Abstract:AIM: To investigate the effect of interleukin-10 (IL-10) gene on experimental autoimmune thyroiditis mice. METHODS: Mice were immunized to induce autoimmune thyroiditis with porcine thyroglobulin (pTg), and thyroids of mice were injected with IL-10 DNA. On d 28 after immunization with pTg, mRNA expression of IL-10 in thyroid glands was detected and thyroid specimens were histopathological studied. RESULTS: The mRNA expression of IL-10 was detected in thyroid glands on d 7 and 14 after injection of IL-10 plasmid DNA or on COS-7 cells 48 h after IL-10 plasmid DNA transfection. In addition, hIL-10 levels in culture media significantly increased 48 h and 72 h after IL-10 plasmid DNA transfection. Infiltration index of lymphocytes (1.1 +/- 0.4) in thyroids of IL-10-treated mice was significantly lower than that of pcDNA3-null-treated mice (2.2 +/- 0.5) (P<0.01). Compared with pcDNA3-null control mice, IL-10-treated mice had lower levels of serum IFN-gamma (P<0.01). CONCLUSION: The direct injection of DNA expression vectors encoding IL-10 into thyroid significantly inhibited development of lymphocytic infiltration of thyroid of autoimmune thyroiditis mice, and alleviated the progression of this disease.

Abstract:Background and aims: We designed this study to evaluate the ability of a plasmid carrying an RU486 regulatory system to induce expression of interleukin-2 (IL-2) gene and to examine the antitumour efficacy of the induced IL-2 gene.Methods: The plasmid pRS-mIL-2,which contains an RU486 inducible system and IL-2 gene was injected into mice. Sera and tissues from liver, spleen, lungs and kidneys were taken to test the properties of the plasmid. To examine the antitumour efficacy of pRS-mIL-2, tumours were established in the liver by direct inoculation of H22 hepatoma cells.Results: The IL-2 levels in serum correlated with the dose of plasmid and RU486. High and sustained IL-2 levels could be achieved by administration of RU486 every day. The mRNA of transgene IL-2 was found only in the liver. Treatment of mice with pRS-mIL-2 plus RU486 resulted in the significant reduction in tumour volume compared with control groups.Conclusions: Tight temporal and spatial control of transgene IL-2 expression can be achieved by a plasmid containing an RU486 inducible system driven by liver specific promoter. pRS-mIL-2 exhibited strong antitumour efficacy following consecutive induction with RU486. Abstract:Hepatocellular carcinoma (HCC) and solid cancers with liver metastases are indications with high unmet medical need. Interleukin-12 (IL-12) is a proinflammatory cytokine with substantial anti-tumor properties, but its therapeutic potential has not been realized due to severe toxicity. Here, we show that orthotopic liver tumors in mice can be treated by targeting hepatocytes via systemic delivery of adeno-associated virus (AAV) vectors carrying the murine IL-12 gene. Controlled cytokine production was achieved in vivo by using the tetracycline-inducible K19 riboswitch. AAV-mediated expression of IL-12 led to STAT4 phosphorylation, interferon-γ (IFNγ) production, infiltration of T cells and, ultimately, tumor regression. By detailed analyses of efficacy and tolerability in healthy and tumor-bearing animals, we could define a safe and efficacious vector dose. As a potential clinical candidate, we characterized vectors carrying the human IL-12 (huIL-12) gene. In mice, bioactive human IL-12 was expressed in a vector dose-dependent manner and could be induced by tetracycline, suggesting tissue-specific AAV vectors with riboswitch-controlled expression of highly potent proinflammatory cytokines as an attractive approach for vector-based cancer immunotherapy.

