodeling. Furthermore, TG increased the levels of pulmonary nitric oxide, endothelial nitric oxide synthase, and cyclic guanosine monophosphate. Lastly, TG increased mitogen-activated protein kinase phosphatase-1 expression and promoted the dephosphorylation of extracellular signal-regulated protein kinases 1/2, p38 mitogen-activated protein kinase, and c-Jun NH2-terminal kinase 1/2. CONCLUSION:TG attenuates MCT-induced pulmonary hypertension, which may involve in part the regulation of nitric oxide and mitogen-activated protein kinase pathways.
Monocrotaline-induced Structural Remodeling of the Intra-Acinar Pulmonary Arteries and Pulmonary Hypertension

Abstract:The monocrofaline-induced structural changes of small pulmonary arteries in rat and their relationship with pulmonary hypertension and right ventricular hypertrophy were observed by determining the right ventricular systolic pressure, and by light and electron microscope and morphometry. One to 38 days after last injection of monocrotaline (MCT), a medial thickening and lumen marrrowing of the circular muscular arteries (CMA), accompanying terminal (TB) and respiratory bronchioles (RB), were found. And there after the lumen of CMA, accompanying TB, became dilated, and its medial thickness (MT) decreased, whereas the histopathologic changes of the partially muscular arteries (PMA), accompanying RB, became severe, their MT increased continuously, and finally reached the peak value on Day 50. At the first day after last MCT treatment, inflammation and muscularization were found in PMA and nonmuscular arteries (NMA), and became more severe with the cause of disease. Therefore, the intra-acinar pulmonary arteries, both CMA and PMA, increased in number while the NMA decreased in number significantly because of the structural remodeling. Four days after MCT treatment, the right ventricular systolic pressure began to rise, and reached its peak value on Day 50. Eight days after MCT injection, right ventricular hypertrophy developed, and became most significant from Day 23 to Day 30. The results suggest that structural remodeling, i.e. muscularization, of intra-acinar pulmonary arteries plays an important role in the development of pulmonary hypertension and right ventricular hypertrophy.
Effects of Lipoxin Receptor Agonist BML-111 on Pulmonary Arterial Hypertension of Rats Induced by Monocrotaline

Abstract:Objective To observe the the effects of BML-111 on pulmonary arterial hypertension rats induced by monocrotaline and its possible mechanisms.Methods Thirty SD rats were randomly divided into group C,group PH and group B.The rats in group PH and group B were given subcutaneous injection with monocrotaline at a dose of 50 μg/g,while group C were given normal saline with the same volume.The rats in group B were given intraperitoneal injection with BML-111 at a dose of 1 mg·kg-1 daily for 4 weeks.The levels of MPAP,RVSP,RV/(LV+S),the concentrations of IL-8,ICAM-1,MCP-1 of pulmonary tissues were mearsured.Results The levels of MPAP,RVSP RV/(LV+S) and the concentrations of IL-8,ICAM-1,MCP-1 in group PH and group B were significantly higher than those of group C(P0.05),meanwhile,the above indexes of group B were significantly higher than those of group PH(P0.05).Conclusion BML-111 can ameliorate pulmonary arterial hypertension of rats induced by monocrotaline,wihch may relate to its reducing the inflammation of pulmonary tissues.
Tetrandrine Blocks Cardiac Hypertrophy by Disrupting Reactive Oxygen Species-Dependent Erk1/2 Signalling

