Abstract:We present an alternative approach to realize structured illumination microscopy (SIM), which is capable for live cell imaging. The prototype utilizes two sets of scanning galvo mirrors, a polarization converter and a piezo-platform to generate a fast shifted, s-polarization interfered and periodic variable illumination patterns. By changing the angle of the scanning galvanometer, we can change the position of the spots at the pupil plane of the objective lens arbitrarily, making it easy to switch between widefield and total internal reflection fluorescent-SIM mode and adapting the penetration depth in the sample. Also, a twofold resolution improvement is achieved in our experiments. The prototype offers more flexibility of pattern period and illumination orientation changing than previous systems. Abstract:Through the interference of several beams with the same phase at the rear pupil, an optical lattice is generated and used as the excitation light field in lattice light-sheet microscopy (LLSM). Although, LLSM offers fast imaging ability through the dithered mode, it suffers from poor axial resolution. On the other hand, the structured illumination microscopy (SIM) mode in LLSM provides a high but anisotropic spatial resolution at the price of a lower imaging speed. In this Letter, we propose a new, to the best of our knowledge, type of optical lattice by defining the composed beam vectors with different phases. The lattice light-sheet difference microscopy (LLSDM) proposed in this work utilizes the new optical lattice along with the conventional one by simply changing the pattern loaded on the spatial light modulator. Through simulations with parameters obtained from the real acquisition procedures and the microscope properties, we examine the feasibility of LLSDM, and obtain nearly isotropic spatial resolution and great alleviation of the sidelobes. These characteristics enable LLSDM to broaden the accessible imaging content of light-sheet microscopes and further facilitate their applications in life sciences. Abstract:In this paper, light sheet microscopy using line scan confocal with bidirectional Bessel beam illumination is presented. By matching the scanning of the Bessel beam with the line-by-line exposure of the sCMOS, we shape the active area of the sensor to a slit and use the slit as a spatial filter to suppress the background noise caused by the side lobe of the Bessel beam. The optimal signal-to-noise ratio is obtained by changing the width of the slit. The images of fluorescent particle samples and zebrafish samples obtained by using the in line confocal and wide field modes are compared to verify the ability of this method to suppress background noise. In addition, three-dimensional imaging is realized by cooperating with the movement of the sample translation stage. The system has the ability of in vivo imaging, which is confirmed by recording the heartbeat of a zebrafish and the flow of cells in blood vessels. Finally, an imaging of high-scattered Drosophila larvae confirms that bidirectional illumination can achieve uniform illumination in high-scattering samples.
Light Sheet Microscopy with High Spatial Resolution Based on Polarized Structured Illumination Beam Modulated by the Phase Mask

Abstract:In this article, we present a method to improve axial resolution and expand the field of view of conventional light sheet fluorescence microscopy (LSFM) by phase modulation of the illumination beam. We added a bisected annular binary phase mask on the illumination path to modulate the radially polarized beam. The central spot of the modulated point spread function is narrower than that of Gaussian focus, although the sidelobe energy is increased. Then, a diffraction grating is used to produce multiple copies of this illumination pattern, and these pattern copies form the light sheet. By matching the numerical apertures of illumination lens and detection lens, the sidelobes on the detection axis can be suppressed in the captured data, although they do still contribute to photobleaching and phototoxicity in the sample. The simulation results show that the proposed method effectively reduces the thickness of the light sheet by 1.3 times, and increase the length of the light sheet by two times compared with conventional LSFM.
Three-dimensional super-resolution imaging of live whole cells using structured illumination microscopy

Abstract:1. PRINCIPLE Structured illumination microscopy (SIM) improves the spatial resolution via shifting the high-frequency information of a sample into the acceptable passband of the imaging system. In conventional SIM systems, a diffraction grating is used to diffract the incident light interfering at the sample plane to produce the structured illumination pattern, which lengthens the imaging speed. Faster SIM can be achieved by replacing the grating with a spatial light modulator (SLM). In recent years, the SIM theory have been greatly developed to enhance the spatial and temporal resolution. However, these developments were mainly focused on two dimensions [1]. Only a few studies have been conducted with three-dimensional (3D) SIM, especially for live-cell imaging. With routine fast biological applications in mind, we present here a more flexible system for 3D SIM based on two galvanometer sets conveniently controlling the incident angle (i.e. pattern period) and azimuth angle (i.e. pattern orientation) of the illumination light. The phase of the 3D structured illumination pattern is shifted by two piezoelectric stages. As the galvanometer and piezoelectric stage allow the pattern to be rotated and phase-shifted at the order of kHz, the imaging speed of our system is high enough to observe the sub-cellular dynamics.

