Transfection of recombinant adenovirus/enhanced green fluorescent protein in adipose tissue-derived stem cells

Abstract:Aim: To investigate the transfection efficiency and effect of adenovirus vector in human adipose tissue-derived stem cells (ADSCs), and verify the gene modification of adenovirus/enhanced green fluorescent protein (EGFP) for in vitro transfection. Methods: The experiment was carried out in the laboratory of Institute of Biological Technique, Southern Medical University from October 2006 to February 2007. With the approval of the hospital ethnic committee, the adipose tissue was from five adult female patients who underwent abdominal liposuction in Department of Plastic Surgery at Nanfang Hospital of Southern Medical University, excluding those of coronal artery disease, hypertension or diabetes. They all orally consented to the use of adipose tissue for research purposes. Human adipose tissues were isolated, and the ADSCs were cultured to the fourth generation and detected by flow cytometry for phenotype identification, then infected with recombinant adenovirus expressing EGFP gene by different multiphcity of infection (MOI=O, 10, 20, 30, 50, 100). The cytotoxicity of adenovirus to ADSCs was evaluated by cell proliferation and cell vitality of in vitro transfection. Results: Human ADSCs samples from 5 patients were successfully cultured, grew well and infected by recombinant adenovirus/EGFP. 1 The cells adhered to the culture plate in 48 hours of primary culture. On the day 13, the cells in shuttle shape coated about 80% of the flask area, and proliferated fast after passage according to 1:2. Three days later, the cells coated the whole bottom of flask. The phenotype of ADSCs at passage 4 were CD29 +, CD34 -, CD44 +, CD106 -, CD133 -, and HLA-DR. 2 The EGFP was initially expressed in the ADSCs 24 hours following transfection. The expression level reached a peak on the day 5. There were still strong expressions of EGFP in the subcultured ADSCs. 3 During the period of the culture, cell proliferation, vitality and differentiation in the transfected and untransfected groups had no statistical difference on the days 1, 3, 5, 7, 9, 11. Conclusion: Human ADSCs can be successfully isolated and cultured. The transfection efficiency of adenovirus to ADSCs is proved to be satisfactory at MOI=50.
