Abstract:Fourteen difluoromethyl-containing 1,4-disubstituted 1,2,3-triazoles were synthesized via a novel copper-catalyzed click-multicomponent reaction of 2,2-difluoro-2-phenylsulfanylethanol, sodium azide and terminal alkynes in the presence of N-(p-toluenesulfonyl) imidazole, tetrabutylammonium iodide and triethylamine, followed by reductive cleavage of the phenylsulfanyl group using tributyltin hydride and azobisisobutyronitrile.

Abstract:Cu-II-catalysed diazo transfer and Cu-I-catalysed azide-alkyne 1,3-dipolar cycloaddition (CuAAC) "click chemistry" were used to synthesis C7N aminocyclitol-derivatised 1,2,3-triazoles. In the course of this work, the -N=N- moiety was transferred onto C7N aminocyclitols such as validamine, valienamine and valiolamine by employing imidazole-1-sulfonyl azide as the diazo transfer reagent with catalysis by Cu-II, Zn-II and Ni-II, in moderate to good yields. The obtained azidocyclitols were coupled with various terminal alkynes under modified Meldal's conditions with good to excellent yields. The stereo- and regiochemistry of the products were confirmed by 2D-NMR (NOESY and HMBC). One-pot syntheses of the corresponding 1,2,3-triazoles, as safer and more efficient procedures, were also investigated and gave moderate to good yields.

Abstract:Incorporation of a trifluoromethyl group with 1,2,3-triazoles motifs was described. We explored a click reaction approach for regioselective synthesis of 1-susbstituted-4-trifluoromethyl-1,2,3-triazoles in which 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) reacts with commercial 2-bromo-3,3,3-trifluoropropene (BTP) to form 3,3,3-trifloropropyne (TFP) in situ. Arising from merits associated with the availability and stability of BTP, and the high efficiencies of CuI/1,10-Phenanthroline (Phen)-catalyzed cycloaddition reactions of azides with alkynes, this readily performed click process takes place to form the target 1,2,3-triazoles in high yields, and with a wide azide substrate scope. The potential value of this protocol was demonstrated by its application to a gram-scale reaction.

Abstract:5-Halo-1,2,3-triazoles are important scaffolds in organic chemistry, but current click reactions cannot produce 1,4,5-trisubstituted 5-chloro-1,2,3-triazoles in a simple way. Herein, we disclosed a Cu(I)-catalyzed interrupted click reaction, using N-chlorophthalimide as an electrophilic chlorine source, enabling the facile synthesis of 1,4,5-trisubstituted 5-chloro-1,2,3-triazoles in one step from readily available terminal alkynes and azides. Complete control of regioselectivities with a broad substrate scope was accomplished by this approach. Furthermore, a novel epoxidation was developed using 5-chloro-triazole as substrate, which could be further applied in various organic transformations. Abstract:N-C bonded (non-bridged) 5-(1,2,3-triazol-1-yl)tetrazoles were synthesized by the Cu(I)-catalyzed 1,3-dipolar azide-alkyne cycloaddition click reaction using 5-azido-N-(propan-2-ylidene)-1H-tetrazole (1). For example, the click reaction of 1 in the presence of CuSO(4)5 H(2)O and Na ascorbate at 65-70 degrees C for 48 h in CH(3)CN/H(2)O co-solvent was found to be limited to only terminal alkynes that have electron-withdrawing groups, CF(3)C[triple chemical bond]CH (2 a) and SF(5)C[triple chemical bond]CH (2 b), giving rise to isopropylidene-[5-(4-trifluoromethyl-1,2,3-triazol-1-yl)tetrazol-1-yl]amine (3 a) and isopropylidene-[5-(4-pentafluorosulfanyl-1,2,3-triazol-1-yl)tetrazol-1-yl]amine (3 b) in 47 % and 66 % yields, respectively. When carried out under conditions using CuI and 2,6-lutidine as catalysts at 0 degrees C for 13 h in CHCl(3), the click reaction was versatile toward alkynes even those having electron-donating groups. Properties of new products were determined and compared with those of 1. Heats of formation, detonation pressures, detonation velocities and impact sensitivities are reported for these new 5-(1,2,3-triazol-1-yl)tetrazoles.

Abstract:The synthesis of 1-monosubstituted 1,2,3-triazoles was achieved using azides and propiolic acid by copper-catalyzed click cycloaddition/decarboxylation, which was easily carried out in N, N-dimethylformamide at room temperature or 60 degrees C with yields ranging from moderate to excellent. The reaction conditions were found to be successful for aryl and vinyl azide reactants, including analogues with various functionalities.

Abstract:A simple and efficient method was developed to synthesize 1,2,3-triazolo quinoxalines derivatives under click conditions. 1,3-Dipolar cycloaddition was carried out where 2-azido methyl quinoxaline and trimethyl silyl acetylene are used as dipole and dipolarophile respectively. The products were characterized by NMR and mass spectroscopic methods.