Abstract:Imaging and tracking three-dimensional (3D) nanoscale organizations and functions of live cells is essential for biological research but it remains challenging. Among different 3D super-resolution techniques, 3D structured illumination microscopy (SIM) has the intrinsic advantages for live-cell studies; it is based on wide-field imaging and does not require high light intensities or special fluorescent dyes to double 3D resolution. However, the 3D SIM system has developed relatively slowly, especially in live imaging. Here, we report a more flexible 3D SIM system based on two galvanometer sets conveniently controlling the structured illumination pattern's period and orientation, which is able to study dynamics of live whole cells with high speed. We demonstrate our microscope's capabilities with strong optical sectioning and lateral, axial, and volume temporal resolution of 104 nm, 320 nm and 4 s, respectively. We do this by imaging nanoparticle and microtubule organizations and mitochondria evolution. These characteristics enable our galvanometer-based 3D SIM system to broaden the accessible imaging content of SIM-family microscopes and further facilitate their applications in life sciences. Abstract:An axial-resolution-enhanced two-photon laser scanning microscopy system is presented in this paper. In the proposed method, we use a spatial light modulator (SLM) for the phase modulation of the excitation light. The axially split point spread function (PSF) is generated by loading a 0-π pattern on the SLM. The final quality-enhanced images are acquired by subtracting the two consecutive images acquired by the original PSF and the split PSF. Because of the fluorescence differential processing, the axial elongation of the particles images is suppressed, and the axial resolution is enhanced accordingly. With the axial resolution enhanced, the overlap between layer images is also reduced, which decreases the background noise of the images and enhances the contrast and image quality of the acquired fluorescence images. The capability of axial resolution and contrast enhancement is successfully demonstrated by both theoretical calculation and experimental results.

Abstract:Wide-field saturated structured illumination microscopy has not been widely used due to the requirement of high laser power. We propose a novel method called laser scanning saturated structured illumination microscopy (LS-SSIM), which introduces high order of harmonics frequency and greatly reduces the required laser power for SSIM imaging. To accomplish that, an excitation PSF with two peaks is generated and scanned along different directions on the sample. Raw images are recorded cumulatively by a CCD detector and then reconstructed to form a high-resolution image with extended optical transfer function (OTF). Our theoretical analysis and simulation results show that LS-SSIM method reaches a resolution of 0.16 λ, equivalent to 2.7-fold resolution than conventional wide-field microscopy. In addition, LS-SSIM greatly improves the optical sectioning capability of conventional wide-field illumination system by diminishing our-of-focus light. Furthermore, this modality has the advantage of implementation in multi-photon microscopy with point scanning excitation to image samples in greater depths.

Abstract:We present a mechanically sheared image acquisition format for upright and open-top light-sheet microscopes that automatically places data in its proper spatial context. This approach, which reduces computational post-processing and eliminates unnecessary interpolation or duplication of the data, is demonstrated on an upright variant of Axially Swept Light-Sheet Microscopy (ASLM) that achieves a field of view, measuring 774 x 435 microns, that is 3.2-fold larger than previous models and a raw and isotropic resolution of ∼420 nm. Combined, we demonstrate the power of this approach by imaging sub-diffraction beads, cleared biological tissues, and expanded specimens.

Abstract:Advances in light-sheet microscopy have enabled the fast three-dimensional (3D) imaging of live cells and bulk specimens with low photodamage and phototoxicity. Combining light-sheet illumination with super-resolution imaging is expected to resolve subcellular structures. Actually, such kind of super-resolution light-sheet microscopy was recently demonstrated using a single-molecule localization algorithm. However, the imaging depth and temporal resolution of this method are limited owing to the requirements of precise single molecule localization and reconstruction. In this work, we present two-photon super-resolution light-sheet imaging via stochastic optical fluctuation imaging (2PLS-SOFI), which acquires high spatiotemporal resolution and excellent optical sectioning ability. 2PLS-SOFI is based on non-linear excitation of fluctuation/blinking probes using our recently developed fast two-photon three-axis digital scanned light-sheet microscope (2P3A-DSLM), which enables both deep penetration and thin sheet of light. Overall, 2PLS-SOFI demonstrates up to 3-fold spatial resolution enhancement compared with conventional two-photon light-sheet (2PLS) microscopy and about 40-fold temporal resolution enhancement compared with individual molecule localization-selective plane illumination microscopy (IML-SPIM). Therefore, 2PLS-SOFI is promising for 3D long-term, deep-tissue imaging with high spatiotemporal resolution.

