Abstract:A series of quinoline-based photo-removable protecting (caging) groups were synthesized for the development of new chemical tools to photo-regulate bioactive molecules in living cells and tissues with improved properties. Compared with the recently developed 8-bromo-7-hydroxyquinolinyl (BHQ) chromophore, change of the bromine substituent to a pyridine group led to a new photo-labile group (3′-PyHQ) with an increased water solubility, a lower self-fluorescence, and a higher photolysis efficiency. It was proposed that the replacement of a halogen group by a pyridine-like heterocycle may provide a general strategy to improve the existing photo-caging groups.

Abstract:Here, a redox-neutral palladium-catalyzed photo-induced radical cascade domino Heck reaction of N -aryl acrylamide with vinyl arenes is described. A diverse range of bioactive oxindoles, featuring an all-carbon quaternary center, were synthesized. The reaction is proposed to proceed via an open-shell intermediate and occurs under mild reaction conditions, exhibiting excellent functional group tolerance. Importantly, the synthesized products can be readily transformed into biologically active molecules, including (±)-physostigmine and (±)-physovenine.

Abstract:Indolocarbazole alkaloids and their derivatives were discovered to have potent protein kinase and topoisomerase I inhibitory activities. Disclosed herein is the photochemical synthesis of the indolocarbazole ring system from N-allenyl-2-iodoanilines. The tandem protocol included visible-light-mediated 5-exo-trig radical cyclization and subsequent radical dimerization, followed by acid-promoted deprotection and intramolecular Mannich cyclization. This strategy showed exceptional functional group tolerance and was successfully applied in the concise synthesis of natural products tjipanazoles B and D.

Abstract:Three series of compounds were tested as photocleavable protective groups (photocages), which are derivatives of 1,2-substituted aminonaphthalene 4 , 1,3-substituted aminonaphthalene 5 , or 5,6-substituted aminoquinolines 6 . Their photophysical properties and photochemical reactivity in releasing alcohols and carboxylic acids were investigated. Moreover, mechanism of photochemical reaction was studied computationally and by transient absorption spectroscopy with time-resolution from femtoseconds to milliseconds. Photocages 4 and 5 are generally fluorescent molecules with high Φ F , whereas quinoline 6 is weekly fluorescent due to photoinduced charge transfer. Derivatives 4 release alcohol with low quantum yield (release of methanol from 4d is Φ R = 0.00010 ± 0.00005), but upon prolonged irradiation can be applied in organic synthesis to deprotect both aliphatic and aromatic alcohols. On the contrary, photocages 5 undergo relatively efficient release of carboxylic acids, (release of acetic acid from 5b is Φ R = 0.22 ± 0.01), in accord with the meta -effect in photochemistry, which is our main discovery that is based on the rational design of the chromophore. The photochemical reaction mechanism involves homolytic cleavage on the singlet excited state surface and formation of radicals which were detected by transient spectroscopy. The meta -effect did not promote the heterolytic cleavage as it was the case with aniline chromophores. The subsequent electron transfer delivers naphthalenmethyl carbocation and released carboxylates. The applicability of photocages 5 in organic synthesis was demonstrated on aliphatic and aromatic carboxylic acids. The potential use in biology was suggested due to their very weak photoinduced antiproliferative effect, which was essayed on three human cancer cell lines (MCF-7 breast carcinoma, HCT 116 colon carcinoma, and H460 lung carcinoma).

Abstract:The development of light-responsive molecular tools enables spatiotemporal control of biochemical processes with superior precision. Amongst these molecular tools, photolabile caging groups are employed to prevent critical binding interactions between a bioactive molecule and its corresponding target. Only upon irradiation with light, the bioactive is released in its 'active' form and is now readily available to bind to its target. Coumarin-derived caging groups constitute one of the most popular classes of photolabile protecting groups, due to their facile synthetic accessibility, ease of tuning photophysical properties via structural modification and rapid photolysis reactions. Herein, we highlight the recent progress made on the development of coumarin-derived caging groups, in which the red-shifting of absorption spectra, improving aqueous solubility and tailoring sub-cellular localisation has been of particular interest.

