Abstract:A ligand-controlled cobalt-catalyzed regioselective hydroboration/cyclization of 1,6-enynes with HBPin was developed by switching the size of the coordinated side arm to afford alkenylboronates and alkylboronates, respectively. Gram-scale reactions could be easily conducted, which is beneficial for further derivatizations. A primary mechanism was proposed on the basis of substrate-controlled experiments and deuterium experiments.

Abstract:A novel Lewis acid-catalyzed [3+2] intramolecular cross-cycloaddition (IMCC) between aziridine 2,2-diesters and conjugated dienes has been developed. This is the first regiospecific IMCC of intramolecular 1,3-dipolar cycloadditions of azomethine ylides with carbon=carbon double bonds, and supplies a general and efficient strategy for construction of structurally complex and diverse aza-[n.2.1] skeletons. The [3+2]IMCC could be carried out under mild conditions and in gram scale. More importantly, 3-alkyl-substituted aziridines were also successful. The excellent structural diversity, the facile operation and the versatile post-modifications will support the applications of the [3+2]IMCC in natural products synthesis and drugs discovery.

Abstract:Here, we reported a cobalt-hydride-catalyzed Markovnikov-type hydroallylation of terminal alkynes with allylic electrophile to access valuable and branched skipped dienes (1,4-dienes) with good regioselectivity. This operationally simple protocol exhibits excellent functional group tolerance and exceptional substrate scope. The reactions could be carried out in gram-scale with TON (turn over number) up to 1160, and the products could be easily derivatized. The preliminary mechanism of electrophilic allylation of α-selective cobalt alkenyl intermediate was proposed based on deuterium labeling experiment and kinetic studies.

Abstract:Bicyclic carbocycles containing a high fraction of Csp3 have become highly attractive synthetic targets because of the multiple applications they have found in medicinal chemistry. The formal cycloaddition of bicyclobutanes (BCB) with two- or three- atom partners has recently been extensively explored for the construction of bicyclohexanes and bicycloheptanes, but applications to the synthesis of medium-sized bridged carbocycles remained more limited. We report herein the formal (4π+2σ) cycloaddition of BCB ketones with silyl dienol ethers. The reaction occurred in the presence of 5 mol% aluminium triflate as a Lewis acid catalyst. Upon acidic hydrolysis of the enol ether intermediates, rigid bicyclo[4.1.1]octanes (BCOs) diketones could be accessed in up to quantitative yields. This procedure tolerated a range of both aromatic and aliphatic substituents on both the BCB substrates and the dienes. The obtained BCO products could be functionalized through reduction and cross-coupling reactions.

Abstract:closo-Boranes and closo-carboranes, clusters with a closed-cage structure, exhibit remarkable inertness and are unique components of products with applications in areas such as medicinal chemistry, fluorescence and materials science. Herein, we report the first transition metal-catalyzed functionalization of the mono-closo-decaborate cluster [1-CB9H10]- by regioselective B-H activation. Using terminal alkenes bearing a wide range of functional groups, dehydrogenative coupling under mild conditions leads to B2/3-dialkenylated products, as verified by X-ray crystallography and NMR studies. This substitution pattern has not been accessible by other methods and enables the straightforward synthesis of new derivatives of the {CB9} cluster.

Abstract:A cobalt-diphosphine catalyst has been found to promote a formal [3 + 2] cycloaddition reaction between a cyclopropanol and an allene via cyclopropanol ring opening, which affords a 3-alkylidenecyclopentanol derivative with high regio- and diastereoselectivities. The reaction tolerates mono substituted, 1,1-disubstituted, and 1,3-disubstituted allenes and various functional groups. The reaction is proposed to proceed through carbometalation of the allene with a cobalt homoenolate followed by intramolecular carbonyl allylation of the resulting allylcobalt species.

Abstract:A novel Lewis acid catalyzed tandem cyclization reaction of internal alkynols and vinyl azides has been achieved to afford a series of products containing a pyran-based indeno[1,2-c]isochromene scaffold in moderate to high yields. This tandem polycyclization protocol provides a straightforward entry to construct the complex polycyclic skeleton through cycloisomerization, formal [4 + 2] cycloaddition, and an elimination process.

Abstract:Herein, we report an efficient photoinduced iron-catalyzed strategy for cross-couplings of alkyl carboxylic and acrylic acids, which provides a powerful tool for the synthesis of a variety of alkenes with polar functional groups. This novel synthetic methodology can also be applied to the preparation of ketones by using α-keto acids. Mechanistic experiments revealed preliminary mechanistic details. Diverse functionalization could be achieved, which may help streamline the synthesis of complex analogues for drug discovery.

