Abstract:PurposeEnvironmental endocrine-disrupting chemicals (EDCs) are a mixture of chemical compounds capable to interfere with endocrine axis at different levels and to which population is daily exposed. This paper aims to review the relationship between EDCs and breast, prostate, testicle, ovary, and thyroid cancer, discussing carcinogenic activity of known EDCs, while evaluating the impact on public health.MethodsA literature review regarding EDCs and cancer was carried out with particular interest on meta-analysis and human studies.ResultsThe definition of EDCs has been changed through years, and currently there are no common criteria to test new chemicals to clarify their possible carcinogenic activity. Moreover, it is difficult to assess the full impact of human exposure to EDCs because adverse effects develop latently and manifest at different ages, even if preclinical and clinical evidence suggest that developing fetus and neonates are most vulnerable to endocrine disruption.ConclusionEDCs represent a major environmental and health issue that has a role in cancer development. There are currently some EDCs that can be considered as carcinogenic, like dioxin and cadmium for breast and thyroid cancer; arsenic, asbestos, and dioxin for prostate cancer; and organochlorines/organohalogens for testicular cancer. New evidence supports the role of other EDCs as possible carcinogenic and pregnant women should avoid risk area and exposure. The relationship between EDCs and cancer supports the need for effective prevention policies increasing public awareness.

Abstract:Environmental diseases are increasing globally and the sources of contamination are multiple: indoor and outdoor air, food, cosmetics, drinking and irrigation water.Endocrine-disrupting chemicals (EDCs) are among the causes of the increasing presence of chronic diseases. There are periods of vulnerability—in particular during pregnancy, childhood, and puberty—when these substances can modify the functioning and establishment of major functions such as brain development, the various endocrine systems, the gonads, and the regulation of fatty tissue.Recent studies show the general population's widespread exposure to endocrine disruptors, either persistent (accumulating over time) or non-persistent (but with permanent exposure). However, the consideration of EDCs in clinical practice remains marginal, even though doing so could reduce the risk of diseases linked to environmental factors and positively influence the natural evolution of a chronic disease when it is diagnosed. The aim of this article is to clarify the concepts associated with the exposome and to list the main scientific data attesting to the effects of endocrine disruptors on health in order to draw up clinical, teaching, and research perspectives.

Abstract:Abstract The concept of a threshold of adversity in toxicology is neither provable nor disprovable. As such, it is not a scientific question but a theoretical one. Yet, the belief in thresholds has led to traditional ways of interpreting data derived from regulatory guideline studies of the toxicity of chemicals. This includes, for example, the use of standard “uncertainty factors” when a “No Adverse Effect Level” (or similar “benchmark dose”) is either observed, or not observed. In the context of endocrine-disrupting chemicals (EDCs), this approach is demonstrably inappropriate. First, the efficacy of a hormone on different endpoints can vary by several orders of magnitude. This feature of hormone action also applies to EDCs that can interfere with that hormone. For this reason, we argue that the choice of endpoint for use in regulation is critical, but note that guideline studies were not designed with this in mind. Second, the biological events controlled by hormones in development not only change as development proceeds but are different from events controlled by hormones in the adult. Again, guideline endpoints were also not designed with this in mind, especially since the events controlled by hormones can be both temporally and spatially specific. The Endocrine Society has laid out this logic over several years and in several publications. Rather than being extreme views, they represent what is known about hormones and the chemicals that can interfere with them.

Abstract:An Endocrine Disrupting Chemical (EDC) is any compound that disrupts the function of the endocrine system in humans and is ubiquitous in the environment either as a result of natural events or through anthropogenic activities. Bisphenol A, phthalates, parabens, pesticides, triclosan, polychlorinated biphenyls, and heavy metals, which are frequently found in the pharmaceutical, cosmetic, and packaging sectors, are some of the major sources of EDC pollutants. EDCs have been identified to have a deteriorating effect on the female reproductive system, as evidenced by the increasing number of reproductive disorders such as endometriosis, uterine fibroids, polycystic ovary syndrome, premature ovarian failure, menstrual irregularity, menarche, and infertility. Studying EDCs in relation to women's health is essential for understanding the complex interactions between environmental factors and health outcomes. It enables the development of strategies to mitigate risks, protect reproductive and overall health, and inform public policy decisions to safeguard women's well-being. Healthcare professionals must know the possible dangers of EDC exposure and ask about environmental exposures while evaluating patients. This may result in more precise diagnosis and personalized treatment regimens. This review summarises the existing understanding of prevalent EDCs that impact women's health and involvement in female reproductive dysfunction and underscores the need for more research. Further insights on potential mechanisms of action of EDCs on female has been emphasized in the article. We also discuss the role of nutritional intervention in reducing the effect of EDCs on women's reproductive health. EDC pollution can be further reduced by adhering to strict regulations prohibiting the release of estrogenic substances into the environment.