Abstract:Objective To investigate the antitumor effects of intrasplenically transplanted interleukin-18 (IL-18) gene-modified hepatocytes on murine implanted liver carcinoma.Methods Embryonic murine hepatocyte cell line (BNL-CL2) was transfected with a recombinant adenovirus encoding IL-18 and used as delivery cells for IL-18 gene transfer. Two cell lines, BNL-LacZ and BNL-CL2, were used as controls. One week after intrasplenic injection of C26 cells ( colon carcinoma line), tumor-bearing syngeneic mice underwent the intrasplenic transplantation of IL-18 gene-modified hepatocyte cell line and were divided into treatment group (BNL IL-18) and control groups ( BNL-LacZ and BNL-CL2). Two weeks later, the serum levels of IL-18, interferon-gamma (IFN-gamma), tumor necrosis factor-alpha (TNF-alpha) and nitric oxide (NO) in the implanted liver carcinoma-bearing mice were assayed, the cytotoxicity of murine splenic cytotoxic T-lymphocytes (CTLs) was measured, and the morphology of the hepatic tumors was studied to evaluate the antitumor effects of the approach.Results In the treatment group, the serum levels of IL-18, IFN-gamma, TNF-alpha and NO increased significantly. The splenic CTL activity increased markedly ( P < 0.01), accompanied by a substantial decrease in tumor volume and the percentage of tumor area and prolonged survival of liver carcinomo-being mice.Conclusions In vivo IL-18 expression by ex vivo manipulated cells with IL-18 recombinant adenovirus is able to exert potent antitumor effects by inducing a predominantly T-cell-helper type 1 (Th1) immune response. Intrasplenic transplantation of adenovirus-mediated IL-18 gene-modified hepatocytes could be used as a targeting treatment for implanted liver carcinoma.
Treatment of Rat Hepatoma by Locally Injection of Murine IL-12 Retrovirus Packaging Cell

Abstract:Objective: To investigate the therapeutic effects of the murine IL-12 (mIL-12) retrovirus packaging cell line on hepatoma injected locally. Methods: The retrovirus vector encoding mIL-12 gene was constructed and transfected into packaging cell line PA317. The cells were then used to treat the rats with experimental orthotopic hepatoma at different time. The therapeutic effects, immune functions of the hosts, pathological and toxicological responses were documented. Results: the results showed that the mIL-12 retrovirus packaging cell line could significantly inhibit the growth of the hepatoma cells injected locally to the hepatoma. The early treatment made the rats survive long, while the medium or late stage treatment could prolong the life time of the rats compared with the bland control group or bland vector control group, though the rats did not survive. The number of NK cells and T cells increased significantly in the treatment group. The effects of the early treatment were superior to those of the medium and late stage treatment. Moreover, the transfection of IL-12 gene locally in the hepatoma tissue could make the hepatoma disappear from other liver lobe. This phenomenon demonstrated that IL-12 could activate the immune cells of the host to kill the untransfected tumor cells. This is very important for IL-12 to be used in gene therapy clinically. Meanwhile, the hepatoma would not recur in the rats that had survived more than 2 months from the early treatment after being re-challenged with tumor cells. Conclusion: the results showed that IL-12 gene injected locally in the hepatoma tissue could enhance the anti-tumor immunity of the host.
Inhibition of metastatic liver tumor with intravascular IL-12 gene therapy

Abstract:OBJECTIVE To explore the treatment and the suppression of hepatic metastases efficacy of intravascular administration of plasmid DNA expressing mIL-12.METHODSTo establish tumor models,the M5076 cells were injected into peritoneal cavity or inoculated into the footpad of mice.The hydrodynamics-based transfection was perfomed for transfer pGEG.mIL-12 into C57BL/6 mice.The different survival rate and the number of matastatic foci were observed in experimental and control group animals.The expression of IL-12 and IFN-γ were assayed by ELISA.The activity of NK cells and lymphocyte proliferation from the spleen cells were measured by 51Cr release assay.RESULTSCompared with mice treated with saline or untreated,the significant elevation of the serum concentration of IL-12 and IFN-γ were detected in mice treated with pGEG.mIL-12.The pGEG.mIL-12 transfection drastically suppressed the subcutaneous as well as hepatic metastatic tumors,resulting in significant prolongation of survival period of the animals.Growth of tumor in treated with pGEG.mIL-12 group was significantly inhibited as compared with that in other groups.Transfection with pGEG.mIL-12 also suppressed peritoneal carcinomatosis and prolonged survival time in mice that had been injected with M5076 cells into the peritoneal cavity.The M5076 cells were resistant to CTL and NK cell cytotoxicity.CONCLUSIONThe high level of IL-12 production elicited by the hydrodynamics-based trasfection of the cytokine gene may be quite effective in inhibiting matastatic and CTL-unsusceptible tumor cells.