Abstract:Light sheet fluorescence microscopy has become a basic tool in biology and medical research with its fast imaging speed, low phototoxicity and high spatiotemporal resolution. Here, we report a novel method to generate multiplane parallel light sheets with a micro structure named isosceles triangular array. The thickness of light sheet in each plane can approach 3.12 mu m along with the working distance of 152.20 mu m directly generated with the isosceles triangular array. We experimentally recorded the multiplane parallel light sheets illumination, which is consistent with the corresponding simulation by using ray-tracing method. To evaluate the imaging quality, we compared the light field generated with and without projected phase mask onto the spatial light modulator by using polystyrene fluorescent microspheres nanoparticles and transgenic zebrafish. This study provides a potential method for the building of volumetric imaging light sheet fluorescence microscopy with high resolution and low phototoxicity.

Abstract:We present the first demonstration of three-photon excitation light-sheet fluorescence microscopy. Light-sheet fluorescence microscopy in single- and two-photon modes has emerged as a powerful wide-field, low photo-damage technique for fast volumetric imaging of biological samples. We extend this imaging modality to the three-photon regime enhancing its penetration depth. Our present study uses a standard conventional femtosecond pulsed laser at 1000 nm wavelength for the imaging of 450 µm diameter cellular spheroids. In addition, we show, experimentally and through numerical simulations, the potential advantages in three-photon light-sheet microscopy of using propagation-invariant Bessel beams in preference to Gaussian beams.

Abstract:The promise of single-objective light-sheet microscopy is to combine the convenience of standard single-objective microscopes with the speed, coverage, resolution and gentleness of light-sheet microscopes. We present DaXi, a single-objective light-sheet microscope design based on oblique plane illumination that achieves: (1) a wider field of view and high-resolution imaging via a custom remote focusing objective; (2) fast volumetric imaging over larger volumes without compromising image quality or necessitating tiled acquisition; (3) fuller image coverage for large samples via multi-view imaging and (4) higher throughput multi-well imaging via remote coverslip placement. Our instrument achieves a resolution of 450 nm laterally and 2 μm axially over an imaging volume of 3,000 × 800 × 300 μm. We demonstrate the speed, field of view, resolution and versatility of our instrument by imaging various systems, including Drosophila egg chamber development, zebrafish whole-brain activity and zebrafish embryonic development - up to nine embryos at a time.

Abstract:Light-sheet fluorescence microscopy (LSFM) has emerged as a powerful method for rapid and optically efficient 3D microscopy. Initial LSFM designs utilized a static sheet of light, termed selective plane illumination microscopy (SPIM), which exhibited shadowing artifacts and deteriorated contrast due to light scattering. These issues have been addressed, in part, by multidirectional selective plane illumination microscopy (mSPIM), in which rotation of the light sheet is used to mitigate shadowing artifacts, and digital scanned light-sheet microscopy (DSLM), in which confocal line detection is used to reject scattered light. Here we present a simple and passive multidirectional digital scanned light-sheet microscopy (mDSLM) architecture that combines the benefits of mSPIM and DSLM. By utilizing an elliptical Gaussian beam with increased angular diversity in the imaging plane, mDSLM provides mitigation of shadowing artifacts and contrast-enhanced imaging of fluorescently labeled samples.

Abstract:Full view observation throughout entire specimens over a prolonged period is crucial when exploring the physiological functions and system -level behaviors. Multi -photon microscopy (MPM) has been widely employed for such purposes owing to its deep penetration ability. However, the current MPM struggles with balancing the imaging depth and quality while avoiding photodamage for the exponential increasement of excitation power with the imaging depth. Here, we present a dual -objective two -photon microscope (Duo -2P), characterized by bidirectional two -photon excitation and fluorescence collection, for long -duration volumetric imaging of dense scattering samples. Duo -2P effectively doubles the imaging depth, reduces the total excitation energy by an order of magnitude for samples with a thickness five times the scattering length, and enhances the signal-to-noise ratio up to 1.4 times. Leveraging these advantages, we acquired volumetric images of a 380-mu m suprachiasmatic nucleus slice for continuous 4-h recording at a rate of 1.67 s/volume, visualized the calcium activities over 4000 neurons, and uncovered their state -switching behavior. We conclude that Duo -2P provides an elegant and powerful means to overcome the fundamental depth limit while mitigating photodamages for deep tissue volumetric imaging. (c) 2024 Chinese Laser Press

Abstract:We developed two-photon scanning patterned illumination microscopy (2P-SPIM) for super-resolution two-photon imaging. Our approach used a traditional two-photon microscopy setup with temporally modulated excitation to create patterned illumination fields. Combing nine different illuminations and structured illumination reconstruction, super-resolution imaging was achieved in two-photon microscopy. Using 2P-SPIM we achieved a lateral resolution of 141 nm, which represents an improvement by a factor of 1.9 over the corresponding diffraction limit. We further demonstrated super-resolution cellular imaging by 2P-SPIM to image actin cytoskeleton in mammalian cells and three-dimensional imaging in highly scattering retinal tissue.