Abstract:It has been widely demonstrated that endocrine disruptors play a central role in various physiopathological processes of human health. In the literature, various carcinogenic processes have been associated with endocrine disruptors. A review of the molecular mechanisms underlying the interaction between endocrine disruptors and the endometrial cancer has been poorly developed. A systematic review was performed using PubMed®/MEDLINE. A total of 25 in vivo and in vitro works were selected. Numerous endocrine disruptors were analyzed. The most relevant results showed how Bisphenol A (BPA) interacts with the carcinogenesis process on several levels. It has been demonstrated how BPA can interact with hormonal receptors and with different transcription proliferative and antiproliferative factors. Furthermore, the effect of Polycyclic aromatic hydrocarbons on Aryl hydrocarbon receptors was investigated, and the role of flame retardants in promoting proliferation and metastasis was confirmed. The results obtained demonstrate how the mechanisms of action of endocrine disruptors are manifold in the pathophysiology of endometrial cancer, acting on different levels of the cancerogenesis process.

Abstract:Acting as hormone mimics or antagonists in the interaction with hormone receptors, endocrine disrupting chemicals (EDCs) have the potentials of disturbing the endocrine system in sex steroid hormone-controlled organs and tissues. These effects may lead to the disruption of major regulatory mechanisms, the onset of developmental disorders, and carcinogenesis. Especially, among diverse EDCs, xenoestrogens such as bisphenol A, dioxins, and di(2-ethylhexyl)phthalate, have been shown to activate estrogen receptors (ERs) and to modulate cellular functions induced by ERs. Furthermore, they appear to be closely related with carcinogenicity in estrogen-dependant cancers, including breast, ovary, and prostate cancers. In in vivo animal models, prenatal exposure to xenoestrogens changed the development of the mouse reproductive organs and increased the susceptibility to further carcinogenic exposure and tumor occurence in adults. Unlike EDCs, which are chemically synthesized, several phytoestrogens such as genistein and resveratrol showed chemopreventive effects on specific cancers by contending with ER binding and regulating normal ER action in target tissues of mice. These results support the notion that a diet containing high levels of phytoestrogens can have protective effects on estrogen-related diseases. In spite of the diverse evidences of EDCs and phytoestrogens on causation and prevention of estrogen-dependant cancers provided in this article, there are still disputable questions about the dose-response effect of EDCs or chemopreventive potentials of phytoestrogens. As a wide range of EDCs including phytoestrogens have been remarkably increasing in the environment with the rapid growth in our industrial society and more closely affecting human and wildlife, the potential risks of EDCs in endocrine disruption and carcinogenesis are important issues and needed to be verified in detail.

Abstract:Endocrine disruptors are natural or man-made chemicals that interfere with the body's endocrine system leading to hormone synthesis and production defects. These chemicals are categorized as plasticizers and cosmetic chemicals, heavy metals, phytoestrogens, pesticides, detergents, surfactants, and flame retardants. Some of the most common endocrine disruptors are dioxins, bisphenol A, phthalates, perchlorate, perfluoroalkyl, and poly-fluoroalkyl substances (PFAs), phytoestrogens, polybrominated diphenyl ethers (PBDEs), polychlorinated biphenyls (PCB), triclosan, atrazine, lead, arsenic, mercury, organophosphate pesticides, and glycol ethers. Epigenetic alterations such as DNA methylation, histone modification, and miRNA regulation have been observed to play a major role in many diseases such as cancer, neurodegenerative diseases, PCOS, cardiovascular diseases, and various other disorders. In recent times, there has been a focus on endocrine-disrupting chemicals in epigenetic alterations. This review concentrates on estrogen and androgen disrupting effects, placental, and fetal effects, thyroid disrupting effects, and transgenerational effects of endocrine disruptors.

Abstract:Recent decades have seen progress in the identification and quantification of a wide array of chemicals with endocrine-active properties. Exposure to these so-called endocrine-disrupting chemicals (EDCs) has been implicated in an increase in certain adverse health effects, and some new prospective birth cohort studies have yielded suggestive results on these exposure-effect relationships. Major research efforts have focused on the EDC exposure of women of child-bearing age, because of concerns about embryonic and fetal susceptibility to these chemicals. Investigations have shown that mothers and children are exposed to a complex mixture of compounds; therefore, studies on the health impact of EDC exposure should not be limited to the individual effects of single agents but should rather consider the cumulative effects of multiple chemicals. There is considerable political debate about the need for measures to reduce or avoid exposure to EDCs. While a tighter regulation of exposure to EDCs is being implemented, health professionals and public health practitioners should acquire knowledge of the problem, recognize exposure, and warn the general population about the health risks.

