Abstract:A yeast strain was isolated from sesame flower and classified as Candida tropicalis, with phenol, resorcin and bengidine as the sole carbon and energy resource. The strain can degrade phenol up to 14.88mmol/L within 48h and the highest phenol concentration to be degraded was 19.13mmol/L. Degradation efficiency of 100% was obtained when the strain was cultivated in NMS medium containing 10.0mmol/L phenol with pH ranging from 4.0 to 9.0 and at temperature ranging from 20℃ to 40℃. The optimum culture conditions for phenol degradation were pH 6.0~8.0, 28℃~30℃, with inoculation ratio ranging from 1% to 3%. Growth and phenol degradation was enhanced when additional nitrogen resource was added into culture medium.

Abstract:In this study, strains were isolated via selected medium from aerobic granules acclimated with phenol synthetic wastewater, which could use phenol as the sole carbon and energy source. Among of them, YA-3 was confirmed to possess the highest phenol tolerance and degradation rate, and identified as Candida tropicalis with the phylogenetic analysis of 26S rDNA ITS sequences. To investigate its degradation properties of phenol, inoculum volume, pH, temperature and second carbon source were optimized in phenol degradation experiment, and the optimal conditions were listed as inoculum ratio 3-7% (VN), pH 4.0-8.0 and 25-35 degrees C. Appropriate dosage of glucose could speed up the proliferation of Candida tropicalis, thus promoted the degradation of phenol. According to the degradation of phenol at different initial concentration, the inhibitory kinetics of phenol degradation was simulated using Haldane model. The model's kinetic parameters were evaluated as the maximum specific growth rate mu(X,max) = 0.418hr(-1), the half-saturation coefficient K-S = 74.69mg / L, the inhibition coefficient for cell growth K-i = 432.3 8mg / L, and the maximum specific phenol degradation rate was 0.447hr(-1). These results demonstrated that Candida tropicalis had high feasibility and engineering value, which could be used as bioaugmentation consortia to shorten the acclimation phase of aerobic granules or remedy phenol contamination in situ.

Abstract:Phenol is a ubiquitous pollutant and can contaminate natural water resources. Hence, the removal of phenol from wastewater is of significant importance. A series of biological methods were used to remove phenol based on the natural ability of microorganisms to degrade phenol, but the tolerance mechanism of phenol-degraded strains to phenol are not very clear. Morphological observation on Candida tropicalis showed that phenol caused the reactive oxygen species (ROS) accumulation, damaging the mitochondrial and the endoplasmic reticulum. On the basis of transcriptome data and cell wall susceptibility analysis, it was found that C. tropicalis prevented phenol-caused cell damage through improvement of cell wall resistance, maintenance of high-fidelity DNA replication, intracellular protein homeostasis, organelle integrity, and kept the intracellular phenol concentration at a low level through cell-wall remodeling and removal of excess phenol via MDR/MXR transporters. The knowledge obtained will promote the genetic modification of yeast strains in general to tolerate the high concentrations of phenol and improve their efficiency of phenol degradation.

Abstract:This study examined the phenol degradation capabilities and oxidative stress responses of Candida tropicalis SHC-03, demonstrating its metabolic superiority and resilience compared to Saccharomyces cerevisiae BY4742 in a culture medium with phenol as the sole carbon source. Through comparative growth, transcriptomic, and metabolomic analyses under different phenol concentrations, this study revealed C. tropicalis SHC-03's specialized adaptations for thriving in phenol as the sole carbon source environments. These include a strategic shift from carbohydrate metabolism to enhanced phenol degradation pathways, highlighted by the significant upregulation of genes for Phenol 2-monoxygenase and Catechol 1,2-dioxygenase. Despite phenol levels reaching 1.8 g/L, C. tropicalis exhibits a robust oxidative stress response, efficiently managing ROS through antioxidative pathways and the upregulation of genes for peroxisomal proteins like PEX2, PEX13, and PMP34. Concurrently, there was significant upregulation of genes associated with membrane components and transmembrane transporters, enhancing the cell's capacity for substance exchange and signal transduction. Especially, when the phenol concentration was 1.6 g/L and 1.8 g/L, the degradation rates of C. tropicalis towards it were 99.47 and 95.91%, respectively. Conversely, S. cerevisiae BY4742 shows limited metabolic response, with pronounced growth inhibition and lack of phenol degradation. Therefore, our study not only sheds light on the molecular mechanisms underpinning phenol tolerance and degradation in C. tropicalis but also positions this yeast as a promising candidate for environmental and industrial processes aimed at mitigating phenol pollution.
Biodegradation of Phenol by Candida Tropicalis Sp.: Kinetics, Identification of Putative Genes and Reconstruction of Catabolic Pathways by Genomic and Transcriptomic Characteristics

