Abstract:Objective:To evaluate the relationship of expression of p53,p21,Ki-67,VEGF and bladder cancer stage,grade and prognosis.Methods:The expressions of p53,p21,Ki-67 and VEGF in forty specimens from 40 patients of bladder cancer were studied by means of immunohistochemistry.The immunostaining results were analyzed in relation to the tumor stage,grade and compared with the clinical parameters of prognosis.Results:Changes in expression of p53,p21,Ki-67 and VEGF were noted in 31(77.5%),22(55%),16(40%)and 17(42.5%)tumors,respectively.Thirty-five patients(87.5%)had at least one marker altered,and 7 patients(17.5%)had all four altered.The patients were followed-up for 51 months by median.Abnormal expressions of p53,p21,Ki-67 and VEGF were found associated with bladder cancer stage and grade.Multivariate analysis indicates that the combined abnormal expressions of four markers or p53(+)/p21(-)provided prognostic information in bladder cancer(P<0.01,P<0.05,respectively).Ki-67 or VEGF wasn't independently associated with prognostic information in bladder cancer.The incremental number of altered markers was independently associated with an increased risk of bladder cancer mortality.Conclusions:Abnormal expressions of p53,p21,Ki-67 and VEGF expression were almost associated with bladder cancer stage and grade.The combined evaluation of p53,p21,Ki-67 and VEGF is useful in prognosis of bladder cancer.
Expression of VEGF and Its Correlation with Prognosis of Small Cell Lung Cancer

Abstract:[Purpose] To investigate the expression of vascular endothelial growth factor(VEGF)and its correlation with prognosis of small cell lung cancer(SCLC).[Methods] Immunohistochemistry technology was used to detect the expression of VEGF protein in 35 cases with SCLC who received surgical treatment from 1998 to 2010,and its correlation with prognosis of SCLC was evaluated. [Results] VEGF expression rate was 45.7% in all SCLC patients and all samples were mild expression.There was no difference in median survival time(MST) between positive and negative VEGF expressions(13 months vs 20 months,P=0.784) in 22 followed-up cases.[Conclusion] Νear half SCLC patients show VEGF mild expression.VEGF expression does not correlate with prognosis. The strategy of antiangiogenesis in SCLC needs to be further studied.

Abstract:Objective To study the vascular endothelial growth factor-C(VEGF-C) and its receptor(VEGFR-3) and their relationship with clinical pathological factors of gastric cancer.Methods Expression of VEGF-C in 20 cases of normal gas-tric tissues and 98 cases of gastric cancer tissues were detected by immunohistochemical staining.Results VEGF-C positive ex-pression rate was 71.4%(70/98) in gastric cancer tissues,and VEGF-C positive expression rates in tissues adjacent to tumors and normal gastric mucosa tissues were 9.2%(9/98) and 5.0%(1/20).The expression of VEGF-C in 98 cases of gastric cancer tissues and patient's gender,tumor diameter,size,location,degree of differentiation,presence of vascular invasion and infiltration depth had no significant correlation,but were significantly correlated with lymph node metastasis,lymphatic vessel invasion,distant metastasis,and TNM clinical stage (P<0.05).Conclusion VEGF-C and its receptor system is an important regulatory factor of lymphangiogenesis,it has very important significance in lymphangiogenesis of gastric cancer region and lymphatic metastasis.

Abstract:Objective To investigate the features of VEGF expression in renal pelvic carcinoma and its correlation with MVD and its clinical significance. Methods The expressions of VEGF and CD34 in the cancerous tissues were detected in 26 patients by immunohistochemistry and MVD was assessed. The relation of VEGF with MVD, that with the clinical stages, and the relation of MVD with the clinical stages were analyzed respectively. Results VEGF was highly expressed in 18 patients and moderately expressed in 8 patients and the expression level was positively related with the clinical stages. There were no significant differences between the MVD value and both the VEGF expression levels and the clinical stages. Conclusions VEGF expression is increased with the advancing stages but MVD dose not differ significantly in difference stages. MVD in the advanced stages does not enhance with the increasing VEGF expression , which may due to an excessive exudation of plasma proteins caused by the elevation of the vascular permeability.
Expression of Vascular Endothelial Growth Factor-C and Its Receptor VEGFR-3 in Cervical Squamous Cell Carcinoma and Its Clinical Significance