Abstract:Despite several modifiable and non-modifiable risk factors of hormone-associated cancers have been established, less heed has been paid to chemicals, those having the potential to thwart the body's normal detox system and affect the endocrine-hormonal pathways. Phthalates are endocrine-disrupting chemicals, most widely manufactured and used indiscriminately in several industries, including processed, ultra-processed and packaged food, single-use plastics, household and personal care/cosmetic products including diapers and electronics. The general population is regularly being exposed to phthalates on contact with these products, especially women and children are most vulnerable. It is therefore highly crucial to monitor and evaluate the biological burden of plasticizing phthalates in humans and understand the potential mechanisms of etiological link between pervasive exposure to phthalates and development of chronic diseases such as cancer through epigenetic and/or genetic alterations. It is also important to identify sustainable and scalable interventions for increasing public awareness, and restricting chronic phthalate exposure to individual and the population at large through relevant policy legislations, particularly in low-income and middle-income countries, such as India.

Abstract:Endocrine-disrupting chemicals (EDCs) increasingly have been a subject of concern and study in the past few decades. These chemicals can interfere directly or indirectly with normal physiology of endocrine system organs or organs under the influence of hormones. EDCs have been shown to cause a range of adverse effects, such as developmental abnormalities, abnormal growth patterns in children, reproductive abnormalities, hormone-sensitive cancers, and alterations in immune function. Some have been shown to contribute to obesity. Most EDCs are synthetically developed compounds that are ubiquitous in food packaging, consumer products, and the environment. They are found in pesticides, herbicides, plastics, solvents, flame retardants, and in the environment as industrial byproducts and waste. These substances are poorly regulated and exposures are not tracked. It is virtually impossible to avoid contact with EDCs in everyday life. Populations at greatest risk of adverse health effects include fetuses, newborns, and pregnant individuals. Comprehensive preconception and prenatal care can help family members and caregivers identify sources of and minimize EDC exposure in newborns and infants. For individuals, the clinical significance of these exposures is unknown and there is no current role for testing. For patients with possible exposure, a pertinent history should be taken and counseling provided to help minimize exposure.

Abstract:We are all exposed to endocrine-disrupting chemicals (EDCs) starting from embryonic life. The fetus and child set up crucial developmental processes allowing adaptation to the environment throughout life: they are extremely sensitive to very low doses of hormones and EDCs because they are developing organisms. Considering the developmental origin of well-being and diseases, every adult organism expresses consequences of the environment in which it developed. The molecular mechanisms through which the main EDCs manifest their effects and their potential association with endocrine disorders, such as diabetes, obesity, thyroid disease and alteration of adrenal hormones, will be reviewed here. Despite 40 years having passed since the first study on EDCs, little is yet known about them; therefore, our purpose is to take stock of the situation to establish a starting point for further studies. Since there is plenty of evidence showing that exposure to EDCs may adversely impact the health of adults and children through altered endocrine function-suggesting their link to endocrinopathies-it is essential in this context to bear in mind what is already known about endocrine disruptors and to deepen our knowledge to establish rules of conduct aimed at limiting exposure to EDCs' negative effects. Considering that during the COVID-19 pandemic an increase in endocrine disruptor effects has been reported, it will also be useful to address this new phenomenon for better understanding its basis and limiting its consequences.

Abstract:Endocrine disrupting chemicals are a group of pollutants that can affect the endocrine system and lead to diseases and dysfunctions across the lifespan of organisms. They are omnipresent. They are in the air we breathe, in the food we eat and in the water we drink. They can be found in our everyday lives through personal care products, household cleaning products, furniture and in children’s toys. Every year, hundreds of new chemicals are produced and released onto the market without being tested, and they reach our bodies through everyday products. Permanent exposure to those chemicals may intensify or even become the main cause for the development of diseases such as type 2 diabetes, obesity, cardiovascular diseases and certain types of cancer. In recent years, legislation and regulations have been implemented, which aim to control the release of potentially adverse endocrine disrupting chemicals, often invoking the precautionary principle. The objective of this review is to provide an overview of research on environmental aspects of endocrine disrupting chemicals and their effects on human health, based on evidence from animal and human studies. Emphasis is given to three ubiquitous and persistent groups of chemicals, polychlorinated biphenyls, polybrominated diphenyl ethers and organochlorine pesticides, and on two non-persistent, but ubiquitous, bisphenol A and phthalates. Some selected historical cases are also presented and successful cases of regulation and legislation described. These led to a decrease in exposure and consequent minimization of the effects of these compounds. Recommendations from experts on this field, World Health Organization, scientific reports and from the Endocrine Society are included.