Abstract:2-Trifluoromethyl and 2-difluoromethyl substituted benzimidazole, benzoxazole and benzothiazole derivatives were efficiently prepared through a one-pot reaction of trifluoroacetic acid and difluoroacetic acid, respectively, with commercially available o-phenylenediamines, 2-aminophenols, and 2-aminobenzenethiols in good to excellent yields. Subsequential bromination of 2-difluoromethyl groups by photolysis with NBS led to the formation of bromodifluoromethyl benzo-1,3-diazoles which may be utilized to prepare the new generation of gem-difluoromethylene linked identical or non-identical twin molecules for drug synthesis.

Abstract:The reactions of α-bromoacetophenones with methylhydrazine in refluxing acetic acid generated 2-methyl-4-aryl-2H-[1,2,3]triazoles in good yields. The method was developed by the reactions of α-bromoacetophenones with phenylhydrazines in the presence of cupric ion, leading to 2,4-diary-2H-[1,2,3]triazoles. The structures were established on the basis of corresponding IR, 1H NMR, and elemental analysis data.

Abstract:A mild and efficient synthesis of 1,4-disubstituted 1,2,3-triazole linked pyrimidines is reported using click reactions. These compounds were prepared by the reaction of 4-propargyl substituted 5-iodo-6-methyl-2(methylthio) pyrimidine with aryl and benzyl azides via copper-catalyzed azide-alkyne cycloaddition reactions in the presence of sodium ascorbate, as a reducing agent. Furthermore, the synthesized compounds were screened against the two bacterial strains pseudomonas aeruginosa and saphylococcus aureus .

Abstract:An effective strategy for constructing substituted 1-benzyl-1 H -1,2,3-triazoles was developed through click reactions of benzylic halides with sodium azide and calcium carbide as sources of nitrogen and acetylene, respectively. The advantages of this method are an easily handled inexpensive source of acetylene, a wide range of substrates, satisfactory yields, and simple workup procedures, which could promote the use of calcium carbide as a sustainable acetylene source in modern industrial chemistry. Abstract:The syntheses of triazole and tetrazole compounds via intramolecular "Click" reaction were described.The intramolecular cycloaddition of alkyne or cyano triple bond with azido group was achieved under heating with or without Cu-powder as the catalyst,and piperazinoltriazole and piperazinoltetrazole were obtained in high yields.The structures were characterized by NMR,IR and MS.

Abstract:A number of enamine-functionalized 1,2,3-triazole derivatives have been prepared via the Cu-catalyzed three-component reaction of terminal alkyne, azide, and 2H-azirine. The reaction proceeds through insertion of vinyl nitrene into the C-Cu bond of the triazolyl-Cu species, providing an efficient and step- and atom-economic approach to the enamine-bearing polysubstituted 1,2,3-triazoles. The resulting triazoles were easily transformed to trisubstituted pyrazoles in the presence of a Rh catalyst.

Abstract:The divergent synthesis of 4‐amino‐1,2,3‐triazoles was accomplished through the Cu‐catalyzed cycloaddition of organic azides with alkynyliodonium(III) salts, followed by Cu‐catalyzed triazole‐amine coupling involving treatment with amines and amides. The in situ formation of 1,2,3‐triazole iodonium salts is crucial to this approach, and the utilization of a Cu‐catalyst facilitates cycloaddition and C‐N bond formation. The synthesis of 1,2,3‐triazolyliodonium(III) salts proposed in this study offers advantages, including mild reaction conditions, a broad substrate scope, and operational simplicity, while also providing efficient access to a complex and diverse range of 4‐amino‐1,2,3‐triazole derivatives.

Abstract:A Cu (I)-catalyzed three-component click reaction of alkyl bromide, terminal alkyne and sodium azide was developed in micro flow reactor. Sodium ascorbate was added as additive to prevent the oxidative coupling of desired product with terminal alkyne. Under optimized condition, various 1,4-disubstituted-1,2,3-triazoles were obtained in 85-95% yield with exclusive 1,4-regioisomeric selectivity. The present smooth procedure greatly accelerated the reaction due to the excellent mixing and mass transfer in micro flow system.

Abstract:A CuI-catalyzed three-component coupling of trifluoromethyl ketone N-tosylhydrazones, alkynes and azides has been developed. The reactionrepresents a straightforward method to access difluoromethylene substituted 1,2,3-triazoles. Mechanistically, it has been proposed that the reaction follows a pathway involving the formation of Cu(I)triazolide intermediate, Cu(I) carbene formation, migratory insertion, and -fluoride elimination. Thetransformation is featured by mild conditions, wide substrate scope and high efficiency.

Abstract:A new methodology for the Cu-catalyzed synthesis of 1H-1,2,3-triazoles from 1,1-dibromoalkenes and sodium azide is presented. Aryl dibromoolefins were efficiently converted into the corresponding 1,2,3-triazoles. A comprehensive number of functional groups were compatible with this reaction. 1,2,3-Triazoles were obtained in moderate to excellent yields.
