Abstract:Over the past decade, Gold complexes have emerged as efficient and mild catalysts for the transformation of substrates possessing alkyne functionality into a range of useful scaffolds. These powerful methods have enabled the development of novel approaches for the total synthesis of biologically active natural products by gold catalysis. In this case, we found that the intramolecular nucleophilic addition of a hydroxyl group to a carbon-carbon triple bond, which activated by a gold catalyst, followed by further useful transformation has proven to be an excellent method for rapid construction of structural diversity of molecular scaffolds. The cortistatins are a family of 11 steroidal alkaloids which exhibit significant biological activities. The intriguing biological properties and their low natural abundance have elevated cortistatins to be a typical target for both partial and total synthesis. Up to now, more than a dozen research groups have published approaches directed toward the synthesis of cortistatins, including one semi-synthesis, five total syntheses and five formal syntheses, as well as a number of synthetic studies about the pentacyclic core and some illuminating model studies. One of the biggest challenges for the synthesis of cortistatins is how to construct the unprecedented oxabicyclo[3.2.1] octane ring system which lies within a complex tetracarbocyclic skeleton. In our previous work, we have developed a gold-catalyzed semi-pinacol rearrangement strategy to diastereoselective synthesis of the oxabicyclo[3.2.1] octane ring system. The wide substrate scope as well as the high diastereoselectivity have made us to apply this method into the asymmetric formal synthesis of Cortistatins. Herein, full details about our efforts towards the formal synthesis of cortistatins were described by employing our developed gold-catalyzed cascade reaction to oxabicyclo[3.2.1] octane ring systems. This route is featured with a novel gold-catalyzed cascade reaction involving intramolecular nucleophilic addition of hydroxyl group to the carbon-carbon triple bond, followed by an oxonium ion initiated semi-pinacol-type 1,2-migration to construct the key oxabicyclo[3.2.1] octane skeleton.

Abstract:Nature produces many molecules (so-called natural products) containing compact scaffolds such as cis-anti-cis-configurated 5/5/5 and 5/6/4 tricycles. But other molecules with more strained structures, such as trans-anti-cis-configurated 5/5/5 tricycles have been rarely found in nature. Accessing all these natural and non-natural molecules will strengthen the power of organic synthesis and benefit many fields such as medicinal chemistry, chemical biology that are heavily dependent on the available quantity of new/important molecules. Herein, we report a new strategy to synthesize molecules with cis-anti-cis-configurated 5/5/5 tricycles, 5/6/4 tricycles and trans-anti-cis-configurated 5/5/5 tricyclic skeletons by [5+2+1]/epoxidation/transannular radical cyclization strategy, where challenging cis and trans 5/8 bicycles with both ene and carbonyl functional groups were accessed by Rh-catalyzed [5+2+1] reaction of ene-vinylcyclopropanes and carbon monoxide, followed by epoxidation of the double bond in the 5/8 bicycles, and Ti(III) mediated epoxide/carbonyl cyclization to build target skeletons. This strategy was further applied to the first total synthesis of (+)-antrodiellin B, the asymmetric total synthesis of (−)-hypnophilin and formal synthesis of (−)-coriolin with cis-anti-cis configurated 5/5/5 skeleton (both hypnophilin and coriolin are highly oxidized and have significant biological activities).

Abstract:A convenient approach to the construction of the 5–6–7 tricarbocyclic fused core structure of cyanthiwigins via a Co-mediated Pauson–Khand reaction as a key step has been developed. The cyathane core intermediate obtained by this strategy was used in the concise synthesis of (±)-5-epi-cyanthiwigin I. The developed chemistry paves the way for the total synthesis of structurally diverse cyanthiwigins.

Abstract:A rhodium-catalyzed [5 + 2 + 1] reaction of exocyclic-ene-vinylcyclopropanes (exo-ene-VCPs) and CO has been realized to access challenging tricyclic n/5/8 skeletons (n = 5, 6, 7), some of which are found in natural products. This reaction can be used to build tetracyclic n/5/5/5 skeletons (n = 5, 6), which are also found in natural products. In addition, 0.2 atm CO can be replaced by (CH2O)n as the CO surrogate to achieve the [5 + 2 + 1] reaction with similar efficiency.