Abstract:Endocrine-disrupting chemicals (EDCs) have significant impacts on biological systems, and have been shown to interfere with physiological systems, especially by disrupting the hormone balance. During the last few decades, EDCs have been shown to affect reproductive, neurological, and metabolic development and function and even stimulate tumor growth. EDC exposure during development can disrupt normal development patterns and alter susceptibility to disease. Many chemicals have endocrine-disrupting properties, including bisphenol A, organochlorines, polybrominated flame retardants, alkylphenols, and phthalates. These compounds have gradually been elucidated as risk factors for many diseases, such as reproductive, neural, and metabolic diseases and cancers. Endocrine disruption has been spread to wildlife and species that are connected to the food chains. Dietary uptake represents an important source of EDC exposure. Although EDCs represent a significant public health concern, the relationship and specific mechanism between EDCs and diseases remain unclear. This review focuses on the disease-EDC relationship and the disease endpoints associated with endocrine disruption for a better understanding of the relationship between EDCs-disease and elucidates the development of new prevention/treatment opportunities and screening methods.

Abstract:Endocrine Disrupting Chemicals (EDCs) or Endocrine Disruptors are unique assemblage or cluster of emerging pollutants as they affect the synthesis, release and transport of hormones. EDCs have been associated with a diverse array of health issues and diseases. EDCs can alter the endocrine system and are involved in carcinogenesis and inducing poly-cystic ovary syndrome (PCOS). The objective of this article is to furnish an outline of research on environmental aspects of EDCs and their effects on human health specially on cancer and PCOS based on evidence from animal and human studies. EDCs include natural compounds such as phytoestrogens and various synthetic chemicals which are utilized by the chemical, agriculture, cosmetic and food industries. Several EDCs may work as carcinogens and causes initiation and advancement of cancer. Uterine and ovarian cancers of female have been associated with exposure to EDC. Bisphenol-A (BPA), an EDC which is found in plastic bottles, household materials, canned food, waste water, beverage containers and thermal paper, can increase risk of breast cancer. Even low levels of BPA exposure may poses threat of prostate cancer in men. Testicular cancer and thyroid cancer could be influenced by EDCs. Comprehensive studies have been conducted by many researchers in the light of toxicity pattern of EDC that render interpretation of impact of EDCs on development of cancer and PCOS inducing capacity in female reproductive system. Abstract:Obesity represents a global health concern, affecting individuals of all age groups across the world. The prevalence of excess weight and obesity has escalated to pandemic proportions, leading to a substantial increase in the incidence of various comorbidities, such as cardiovascular diseases, type 2 diabetes, and cancer. This chapter seeks to provide a comprehensive exploration of the pathways through which endocrine-disrupting chemicals can influence the pathophysiology of obesity. These mechanisms encompass aspects such as the regulation of food intake and appetite, intestinal fat absorption, lipid metabolism, and the modulation of inflammation. This knowledge may help to elucidate the role of exogenous molecules in both the aetiology and progression of obesity.

Abstract:Exposure to Endocrine Disrupting Chemicals (EDC) has been linked with several adverse outcomes. In this review, we examine EDCs that are pervasive in the environment and are of concern in the context of human, animal, and environmental health. We explore the consequences of EDC exposure on aquatic life, terrestrial animals, and humans. We focus on the exploitation of genomics technologies and in particular whole transcriptome sequencing. Genome-wide analyses using RNAseq provides snap shots of cellular, tissue and whole organism transcriptomes under normal physiological and EDC perturbed conditions. A global view of gene expression provides highly valuable information as it uncovers gene families or more specifically, pathways that are affected by EDC exposures, but also reveals those that are unaffected. Hypotheses about genes with unknown functions can also be formed by comparison of their expression levels with genes of known function. Risk assessment strategies leveraging genomic technologies and the development of toxicology databases are explored. Finally, we review how the Adverse Outcome Pathway (AOP) has exploited this high throughput data to provide a framework for toxicology studies.

Abstract:Endocrine disruptors (EDs) are defined as environmental pollutants capable of interfering with the functioning of the hormonal system. They are environmentally distributed as synthetic fertilizers, electronic waste, and several food additives that are part of the food chain. They can be considered as obesogenic compounds since they have the capacity to influence cellular events related to adipose tissue, altering lipid metabolism and adipogenesis processes. This review will present the latest scientific evidence of different EDs such as persistent organic pollutants (POPs), heavy metals, “nonpersistent” phenolic compounds, triclosan, polybrominated diphenyl ethers (PBDEs), and smoke-derived compounds (benzo -alpha-pyrene) and their influence on the differentiation processes towards adipocytes in both in vitro and in vivo models.