Abstract:To investigate the feasibility and efficacy of liver gene therapy mediated by intrasplenic transplantation of genetically modified hepatocytes, the normal mouse liver cell line BNL CL. 2 cells were introduced with Neo-resistant (NeoR) gene or interleukin-2 (IL-2) genein vitro, and transplanted intrasplenically into normal syngeneic mice (2 x 106 cell/mouse); subsequently, the expressions of the introduced genesin vim were detected. The RT-PCR results showed that NeoR mRNA expressions were detectable in livers 24 h after transplantation and lasted over 11 weeks. Moreover, The NeoR mRNA was detected to be expressed temporarily in spleens (24 h–1 week) and lungs (24–96 h) after transplantation. After intrasplenic transplantation of IL-2 gene-modified BNL CL.2 cells, the stable expressions of IL-2 mRNA in the livers of transplanted mice were detectable by RT-PCR (24 h–11 weeks), and certain levels of IL-2 (5–40 pg/mL) remained in the peripheral blood. When IL-2 gene-modified BNL CL. 2 cells were transplanted intrasplenically to treat the metastatic liver colon carcinoma-bearing mice, the survival time of the treated mice was significantly prolonged. The data indicate that intrasplenic transplantation of genetically modified hepatocytes could allow for oriental distribution in host livers and long-term survival of the transplanted liver cells, and effective expression of exogenous genesin vim, suggesting that this can be a candidate approach to liver-directed gene therapy.
Experimental Study of Interleukin 12 Gene Therapy on Colon Cancer

Abstract:Objective To investigate the effects of interleukin 12 gene transfer on tumor growth, survival rate of tumor bearing mice. Methods Tumor bearing murine models were established with colonic cancer cells and PA317 mIL 12 carrying mIL 12 was constructed. Chemotherapy, radioimmunological treatment(RIT),gene treatment(GT) and combined RIT and GT were given in each group.Tumor volume and weight were measured, and survival rates were analyzed to compare their anti tumor effects. Results The packaging cell system(PA317 mIL 12) carrying pGCXEXPN mIL 12 expressed mIL 12. The concentration of mIL 12 in the supernatants reached 27 ng/106 cells after 48 hours.After treating with different schemes for 3 weeks, the tumor volume and tumor weight were significantly lower, and the survival rates of tumor bearing mice were significantly higher in GT group and combined GT and RIT group than those in chemotherapy and RIT groups. Conclusions Direct intra tumor injection of PA317 mIL 12 can effectively inhibit the growth of colon cancer. IL 12 gene transfer can also prolong the survival time of tumor bearing mice.
Study on IL-12 eukaryotic expression vector inhibiting the growth of transplanted liver cancer by two different methods

Abstract:Objective:To compare with the effect of two different methods mediate IL-12 recombinant expression plasmids inhibiting the growth of transplanted liver cancer.Methods:Mouse hepatoma cell line H22 was transplanted subcutaneously into mice,complex of IL-12 plasmids and lipofectin were injected into abdominal cavity of mice or intratumor,one time every week,two times totally.After administration,the sizes of tumor were messured two times every week.The concentration of IL-12 and IL-2 in serum were detected by ELISA,the cytotoxity activity of NK cells were detected by MTT.Results:The sizes of tumor in the group of direct intratumor injection were significantly smaller than that in the group of abdomen cavity injection(P0.05).The concentration of IL-12 and IL-2 in serum had obviously periodic variation,but the cytotoxity activity of NK cells hadn\u0027t obvious variant between two groups.Conclusion:Complex of IL-12 plasmids and lipofectin injected into intratumor directly can effectively inhibit the growth of transplanted liver cancer.
Combined Interleukin 12 and Interleukin 2 Gene Therapy for Hepatocellular Carcinoma in A Rat Model

Abstract:Objective:To determine the feasibility and efficacy of combined murine interleukin 12 (mIL-12) and human interleukin 2 ( hIL-2) gene therapy for hepatocellular carcinoma in a rat model. Methods; The retroviral vector encoding mIL-12/hIL-2 gene was constructed and then transfected into packaging cell line. The cells were injected into rats in the established hepatoma at different time points. The therapeutic effect, immune function and toxicological response were evaluated. Results:Intratumoral injection of recombinant retroviral vector encoding mIL-12/hIL-2 gene resulted in marked hepatoma regression. The 35 d survival rates of the subgroups treated on the first,third,fifth and seventh day after tumor implantation were 100% , 100% , 30% and 10% respectively. The average survival time of the IL-12 + IL-2 treatment group was superior to those of the physiological saline group (P 0. 01) , retroviral empty vector group(P0.01 ) , IL-2 treatment group(P 0. 01) and IL-12 treatment group( P 0. 01). The immunological study showed that the number of hepatoma infiltrating lymphocytes was increased in the IL-12 + IL-2 treatment group. Conclusion; The retroviral packaging cell line encoding mIL-12 and hIL-2 gene via intratumoral injection inhibits the growth of hepatocellular carcinoma significantly. The therapeutical effects of early administration is superior to that of later one.
[Treatment of Hepatocellular Carcinoma by Transfecting Interleukin-12 and Interleukin-2 Fusion Gene Intrasplenically, an Experimental Study].