Abstract:Cranomycin and jogyamycin, two aminocyclopentitol natural products, possess complex structures and potential medicinal properties. This review describes synthetic studies about the process of making an advanced intermediate of cranomycin and jogyamycin. This highly functionalized intermediate, featuring three contiguous amine-substituted stereocenters, was constructed from cyclopentadiene through a series of reactions including the nitroso Diels-Alder reaction, nitrogen radical cyclization reaction, 1,2-nitrogen migration, and stereoselective nitrogen addition.

Abstract:Spirocyclic pyrrolidinoindolines are privileged scaffolds present in a growing number of alkaloids with potential biological importance. A concise and efficient construction of a tetracyclic spirocyclic pyrrolidinoindoline via gold-catalyzed intramolecular tandem cyclization of indole-ynamide in a single step is reported. Meanwhile, the corresponding tricyclic spirocyclic pyrrolidinoindoline enone was generated in one pot through a key 1,5-hydride shift. This method provides a straightforward entry to structurally diverse polycylic pyrrolidinoindoline alkaloids. Abstract:Report here is the development of a [5 + 2 + 1]/ene strategy for the synthesis of molecules with linear tricyclic 5/5/5 and 6/5/5 skeletons widely found in natural products. Abstract:The polysubstituted and spirocyclic cyclobutanes were efficiently constructed by the utilization of a novel skeletal contraction strategy which only took one-step synthesis from the readily accessible pyrrolidines. The mechanistic studies indicated that a key intermediate N-aminated pyrrolidine was formed initially and subsequent extrusion of a nitrogen via a radical pathway completed the transformation. This practical methodology was further highlighted by the concise, formal synthesis of the cytotoxic natural product piperarborenine B.

Abstract:A remarkable TFA-mediated method for the construction of a biologically interesting tricyclic ketal skeleton was uncovered by starting from a variety of readily available acylphloroglucinol and diacylphloroglucinol substrates. This approach, which mimics a biosynthetic olefin isomerization/hemiacetalization/dehydration/(3 + 3) type cycloaddition sequence through a 2H-furan-1-ium intermediate, establishes a viable synthetic strategy for efficient synthesis of bullataketals analogs.

Abstract:An efficient protocol for the direct transformation of chrornan-4-ones to tricyclic fused pyridines with the skeleton of cassiarins, a family of alkaloids with antimalarial activity, was developed. Also, a general strategy for modular construction of polyheterocydes with diverse natural product-like skeletons was developed by using ketone-alkyne bifunctional substrates. These reactions involved a one-pot cascade oximation of ketones, rhodium-catalyzed C-H activation, and intermolecular/intramolecular alkyne annulations under mild conditions with high atom, step, and redox economy.

Abstract:Euphorikanin A is a diterpenoid possessing a highly congested and unprecedented 5/6/7/3-fused tetracyclic ring skeleton. To access the challenging chemical structure of Euphorikanins, an efficient total synthetic approach is described. The stereoselective synthesis of the core structure of Euphorikanin A has been achieved from a simple dienyne building block, and a domino ring-closing metathesis (RCM) strategy was used for the gram-scale synthesis of the highly strained Euphorikanin A core. This paves the way for the synthesis of structurally diverse Euphorikanins.

Abstract:A new Ru-catalyzed C-H activation/cyclization reaction for the synthesis of 3-C-glycosyl isocoumarins and 2-glycosyl-4H-chromen-4-ones with carbonyl sulfoxonium ylide glycogen are reported. In this catalytic system, benzen acid and its derivatives were reacted with carbonyl sulfoxonium ylide glycogen to yield isocoumarin C-glycosides, whereas 2-hydroxybenzaldehyde substrates were reacted to afford chromoen C-glycosides. These reactions were characterized by mild reaction conditions, broad substrate scope, high functional group compatibility, and high stereoselectivity to yield several high-value isocoumarins and chromones skeleton–containing C-glycosides. The strategy and methods were effectively applied to large scale reaction, and complex natural products late-stage modification.

Abstract:Vilmoraconitine belongs to one of the most complex skeleton types in the C<sub>19</sub>-diterpenoid alkaloids, which architecturally features an unprecedented heptacyclic core possessing a rigid cyclopropane unit. Here, we report the first total synthesis of vilmoraconitine relying on strategic use of efficient ring-forming reactions. Key steps include an oxidative dearomatization-induced Diels-Alder cycloaddition, a hydrodealkenylative fragmentation/Mannich sequence, and an intramolecular Diels-Alder cycloaddition.