Abstract:Objective To investigate the expression of vascular endothelial growth factor-C and its receptor VEGFR-3 in cervical squamous cell carcinomas and its clinical implication.Methods The detection of expression of VEGF-C and its receptor VEGFR-3 was immunohistochemically performed in 10 cases of normal cervical tissue,12 of CIN Ⅰ,12 of CIN Ⅱ,20 of carcinoma in situ and 66 of cervical invasive squamous cell carcinomas. Results The positive expressive rate of VEGF-C was 75.8% in cervical invasive squamous cell carcinomas,while that of VEGF-C expression was 30.0%,41.7%,41.7% and 45.0% in normal cervical tissue,CIN Ⅰ,CIN Ⅱ and carcinoma in situ,respectively.The difference between the invasive squamous cell carcinoma and the normal cervical tissue in VEGF-C expression was statistically significant(P0.01).The expression level of VEGF-C in cervical squamous carcinoma was positively correlated with the positive number of stromal lymphatic vessels stained by VEGFR-3.Enhanced expressions of VEGF-C in cervical squamous cancer cells and stromal VEGFR-3-positive lymphatic proliferation were both positively associated with poor clinical stages and regional lymph node metastasis.Conclusions The enhanced expression of VEGF-C and proliferation of stromal lymphangiogenesis play an important role in the tumoriogenesis and progression of cervical squamous cell carcinoma,and VEGF-C and VEGFR-3 might be valuable markers for predicting the infiltrating potential and lymph node metastasis of cervical squamous cell carcinoma.
Correlation of the Expression of VEGF-C with CEA and Survivin and Their Correlation in Rectal Cancer

Abstract:OBJECTIVE: To investigate the correlation of the expression of VEGF-C with CEA and Survivin in rectal cancer,and their values for determining the clinicopathological characteristics and prognosis of rectal cancer.METHODS: Immunohistochemical SP stainingwasused to detect the expressions of VEGF-C,CEA and Survivin proteins in 82 rectal cancer tissues and 30 adjacent normal tissues.Their relationship with the pathological characteristics of rectal cancer was analyzed.The correlation of abnormal expressions of VEGF-C and CEA and Survivin was analyzed.RESULTS: In rectal cancer tissues,the positive rate of VEGF-C,CEA and Survivin protein were 45.1%,56.1% and 47.6%,respectively.In tissues adjacent to cancer the positive rates of VEGF-C,CEA and Survivin expression were 10.5%,14.0% and 5.0%,respectively.The expression of VEGF-C had positive correlation with expression of CEA(r=0.287,P=0.024) and Survivin(r=0.320,P=0.018).The expression of VEGF-C,CEA and Survivin was associated with TNM staging(P0.05).However,they all no correlated with ages,gender,tumor size and pathological classification(P0.05).CONCLUSIONS: VEGF-C and CEA,Survivin is highly expressed in rectal cancer patients,and they are all corelated with cancer invasion and metastasis.The three joint detection can be used for rectal cancer stage of disease extent and prognosis of reference.The prognosis of rectal cancer may have some clinical significance.

Abstract:Objective To investigate the expression of vascular endothelial growth factor(VEGF) and kinase domain containing receptor(KDR) in human prostate cancer(PCa) and their relationship with Gleason Score.Methods The expression of VEGF and KDR were dectected by immunohistochemistry method in 20 Pca,15 benign prostatic hyperplasia(BPH) and 10 normal prostate(NP).The PCa was divided into high Gleason score group(Gleason score≥7,n=12) and low Gleason score group(Gleason score7,n=8).Results The positive rate of VEGF and KDR in PCa were significantly higher than BPH and NP(P0.05),and the positive rate of VEGF and KDR were significantly higher in high Gleason score group than low Gleason score group(VEGF:83.3% vs 37.5%,KDR:91.7% vs 62.5%,P0.05).Conclusion The high expression of VEGF and KDR in PCa may be related to tumor pathological gleason score.

