Localization of a Plant Virus Combining in Situ Reverse Transcription-Polymerase Chain Reaction with a Light Microscope

Abstract:It is important to develop an optimized technique for location of plant viruses. In the study, a plant virus, Strawberry mottle virus (SMoV), was selected to study for location of virus. Combining in situ reverse transcription-polymerase chain reaction (RT-PCR) with a light microscope, a sensitive system for localization of plant virus in tissue level, was developed. By observation with the light microscope after in situ RT-PCR, a good deal of alcian blue precipitate could be found in palisade tissues, it suggested that SMoV could be located in the tissues. Because the chloroplasts were also in existence in palisade tissues, the virus was likely to tamper with the reproduction of the chloroplast cell, so it would tamper potentially with the photosynthesis and development of plant leaflets. To our knowledge, this is first report for location of strawberry virus in tissue. The system for location of virus may be developed to apply to other plant viruses.