Abstract:Background and purpose: Tetrandrine, a well‐known naturally occurring calcium antagonist with anti‐inflammatory, antioxidant and anti‐fibrogenetic activities, has long been used clinically for treatment of cardiovascular diseases such as hypertension and arrhythmia. However, little is known about the effect of tetrandrine on cardiac hypertrophy. The aims of the present study were to determine whether tetrandrine could attenuate cardiac hypertrophy and to clarify the underlying molecular mechanisms.Experimental approach: Tetrandrine (50 mg·kg−1·day−1) was administered by oral gavage three times a day for one week and then the mice were subjected to either chronic pressure overload generated by aortic banding (AB) or sham surgery (control group). Cardiac function was determined by echocardiography.Key results: Tetrandrine attenuated the cardiac hypertrophy induced by AB, as assessed by heart weight/body weight and lung weight/body weight ratios, cardiac dilatation and the expression of genes of hypertrophic markers. Tetrandrine also inhibited fibrosis and attenuated the inflammatory response. The cardioprotective effects of tetrandrine were mediated by blocking the increased production of reactive oxygen species and the activation of ERK1/2‐dependent nuclear factor‐κB and nuclear factor of activated T cells that occur in response to hypertrophic stimuli.Conclusions and implications: Taken together, our results suggest that tetrandrine can improve cardiac function and prevent the development of cardiac hypertrophy by suppressing the reactive oxygen species‐dependent ERK1/2 signalling pathway.
Polydatin Glycosides Improve Monocrotaline-Induced Pulmonary Hypertension Injury by Inhibiting Endothelial-To-Mesenchymal Transition

Abstract:Objective: To study the effect of polydatin on the injury of pulmonary arterial hypertension (PAH) induced by monocrotaline (MCT). Methods: SD rats were induced to develop PAH injury by a single subcutaneous injection of MCT (60 mg/kg). From the second day, rats in the administration group were orally given sildenafil (20 mg/kg) and polydatin (30 or 60 mg/kg) for 3 weeks. At the end of the experiment, right ventricular hypertrophy (RVH) index of SD rats was calculated, pathological damage was assessed by HE staining, transcription levels of target genes were detected by RT-PCR and Elisa, and expression levels of Endothelial-to-mesenchymal transition (EndMT) related proteins were detected by immunohistochemistry (IHC) and immunofluorescence (IF). Finally, molecular docking analysis was used to verify the interaction of polydatin on the main targets. Results: Polydatin could significantly restore the body function, reduce MCT-induced PAH injury, reduce serum biochemical indices; polydatin could effectively inhibit EndMT process by decreasing the expression of N-cadherin, β-catenin and vimentin; polydatin could down-regulate TAGLN expression and increase PECAM1 expression to reduce pulmonary vascular remodeling. The interaction between polydatin and EndMT target was confirmed by molecular docking operation. Conclusion: Pharmacological experiments combined with Combining molecular docking was first used to clarify that polydatin can reduce the pulmonary endothelial dysfunction and pulmonary vascular remodeling induced by MCT by inhibiting EndMT. The results of the study provide new ideas for the further treatment of PAH injury.

Effects of T-type Calcium Channel Inhibitors on Monocrotaline-Induced Pulmonary Arterial Hypertension in Rats

Abstract:Objective To investigate the effects of T-type calcium channel inhibitors (ProTx-1,micromolar Ni2+ and Mibefradil) on Monocrotaline (MCT)-induced pulmonary arterial hypertension (PAH) in rats.Methods Forty male Sprague-Dawley rats were randomly divided into 5 groups:normal control group,MCT group,ProTx-1 group,micromolar Ni2+ group and Mibefradil group (8 cases in each group).The right ventricular systolic pressure (RVSP),the right ventricular hypertrophy index (RVHI),and the index of pulmonary vascular remodeling(MA％) were measured on day 28 after MCT-treatment.Western blot was used to detect the expression of proliferating cell nuclear antigen(PCNA) and Cleaved Caspase-3 in pulmonary artery.Results (1)RVSP and RVHI in MCT group were significandy higher than those in the other 4 groups (F =21.55,P ＜ 0.01;F =15.63,P ＜ 0.01).The two indexes in 3 intervention groups were higher than those in normal control group (all P ＜ 0.05),nevertheless,significantly lower than those in MCT group,and 3 intervention groups showed no significant differences (all P ＞ 0.05).(2) MA％ in normal control group [(23.43 ± 1.95) ％] was lower than that in MCT group [(80.42 ± 4.30) ％],ProTx-1 group [(60.35 ± 3.83)％],micromolar Ni2+ group[(62.44 ± 3.81)％] and Mibefradil group[(58.66 ± 4.23)％] (F =216.2,P ＜ 0.01);3 intervention groups showed no significant differences (all P ＞ 0.05),however,they were all significantly lower than that in MCT group.(3) The expression of PCNA in MCT group was higher than that in normal control group,meanwhile,3 intervention groups were significantly lower than that in MCT group.The expression of Cleaved Caspase-3 in MCT group was higher than that in normal control group,nevertheless,3 intervention groups showed no significant changes compared with MCT group,respectively.Conclusions T-type calcium channel inhibitors could ameliorate the progression of MCT-PAH in rats,mainly through suppressing the proliferation of pulmonary artery smooth muscle cells.
Effect of Tetrandrine on Endothelin Expression in the Lungs and Clinical Significance Thereof:Experiment with Rat Models with Nitrofen-Induced Congenital Diaphragmatic Hernia

