Abstract:We report a non-axial-scanning second harmonic imaging technique, in which the chromatic aberration of a Fresnel lens is exploited to focus different wavelengths of a fundamental beam into different axial positions to effectively realize axial scanning. Since the second harmonic signals at different axial positions are generated by different fundamental wavelengths and hence accordingly have different center wavelengths, they can be resolved and detected in parallel by using a spectrometer without axial mechanical scanning. We have demonstrated a system capable of achieving about 8 μm effective axial scanning range. Proof-of-concept imaging results are also presented.

Abstract:An axial-resolution-enhanced two-photon laser scanning microscopy system is presented in this paper. In the proposed method, we use a spatial light modulator (SLM) for the phase modulation of the excitation light. The axially split point spread function (PSF) is generated by loading a 0-π pattern on the SLM. The final quality-enhanced images are acquired by subtracting the two consecutive images acquired by the original PSF and the split PSF. Because of the fluorescence differential processing, the axial elongation of the particles images is suppressed, and the axial resolution is enhanced accordingly. With the axial resolution enhanced, the overlap between layer images is also reduced, which decreases the background noise of the images and enhances the contrast and image quality of the acquired fluorescence images. The capability of axial resolution and contrast enhancement is successfully demonstrated by both theoretical calculation and experimental results.

Abstract:Nonlinear focal modulation microscopy (NFOMM) is a really simple and effective super-resolution imaging method based on phase modulation with high-intensity illumination, which can extend the effective spatial-frequency bandwidth of the imaging system and achieve a transverse resolution of similar to 60 nm (similar to lambda/10). However, multiple images under different illumination pattern is needed, which result in limited imaging speed. A novel interleaved reconstruction method is proposed to increase the frame rate without any change in the raw data of NFOMM. Since allowing easy integration with confocal microscopes, we anticipate this method will be a useful observation tool in the biological community and other research field.

Abstract:The optical system is designed here, to miniaturize the mechanical scanning and number of phase patterns illumination in lensless microscopy. The random phase mask is positioned in vertical column before the sample, which encoded the sample information and resulted into a final image at the sensor plane. To improve the signal to noise ratio and resolution of final image, the axial direction scanning of sample and sensor (SS) is performed while keeping them mutually immovable. This operation gathered the lost information of sample in between the sample and sensor planes. This axial motion of SS is performed with periodic steps, which would help in the reconstruction process during post processing. The final image has resolution of about 4 µm and field of view of 15 mm 2 . There are numerous advantages of such type of techniques, including portability and fast processing of data.

Abstract:A new frequency-shifting confocal microscopy (FSCM) illuminated by an azimuthally polarized Bessel-Gaussian (BG) beam is investigated. The solid excitation spot is produced by the BG beam modulated by a spiral phase plate, and the donut excitation spot is directly obtained by the same BG beam. Through vector diffraction theory and two-view RL reconstruction algorithm, the optical transfer functions of two confocal imaging modes and the simulation imaging of FSCM are presented. The results show that, two illumination modes produced by the azimuthally polarized BG beam can enhance the spatial resolution of FSCM, the spatial resolution of reconstructed image is mainly depended on the illumination mode with higher frequency transfer efficiency, the small pinhole is helpful to improve the contrast and spatial resolution of image. When the iterations number is about 100, the reconstructed image has good quality. This FSCM is helpful to quickly realize super resolution and high contrast in cell imaging.

Abstract:The application of optical microscopy in four-dimensional (spatial and temporal) super-resolution imaging poses challenges because of the requirement of a long acquisition time or high illumination intensity. In this paper, we introduce simultaneous two-angle axial ratiometry (STARII) for <20 nm axial super-resolution imaging and for fast and long-term imaging of live cells up to hundreds of frames per second. This method involves recording two raw images in two incident angle channels in the context of evanescent wave illumination and obtaining the corresponding intensity ratio. Furthermore, we demonstrate the combination of STARII with the lateral super-resolution method to resolve three-dimensional nanoscale structures of microtubules and to visualize the long-term dynamical plasma membrane curvature and fast remodeling of endoplasmic reticulum tubule meshwork and three-way junctions. These demonstrations indicate an important potential application of STARII in investigating nanoscale cellular complex processes in the native state.

