Abstract:Myrica rubra pomace accounts for 20% of the fruit’s weight that is not utilized when it is juiced. The pomace contains bioactive phenolic substances such as anthocyanins and flavonoids. To improve the utilization value of Myrica rubra pomace, an optimized extraction method for the residual polyphenols was developed using response surface methodology (RSM). The resulting extract was analyzed by high performance liquid chromatography (HPLC), and the in vitro hypoglycemic activity and antioxidant activity of the polyphenolic compounds obtained were also investigated. The optimum extraction conditions (yielding 24.37 mg·g−1 total polyphenols content) were: extraction temperature 60 °C, ultrasonic power 270 W, ethanol concentration 53%, extraction time 57 min, and solid to liquid ratio 1:34. Four polyphenolic compounds were identified in the pomace extract by HPLC: myricitrin, cyanidin-O-glucoside, hyperoside, and quercitrin. DPPH and hydroxyl radical scavenging tests showed that the Myrica rubra polyphenols extract had strong antioxidant abilities. It is evident that the residual polyphenols present in Myrica rubra pomace have strong hypoglycemic activity and the juiced fruits can be further exploited for medicinal purposes.

Abstract:The chemical constituents of Plantaginis Semen with hypoglycemic effect was investigated in this paper. The previous results of the in vivo hypoglycemic effect showed that 60% ethanol extract of Plantaginis Semen decreased the levels of FBG and improved the glucose tolerance in high fat diet(HFD)-induced diabetic C57BL/6 mice. Then, in the present study, the above potential bioactive extract was separated and purified by silica gel, ODS, Sephadex LH-20 column chromatography, medium pressure liquid chromatography(MPLC)and preparative HPLC. The structures of isolated compounds were identified by physicochemical properties and spectral analyses. Eight compounds were obtained and identified as 4, 4a, 5, 7a-tetrahydro-7-(hydroxymethyl)cyclopenta[c]pyran-3(1H)-one(1), iridolactone(2), pedicularislacton(3), rehmaglutin C(4), geniposidic acid(5), p-hydroxylphenylglycerol(6), 1, 2-benzenediol-4-(2-hydroxyethyl)(7), and 3-buten-2-one-4-[3-(β-D-glucopyranosyloxy)-4-hydroxyphenyl](8). Among them, compounds 1-5 were iridoids, and 6-8 were phenolic acids. Compound 1 was a new natural product, and compounds 2-4, 6 and 8 were isolated from the Plantaginaceae family for the first time.

Abstract:AIM:To investigate the bioactive compounds from Arctium lappa.METHODS:The compounds were obtained by various chromatographic methods including silica gel and sephadex LH-20 and their structures were elucidated on the basis of physical parameters and spectral data including IR,UV,NMR and MS.RESULT:Eight compounds were determined as matairesinol(Ⅰ),arctigenin(Ⅱ),lappaol A(Ⅲ),lappaol F(Ⅳ),lappaol C(Ⅴ),arctignan E(Ⅵ),arctiin(Ⅶ)and lappaol H(Ⅷ).CONCLUSION:All the compounds from the active fraction of Arctium lappa are lignans which might be associated with its hypoglycemic activities.

Abstract:The constituents were isolated and purified by the silica gel and semi-preparative HPLC, and their structures were elucidated by NMR spectral and MS data. Fifteen compounds were isolated from the ethyl acetate fraction of 95% ethanol extract from the leaves of Garcinia xanthochymus, and identified as 5, 7, 4'-trihydroxy-6-(3-hydroxy-3-methylbutyl)-flavone(1), 1,5-dihydroxy-3-methoxyxanthone(2), 1, 3-dimethoxy-5-hydroxy xanthone(3), kaempferol(4),(2S,3S)-trans-dihydrokaempferol(5), 3, 24, 25-trihydroxytirucall-7-ene(6), 4-hydroxycinnamic acid(7), isovanillic acid(8),(Z)-2-(2,4-dihydroxy-2, 6, 6-trimethylcyclohexylidene)acetic acid(9), volkensiflavone(10), morelloflavone(11), 3, 8″-biapigenin(12), bilobetin(13), fukugiside(14), GB2a glucoside(15). Compound 1 is a new compound, compounds 5, 6, 9 and 13 are isolated from the genus Garcinia for the first time, and compounds 4, 7-8, 10-12, 14 and 15 are firstly found from this plant. α-Amylase inhibitory activities of 10 compounds were determined using starch azure as the substrate, and the results show that compound 13 has the inhibitory activities against α-amylase, IC₅₀ values of compound 13 and acarbose are 8.12, 4.32 μmol•L⁻¹ respectively.