Abstract:A bis(cycloocta-1,5-diene)nickel-catalyzed highly regio- and (E)-stereoselective hydrocarboxylation of 2-alkynylanilines under very mild conditions has been developed to afford (E)-[2-(o-aminophenyl)]acrylic acids, which could easily be converted to (E)-3-alkylidene-2-indolinones with important potential bioactivity. The stereoselective syntheses of two biologically active molecules, (E)-3-benzylidene-2-indolinone (chemo-preventive potential in inducing NQO1 activity) and (E)-3-(3-methylbutylidene)-2-indolinone (a natural product isolated from Cimicifuga foetida with cytotoxic activity against HL-60 cells) are presented as examples.

Abstract:A new photoinduced acylation/arylation of N‐(arylsulfonyl)acrylamide with acyl oxime esters through a nitrogen‐centered radical protocol has been disclosed for efficient preparation of functionalized indolin‐2‐ones, which proceeds through a cascade acyl radical addition of carbon‐carbon double bond, aryl migration, desulfonylation, and intramolecular cyclization sequence. This redox‐neutral strategy features excellent selectivity, good substituent tolerance, and wide substrate scope.

Abstract:A series of C‐8 substituted indeno[1,2‐g]coumarin‐based photoremovable protecting groups (PPGs) were synthesized. para‐Substituted benzoic acids were employed as leaving groups to evaluate their photolytic efficiency. Substitution of phenyl groups was proved to have negative impacts on photochemical properties of the PPGs, including but not limited to: retarded photolysis course, decreased uncaging quantum yield, and unsatisfactory cargo release yield. Electron‐donating diethylamino substituted PPG 3d, a structural analogue of the widely used 7‐diethylaminocoumarin PPG (DEACM), exhibited red‐shifted absorption maximum and improved optical properties. Photochemical characterization revealed that PPG 3d not only showed comparable photolytic efficiency to DEACM at 365 nm and 405 nm, but also demonstrated superior sensitivity towards 465 nm wavelength, to which DEACM is unable to absorb and therefore, non‐responsive. The >450 nm photosensitivity makes 3d a complement to DEACM for long wavelength excitation and a promising PPG for biological applications.

Abstract:A series of quinoline-based photo-removable protecting groups for photo-regulation of thrombin aptamer (HD1) activity were synthesized with improved caging and uncaging efficiency. Among them, 8-bromo-2-diazomethyl-7-hydroxyquinolinyl (BHQ-diazo, 1) chromophore was found to cage the HD1 with highest caging and restoration efficiency. Moreover, on the basis of the RP-HPLC and SPR analysis, BHQ was demonstrated to regulate HD1s specific affinity to target molecule with 3-fold photolysis sensitivity and about 40% percent higher uncaging efficiency than Bhc (6-bromo-7-hydroxycoumarin-4-ylmethyl) group. It was proposed that the development and use of quinoline derivative may provide a general strategy to photo-regulate oligonucleotide's activity with improved caging and uncaging efficiencies by the convenient non-site-specific caging method.

Abstract:The direct C(sp3)-H functionalization reactions enable the development of green and streamlined synthetic routes for structurally complex molecules by circumventing the pre-activation of the substrates. Herein, we disclose an atom- and step-economic synthetic approach for rapid access to both α-(2-indolyl) ketones and α-(3-indolyl) ketones, which are privileged structures in bioactive molecules and organic synthesis, however, generally prepared by bespoke multi-step strategies. The reaction proceeded via direct acylation of indole benzylic C(sp3)-H bond under the NHC and photocatalyst catalysis. The mild conditions, broad substrate scope, and predictable regioselectivity among multiple potential reactive sites enable efficient access to α-indolyl ketones from readily available 2- or 3-alkyl substituted indoles. Furthermore, the competitive reaction at the 2- or 3-benzylic position of the 2,3-dialkyl substituted indoles was systematically studied. Further transformation and bioactivities studies of the obtained α-(2- or 3-indolyl) ketones highlight the potential utility of this method.