Abstract:We propose a novel imaging method that enables the enhancement of three-dimensional resolution of confocal microscopy significantly and achieve experimentally a new fluorescence emission difference method for the first time, based on the parallel detection with a detector array. Following the principles of photon reassignment in image scanning microscopy, images captured by the detector array were arranged. And by selecting appropriate reassign patterns, the imaging result with enhanced resolution can be achieved with the method of fluorescence emission difference. Two specific methods are proposed in this paper, showing that the difference between an image scanning microscopy image and a confocal image will achieve an improvement of transverse resolution by approximately 43% compared with that in confocal microscopy, and the axial resolution can also be enhanced by at least 22% experimentally and 35% theoretically. Moreover, the methods presented in this paper can improve the lateral resolution by around 10% than fluorescence emission difference and 15% than Airyscan. The mechanism of our methods is verified by numerical simulations and experimental results, and it has significant potential in biomedical applications.

Abstract:SummaryWe report on a chromatic axial scanning method for two‐photon excitation fluorescence imaging. Effective axial scanning is achieved by incorporating a Fresnel lens in the system, which has large chromatic aberration and can therefore focus the excitation beam to different axial positions depending on its wavelength. We experimentally demonstrated this technique and used it to image the cross‐section of fluorescent microspheres.

Abstract:We demonstrate nonlinear focal modulation microscopy (NFOMM) to achieve superresolution imaging. Traditional approaches to superresolution that utilize point scanning often rely on spatially reducing the size of the emission pattern by directly narrowing (e.g., through minimizing the detection pinhole in Airyscan, Zeiss) or indirectly peeling its outer profiles [e.g., through depleting the outer emission region in stimulated emission depletion (STED) microscopy]. We show that an alternative conceptualization that focuses on maximizing the optical system's frequency shifting ability offers advantages in further improving resolution while reducing system complexity. In NFOMM, a spatial light modulator and a suitably intense laser illumination are used to implement nonlinear focal-field modulation to achieve a transverse spatial resolution of ∼60 nm (∼λ/10). We show that NFOMM is comparable with STED microscopy and suitable for fundamental biology studies, as evidenced in imaging nuclear pore complexes, tubulin and vimentin in Vero cells. Since NFOMM is readily implemented as an add-on module to a laser-scanning microscope, we anticipate wide utility of this new imaging technique.

Abstract:We have developed a second harmonic photoacoustic microscopy (SH-PAM) for subdiffraction-limited imaging based on nonlinear thermal diffusion. When a sine-modulated Gaussian temperature field is introduced by a laser beam, the temperature dependence of the thermal diffusivity induces a nonlinear photoacoustic (PA) effect and thus results in the production of second harmonic PA signals. We demonstrate through both simulation and experiment that the second harmonic PA images can be reconstructed with a lateral resolution exceeding that of conventional optical resolution PA microscopy. The feasibility of SH-PAM was verified on phantom samples. Amphioxus zygotes and germinated pollens have been studied by SH-PAM to demonstrate its biomedical imaging capability. This method expands the scope of conventional PA imaging and opens up new possibilities for super-resolution imaging, prefiguring great potential for biological imaging and material inspection.

Abstract:We experimentally demonstrate a microwave far-field imaging modality with the transverse resolution exceeding the diffraction limit through the use of a single layer of highly nonlinear metamaterial. The harmonic fields of the nonlinear metamaterial surface allow the far-field propagation of wavefronts with spatial frequencies several times higher than that of the fundamental field. Near-field images can therefore be mathematically recovered from the far-field patterns of the harmonic fields.

Abstract:Second harmonic generation (SHG) microscopy is a valuable tool for optical microscopy. SHG microscopy is normally performed as a point scanning imaging method, which lacks phase information and is limited in spatial resolution by the spatial frequency support of the illumination optics. In addition, aberrations in the illumination are difficult to remove. We propose and demonstrate SHG holographic synthetic aperture holographic imaging in both the forward (transmission) and backward (epi) imaging geometries. By taking a set of holograms with varying incident angle plane wave illumination, the spatial frequency support is increased and the input and output pupil phase aberrations are estimated and corrected -- producing diffraction limited SHG imaging that combines the spatial frequency support of the input and output optics. The phase correction algorithm is computationally efficient and robust and can be applied to any set of measured field imaging data.