Abstract:Objective To evaluate the expression of vascular endothelial growth factor (VEGF)-C in colorectal carci- noma (CRC) and to elucidate its relationship to the clinicopathological features and prognosis in patients with CRC. Methods From Jan 1996 to Jan 1998,69 patients with pathologically confirmed CRC who received curative surgery and routine follow-up were included in this study.The expression of VEGF-C protein in relation to mierovessel density (MVD) in 69 surgical specimens of CRC and in 20 normal colorectal tissues were examined by the EnVision method of immuno- histochemistry (IHC).Results The VEGF-C protein expression was detected in the cytoplasm of 94． 2%(65/69) of the CRC specimens and 30%(6/20) of the normal specimens.The VEGF-C expression level was much higher in the tumor tis- sue than that in the corresponding normal tissue (P＜0． 01) .The VEGF-C expression level was correlated significantly with the lymphatic invasion (P=0． 011) ,MDV and clinical staging of CRC (P=0． 006) .No significant correlation was found be- tween VEGF-C expression and patients' age,gender,depth of invasion,metastasis,tumor differentiation and site of the tumor.Patients with high expression of VEGF-C had shorter overall survival and disease-free survival.Conclusions The VEGf-C expression is up-regulated in CRC and higher expression level of VEGF-C might predict a poorer prognosis in patients with CRC.
Increased expressions of VEGF and VEGF-C in prostate cancer tissue are associated with tumor progression

Abstract:Objective To investigate the expressions of VEGF and VEGF-C between PCa tissues and adjacent benign tissues, and explore the roles of VEGF and VEGF-C in the progression process of PCa. Methods Seventy-one primary prostatic adenocarcinoma cases were included in this study and an immunohistochemical approach was adopted to detect the expressions of CD34, VEGF and VEGF-C in each sample. Then a statistic analysis was performed according to the experimental data. Results Significantly up-regulated expressions of VEGF and VEGF-C were both found in malignant epithelium/cancer cells compared with adjacent benign epithelium(P<0.01, respectively). Patients in stage D had a significantly higher score than patients in stage A, B or C when comparing VEGF-C expression in tumor area(P<0.01). In addition, significant correlations were observed between Jewett-Whitmore staging and VEGF-C(rs=0.738, P<0.001), and MVD and VEGF(rs=0.492, P<0.001). Conclusions Increased expressions of VEGF and VEGF-C were closely associated with progression of prostate cancer. The main contribution of VEGF for PCa progression was to stimulate angiogenesis, while the chief role of VEGF-C was to enhance lymphangiogenesis.

Abstract:Objective To investigate the expression of vascular endothelial growth factor receptor-3 (VEGFR-3) in tissues of non-small cell lung cancer.(NSCLC) and explore the relationship of VEGFR-3 to the different types, stage in tumor node metastasis (TNM) and prognosis of NSCLC.The expressions of VEGFR-3 in surgically resected tumor specimens collected from 50 cases with NSCLC and pulmonary inflammatory pseudotumor specimens from 20 cases served as controls were detected by immunohistochemical method.①The positive expression rate of VEGFR-3 in NSCLC (60%) was significantly higher than that in control group (10.0%)(P0.05). ②The elevation of positive VEGFR-3 expression accompanied the increase of TNM stage, there were statistical differences between I and Ⅲa stage(P0.05)as well as Ⅰ and Ⅲb stage of lung cancers(P0.05), also statistical difference was found between its Ⅱ and Ⅲb stage(P0.05). ③The NSCLC patients with positive expression of VEGFR-3 protein had a markedly shorter survival time than those with negative expression.[Conclusion]The expression of VEGFR-3 in tumor tissues of NSCLC is correlated with its TNM stage and prognosis. VEGFR-3 detection can be used as a criterion for evaluation of prognosis in NSCLC patients.
Correlation of VEGF-C and VEGFR-3 Expressions with Lymph Node Metastasis in Breast Cancer