Abstract:The acyclic alkyl aromatic ketones, 2-isobutyl-4-methyl-1-arylpentan-1-one derivatives were designed and synthesized for photochemical investigations. Irradiation of such compounds in acetonitrile solution led to the Yang cyclization and Norrish type II cleavage photoproducts with a ratio of 1:1, whereas the reaction conducted in the solid state using the ionic chiral auxiliary method led to only the Yang cyclization product with as high as 99% ee. Furthermore, the conformational transitions were first observed in the reaction.

Abstract:In this Letter, a pair of novel naphthalimide derivatives with long-wavelength emission (>600 nm) and larger Stokes shift (>140 nm) have been developed through the photochemical cycloaromatization, in which intramolecular radical-induced 1,3-aromatic hydrogen transfer might be occurred. Abstract:Development of a new catalytic and straightforward strategy to construct C–N bonds is playing a pivotal role in synthetic chemistry. Here, we report a photocatalysed protocol to access direct C–H amidation of indoles, enabled by a rationally designed tert -butyl alkyl((perfluoropyridin-4-yl)oxy)carbamate. A series of biologically important aminoindoles were prepared under mild conditions with excellent regioselectivity and broad substrate scope.

Strategies based on nido-carborane embedded indole fluorescent polymers: their synthesis, spectral properties and cell imaging studies

Abstract:The continuous preparation scheme EPO-Poly-indol-nido-carborane (E-P-INDOLCAB), L100-55-Poly-indol-nido-carborane (L-P-INDOLCAB), RS-Poly-indol-nido-carborane (S-P-INDOLCAB), and RL-Poly-indol-nido-carborane (R-P-INDOLCAB) were used to prepare the four types of acrylic resin-coated nido-carborane indole fluorescent polymers. After testing their spectral properties and the fluorescence stability curve trend at various acidic pH values (3.4 and 5.5, respectively), L-P-INDOLCAB and S-P-INDOLCAB were determined to be the best polymers. The stable states of the two polymers and the dispersion of the nanoparticles on the system's surface during Atomic Force Microscope (AFM) test are shown by the zeta potentials of -23 and -42 mV. The dispersion of nanoparticles on the system's surface and the stable condition of the two polymers were examined using zeta potential and atomic force microscopy (AFM). Transmission electron microscopy (TEM) can also confirm these findings, showing that the acrylic resin securely encases the interior to form an eyeball. Both polymers' biocompatibility with HELA cells was enhanced in cell imaging, closely enclosing the target cells. The two complexes displayed strong inhibitory effects on PC-3 and HeLa cells when the concentration was 20 ug/mL, as validated by subsequent cell proliferation toxicity studies.

Abstract:The introduction of aryl groups and/or alkyl side chains to the periphery of a conjugated skeleton can modulate the pattern of molecular aggregation in solution and facilitate its aggregation-induced emission (AIE) performance. In this study, novel three compounds of 4,4′-binaphthalimidyl derivatives (BNIC n ) were synthesized successfully in which two 1,8-naphthalimide (NI) units were connected by a rotatable single bond at their 4-sites, and two carboxyalkyl chains (number of carbon atoms in the carboxyalkyl chains ( n ) = 2, 6, and 11) were linked with the nitrogen atoms of NI. BNIC n has weakly fluorescent in dilute solutions but become highly emissive in aggregate states. Interestingly, they exhibited carboxyalkyl-dependent AIE properties. The AIE performance of BNIC 2 with the shortest carboxyalkyl chain is the strongest among those of the three compounds. The aggregation was also confirmed by the dynamic light scattering (DLS) tests. Moreover, the fluorescence intensities enhanced with the increasing viscosity of the solutions, verifying that the restriction of intramolecular motions (RIM) leads to the occurrence of AIE. Furthermore, the interactions such as π-π stacking of NI groups, and the intermolecular hydrogen-bonding of the carboxyl groups at the terminal of the side chains of BNIC n was respectively verified by 1 H NMR and FTIR. Thus, a possible self-assemble mode of BNIC n is that the stacking NI segments of BNIC n are located by the intermolecular hydrogen bonds of carboxyl groups at the terminal of the side chains of BNIC n enacting an anchor-like role during aggregation process For BNIC 2 with carboxymethyl group, the shortest spacer of methylene group resulted in more effectively restriction for the intramolecular vibration and rotation, leading to a more efficient AIE performance. Meanwhile, BNIC 11 can form thermal-reversible fluorescence hydrogel with ethylenediamine as well as BNIC 2 can be applied in cellular imaging. Thus, an effective strategy to achieve and adjust the AIE performance of NI derivatives is proposed.