Abstract:Fourier ptychographic microscopy (FPM), as an emerging computational imaging method, has been applied to quantitative phase imaging with resolution bypassing the physical limit of the detection objective. Due to the weak illumination intensity and long image acquisition time, the achieved imaging speed in current FPM methods is still low, making them unsuitable for real-time imaging applications. We propose and demonstrate a high-speed FPM method based on using laser illumination and digital micro-mirror devices for illumination angle scanning. In this new, to the best of our knowledge, FPM method, we realized quantitative phase imaging and intensity imaging at over 42 frames per second (fps) with around 1 µm lateral resolution. The quantitative phase images have revealed membrane height fluctuations of red blood cells with nanometer-scale sensitivity, while the intensity images have resolved subcellular features in stained cancer tissue slices.

Abstract:The ability to image at high speeds is necessary for biological imaging to capture fast-moving or transient events or to efficiently image large samples. However, due to the lack of rigidity of biological specimens, carrying out fast, high-resolution volumetric imaging without moving and agitating the sample has been a challenging problem. Pupil-matched remote focusing has been promising for high NA imaging systems with their low aberrations and wavelength independence, making it suitable for multicolor imaging. However, owing to the incoherent and unpolarized nature of the fluorescence signal, manipulating this emission light through remote focusing is challenging. Therefore, remote focusing has been primarily limited to the illumination arm, using polarized laser light to facilitate coupling in and out of the remote focusing optics. Here, we introduce a novel optical design that can de-scan the axial focus movement in the detection arm of a microscope. Our method splits the fluorescence signal into S and P-polarized light, lets them pass through the remote focusing module separately, and combines them with the camera. This allows us to use only one focusing element to perform aberration-free, multi-color, volumetric imaging without (a) compromising the fluorescent signal and (b) needing to perform sample/detection-objective translation. We demonstrate the capabilities of this scheme by acquiring fast dual-color 4D (3D space + time) image stacks with an axial range of 70 μm and camera-limited acquisition speed. Owing to its general nature, we believe this technique will find its application in many other microscopy techniques that currently use an adjustable Z-stage to carry out volumetric imaging, such as confocal, 2-photon, and light sheet variants.

Synthetic aperture phase imaging of second harmonic generation field with computational adaptive optics

Abstract:Second-harmonic generation (SHG) microscopy provides label-free imaging of biological tissues with unique contrast mechanisms, but its resolution is limited by the diffraction limit. Here, we present the first experimental demonstration of super-resolution quantitative phase imaging of the SHG field based on synthetic aperture Fourier holographic microscopy. We discuss the mathematical model of synthetic-aperture imaging of SHG fields, as well as the computational adaptive optics technique for correcting sample-induced aberration. We demonstrate proof-of-concept experiments where SHG targets are embedded within a thick scattering medium to validate the performance of the proposed imaging technique. It is shown to be able to overcome the conventional Abbe diffraction limit even in the complex aberrations and strong multiple scattering. We also demonstrate SHG-based super-resolution deep-tissue phase imaging of ex-vivo zebrafish muscle tissue.

Abstract:Third-harmonic generation microscopy is a powerful label-free nonlinear imaging technique, providing essential information about structural characteristics of cells and tissues without requiring external labelling agents. In this work, we integrated a recently developed compact adaptive optics module into a third-harmonic generation microscope, to measure and correct for optical aberrations in complex tissues. Taking advantage of the high sensitivity of the third-harmonic generation process to material interfaces and thin membranes, along with the 1,300-nm excitation wavelength used here, our adaptive optical third-harmonic generation microscope enabled high-resolution in vivo imaging within highly scattering biological model systems. Examples include imaging of myelinated axons and vascular structures within the mouse spinal cord and deep cortical layers of the mouse brain, along with imaging of key anatomical features in the roots of the model plant . In all instances, aberration correction led to significant enhancements in image quality.

Abstract:We report on a novel chromatic confocal microscope system using supercontinuum white light generated from a photonic crystal fiber. The chromatic aberration of a pair of singlet lenses is employed to focus the different spectral components of the supercontinuum at different depth levels. An effective depth scanning range of 7 microm is demonstrated. The corresponding depth resolution is measured to be less than 1 microm (FWHM).