Abstract:Objective To study the expressions of vascular endothelial growth factor C(VEGF-C) and vascular endothelial growth factor receptor 3(VEGFR-3) in breast cancer and their correlations with lymph node metastasis.Methods Immunohistochemical SP method was adopted in detecting VEGF-C and VEGFR-3 protein expressions in 60 cases of breast cancer and 15 cases of breast fibroadenoma as controls.Results The positive rate and degree of VEGF-C and VEGFR-3 expressions in breast cancer were significantly higher than those in the control group(P0.05).VEGF-C and VEGFR-3 were conspicuously higher in lymph-node-positive group than in lymph-node-negative group(P0.05).The expressions of VEGF-C and VEGFR-3 were positively correlated with axillary lymph node metastasis;VEGF-C expression was positively associated with VEGFR-3.However,there were no significant differences in pathologic features,tumor size and clinical stage.Conclusion The expressions of VEGF-C and VEGFR-3 were higher in breast cancer,and they promoted lymph node metastasis.

Abstract:Objective:To evaluate expression of HIF-1α and VEGF in prostate cancer, and study relationship between their expression and the pathological grade, clinical stage in prostate cancer.Methods:HIF-1α and VEGF were detected by immunohistochemistry method in tissues of prostate cancer and BPH.Results:HIF-1 and VEGF levels in prostate cancer were much higher than those in BPH; the degree of cancer malignant and invasion positively correlated with the expression of HIF-1α VEGF; HIF-1α level positively correlated with VEGF level.Conclusions:HIF-1α and VEGF are good molecular markers of prostate cancer which can be used to assist diagnosis and predict prognosis; HIF-1αis one of important regulator of VEGF.

Abstract:Objective:The aim of this study was to determine VEGFR-3 in BTCC and its correlation with clinical findings and the expression of VEGF-C. The prognostic value of VEGF-C and VEGFR-3 related to progression of BTCC will be also evaluated.Methods:We assessed the levels of expression of VEGF-C and VEGFR-3 by RT-PCR and immunohistochemistry in 36 samples of BTCC and 8 normal bladder tissues.Results:VEGFR-3 was stained in tumor cells. Immunoreactivity for VEGFR-3 became significantly stronger with progressive lesions in tumor. The expression level of VEGFR-3 corresponded with that of VEGF-C in carcinoma cells. The expression levels of VEGF-C and VEGFR-3 in carcinoma cells were significantly corelated to lymphatic invasion.Conclusions:The present results suggest that an autocrine loop of VEGF-C via VEGFR-3 accelerates cells lymphatic invasion. Therefore, it will raise the possibility of anticarcinoma therapy which specifically targets VEGF-C/ VEGFR-3 interaction in BTCC.

Abstract:OBJECTIVE:To investigate the expression and prognostic value of vascular endothelial growth factor receptor 3 (VEGFR-3) in human breast cancer.METHODS:A tissue microarray containing cores was constructed from 173 female patients with primary breast cancers (experimental group) and 19 female patients with benign breast lesions (control group) between March 2004 and June 2007. HE staining was performed to determine the quality of tissue microarray and immunohistochemistry staining was used to detect VEGFR-3 expression in the tissue microarray. Statistical analysis was performed to show the association between VEGFR-3 expression and clinicopathologic parameters.RESULTS:Histological examination showed that the tissue microarray had good quality, which could represent the histological characteristics of breast cancer and benign breast lesions. VEGFR-3 was expressed in 55.5% (96/173) breast cancer patients of experimental group, while the expression was negative in lesions of control group. VEGFR-3 expression rate had no significant relation with age, estrogen receptor, and progesterone receptor (P > 0.05). VEGFR-3 expression rate correlated positively with tumor size and pathological staging compared to VEGFR-3 negative tumors (P < 0.05). The positive expression rate of VEGFR-3 in breast cancer with lymph node metastasis was significantly higher than that without lymph node metastasis (P < 0.05). Furthermore, VEGFR-3 positive staining correlated with human EGF receptor 2 expression (P < 0.05).CONCLUSION:The expressions of VEGFR-3 in human breast cancer have close relations with lymph node metastasis. Thus, VEGFR-3 may be a potential prognostic indicator and a new anti-cancer target for breast cancer.

