Abstract:A novel and concise approach for the construction of the core structure of maoecrystal V (1) has been developed. Utilizing the lead-mediated arylation of beta-ketoesters and oxidative dearomatization/IMDA reaction as key steps, the two consecutive all-carbon quaternary centers (C-9 and C-10) were constructed in a stereoselective manner. The developed chemistry paves the way for the total synthesis of this fascinating natural product.

Abstract:Get to the core: The synthesis of an advanced structure of the recently reported cytotoxic diterpenoid maoecrystal V (1) has been accomplished. The key carbon–carbon bond-forming events feature an intramolecular Diels–Alder reaction, a cyclopropanation/reductive cleavage sequence, and an intramolecular alkylative lactonization reaction. Abstract:Maoecrystal V (1) is a novel diterpenoid, which was originally isolated from the leaves of the Chinese medicinal herb Isodon eriocalyx in 2004 by Sun et al.1 It has been found to be selectively cytotoxic towards HeLa cells, with an IC50 value of 20 ng mL(-1) . Significant research efforts have been devoted to the synthesis of maoecrystal V because of its intriguing biological properties, rarity in nature, and complex structural features. Herein, we describe our recent investigations, which have culminated in the total synthesis of (±)-maoecrystal V. The current strategy involved three key steps for the successful construction of the key tetrahydrofuran oxa-bridge skeleton, including a Wessely oxidative dearomatization, a novel intramolecular Diels-Alder reaction, and a Rh(II) -catalyzed OH insertion reaction.

Abstract:The evolution of a program directed at the enantioselective total synthesis of maoecrystal V, a highly modified ent-kauranoid, is described. An early stage chiral auxiliary-directed asymmetric C-H functionalization for the construction of a key benzofuran intermediate enabled the first asymmetric synthesis of the natural enantiomer of maoecrystal V, confirming the assigned stereochemistry. A divergent course of the central intramolecular Diels-Alder reaction, which is dependent on the nature of the dienophile, initially led to the development of an unanticipated and previously unknown isomer of maoecrystal V, which we named maoecrystal ZG. In light of the reported selective and potent cytotoxic activity of maoecrystal V, the cytotoxic properties of maoecrystal ZG were also investigated.

Abstract:Maoecrystal V is a C-19 diterpenoid natural product with rare caged skeleton. The main difficulties of synthesis were forecasted through structure analysis: (1) construction of highly compact five-membered ring; (2) installing continuous quaternary carbon chiral center; and (3) construction of [2.2.2] bridge ring. For these reasons, total synthesis of Maoecrystal V should conquer enormous challenges in both strategic design and practical reaction exploration. Exploring meritorious methods for basic skeleton construction of Maoecrystal V is the foundation of total synthesis. It is necessary to build up a model in order to provide worthy information for further total synthesis study.

Abstract:Total Synthesis Maoecrystal V is a complex natural product with distinguished anticancer activity. The biological synthesis of maoecrystal V has been proposed to take place via the acid-catalyzed cyclization of geranylgeranyl pyrophosphate (GGPP) as a key step. In their Full Paper on page 903 ff., Zhen Yang, Jian-Xian Gong et al. report on the total synthesis of maoecrystal V in 18 steps from 2,2-dimethylcyclohex-3-enone via an intramolecular Diels–Alder reaction and an Rh-catalyzed OH insertion reaction as key steps. This chemistry could be used to achieve the synthesis of several analogues of maoecrystal V (see the Back Cover).

Abstract:This work reports the total syntheses of Isodon diterpenes trichorabdal A and maoecrystal Z via a common bicyclo[3.2.1]octane intermediate, which chemically bridges the two distinct structures through a retro-aldol/aldol reaction cascade. Other synthetic features include an efficient cross-ring radical cyclization for rapid construction of an all carbon quaternary center commonly seen in Isodon diterpenoids, a challenging Ueno-Stork cyclization on to a sensitive 1,6-enone system, as well as a serial of chemoselective functional group manipulations in complex chemical environments.

