Abstract:Spiro beta-lactams are important scaffolds in many bioactive compounds and natural products. The development of new synthetic methods for the formation of structurally diversified chiral spiro beta-lactams is highly important for the discovery of novel bioactive structures. In this work, a copper(I)-catalyzed asymmetric Kinugasa/aldol-type cascade reaction of ketone-tethered propiolamides and nitrones is developed and a set of novel chiral spiro beta-lactams with three contiguous stereogenic centers was achieved in satisfactory yields with high diastereo- and enantioselectivity. Control experiments suggested that a retro-aldol/aldol equilibrium process of the spirobilactam products is critical for the acquisition of high diastereoselectivity.

Abstract:Here, we report a chiral copper(II)-bisoxazoline-catalyzed enantioselective ring opening of cyclic diaryliodonium salts with heteroaryl thiols. The readily available 2-mercaptoben-zoxazole and 2-mercaptobenzothiazole derivatives reacted efficiently with cyclic diaryliodonium salts and afforded various axially chiral biaryls bearing iodine and sulfur functional groups in excellent yields and enantioselectivities. The products were further transformed into diverse enantiopure alkyl biaryl sulfides, which can be employed as chiral ligands.

Abstract:A mild copper-catalyzed asymmetric Kinugasa/Michael addition cascade process is developed. The reaction of α, β-unsaturated ester-tethered propiolamides with nitrones provides an efficient protocol for the construction of functionalized chiral 2,6-diazaspiro[3.4]octane-1,5-dione products in satisfactory yields and with high enantio- and diastereoselectivities.

Abstract:β-Lactams are important structural motifs because of their ubiquity in natural products and pharmaceuticals. We report herein a Cu-catalyzed intramolecular oxidative C(sp3 )-H amidation for the synthesis of β-lactams using tBuOOtBu. This method is based on Kharasch-Sosnovsky amidation and does not require prefunctionalization of C(sp3 )-H bonds or the installation of a directing group, thereby allowing for the straightforward synthesis of β-lactams. Our intramolecular functionalization protocol can be extended to diverse benzylic C(sp3 )-H bonds and shows excellent functional-group tolerance.

Abstract:Spiro rings such as spiroxindoles and spiroindanones are core skeletons in many pharmaceutically active molecules and natural products. Herein, we developed a copper-catalyzed cascade silylation-cyclization reactions of 1,6-diynes which enabled the rapid construction of silyl-functionalized spirocyclic compounds. A variety of racemic and optical pure spirocyclic oxindoles, indanone, quinolinone, and tetralone derivatives were obtained in high yields with excellent enantioselectivities and high stereoselectivities.

Abstract:Despite extensive efforts to develop γ-lactamization reactions for pyrrolidinone synthesis using either cyclometallation, C−H insertion, or radical C−H abstraction strategies, γ-lactamization reactions of aliphatic amides using practical catalysts and common protecting groups remain extremely rare. Herein we report copper-catalyzed γ-C(sp3)−H lactamization and iminolactonization of tosyl-protected aliphatic amides using inexpensive Selectfluor as the sole oxidant. A switchable selectivity of γ-Lactams or γ-iminolactones can be obtained by using two different sets of reaction conditions. Notably, structurally diverse spiro-, fused-, and bridged-lactams and iminolactones, as well as isoindolinones are accessible by this method. Further derivatization of the γ-lactam products enables the synthesis of a range of biologically important motifs, including γ-amino acids, δ-amino alcohols, and pyrrolidines.

Abstract:A new protocol for amide-directed Cu-catalyzed aminoalkylation of unactivated alkenes using cyclobutanone oxime esters as alkyl radical donors is developed. Both primary and secondary alkyl groups can be selectively installed at the C4 position of terminal or <i>cis</i>-internal 3-alkenamides in moderate to good yield. This reaction offers a useful method for the diastereoselective synthesis of β-lactams bearing 4-cyanoalkyl β-substituents. The use of a weakly coordinating counteranion as the Cu catalyst is critical for the formation of β-lactam products.