Abstract:OBJECTIVE To study the inhibitory effect of retroviral packaging cells injected intrasplenically encoding mouse interleukin-12 (mIL-12) and human interleukin-2 (hIL-2) fusion gene on the growth of hepatocellular carcinoma. METHODS The retroviral vectors encoding mIL-12 gene, hIL-2 gene, and mIL-12 and hIL-2 genes, GCIL12EXPN, GCXEIL2PN, and GCIL12EIL2PN were constructed and then transfected into the retroviral packaging cells PA317 to construct cells PA317-GCIL12EXPN, PA317-GCXEIL2PN, and PA317-GCIL12EIL2PN. Rat hepatocellular carcinoma cells CBRH3 were implanted into the livers of Wistar rats to establish hepatoma animal model. Then the rats were divided into 5 groups to be injected intrasplenically with normal saline one day after the implantation (0.8 ml/rat, group I, n = 10), blank vector PA317-GCXEXPN one day after the implantation (10(7) cells/rat, group II, n = 10), PA317-GCIL12EXPN containing IL-2 gene (1 x 10(7) cells/rat 1, 3, 5, or 7 days after the implantation, group III, n = 40), PA317-GCXEIL2PN containing mIL-12 gene (1 x 10(7) cells/rat 1, 3, 5, or 7 days after the implantation, group IV, n = 40), and PA317-GCIL12EIL2PN containing IL-12-IL-2 fusion gene (1 x 10(7) cells/rat 1, 3, 5, or 7 days after the implantation, group V, n = 40) respectively. The rats surviving longer than 2 months were re-injected with hepatocellular carcinoma cells. The therapeutic effect, immune function and toxic effect were evaluated. CT was conducted on the liver before and after the experiment. Laparotomy was performed 3 and 7 days after treatment to resect some of the carcinoma tissues to undergo pathological examination and OX8 immunohistostaining. Serum mIL-12 and hIL-2 were detected one day before and 3, 7, 30, and 60 days after treatment. RESULTS The average survival times of the rats treated with IL-12-IL-2 fusion gene at the first, third, fifth and seventh day after tumor implantation were 53.3 +/- 3.7 days, 49.3 +/- 4.2 days, 31.0 +/- 2.1 days, and 24.3 +/- 1.4 days respectively, longer than those treated with IL-2 gene (25.0 +/- 2.5 days, 23.5 +/- 2.0 days, 18.3 +/- 2.4 days, and 12.0 +/- 1.8 days respectively, P < 0.001), and those treated with IL-12 gene (39.0 +/- 4.8 days, 32.0 +/- 3.9 days, 23.0 +/- 2.5 days, and 19.4 +/- 2.1 days respectively, P < 0.001). Long survival (>or= 60 days) rate in the rats treated with IL-12-IL-2 fusion gene on the first and third day was 30%. The serum mIL-12 and hIL-2 levels in these rats remained high on the 60th day after treatment. The pathological study showed that the number of infiltrating lymphocytes in liver tumor tissues was increased in the IL-12-IL-2 fusion gene treatment group. CONCLUSION The retroviral packaging cell line injected intrasplenically encoding mIL-12 and hIL-2 fusion gene inhibits the growth of hepatocellular carcinoma significantly in rats. The therapeutical efficacy of early administration is superior to that of late one.

Abstract:OBJECTIVE:To investigate the therapeutic effect of cationic liposome-mediated interleukin-12 gene delivery on established murine melanoma in vivo. METHODS:The lipofectin encapsulated pCmIL-12 plasmid was given to C57BL/6 mice on the day 3,5,7,9 after inoculation of B16 melanoma cells. The tumor size, the survival time of mice and the NK cell activity were observed. RESULTS:The pCmIL-12 plasmid coupled with cationic liposome inhibited the tumor growth and improved the survival of mice bearing established melanoma. The activity of NK cells was also enhanced after interleukin-12 gene delivery in vivo. CONCLUSION:Cationic liposome-mediated interleukin-12 gene delivery has significantly therapeutic effects on mice melanoma in vivo.

