Abstract:The copper-catalyzed cyclization of activated alkenes with cyclobutanone O-acyl oximes under redox-neutral conditions has been reported. This facile protocol provided an efficient approach to a variety of cyanoalkylated oxindoles and dihydroquinolin-2(1H)-ones with a broad substrate scope and excellent functional group tolerance. In this reaction, sequential C-C bond cleavage, radical addition, and cyclization processes were involved, wherein multiple bonds were constructed in a one-pot reaction. Mechanistic studies suggest that the reaction probably proceeded via a radical pathway.

Abstract:A copper‐catalysed (diacetoxyiodo)benzene‐promoted aerobic esterification reaction of acetoacetamides was developed for the synthesis of oxamates, which are useful precursors in synthetic organic chemistry. This practical and mild synthetic approach proceeded at 25 °C under open‐air conditions and afforded methyl 2‐oxo‐2‐(phenylamino)acetates in good to excellent yields combined with C−C σ‐bond cleavage and formal oxidative C−H bond functionalization. A mechanism is proposed.

Abstract:A photoinduced copper-catalyzed alkoxyl triggered C−C bond cleavage/aminocarbonylation cascade is presented. Through adjusting the structure of alkoxyl radical precursors, functionalized lactones and keto-amides were synthesized with good yields and excellent functional group tolerance under redox-neutral conditions. Notably, this protocol enables the integration of lactone fragments with many amine drugs and drug fragments.

Abstract:Oxazoles are among the most important heterocyclic scaffolds in the fields of natural products and medicinal chemistry. Herein is developed a tandem reaction for the synthesis of a diverse array of 4,5-difunctionalized oxazoles utilizing easily-accessible ethyl 2-isocyanoacetate and aldehydes (26 examples, 31-83% yields). This cascade reaction is facilitated by catalytic CuBr and molecular oxygen as the oxidant. The process involves a catalytic cycloaddition oxidative dehydroaromatization mechanism. The broad aldehyde substrate scope, mild reaction conditions, and atom economy make this protocol an attractive alternative to access functionalized oxazoles.

Abstract:A simple and efficient copper-catalyzed approach to quinazoline derivatives has been developed, and the protocol uses readily available substituted (2-bromophenyl)methylamines and amides as the starting materials, and the cascade reactions were performed under air via sequential Ullmann-type coupling and aerobic oxidation without addition of any ligand or additive. The present method provides a convenient and practical strategy for synthesis of quinazoline derivatives.

Abstract:Copper(I)-catalyzed N-O bond formation reactions through vinyl nitrene-mediated pathway were described. The reactions of N-alkoxylbenzamides and 2H-azirines afforded alpha-amino oxime ethers in good to excellent yields at room temperature, which involved the cleavage of C-N and N-O bonds and the construction of new N-O and C-N bonds. It offers an efficient, regio- and stereoselective synthetic route for alpha-amino oxime ethers.

Abstract:A copper-catalyzed bisannulation reaction of malonate-tethered <i>O</i>-acyl oximes with pyridine, pyrazine, pyridazine, and quinoline derivatives has been developed for the concise synthesis of structurally novel dihydroindolizine-fused pyrrolidinones and their analogues. The present reaction shows excellent regioselectivity and stereoselectivity. Theoretical calculations reveal that the coordination effect of the carbonyl group in the nucleophilic substrate determines the excellent regioselectivity. Further functionalization of the generated dihydroindolizine-fused pyrrolidinone could be easily realized through substitution, Michael addition, selective aminolysis, and hydrolysis reactions.

Abstract:Copper(II)-mediated unprecedented intermolecular radical [3+2] annulation of N,N-dimethyl enaminones has been developed. The protocol is promoted simply by copper(II) chloride to access 5-acyl-3-furancarboxaldehydes with acceptable to good yields and broad substrate scope. This reaction allows the formation of multiple new bonds, including C(sp2)−O bond between two nucleophilic sites, C(sp2)−C(sp2) bond and C=O bond, through a radical cyclization process. Moreover, gram-synthesis and application research show the potential application value of this transformation in industry.

Abstract:A new copper-catalyzed aerobic synthesis of 4-oxobutanenitriles by nitrogen-migration cyanation and oxygenation of γ,δ-unsaturated keto oximes at room temperature is developed. By means of the copper(0) powder catalyst, a nitrogen-migration cyanation and oxygenation of unsaturated keto oximes with O2 via a radical intramolecular 5-exo-trig cyclization, C=C double bond cleavage and ring-opening process to assemble 4-oxobutanenitriles. Mechanistic studies indicate that these processes include radical initiating the reaction from the oxime side and the superoxide radical formation.
