DRAGEN optimizes genomic variant detection

Abstract:Software development for comprehensive variant detection would enhance our understanding of the human genome. Behera et al. introduce a method, dynamic read analysis for genomics (DRAGEN), to identify all types of genomic variant at scale and accuracy on Illumina's whole-genome sequencing platform. The DRAGEN variant calling pipeline improves the accuracy of variant detection and enables more comprehensive insights by implementing optimized concepts, such as multigenome mapping with pangenome references, and variant calling with machine learning-based filtering. DRAGEN enables the detection of the entire landscape of variations including single-nucleotide variations (SNVs), copy number variations (CNVs), structural variations (SVs), short tandem repeats (STRs), and accurate genotyping of clinically relevant genes. The DRAGEN multigenome mapper outperforms existing pangenome implementations in accuracy and speed, and is incorporated for CNV and SV calling, showing substantial improvements in variant detection compared with other state-of-the-art methods. DRAGEN allows the handling of all different variant classes across various human populations when applied to large datasets such as the 1000 Genome Project, providing the potential for comprehensive genome analysis and future research on medically relevant genes.