Abstract:OBJECTIVE:To study the anti-tumor effects of combined IL-12 and granalocyte-macrophage-colong scimulating factor (GM-CSF) gene therapy on murine hepatocellular carcinoma.METHODS:Twenty-four mice received subcutaneous inoculation of 1 x 10(6) BNL hepatoma cells were randomly divided into the following four groups with different cytokine encoding plasmids (6 mice for each group): (1)pXX-GM-CSF 12.5 microg and pXX-IL-12 12.5 microg; (2)pXX-IL-12 25 microg; (3)pXX-GM-CSF 25 microg; (4)pXX-Neo 25 microg. The plasmids were given through tail vein using a versatile hydrodynamics-based DNA delivery method on day 3 and day 6 after tumor challenge. The growth of tumor and cellular immune response were observed intensively. The changes in serum concentration of IL-12, GM-CSF, and IFN-gamma after plasmids injection were also observed.RESULTS:Co-delivery of IL-12 and GM-CSF could mount stronger anti-tumor effects, longer term enhanced IL-12 expression and lower level of IFN-gamma than did IL-12 alone.CONCLUSIONS:Combined IL-12 and GM-CSF can render a strong anti-tumor effect as well as a potential to lower the side effects.

Abstract:Among various immunotherapeutic approaches, interleukin-12 (IL-12) is particularly appealing because of its superior antitumor effects, which have been demonstrated in preclinical as well as clinical studies. However, IL-12 therapy was often accompanied by severe side effects due mainly to the supranormal induction of interferon-γ. To optimize the therapeutic efficacy and lower the side effects of IL-12, we have investigated the antitumor activity of combined IL-12 and granulocyte–macrophage colony-stimulating factor (GM-CSF) gene therapy in a highly malignant and poorly immunogenic murine hepatocellular carcinoma model. Using a versatile hydrodynamics-based DNA delivery method, we showed that the combined gene delivery of IL-12 and GM-CSF induced very strong antitumor cellular immunity and achieved significant therapeutic efficacy, whereas each cytokine gene alone yielded appreciable but less effects. We also observed that the combined therapy induced lower levels of interferon-γ than did IL-12 alone. These results suggest that combined IL-12 and GM-CSF therapy can render a stronger antitumor effect as well as lowering potential side effects. Cancer Gene Therapy (2001) 8, 751–758
The Adoptive Theraputic Effect of Interleukin-12 Gene Transfected Splenocytes from the Mice Immunized by Immunogene Tumor Vaccine on Implanted Liver Cancer

Abstract:Objective To find the feasibility and antitumor effect of mIL-12 gene transfected splenocytes derived from IL-2 and B7-1 geneco-transfected liver cancer vaccines immunized mice.Methods The murine liver cancer cell strain Hepal-6 cotransfected with both human IL-2 and B7-1 gene via racombinant adenovirus vectors (200 PFU/cell)were treated with 80 mg/L mytomycin (MMC) to prepare liver cancer cell vaccines.The splenocytes isolated from the mice immunized with these vaccines were transfected with mIL-12 gene or BGFP gene or nontransfected and injected into the mice xenograft liver cancer with tumor diameter being 1 cm) respectively.The adoptive anticancer effect was investigated.Results The increasing volume of the tumor injected with mIL-12 gene transfected splenocytes intratumorally was significantly smaller than in the control groups [The increasing volume of the tume in the mIL-12 gene transfected,BGFP gene transfected,nontransfected splenocytes and saline injected groups was (0.08±0.05),(3.46±0.15),(3.56±0.23) and (8.12±0.54) cm3,respectively, P 0.05].Conclusion Significant tumor inhibitory effect was demonstrated in the immuno-activated splenocytes derived from IL-2 and B7-1 gene co-transfected liver cancer vaccines immunized mice and was further improved while modified with mIL-12 gene.It is suggested that it would be more efficiency if gene therapy,specific active immunotherapy in combination with adopive cellular immunotherapy were adopted.