Abstract:Objective:To observe the relationship between VEGF and angiostatin in patients with carcinoma of prostate and benign hyperplasia of prostate.Methods:30 carcinoma of prostate tissue and 30 benign hyperplasia of prostate tissue were tested by using immunohistochemicaI and Western blotting technology.Results:The positive staining average grey value of angiostatin in carcinoma of prostate was increased significantly compared with that of benign hyperplasia of prostate,but the positive staining average grey value of VEGF in carcinoma of prostate was decreased compared with that of benign hyperplasia of prostate.The negative correlation was found between A S and VEGF (r=-0.560 P<0.05;).Conclusion:The level of angiostatin expression is reduced in carcinoma of prostate;however,the level of VEGF expression is increased significantly in carcinoma of prostate,and the negative correlation was found between AS and VEGF in carcinoma of prostate).AS and VEGF may effect development and growth mechanism of carcinoma of prostate in biology.
Expression of VEGF-C correlates with a poor prognosis based on analysis of prognostic factors in 73 patients with esophageal squamous cell carcinomas.

Abstract:Objective: The purpose of this study was to specifically investigate the clinicopathological role of expression of vascular endothelial growth factor-C (VEGF-C) as well as the correlation with clinical outcomes in esophageal squamous cell carcinomas (ESCCs). Methods: Seventy-three patients with ESCC resected in our institute were included in this study. Formalin-fixed paraffin-embedded specimens were stained for VEGF-C and the correlation between the staining, its clinicopathological parameters and its prognostic power were analyzed statistically. Results: Of the 73 ESCC patients studied, 39 cases (53.4%) were strongly positive for VEGF-C. Six cases (8.2%) were negative and 28 cases (38.4%) revealed unclear weak reactions. All 34 cases were included in the negative group (46.6%). VEGF-C expression correlated with histological grade (P = 0.005), depth of tumor invasion (pT) (P = 0.021), lymph node metastasis (pN) (P = 0.002) and lymphatic invasion (P = 0.008). The median overall survival of 39 patients who had positive staining for tumor cell VEGF-C and 34 patients who had negative staining were 10.4 months (95% CI, 6.9-13.9 months) and 28.5 months (95% CI, 12.6-44.4 months), respectively (P = 0.003). In univariate analysis by log-rank test, histological grade, pN, stage, lymphatic invasion and VEGF-C were significant prognostic factors (P=0.047, 0.007, 0.018, 0.002 and 0.003, respectively.). In multivariate analysis, high VEGF-C expression (P = 0.0451) maintained its independent prognostic influence on overall survival, as well as pN status (P = 0.0029). Conclusions: Expression of VEGF-C is related to histological grade, pT, pN and lymphatic invasion, and is a prognostic indicator for ESCC.

Abstract:Objective To detect signal transducer and activator of transcription 3(Stat3)and vascular endothelial growth factor(VEGF)expression and their roles in development and metastasis of gastric cancer.Methods Immunohistochemistry was used to detect the expression of VEGF and Stat3 in normal gastric tissue, gastric carcinoma,lymph node metastasis and VEGF expression of Stat3.Results The positive expression rate of Stat3 and VEGF in gastric carcinoma was significantly higher than that in control group[64.6％(42/65)vs 0,76.9％(50/65)vs 0,all P ＜ 0.05].The positive expression rate of Stat3 and VEGF was different in different histological types of gastric cancer; the positive expression rate of Stat3 and VEGF in undifferentiated carcinoma patients was 90.0％,90.0％,respectively.The positive expression rates of Stat3 and VEGF in different histological grade and the different clinical stages of gastric carcinoma were different. The positive expression rates of Stat3 and VEGF in poorly differentiated group and undifferentiated group was significantly higher than that in well-differentiated group and moderately differentiated group; Ⅲ + Ⅳ period [78.6％(22/28),92.9％(26/28)]was significantly higher than the phaseⅠ + Ⅱ [49.0％(24/49),67.4％(33/49)](P ＜ 0.05).Stat3 and VEGF expression were associated with lymph node metastasis(P ＜ 0.01).Conclusions Stat3 and VEGF are involved in gastric carcinogenesis,tumor metastasis and cancer prognosis.VEGF and stat3 are positively correlated with tumor transfer and invasion.
Expressions Correlation of P53,vegf,cox-2 and EGFR in Bladder Cancer and Their Clinical Significance