Abstract:OBJECTIVE:To investigate the effects of tetrandrine on endothelin expression in the lungs and its clinical significance.METHODS:25 pregnant female SD rats were randomly divided into 5 equal groups: Groups A to D were fed with nitrofen to cause congenital diaphragmatic hernia (CDH) in the fetuses. Group A was injected with normal saline, Group B with dexamethasone (DXM), Group C with tetrandrine, and Group D with DXM + tetrandrine. Group E was control Group. On day 21.5 of pregnancy the fetuses were delivered by cesarean section and killed. Microscopy was used to observe the CDH formation, and the relative wall thickness (RWA) and relative wall area (RWA) of pulmonary arterioles. The lung/body weight ratio, and relative integrated optical density (IOD) of pulmonary arteriole and bronchiole were observed. The expression of endothelin in the lung tissues was detected by immunohistochemistry.RESULTS:9 rats of Group A-D produced 57 fetuses with CDH with a CDH arte of 45.2%. The lung/body weight ratios, and RWA values of Group A-D were all significantly lower than that of Group E (all P < 0.05). The RWT of pulmonary arteriole was significantly lower in Groups B and C compared with Group E (both P < 0.05). The RWT and RWA of Group A were significantly lower than those of Group B-D (all P < 0.05). The values of relative IOD of pulmonary tissues and of pulmonary arteriole of Group A-D were all significantly lower than those of Group E (all P < 0.05). A positive correlation existed between the relative IOD of endothelin in pulmonary arteriole and in bronchiole (P < 0.01), and among the RWT and RWA of pulmonary arteriole, and relative IOD (all P < 0.01).CONCLUSION:Tetrandrine improves the pulmonary hypoplasia and degrades the pulmonary hypertension.
Preventive effect of sildenafil on right ventricular function in rats with monocrotaline-induced pulmonary arterial hypertension

Abstract:The present study aimed to evaluate the preventive effect of sildenafil treatment on pulmonary hypertension (PH) induced by monocrotaline (MCT) in rats. Fifty-four 12-week-old male Sprague-Dawley rats were injected with MCT or saline solution (MCT-injected rats: n=36; saline: n=18). Serial echocardiography and right ventricular systolic pressure (RVSP) measurements via a cardiac catheter were performed at 2, 4 and 6 weeks after the injection. After injection of MCT, rats received oral sildenafil (MCT/sildenafil group: n=18) or no treatment (MCT group: n=18) until undergoing echocardiography and cardiac catheterization. RVSP in the MCT/sildenafil group was lower than that in the MCT group at 4 (P<0.001) and 6 weeks (P<0.001). The septal curvature was improved in the MCT/sildenafil group compared with the MCT group. This finding showed that sildenafil prevented flattening of the interventricular septum because of right ventricular pressure overload. The ratio of peak trans-tricuspid early diastolic wave velocity to active filling with atrial systolic velocity showed that sildenafil improved diastolic function. Tricuspid annular plane systolic excursion and tricuspid annular systolic velocity in the MCT/sildenafil group did not show preserved myocardial contraction after administration of sildenafil. Administration of sildenafil leads to a reduction in RVSP and improvement in cardiac function in rats with PH induced by MCT. The vasodilatory action of sildenafil improves right ventricular diastolic function, but the intrinsic, positive, inotropic effect of sildenafil is minimal.