Abstract:Objective To investigate the effectiveness of gene therapy in vivo for hepatocellular carcinoma using double suicide genes combined with IL-12 gene. Methods SCID mice were subcutaneoulsy injected with HepG2 cells, 5×105 (in 0. 2 ml) in each flank, and intraperitoneally (ip) injected with 2×107(in 0. 5 ml) human PBL. When tumor reached a diameter of 0. 6 cm, the mice were separated into four groups and intratumoral injected with 5×105 (in 0. 1 ml) MMC-treated HepG2-IL-12 cells, 5× 105(in0.1 ml) HepG2-pALBeAFP-CD/TK cells or 5×105 (in 0. 05 ml) HepG2-pALBeAFP-CD/TK cells + 5×105 (in 0. 05 ml) MMC-treated HepG2-IL-12 cells in the left flank, and ip injected with PBS or GCV 100mg/kg+ 5-FC 500 mg/kg for 10 days: group Ⅰ, HepG2-IL-12 + GCV, 5-FC; and group Ⅱ . HepG2-pALBeAFP-CD/TK + PBS; group Ⅲ , HepG2-pALBeAFP-CD/ TK+GCV,5-FC; and groupⅣ , HepG2-pALBeAFP-CD/TK + HepG2-IL-12 + GCV, 5-FC. Tumor size determination and routine pathological study were performed on day 5 after treatment. Results On the left flank, there was a decrease of tumor volume of 20. 5% , 54. 9% , and 87. 6% on day 30 in group Ⅰ, Ⅲ and Ⅳ, compared with group Ⅱ, respectively. And there was distinct difference among group Ⅰ , Ⅲ and Ⅳ. On the right flank, tumor exhibited palpable volume reduction of 23. 3% , 24. 9% and 45. 0% , respectively in group Ⅰ , Ⅲand Ⅳ , compared to group Ⅱ . Conclusion The pALBeAFP-CD/TK-GCV. 5-FC system can efficiently treat HCC in vivo. In immune competent mouse, local expression of IL-12 can enhance the therapy effect and distal bystander effect of double suicide gene therapy.

Abstract:AIM:To test the efficacy of gene therapy in rat liver tumor.METHODS:A retroviral vector GCIL12EIL2PN encoding human IL-2 (hIL-2) and mouse IL-12 (mIL-12) fused gene and its packaging cell were constructed. The packaging cell lines contained of IL-2 and/or IL-12 genes were injected intrasplenically to transfect splenocyte at different time. The therapeutic effect, immune function and toxic effect were evaluated.RESULTS:The average survival times of the 4 groups using IL genes at days 1, 3, 5 and 7 after tumor implantation were 53.3+/-3.7, 49.3+/-4.2, 31.0+/-2.1 and 24.3+/-1.4 d respectively in IL-2/IL-12 fused gene group, 25.0+/-2.5, 23.5+/-2.0, 18.3+/-2.4 and 12.0+/-1.8 d respectively in IL-2 gene treatment group, and 39.0+/-4.8, 32.0+/-3.9, 23.0+/-2.5 and 19.4+/-2.1 d respectively in IL-12 gene treatment group (P<0.01, n=10). In the IL-12/IL-2 fused gene treatment group, 30% of rats treated at days 1 and 3 survived more than 60 d and serum mIL-12 and hIL-2 levels were still high at day 3 after treatment. Compared with IL alone, NK cell activity was strongly stimulated by IL-2/IL-12 gene. Microscopy showed that livers were infiltrated by a number of lymphocytes.CONCLUSION:IL-2 and/or IL-12 genes injected directly into spleen increase serum IL-2 and IL-12 levels and enhance the NK cell activity, which may inhibit the liver tumor growth. The therapy of fused gene IL-2/IL-12 is of low toxicity and relatively high NK cell activity. Our data suggest that IL-2/IL-12 fused gene may be a safe and efficient gene therapy for liver tumor. The gene therapy should be administrated as early as possible.

