Abstract:An enantioselective one-pot Michael/Michael/Henry/hemiacetalization reaction between alpha,beta-unsaturated aldehydes, alpha-ketoamides, and nitroalkenes under mild conditions catalyzed by a diarylprolinol silyl ether has been developed. The sequential methodology provides a direct approach to a wide range of fully substituted chiral oxabicyclo[2.2.2]octanes with seven contiguous stereocenters in moderate to excellent yields (up to 99%), high to excellent diastereoselectivities (up to >25:1 dr), and high to excellent enantioselectivities (up to 99% ee).

Abstract:A good diastereo- and enantioselective 1,4-addition Michael reaction catalyzed by a chiral copper complex was developed in aqueous media. A series of nitro-containing pyrrolidones could be gained in high yields with excellent diastereoselectivities and good ee values by virtue of this developed method. It affords a facile access to construct carboncarbon bonds with water and air tolerance. Furthermore, the gram scale synthesis was conducted successfully to give rise to the corresponding products.

Abstract:The γ‐lactam ring is a prominent feature in medicinal chemistry, and its synthesis has garnered significant interest due to its valuable properties. Among the γ‐lactams, 2‐oxopyrrolidine‐3‐carbonitrile derivatives stand out as versatile synthons that can be readily transformed into a variety of other functional groups. In this work, we successfully synthesized highly functionalized 3‐cyano‐2‐pyrrolidinones with moderate to good overall yields using the Ugi reaction followed by intramolecular Michael addition. The process demonstrated excellent diastereoselectivity and showed good tolerance to a range of isonitriles and carbonyl compounds.

Abstract:A new series of mono-N-sulfonamides of C2-symmetrical 2′-bipyrrolidine have been designed, synthesized, and successfully applied as the organocatalysts to the enantio- and diastereoselective Michael addition of 4H-thiopyran-4-one to a variety of nitroolefins. Catalyst 10 was identified in the examinations to exhibit more superior catalytic ability than the traditional chiral pyrrolidine catalysts, giving high chemical yields, excellent diastereo- and enantioselectivities (up to 99:1 dr and 98% ee). The tert-butyl sulfonamide functionality of catalyst 10 is speculated to play crucial roles in the transition state as both bulky group and hydrogen-bond acceptor. Application of a representative Michael adduct has been also studied and proven to be useful for the diversity-oriented synthesis of enantiopure multiring heterocycles through a unique common bicyclic nitrone intermediate.

Abstract:Hochsubstituierte α,β-ungesättigte Cyclohexenone, ein zentrales Strukturmotiv in vielen Naturstoffen, sind mit der Titelreaktion zugänglich (siehe Schema). Die Diastereoselektivität bezüglich der beiden stereogenen Zentren (4- und 6-Position) ist hoch. Abstract:Bifunctional secondary amine-thiourea organocatalysts were successfully applied in the stereocontrolled synthesis of barbiturate-fused tetrahydropyrano scaffolds. Compared with typically used tertiary amine-thiourea organocatalysts, the developed catalysts exhibited excellent catalytic performance in the domino Michael-Michael reaction between N, N'-dimethylbarbituric acid and Morita-Baylis-Hillman acetates of nitroalkenes to yield pharmaceutically important heterocycles in good yields with excellent enantioselectivities. Moreover, this catalytic protocol can also be applied to synthesize biologically active pyranocoumarin compounds.

Abstract:A highly enantioselective Michael addition of alpha-substituted cyanoacetates to vinyl ketones was accomplished in the presence of simple bifunctional thiourea/tertiary amine organocatalysts. A number of alpha-aryl or alkyl cyanoacetates have been successfully applied to give multifunctional compounds with an all-carbon-substituted quaternary stereocenter in excellent enantioselectivities (82-97 % ee) and yields (61-99 %). The optical pure adducts could be smoothly converted to variously structured beta(2,2)-amino acid esters. Moreover, an interesting reaction model involving multiple hydrogen-bonding interactions amongst the thiourea/tertiary amine catalyst and the reactants has been proposed based on the absolute configuration of the adduct and computational studies.

Abstract:A highly enantioselective intramolecular 6-exo-trig aza-Michael addition was developed to afford chiral 3-substituted 1,2-oxazinanes in high yields (up to 99% yield) and good enantioselectivities (up to 98/2 er). Abstract:Highly enantioselective catalytic conjugate additions of 3-substituted benzofuran-2(3H)-ones to 1,1-bis(phenylsulfonyl)ethylene in the presence of catalysts based on Cinchona alkaloids and containing tertiary amine and thiourea groups have been developed. Good to excellent stereoselectivities (up to 99?% ee) could be achieved. An interesting effect of substituent positions on stereoselectivities was observed, and a theoretical study to explain this phenomenon was undertaken.