Abstract:Photoaffinity labeling is a widely used technique for studying ligand-protein and protein-protein interactions. Traditional photoaffinity labels utilize nonspecific C-H bond insertion reactions mediated by a highly reactive intermediate. Despite being the most widely used photoaffinity labels, diazirines exhibit limited compatibility with downstream organic reactions and suffer from storage stability concerns. This study introduces oxadiazolines as innovative and complementary photoactivatable labels for addition to the toolbox and demonstrates their application in vitro and through in cellulo labeling experiments. Oxadiazolines can be easily synthesized from ketone moieties and cleaved with 302-330 nm light to cleanly liberate a diazo reactive moiety that can covalently modify nucleophilic amino acid residues. Notably, oxadiazolines are compatible with various organic reaction conditions and functional groups, allowing for the exploration of a large chemical space. Several known inhibitors featuring the oxadiazoline functionality were prepared without affecting their binding affinity. Furthermore, we confirmed the ability of oxadiazolines to form covalent bonds with proteins upon UV-irradiation, both in vitro and in cellulo, yielding comparable results to those of the matched diazirine compounds.
Competition between cyclization and unusual Norrish type I and type II nitro-acyl migration pathways in the photouncaging of 1-acyl-7-nitroindoline revealed by computations

Abstract:Abstract The 7-nitroindolinyl family of caging chromophores has received much attention in the past two decades. However, its uncaging mechanism is still not clearly understood. In this study, we performed state-of-the-art density functional theory calculations to unravel the photo-uncaging mechanism in its entirety, and we compared the probabilities of all plausible pathways. We found competition between a classical cyclization and an acyl migration pathway, and here we explain the electronic and steric reasons behind such competition. The migration mechanism possesses the characteristics of a combined Norrish type I and a 1,6-nitro-acyl variation of a Norrish type II mechanism, which is reported here for the first time. We also found negligible energetic differences in the uncaging mechanisms of the 4-methoxy-5,7-dinitroindolinyl (MDNI) cages and their mononitro analogues (MNI). We traced the experimentally observed improved quantum yields of MDNI to a higher population of the reactants in the triplet surface. This fact is supported by a more favorable intersystem crossing due to the availability of a higher number of triplet excited states with the correct symmetry in MDNI than in MNI. Our findings may pave the way for improved cage designs that possess higher quantum yields and a more efficient agonist release.

Abstract:Herein, we have developed a sequential visible-light-promoted Wolff rearrangement of 1-diazonaphthalen-2(1 H )-ones, followed by capturing the in situ generated ketene intermediates with various alcohols, producing diverse 1 H -indene-3-carboxylates in moderate to good yields under mild reaction conditions. The broad substrate scope, high functional group tolerance, and robust conditions make the resulting derivative a versatile platform for the synthesis of plenty of bioactive molecules.

Abstract:3-Acyl indoles play important roles in both organic synthesis and diverse types of functional molecules. Herein, a facile nitrogen heterocyclic carbene (NHC) and photocatalyst cooperatively-catalyzed 3-acylation of indoles was disclosed. The reaction proceeded via radical cross-coupling of indole-based aryl radical cations with NHC-bound ketyl radical species, which are less explored in radical NHC catalysis. The reaction exhibits mild reaction conditions, broad substrate scope, and good functional group tolerance. Mechanistic studies support our proposed reaction pathway. The synthesis of structurally diverse analogs of an aldose reductase inhibitor and antibacterial activity investigation further demonstrated the utility of the current acylation reaction.