Abstract:Objective: To study the effects of intrasplenic injection of IL-12 and/or IL-2 genes on serum levels of IL-2 and IL-12 and T cell activity in induced liver cancer in rats. Methods: Forty-eight rats with diethylnitrosamine induced liver cancer were equally divided into 4 groups: mock-infected group, IL-2 group, IL-12 group and IL-2/IL-12 group. Retroviral vectors encoding IL-2 and/or IL-12 genes were constructed and were used to transfect packaging cell lines PE501 and PA317. The tansfected PE501 and PA317 cells were injected into rat spleen at different time points and the serum levels of IL-2 and IL-12, T cell activity and toxic effect were determined. Results: The serum levels of IL-2 and IL-12 were obviously increased in IL-2 and IL-12 group, respectively, and the increase was even more obvious in IL-2/IL-12 group. Pathological study showed that there was an increase in lymphocytes infiltration of liver tissues in IL-2/IL-12 group. Compared with mock-infected group, T cell activity markedly increased in IL-2 and IL-12 group, and that in IL-2/IL-12 group was even more active. Moreover, the acting time of IL-2/IL-12 was longer than that of IL-2 or IL-12 alone. Conclusion: Direct injection of IL-2 and/or IL-12 genes into spleen can increase the T cell activity. Combined gene therapy with IL-2 and IL-12 is superior to IL-2 or IL-12 gene alone.

Abstract:In our previous studies using gene gun-mediated delivery of interleukin 12 (IL-12) cDNA in vivo, we observed T-cell-mediated regression of established murine tumors and demonstrated the induction of systemic immunity in test animals. In this study, we further characterized the antitumoral and anti-metastatic effect of this gene therapy approach by employing two murine metastatic mammary tumor models: the immunogenic TS/A adenocarcinoma and the weakly immunogenic 4T1 adenocarcinoma. In the TS/A model, gene transfer into the skin overlying an established intradermal tumor with an IL-12 cDNA expression vector resulted in complete tumor regression in 50% of mice followed by the development of immunological memory. In contrast, the growth of the intradermal 4T1 tumors was not affected by the IL-12 gene therapy protocol. However, this treatment resulted in a substantial reduction of spontaneous metastases in the lungs of 4T1 tumor-bearing mice and significantly prolonged their survival time. T cells were not required for this anti-metastatic effect, because it was also observed in nude mice and in mice depleted of CD4+ and CD8+ T cells. Tumor-draining lymph node cells obtained from 4T1 tumor-bearing mice treated with IL-12 cDNA exhibited increased natural killer (NK) activity and produced enhanced levels of interferon-γ (IFN-γ) compared with similar mice treated with luciferase cDNA. In addition, in vivo depletion of NK cells or neutralization of IFN-γ resulted in partial suppression of the anti-metastatic effect of IL-12 gene therapy, suggesting the involvement of both NK cells and IFN-γ in this effect.

Abstract:Objective To investigate the inducible ability of plasmid DNA carrying a RU486 regulatory system.Methotis Plasmid pRS22 containing RU486 regulatory system,liver specific promoter and transgene IL-12 was injected into mice by hydrodynamic injection.RU486 was injected intraperitoneally into mice at difierent time points after plasmid administration.The IL-12 1evel in serum was tested by an ELISA kit.The distribution and inducible expression of pRS22 in mice were assayed by measuring DNA,RNA and protein levels by PCR,RT-PCR and immunohistochemical staining.Resuits To determine the duration of the activity of plasmid pRS22,mice injected with 10μg of pRS22 were treated repeatedly with 250μg/kg of RU486 per 7 days after hydrodynamic injection of plasmid.IL-12 expression in serum abruptly increased to peak was detected at 10 h after induction and declined to baseline on day 6.Though peak values of IL-12 decreased gradually after each induction,IL-12 in serum could be induced until 15 weeks after plasmid administration.A total of 5μg of pRS22 was injected into mice to detect the effect of different induction manners on the IL-12 expression.The mice were treated with RU486 per day or per 2 days iil 6 days,respectively.The induction per 2 days resulted in a wavelike pattern of serum IL-12 expression with peak lev-els on the day of induction alternating with lower values on the following day.In contrast,sustained levels of IL-12 could be achieved by administering RU486 per day.Plasmid DNA and GLp65 mRNA were detected in liver of mice with or without RU486 until at least day 28 after plasmid administration.However,IL-12 p35 mRNA was detected only in the liver of mice with RU486 induction.IL-12 immunohistochemical staining in liver demonstrated that IL-1 2 expressed predominantly in the hepatocytes near the surface of liver or between the central vein and portal area after induction with RU486.In contrast,no IL-12 expression was observed in the hepatocytes after induction with sesame oil.Conclusion Tight temporal and spatial control of transgene IL-12 expression could be achieved by RU486 regulatory system driven by liver specific promoter.