Abstract:Twenty compounds were obtained from Glycyrrhiza glabra L., which were divided as fatty alcohols (1 and 5), sterols (2, 4 and 8), triterpenes (3, 6 and 7), flavones (9, 12-15), coumarins (10, 11, 19 and 20) and stilbenes (16-18). The chemotaxonomic significance of G. glabra and the other species among Fabaceae was also discussed. As a result, genus Glycyrrhiza may have close relationships with genera Caesalpinia and Dalbergia, and Trib. Galegeae may have close relationships with Caesalpinieae, Desmodieae, Dipterygeae and Millettieae.

Abstract:Objective:To investigate the chemical constituents of Glechoma longituba.Methods:The dried herbs of Glechoma longituba were powdered and extracted with 95 ％ ethanol,then partitioned with petroleum ether,ethyl acetate and n-butanol.The ethyl acetate fraction was separated by silica gel,Sephadex LH-20 and preparative HPLC.In additional,the structures of isolation were identified on the basis of their spectral data and physicochemical properties.Results:Thirteen compounds were respectively obtained and identified as apigenin-7-O-glucoside (1),genkwanin (2),oleanolic acid (3),4-hydroxy-2-methyl-phenyl-acetonitrile (4),acacetin (5),grevillic acid (6),caffeic acid(7),ferulic acid(8),apigenin(9),quercetin(10),5-hydroxymethylfurfural(11),chlorogenic acid(12) and formononetin(13).Conclusion:Compounds 4,6,11 ～ 13 are isolated from this plant for the first time.

Abstract:Helicteres angustifolia L. (H. angustifolia), a well‐known traditional Chinese medicine, has been demonstrated to have hypoglycemic activity. We found that the EtOAc extract of H. angustifolia (HAEF) showed stronger α‐glucosidase inhibitory activity than that of positive control. Furthermore, the hypoglycemic activity of HAEF was evaluated in streptozotocin (STZ)‐induced type 2 diabetes mellitus (T2DM) rats. The results demonstrated that HAEF reduced the drinking quantity, feeding quantity, and controlled weight loss in diabetic rats. Besides, the fasting blood glucose (FBG), viscera index, and the area under time‐blood glucose curve (AUC) were significantly decreased, and the oral glucose tolerance was also improved after 5 weeks. Then, the high‐performance liquid chromatography with quadrupole time of flight tandem mass spectrometry (HPLC‐Q‐TOF‐MS/MS) method was performed for qualitative analysis of the chemical constituents in HAEF. Twenty‐one compounds were identified from in HAEF. Four compounds were further isolated from HAEF and subjected to α‐glucosidase inhibition experiments. At the end, molecular docking was empolyed simulate the interaction of three compounds with α‐glucosidase. This is the first report on major hypoglycaemic components has been identified in the roots of H. angustifolia. These findings provide a material basis for the use of H. angustifolia in the treatment of diabetes.

Abstract:To study the chemical constituents in the twigs and leaves of Melodinus hemsleyanus. The chemical constituents were isolated by silica gel, ODS and Sephadex LH-20 column chromatographies. Their structures were determined by chemical and spectroscopic methods. Fourteen compounds were isolated from the 95% ethanol extract of the twigs and leaves of M. hemsleyanus. Their structures were identified as 11-hydroxytabersonine (1), venalstonid (2), sandine (3), oleanolic acid (4), ursolic acid (5), betulin (6), (+)-pinoresinol (7), (-)-syringaresinol (8), 8-hydroxypinoresinol (9), (-)-latifolin (10), cirsilineol (11), loganin (12), dibutylterephthalate (13), and beta-sitosterol (14). Compounds 4-14 were obtained from this plant for the first time.

