The Effects of VEGF and Smac/DIABLO on Apoptosis and Chemosensitivity of Human Glioma Cells

Abstract:Objective: To investigate the roles of vascular endothelial growth factor (VEGF) and second mitochondrial activator of caspase (Smac) / direct IAP binding protein with low PI (DIABLO) in occurrence and development of glioma. Methods: The expression of VEGF gene was down-regulated in glioma U251 cells by lentivirus-mediated RNA interference. The expressions of VEGF and Smac/DIABLO mRNAs and protein were analyzed by real-time fluorogenic quantitative-PCR and Western blotting, respectively. The expression of phosphorylated extracellular regulated protein kinase (p-ERK) was detected by Western blotting. The cell cycle distribution induced by VEGF gene silencing was analyzed by flow cytometry, and the chemosensitivity of U251 cells was determined by MTT method. Results: The expression levels of VEGF and Smac/DIABLO mRNA and protein were down-regulated by VEGF gene silencing, and the expression level of pERK was down-regulated, which suggested that VEGF gene silencing was an upstream regulator for Smac/DIABLO and it could modulate the expression level of Smac/DIABLO by activation of the ERK signaling pathway. Furthermore, VEGF gene silencing resulted in the arrest of cell cycle at S phase and enhanced the effect of cisplatin on apoptosis of U251 cells. Conclusion: VEGF can influence the distribution of cell cycle and chemosensitivity to cisplatin in glioma. Smac/DIABLO is involved in the VEGF signaling pathway.