Abstract:The title complex, [Mn3O(C6H5COO)(6)(py)(3)]. 2CH(3)CN (py = pyridine), has been prepared. Single-crystal X-ray analysis revealed a mu(3)-oxo-bridged (Mn2MnII)-Mn-III structure, in which each manganese atom has a distorted octahedral environment. The Mn(1) atom is readily assigned as a Mn-III centre from the Mn-(mu(3)-O) distance [1.819(5) Angstrom]. Abstract:Cranomycin and jogyamycin, two aminocyclopentitol natural products, possess complex structures and potential medicinal properties. This review describes synthetic studies about the process of making an advanced intermediate of cranomycin and jogyamycin. This highly functionalized intermediate, featuring three contiguous amine-substituted stereocenters, was constructed from cyclopentadiene through a series of reactions including the nitroso Diels-Alder reaction, nitrogen radical cyclization reaction, 1,2-nitrogen migration, and stereoselective nitrogen addition.

Abstract:Epitaxial growth of MOF-on-MOF composite is an evolving research topic in the quest for multifunctional materials. In previously reported methods, the core-shell MOFs were synthesized via a stepwise strategy that involved growing the shell-MOFs on top of the preformed core-MOFs with matched lattice parameters. However, the inconvenient stepwise synthesis and the strict lattice-matching requirement have limited the preparation of core-shell MOFs. Herein, we demonstrate that hybrid core-shell MOFs with mismatching lattices can be synthesized under the guidance of nucleation kinetic analysis. A series of MOF composites with mesoporous core and microporous shell were constructed and characterized by optical microscopy, powder X-ray diffraction, gas sorption measurement, and scanning electron microscopy. Isoreticular expansion of microporous shells and orthogonal modification of the core was realized to produce multifunctional MOF composites, which acted as size selective catalysts for olefin epoxidation with high activity and selectivity.

Abstract:Metal-organic frameworks (MOFs) are a class of porous crystalline materials constructed of metal centres with organic linkers, creating one-, two-, or three-dimensional well-organized frameworks with very high surface areas. The study of MOFs has become one of the research hot spots in many fields, owing to the broad potential applications projected for these materials in various areas. It is well recognized that synthesis strategies dictate the structure and thus the properties and performance of the resulted MOFs. This chapter provides a comprehensive up-to-date overview on the modulated synthesis strategies for MOFs. The ability to control crystal morphology and size by a number of modulated synthesis methods is illustrated by the zirconium-terephthalate-based MOF, the UiO-66, and a number of other MOFs.

Abstract:Crystal engineering of metal-organic frameworks (MOFs) has allowed the construction of complex structures at atomic precision, but has yet to reach the same level of sophistication as organic synthesis. The synthesis of complex MOFs with multiple organic and/or inorganic components is ultimately limited by the lack of control over framework assembly in one-pot reactions. Herein, we demonstrate that multi-component MOFs with unprecedented complexity can be constructed in a predictable and stepwise manner under simple kinetic guidance, which conceptually mimics the retrosynthetic approach utilized to construct complicated organic molecules. Four multi-component MOFs were synthesized by the subsequent incorporation of organic linkers and inorganic clusters into the cavity of a mesoporous MOF, each composed of up to three different metals and two different linkers. Furthermore, we demonstrated the utility of such a retrosynthetic design through the construction of a cooperative bimetallic catalytic system with two collaborative metal sites for three-component Strecker reactions.

Abstract:A new synthetic approach is developed and nine distinct organic functionalized oxomolybdenum complexes, 1-9, are synthesized and fully characterized. Specifically by altering the functional groups on imidazoles we succeeded in making a series of crystalline structures with unique underlying topologies and/or functionalities. These imidazole derivatives can act as ligands, structural directing agents, counter cations and simultaneously, solvents where the reactions and the crystallization occur.