Abstract:Through the combined use of computational (density functional theory) and experimental studies, a new [Rh(CO)(2)Cl](2) catalyzed two-component [5+2+1] cycloaddition of ene-vinylcyclopropanes and CO for the synthesis of fused bicyclic cyclooctenones has been designed and experimentally realized. The key point behind this design is to turn a disfavored reductive elimination of (sp(3))C-Rh-C(sp(3)) to an easier migratory reductive elimination of (sp(3))C-Rh-CO-C(sp(3)) by the introduction of CO. These reactions proceed in good yields for substrates with various tether types and substituents, providing a flexible, convenient, efficient, and stereocontrolled method for the construction of bicyclic cyclooctenones.

Abstract:Catalytic cross-coupling by transition metals has revolutionized the formation of carbon–carbon bonds in organic synthesis. However, the challenge of forming multiple alkyl–alkyl bonds in crowded environments remains largely unresolved. Here we report the regioselective functionalization of olefins with sp 3 -hybridized organohalides and organozinc reagents using a simple (terpyridine)iron catalyst. Aliphatic groups of various sizes are successfully installed on either olefinic carbon, furnishing a diverse array of products with congested cores featuring carbon- or heteroatom-substituted stereocentres. The method enables access to valuable but synthetically challenging C( sp 3 )-rich molecules, including alicyclic compounds bearing multiple contiguous stereocentres, through annulation cascades. Mechanistic and theoretical studies suggest a stepwise iron-mediated radical carbometallation pathway followed by outer-sphere carbon–carbon bond formation, which potentially opens the door to a broader scope of transformations and new chemical space.

Abstract:Catalytic enantioselective functionalization of cyclobutenes constitutes a general and modular strategy for construction of enantioenriched complex cyclobutanes bearing multiple stereogenic centers, as chiral four-membered rings are common motifs in biologically active molecules and versatile intermediates in organic synthesis. However, enantioselective synthesis of cyclobutanes through such a strategy remained significantly limited. Herein, we report a series of unprecedented cobalt-catalyzed carbon-carbon bond forming reactions of cyclobutenes that are initiated through enantioselective carbometalation. The protocols feature diastereo- and enantioselective introduction of allyl, alkynyl, and functionalized alkyl groups. Mechanistic studies indicated an unusual 1,3-cobalt migration and subsequent β-carbon elimination cascade process occurred in the allyl addition. These new discoveries established a new elementary process for cobalt catalysis and an extension of diversity of nucleophiles for enantioselective transformations of cyclobutenes.

Abstract:Cyclopropane is one of the most reactive functionalities owing to its intrinsic ring strain. Transition-metal catalysis and Lewis acid catalysis have been extensively used in ring openings of cyclopropanes; however, Lewis base-catalyzed activation of cyclopropanes remains largely unexplored. Abstract:Transition-metal-catalyzed cross-electrophile coupling has emerged as a reliable method for constructing carbon–carbon bonds. Herein, we report a general method, cobalt-catalyzed reductive alkynylation, to construct C(sp)-C(sp3) and C(sp)-C(sp2) bonds. This presented reaction has a broad substrate scope, enabling the efficient cross-electrophile coupling between alkynyl bromides with alkyl halides and aryl or alkenyl (pseudo)halides. This presented reaction is conducted under mild conditions, tolerating many functional groups, thus suitable for the modification and synthesis of biologically active molecules.

Abstract:A highly enantioselective cobalt-catalyzed desymmetrizing reductive cyclization of alkynyl cyclodiketones has been developed. Under mild reaction conditions by employing HBpin as reducing agent and ferrocene-based PHOX as chiral ligand, a series of polycyclic tertiary allylic alcohols bearing contiguous quaternary stereocenters are achieved in moderate to excellent yields with excellent enantioselectivities (up to 99%). Broad substrate scope and high functional group compatibility are observed in this reaction. A CoH-catalyzed pathway involving alkyne hydrocobaltation followed by nucleophilic addition to C=O bond is proposed. Synthetic transformations of the product are conducted to demonstrate the practical utilities of this reaction.

Abstract:We have developed an efficient annulation of benzamides with allenes using cobalt and photoredox dual catalysis under an oxygen atmosphere. This reaction provides a mild and environmentally friendly method for the construction of isoquinolinone scaffolds in good to excellent yields, demonstrating broad substrate scopes, high regioselectivity, and good functional group compatibility. Notably, the transformation features an alternative strategy for the regeneration of a cobalt catalyst with the aid of Eosin Y. Preliminary mechanistic studies reveal that a radical-mediated cascade annulation is involved in this reaction.