Abstract:A copper-catalyzed intramolecular asymmetric double C-arylation reaction was developed. The method provides a facile approach to chiral spiro bis-oxindoles in high yields and with good to excellent enantioselectivities. It also shows a broad substrate scope and good functional group tolerance. Density functional theory (DFT) calculations were conducted and revealed that the enantioselectivity is determined at the oxidative addition of Cu(I) into the second C-I bond.

Abstract:A photoinduced copper-catalyzed alkoxyl triggered C−C bond cleavage/aminocarbonylation cascade is presented. Through adjusting the structure of alkoxyl radical precursors, functionalized lactones and keto-amides were synthesized with good yields and excellent functional group tolerance under redox-neutral conditions. Notably, this protocol enables the integration of lactone fragments with many amine drugs and drug fragments.

Abstract:A copper-catalyzed efficient, operationally simple, general method for straightforward syntheses of spirocyclic compounds employing EMCCs (ethynyl methylene cyclic carbamates) as precursors reacting with different nucleophilic reagents was firstly reported. This transformation features mild reaction conditions, broad functional group tolerance, and an extensive substrate scope, thereby facilitating the efficient construction of an array of spirocyclic compounds. In addition, a scale-up experiment further highlighted the synthetic utility.

Abstract:In this paper, a non‐Cp* cobalt‐catalyzed [4 + 1] spirocyclization of aromatic amides with cyclic diazo compounds for synthesis of pentatomic spirolactams is presented. The reaction features with high regio/chemoselectivity by using cheap and commercially available base metal catalyzed C−H/N−H bond functionalization. Moreover, the practicability of this developed protocol is further demonstrated by the gram‐scale synthesis and removal of the directing group.

Abstract:A novel Cu(OAc)2·H2O catalyzed coupling reaction of 2‐iodobenzamides with malononitrile to afford 2‐substituted‐3‐amino‐4‐cyano‐isoquinoline‐1(2H)‐ones is described. The reaction proceeded in DMSO at 90°C for 5 h in nitrogen without external ligands. β‐Lactams, as nitrogen‐containing heterocycles with distinctive biological activities, have made significant contributions to the treatment of infectious diseases. This study which used inexpensive copper salts as catalysts, trichloroacetonitrile as a radical precursor, and potassium carbonate as base offers a concise route for the synthesis of β‐lactam compounds substituted with potentially pharmacologically active dichloroacetyl moieties. Preliminary mechanistic studies indicate that unactivated alkenes undergo sequential intermolecular radical addition and intramolecular amidation reactions. The copper salts undergo catalytic cycles involving Cu(I)/Cu(II)/Cu(III) species.

Abstract:A copper(II)-catalyzed protocol to construct trans-configured β-lactams and spirocyclic β-lactams from oximes and methyl propiolate has been developed, which features excellent substrate flexibility and diastereoselectivity (up to >99:1 dr). In situ FT-IR mechanistic experiments support that ketene species might be involved in the formation of β-lactams.

Abstract:AbstractAn unprecedented enantioselective synthesis of spiro‐γ‐lactams via a sequential C−H olefination/asymmetric [4+1] spirocyclization under a simple CoII/chiral spiro phosphoric acid (SPA) binary system is reported. A range of biologically important spiro‐γ‐lactams are obtained with high levels of enantioselectivity (up to 98 % ee). The concise, asymmetric synthesis of an aldose reductase inhibitor was successfully achieved. Notably, contrast to previous reports that relied on the use of cyclopentadienyl or its derivatives (achiral Cp*, CptBu, or chiral Cpx) ligated CoIII complexes requiring tedious steps to prepare, cheap and commercially available cobalt(II) acetate tetrahydrate was used as an efficient precatalyst.

Abstract:An expedient and efficient carbonylative protocol proceeded via palladium‐catalyzed tandem cyclic ketonization and carbonylative spirolactonization has been developed. This carbonylation reaction is compatible with a wide range of hydroxyalkyl ketone tethered benzyl chlorides bearing different functional groups and can be proceeded under comericially available Pd2(dba)3/BINAP catalyst system, providing a straithfowrd and practical approach to a broad range of [6,6]‐oxaspirolactones. The synthesized [6,6]‐oxaspirolactones are commonly found as core structures in natural products and can potentially serve as C2‐symmetric ligand synthons.