Abstract:High-speed 3D microscopic imaging methods have led to numerous biological discoveries. In this Letter, we present a sync-free light sheet microscope (LSM) based on a static phase mask that eliminates the need for coupling the detection plane, thereby enabling high-speed volumetric imaging that is only limited by the speed of cameras, e.g., similar to 1000 cross sections/s. In the sync-free design, the emission signals are first guided to the back of a galvanometric mirror, which laterally scans the emissions across a 2D phase mask, converting it to axial scanning that automatically compensates the focal shifts in the detection optics. Parametric models are developed to guide the phase mask design as well as to relate the axial scanning depth and magnification to design parameters. To quickly evaluate the different mask designs, a liquidcrystal-based spatial light modulator (LC-SLM) is used in the system. In the experiments, we scanned pollen and tissue samples via both the 2D phase mask and a piezoelectric objective scanner. The results show that the new method can generate clear images with comparable quality throughout the scanning range. The overall efficiency of the LSM is similar to 10%. It is worthwhile to note that the efficiency will be significantly improved by replacing the LC-SLM with a custom-made lens. The new method realizes a compact LSM for high-speed 3D imaging that may find important applications in in vivo biological imaging. (C) 2017 Optical Society of America
Laterally Swept Light-Sheet Microscopy Enhanced by Pixel Reassignment for Photon-Efficient Volumetric Imaging

Abstract:In light-sheet fluorescence microscopy, the axial resolution and field of view are mutually constrained. Axially swept light-sheet microscopy (ASLM) can decouple the trade-off, but the confocal detection scheme using a rolling shutter also rejects fluorescence signals from the specimen in the field of interest, which sacrifices the photon efficiency. Here, we report a laterally swept light-sheet microscopy (LSLM) scheme in which the focused beam is first scanned along the axial direction and subsequently laterally swept with the rolling shutter. We show that LSLM can obtain a higher photon efficiency when similar axial resolution and field of view can be achieved. Moreover, based on the principle of image scanning microscopy, applying the pixel reassignment to the LSLM images, hereby named iLSLM, improves the optical sectioning. Both simulation and experimental results demonstrate the higher photon efficiency with similar axial resolution and optical sectioning. Our proposed scheme is suitable for volumetric imaging of specimens that are susceptible to photobleaching or phototoxicity.

Abstract:Light-sheet microscopy stands out as a powerful tool in biological imaging due to its exceptional performance in fluorescence imaging. However, achieving both high sectioning performance and a vast field of view (FOV) poses a fundamental challenge in conventional light-sheet microscopy. The light-sheet thickness is typically constrained to 1 mu m for a wide FOV, potentially compromising resolution. To address this limitation, an axial scanning light-sheet microscopy (ASLM) technique integrated with aberration-free tunable foci to enable high-NA excitation while maintaining a generous FOV, is introduced. The proposed scheme successfully achieves isotropic resolution of 280 nm in a 3D imaging system, encompassing a FOV of 80 x 80 mu m and an impressive imaging speed of 80 ms per frame. These remarkable characteristics underscore the immense potential of ASLM for high- spatiotemporal resolution imaging. Light-sheet microscopy is a powerful tool in biological imaging, but achieving high sectioning performance and large field of view (FOV) is still a challenge. Abstract:Super-resolution three-dimensional (3D) optical microscopy has incomparable advantages over other high-resolution microscopic technologies, such as electron microscopy and atomic force microscopy, in the study of biological molecules, pathways and events in live cells and tissues. We present a novel approach of structured illumination microscopy (SIM) by using a digital micromirror device (DMD) for fringe projection and a low-coherence LED light for illumination. The lateral resolution of 90 nm and the optical sectioning depth of 120 μm were achieved. The maximum acquisition speed for 3D imaging in the optical sectioning mode was 1.6×10 7 pixels/second, which was mainly limited by the sensitivity and speed of the CCD camera. In contrast to other SIM techniques, the DMD-based LED-illumination SIM is cost-effective, ease of multi-wavelength switchable and speckle-noise-free. The 2D super-resolution and 3D optical sectioning modalities can be easily switched and applied to either fluorescent or non-fluorescent specimens.
