Abstract:Molecules containing vicinal all-carbon quaternary stereocenters are found in many secondary metabolites, and they exhibit a variety of biological and pharmacological activities. However, the construction of such a structural motif remains a significant challenge in natural product synthesis. Only in recent years have considerable efforts been made to construct vicinal quaternary stereocenters in a single-step operation. In this review, we focus on the different types of methods that have been successfully used in the total synthesis of natural products. Based on the classified reactions for the simultaneous generation of vicinal all-carbon quaternary stereocenters, the total syntheses of the natural products are discussed, placing emphasis on the diastereoselective preparation of vicinal quaternary carbon centers and the subsequent total syntheses.

Abstract:Asymmetric catalytic carbene reactions have been well documented in the last few decades for the expeditious assembly of chiral molecules with structural diversity. However, the enantioselective construction of all‐carbon quaternary centers remains a challenge in this area. In this review article, two types of asymmetric carbene reactions that beyond cyclopropanation, cyclopropenation, and Büchner reaction, have been summarized for the construction of all‐carbon quaternary centers: 1) using carbene species as a 1C synthon that reacts with a trisubstituted prochiral center; 2) sequential installation of two different C‐C bonds on the carbene position, which features a gem‐difunctionalization reaction. Especially, the asymmetric metal carbene gem‐dialkylation process, which has emerged as a practical and versatile method for the expeditious assembly of complex architectures from readily available chemical resources, is a complementary approach for the expeditious assembly of all‐carbon quaternary centers.

Abstract:Herein, we report an unprecedented strategy for the simultaneous construction of a remote C-N stereogenic axis and three contiguous stereogenic carbon centers via silver-catalyzed desymmetrizative [3 + 2] cycloaddition of prochiral N-aryl maleimides with activated isocyanides. This method features operational simplicity, wide substrate scope, high efficiency, and good to excellent stereoselectivity. Notably, it represents the first example of catalytic enantioselective synthesis of C-N atropisomers with the use of activated isocyanides.

Abstract:Acyclic quaternary carbon stereocenters occur frequently in natural products, bioactive molecules, and pharmaceutical compounds. Construction of a carbon stereogenic center attached to four different carbons with defined spatial arrangement is a daunting challenge in asymmetric catalysis. Significant efforts have been directed towards the stereoselective construction of such acyclic quaternary carbon stereocenters. In particular, catalytic generation of acyclic quaternary carbon stereocenters through functionalization of unsaturated hydrocarbons is an extremely attractive approach because unsaturated hydrocarbons are easily accessible both in industry and in organic synthesis. Abstract:Quaternary carbon stereocenters are common structural motifs in organic synthesis. The construction of these stereocenters in a catalytic and enantioselective manner remains a prominent synthetic challenge. In particular, methods for the synthesis of alkyne-substituted quaternary carbon stereocenters are very rare. Previous catalytic systems for hydroalkynylation of alkenes create tertiary stereocenters. We describe here an iridium catalyzed asymmetric hydroalkynylation of nonactivated trisubstituted alkene. The hydroalkynylation of β,γ-unsaturated amides occurs with high regio- and enantioselectivities to afford alkyne-substituted acyclic quaternary carbon stereocenters. Computational and experimental data suggest that the enantioselectivity is not only determined by the facial selectivity of the alkene but also by an alkene isomerization process. This strategy provides an efficient method to access alkyne-substituted acyclic quaternary carbon stereocenters with minimally functionalized starting materials.

Abstract:The catalytic enantioselective assembly of spirocyclic molecules featuring a spiro quaternary carbon stereocenter is currently of great interest because such privileged 3D structures are widely present in natural products that exhibit a broad spectrum of biological and pharmacological activities. This review summarizes the advances based on six major synthetic strategies and showcases the reaction mechanisms in detail. The advantages and limitations of each synthetic strategy are presented, and the remaining synthetic opportunities are outlined.

Abstract:Abstract Acyclic quaternary stereocenters are widely present in a series of biologically active natural products and pharmaceuticals. However, development of highly efficient asymmetric catalytic methods for the construction of these privileged motifs represents a longstanding challenge in organic synthesis. Herein, an efficient chiral phosphoric acid catalyzed direct asymmetric addition of α-alkynyl acyclic ketones with allenamides has been developed, furnishing the acyclic all-carbon quaternary stereocenters with excellent regioselectivities and high enantioselectivities. Extensive and detailed experimental mechanistic studies were performed to investigate the mechanism of this reaction. Despite a novel covalent allyl phosphate intermediate was found in these reactions, further studies indicated that a S N 2-type mechanism with the ketone nucleophiles is not very possible. Instead, a more plausible mechanism involving the elimination of the allyl phosphate to give the α,β-unsaturated iminium intermediate, which underwent the asymmetric conjugate addition with the enol form of ketone nucleophiles under chiral anion catalysis, was proposed. In virtue of the fruitful functional groups bearing in the chiral products, the diverse derivatizations of the chiral products provided access to a wide array of chiral scaffolds with quaternary stereocenters.