Abstract:AIM To study chemical constituents of the active fractions of Yanning Granules(Commelina communis,Hedyotis diffusa,etc.).METHODS The isolation was performed on chromatographic techniques,and compounds were identified with 1H-NMR,13C-NMR and their physical-chemical properties.RESULTS Seven compounds were isolated and identified,including five flavonoids,one triterpenoid and one aromatic acid,identified as oleanolic acid(1),apigenin(2),luteolin(3),quercetin(4),kaempferol(5),rutin(6),vanillic acid(7).CONCLUSION All of these seven compounds are obtained from Yanning Granules for the first time.

Abstract:The chemical constituents from polarity part in the roots of Angelica dahurica var. formosana cv. Chuanbaizhi were studied in this paper. The compounds were separated and purified by repeated column chromatographic methods on silica gel and HPLC, and the chemical structures of compounds were determined by spectral data analyses. Fourteen compounds were obtained and identified as tert-O-β-D-glucopyranosyl-(R)-byakangelicin (1), (2"S) -3"-O-β-D-glucopyranosyl-oxypeucedanin hydrate (2), marmesinin (3), sec-O-β-D-glucopyranosyl-byakangelicin (4), isofraxidin-7-O-β-D-glucopyranoside (5), benzyl-O-β-D-glucopyranoside (6), 8-O-β-D-glycopyranosylxanthotoxol (7), prenyl-O-β-D-glucopyranoside (8), scopolin (9), (2' R) -5'-hydroxymarmesin-5'-O-β-D-glucopyranoside (10), (2'S,3'R) -3'-hydroxymarmesinin (11), skimmin (12), benzyl-O-β-D-apiofuranosyl-(1"--> 6')-β-D-glucopyranoside (13), and decuroside IV (14). Abstract:Objective To study on the chemical constituents of the mycelia of endophytic Guignardia mangiferae from Smilax glabra.Methods The compounds were isolated and purified by various chromatographic methods such as silica gel,reverse silica gel,sephadex-LH20 and preparative TLC.Their structures were determined extensive analysis of their spectroscopic data as well as by comparison with literature reports.Results Seven compounds were isolated and identified as cerebroside D(1),adenosine(2),(22E,24R)-24-methylcholesta-5,22-diene-3β-ol(3),ergosterol peroxide(4),(22E,24R)-5α,8α,-epidioxyergosta-6,9,22-triene-3β-ol(5),22E,24R-5α,6α-epoxyergosta-8,22-diene-3β,7α-diol(6) and cerevisterol(7).Conclusion All the compounds are isolated from the genus of Guignardia for the first time.

Abstract:A water soluble extract of M. charantia Var. abbreviata Ser. and its hypoglycemic activity are studied. Cut-off membranes are used to separate the active extract into two fractions which are orally administrated to the alloxan-induced diabetic rats for their hypoglycemic activity. The results show that the fraction of molecular weight less than 10 kdal has higher hypoglycemic activity than the one of molecular weight more than 10 kdal. And the fraction of molecular weight less than 10 kdal is very obvious.

Abstract:Extensive pharmacological research has demonstrated that Clerodendranthi Spicati Herba has an obvious anti-hyperglycemic effect via alpha-glucosidase inhibitory activity. However, the anti-hyperglycemic active fraction and its metabolic behavior in vivo have not been elaborated clearly. In this study, ultra-high-performance liquid chromatography coupled to quadrupole time of flight tandem mass spectrometry with data filtering strategy, including mass defect screening, diagnostic product ions and neutral loss identification, was established for chemical and metabolic profiling of anti-hyperglycemic active fraction from Clerodendranthi Spicati Herba. A total of 28 methoxylated flavonoids and 61 diterpenoids were rapidly identified. Four main known methoxylated flavonoids were purified and unambiguously identified by nuclear magnetic resonance analysis. Thirty-one absorbed diterpenoids, 12 absorbed methoxylated flavonoids, and 56 methoxylated flavonoids metabolites were identified in rat plasma, urine, bile, and feces after oral administration of anti-hyperglycemic active fraction. The methoxylated flavonoids were predominantly metabolized by demethylation, sulfation, and glucuronidation. Glucuronidation metabolites found in bile and urine after demethylation were dominant metabolites. Diterpenoids were absorbed into the blood mainly in the form of prototypes and excreted through bile and urine. These results indicated that methoxylated flavonoids and diterpenoids were responsible for alpha-glucosidase inhibitory activity, which might provide novel drug candidates for the management of diabetes mellitus.