Abstract:Objective This study was designed to investigate the expressions of p53,VEGF,COX-2 and EGFR in bladder cancer tissues,and to explore the relationship between the expressions and clinicopathological parameters in patients with bladder cancer.Methods The expressions of p53,VEGF,COX-2 and EGFR in 163 cases of bladder cancer were assessed by immunohistochemistry,and their correlations with clinicopathological factors were statistically analyzed.Results The expressions of p53,VEGF and EGFR correlated with the clinical stage(P0.05),but not with the expression of COX-2(P0.05).In addition,the expression intensity of p53 and EGFR increased with higher clinical stage(P0.05).The expression rates and intensity of p53 were associated with pathologic grade(P0.05).The expression intensity of p53 had a positive correlation with the expression intensity of VEGF,COX-2 and EGFR(P0.05),and the expression intensity of COX-2 had a positive correlation with the expression intensity of VEGF and EGFR(P0.05).There was significant difference between single index expression(p53,VEGF,EGFR) and co-expression(p53 combined with VEGF,VEGF combined with EGFR) in bladder transitional cell carcinoma of different clinical stages.Conclusions The abnormal expression of p53,VEGF,COX-2 and EGFR might play a certain role in the course of the genesis and development of bladder transitional cell carcinoma.Combined detection of p53,VEGF and EGFR may help to confirm the clinical stage of bladder transitional cell carcinoma.
Meta-analysis of expression and clinical significance of VEGF-C and VEGFR-3 in gastric cancer patients

Abstract:Objective To reveal the expression pattern of VEGF-C and VEGFR-3 in Chinese gastric cancer patients by meta-analysis,with the hope of finding their relation with clinical features and prognosis for the purpose of directing individualized treatment.Methods All the studies on VEGF-C,VEGFR-3 and gastric cancer included in CNKI during years 2000 to 2009 were reviewed.According to certain criteria,raw data of qualified studies were collected for meta-analysis.Results(1)The expression rates of VEGF-C and VEGFR-3 in gastric cancer were 40.7%-73.6% and 35.4%-61.6% respectively,significantly higher than that in normal gastric mucosa.The elevated expression of VEGF-C correlated with high level of VEGFR3.(2)The high VEGF-C level was related with serosa infiltration,lymph node metastasis,late TNM staging and low 5-year survival rate,and was related to poor differentiation in certain sub-groups.(3) The high VEGFR-3 level was related with serosa infiltration,lymph node metastasis and poor differentiation,while no relation with clinical staging and 5-year survival rate.(4) Age,gender,location of cancer and Bowman′s type were irrelevant to VEGF-C and VEGFR-3 expression in gastric cancer.Conclusions The expression of VEGF-C and VEGFR-3 reflects biological features of Chinese gastric cancer,and has promising prognostic and therapeutic value.
Correlation of CXCR4 and VEGF-C expressions with lymph node metastasis in gastric cancer

Abstract:Objective:To investigate the relationship of the expressions of chemokine CXC receptor 4(CXCR4) and vascular endothelial growth factor C(VEGF-C) with lymph node metastasis in gastric cancer.Methods:Immuohistochemistry was used to detect the expressions of CXCR4 and VEGF-C in 86 samples of human gastric cancer.The relationship between CXCR4 and VEGF-C expressions and the clinical pathological diameters was statistically analyzed.Results:The total positive rate of CXCR4 expression in these samples was 66.28%,and it was higher in the cases with lymph node metastasis than that without lymph node metastasis(83.33% vs 23.26%,P<0.01).Meanwhile,the total positive rate of VEGF-C expression was 73.26%,and was higher in the cases with lymph node metastasis than that without lymph node metastasis(91.67% vs 5.81%,P<0.01).The positive expression of CXCR4 was consistent with the positive expression of VEGF-C(γ=0.498,P<0.05),and the expression levels of CXCR4 and VEGF-C were closely correlated with histological type,cellular differentiation,invasion depth and lymph node metastasis(P<0.01～0.05).Conclusion:CXCR4 and VEGF-C expressions may become predictors for the lymphatic metastasis of gastric cancer.