Abstract:A Pd(II)/N,N '-disulfonyl bisimidazoline-catalyzed asymmetric 1,4-conjugate addition reaction of low-cost arylboronic acids with readily available beta-substituted cyclic enones is described, providing a straightforward way of constructing cyclic all-carbon quaternary stereocenters with high enantioselectivity, in which >= 96% ee was obtained in most cases. The reaction proceeded without the protection of inert gas, making the operation process simple. Theoretical calculations have been applied to understand the origins of enantioselectivity.

Abstract:The enantioselective construction of all-carbon quaternary stereocenters is one of the most challenging fields in asymmetric synthesis. An asymmetric desymmetrization strategy offers an indirect and efficient method for the formation of all-carbon stereocenters. An enantioselective formation of cyano-bearing all-carbon quaternary stereocenters in 1,2,3,4,-tetrahydroquinolines and 2,3,4,5-tetrahydro-1H-benzo[b]azepines by copper-catalyzed desymmetric N-arylation is demonstrated. The cyano group at the prochiral center plays a key role for the high enantioselectivity and works as an important functional group for further transformations. DFT studies provide a model which successfully accounts for the origin of enantioselectivity.

Abstract:Herein, we report the discovery of the catalytic asymmetric conjugated addition of allylstannanes to indol-2-ones promoted by Ni(II)/chiral N,N′-dioxide. This method enables the stereodivergent construction of contiguous quaternary carbon centers (CQCCs), which can be readily achieved by a suitable combination of a chiral catalyst with stereospecific allylstannanes. This protocol allows for a unified access to all stereoisomers of 3,3-disubstituted oxindoles with CQCCs in good to excellent enantioselectivities and diastereoselectivities.

Abstract:The formation of sterically hindered C-C bond represents a great challenge in modern synthetic organic chemistry. A particularly challenging issue is the construction of all-carbon quaternary stereocenters. Herein, a ceric ammonium nitrate (CAN)-mediated intramolecular oxidative cross-coupling of silyl ethers for direct construction of valuable polycyclic scaffolds is described. The reaction enables sterically congested vicinal all-carbon quaternary and tertiary stereocenters to be installed diastereoselectively. The developed method provides a concise and efficient approach for ligation of two different segments through a compact C - C bond formation, which has potential applications in the synthesis of complex molecules as well as sterically congested natural products.

Abstract:Optically active molecule architectures stand as an important class of organic compounds and occupy a key role in academic and industrial communities. Particularly, the molecules bearing quaternary carbon are of vital importance because of its favorable conformation and valuable three‐dimensional molecules, which frequently play a key role in a broad spectrum of functional materials, pharmaceutical relevant natural molecules, and agrochemicals. Over the past few decades, a large amount of synthetic strategies for the enantioselective construction of compounds with chiral quaternary carbon centers have been the focus of a number of research initiatives. In this review, the state‐of‐the‐art toward the synthesis of enantioenriched molecules bearing quaternary stereocenters are summarized, which could be segmented into four categories: 1) Construction of optically active quaternary carbon centers by addition to prochiral sp2 carbon; 2) Construction of optically active all‐carbon quaternary stereocenters via substitution at non‐chiral tetra‐substituted carbon; 3) Construction of optically active all‐carbon quaternary stereocenters via kinetic resolution; 4) Construction of optically active all‐carbon quaternary stereocenters via desymmetrization reactions.

Abstract:Comprehensive Summary The construction of acyclic quaternary carbon stereocenters, which are ubiquitous in many bioactive compounds, pharmaceuticals and natural products, has been a long persuit in synthetic organic chemistry. Among numerous methods, enantioselective nickel‐catalysis has attracted incremental attention in recent years. This review summarizes the recent development in the asymmetric strategies, research progress, mechanistic investigations for the generation of acyclic quaternary carbon stereocenters via enantioselective nickel catalysis. Key Scientists In 2006, the Zhou group and RajanBabu group realized the nickel‐catalyzed enantioselective hydrovinylation of α‐substituted styrenes and ethylene to construct acyclic quaternary carbon stereocenters (QCSs) by using spiro‐ and binaphthyl phosphoramidite ligands, respectively. In 2016, Watson developed a nickel‐catalyzed Suzuki‐Miyaura arylation to generate acyclic QCSs via chirality transfer reaction. In 2017, the Feng and the Fu group developed nickel‐catalyzed enantioselective conjugated/Michael additions to build acyclic QCSs, respectively. In 2020, the Fang group made the first success in the construction of QCSs by nickel‐catalyzed hydrocyanation of alkenes and allenes. In 2021, Gregory C. Fu and co‐workers disclosed a nickel‐catalyzed enantioselective α‐functionalization of carbonyl compounds that could form acyclic QCSs. At the same time, the Shi group constructed chiral acyclic QCSs via nickel‐catalyzed functionalization of alkenes. In 2023, Kleij and co‐workers applied a new strategy for Ni‐catalyzed regio‐ and enantioselective homoallylic coupling to construct acyclic QCSs. In the same year, Tao and co‐workers demonstrated a novel strategy that is dinickel‐catalyzed enantioselective α‐alkylation of carbonyl compounds with alkyl iodides.

Abstract:Comprehensive SummaryUnder the catalysis of chiral Cu(II) complex, C2 and C3 alkylation of pyrrole and C3 alkylation of indole were realized simultaneously. The chiral oxindole skeleton with an all‐carbon quaternary stereogenic center at the C3‐position could be obtained exclusively with high yields (up to 95%) and excellent enantioselectivities (up to > 99%).

Abstract:Multiple contiguous quaternary carbon stereocenters (CQS) are highly challenging, yet important structural motifs in organic synthesis. Here, we describe a visible light induced catalytic [2 + 2] cycloaddition approach that constructed up to four CQS in a pentacyclic fused ring system diastereoselectively, from the readily accessible dienamides with pendent heteroaryls. Variously substituted dienamides have been cyclized with heteroaryls to provide a range of novel CQS-containing scaffolds (26 examples, up to 96% yield and >20:1 dr ratio). Mechanistic studies revealed that it may proceed through an uncommon β -C radical initiated 7- endo cyclization from the biradical intermediate.

Abstract:Pyrazolone moiety bearing an all‐carbon quaternary stereogenic center is frequent in biologically active products and pharmaceuticals. The catalytic route for the construction of a quaternary carbon center possesses challenges. In this report, catalytic asymmetric Michael addition of 4‐monosubstituted‐pyrazol‐5‐ones to simple enones catalysed by primary amine‐Brønsted acid composite has been developed. An array of pyrazolone derivatives bearing vicinal all carbon quaternary and tertiary stereocenters, were obtained in moderate to good diastereoselectivities (up to 92:8 dr) and good to excellent enantiomeric excess (up to 99% ee). In addition, antipode for enantiomers of the pyrazolone derivatives were also be realized in good to high stereoselectivities (up to 92:8 dr and up to 98% ee).

Abstract:Contiguous all carbon quaternary stereocenters are common structural units in bioactive natural products with significant synthetic challenges. In this paper, we reviewed our recent progresses in total synthesis of complex natural products containing contiguous all carbon quaternary stereocenters firstly. Then, four different synthetic strategies concerning the construction of contiguous all carbon quaternary stereocenters (≥3) were summarized. Finally, the total synthesis of waihoensene and crinipellins reported by us and other groups were taken as examples to clarify the advantages and disadvantages of various synthetic methodologies and synthetic strategies.

Abstract:The first highly enantioselective asymmetric decarboxylative addition of β-keto acids with 3-alkenyl-oxindoles bearing an all-carbon quaternary stereocenter have been developed. The relevant products were acquired in 49–98% yields with 88–98% enantioselectivities in the presence of 0.04–1.0 mol % of chiral rhodium catalyst. Abstract:AbstractThe diastereo‐ and enantioselective construction of vicinal all‐carbon quaternary stereocenters is a formidable task. We here report a synergistic bimetallic catalysis via a chiralN,N′‐dioxide‐europium (Eu) complex and a phosphoramidite‐iridium (Ir) catalyst for the asymmetric allylation of oxindole derivatives by using challenging trisubstituted allylic esters. The allylated products bearing vicinal all‐carbon quaternary stereocenters were obtained with good diastereoselectivities (up to 20 : 1 dr) and excellent enantioselectivities (up to 99 % ee). Control experiments showed that the complementary diastereomers of the products were not accessible by the change of the stereochemistry of the chiralN,N′‐dioxide ligand.