Abstract:Candida tropicalis sp. was isolated with predominant biodegradation capability to phenol compounds, even with high concentration or in acid environment. The biodegradation of phenol was evaluated at the following concentrations 10-1750 mg L-1, the strain exhibited well biodegradation efficiency. The maximum specific growth rate was 0.660 h-1 and the specific biodegradation rates was 0.47 mg (phenol) [(mg (VSS) h]-1. Differentially expressed genes were screened out, and results revealed a complete process of energy and carbon metabolism. The genes' arrangements and phylogenetic information showed the unique genetic characteristics of the strain. Catabolic pathways were reconstructed and some key phenol-degrading genes were obviously upregulated, including pheA, catA, OXCT and fadA. A notable detail that CMBL encoding carboxymethylenebutenolidase was speculated to be involved in a shortened pathway of phenol biodegradation, thereby contributing to the reconstruction of the novel phenol catabolic pathway through the hydrolases of dienelactone. Finally, key enzymes were verified by the analysis of specific activity.

Abstract:Abstract Background: Phenolics derived from lignocellulose severely influence cell growth and fermentation ability in yeast. Due to its high tolerance for various stresses, Candida tropicalis has recently emerged as a promising strain for the degradation of industrial sewage. Great efforts have been made on studying the degradation machineries of phenol by C. tropicalis, but the global mechanisms underlying its tolerance of high concentrations of phenolics remain ambiguous. Since the substructure and toxicity of phenol are similar to those of other phenolic compounds derived from lignin hydrolysis, we treated C. tropicalis cells with phenol as a representative substance for the phenolic compounds. Results: We found that after treatment with 0.5 g/l, 1.0 g/l, and 2.0 g/l phenol, the growth of C. tropicalis SHC-03 was inhibited, but recovered completely after a lag phase. Although we found that C. tropicalis could degrade phenol when phenol was used as the sole carbon source, it could not degrade phenol in YPD medium, implying that increased tolerance for phenol contributed to the recovery of cell growth. Morphological observation demonstrated that phenol could induce reactive oxygen species (ROS) accumulation, mitochondrial membrane damage, and damage to the endoplasmic reticulum (ER), but did not affect the structural stability of chromatin. On the basis of transcriptome data and cell wall susceptibility analysis, a number of genes related to DNA repair, DNA replication, heat shock protein (HSP)-mediated proteasomal degradation, autophagy, accumulation of fatty acids, cell wall remodeling, and MDR/MXR transporters were found. These genes may play key roles in the increased tolerance of C. tropicalis to phenol stress. Conclusion: C. tropicalis appears to prevent phenol-induced cell damage through maintenance of high-fidelity DNA replication, intracellular protein homeostasis, organelle integrity, and keep the intracellular phenol concentration at a low level through cell-wall remodeling and removal of excess phenol via MDR/MXR transporters. The knowledge obtained from this research provided us with a global understanding of phenolic tolerance mechanisms in C. tropicalis and will promote the genetic modification of yeast strains to develop biological products using lignocellulosic hydrolysates as a carbon resource.
Identification of High-Strength Phenol Degrading Strain Candida Tropicalis Z-04 and Degradation Condition Optimization

Abstract:A phenol-degrading strain,capable of tolerating high-strength phenol up to 2.0g/L,was isolated from phenol-degrading aerobic granules.According to its morphological and ITS gene analysis,the strain was identified as the genus Candida tropicalis,and was named Candida tropicalis Z-04.A Central Composite Design(CCD) was employed to optimize the phenol degrading conditions.The obtained experiments data were analyzed by Design-Expert software,base on which,the effect of influence factors and their interaction on phenol degradation efficiency were investigated.The results showed that the optimal degradation conditions were: yeast extract 0.41 g/L,phenol 1.03 g/L,inoculum size 1.43%(volume ratio) and temperature 30.04 ℃,and the maximum removal efficiency of phenol(99.10%) was obtained under the optimum conditions.

Abstract:Strain Candida tropicalis was isolated from acclimated activated sludge, and was identified as a member of the genus Candida. Phenol biodegradation using a pure culture of C. tropicalis was studied. The results showed that C. tropicalis has pretty high phenol degradation potential, which could thoroughly degrade the phenol of 2000mgl−1 in the mineral salt medium within 66h. High inoculum volume lessened phenol toxic property for the cells and increased phenol biodegradation velocity. However, for a certain starting inoculum, with the step increase of phenol concentration, substrate inhibition was obviously enhanced, and more phenol consumption was not assimilated by cell for growth, but was used to counteract strong substrate inhibition. In addition, the cell growth and phenol degradation intrinsic kinetics of C. tropicalis in batch cultures were also investigated over a wide range of initial phenol concentrations (0–2000mgl−1) using Haldane model. The results received in these experiments demonstrated that the Haldane kinetic model adequately described the dynamic behavior of the phenol biodegradation by the strain of C. tropicalis.

Abstract:A bacterial strain DDT-6 (D6) capable of utilizing dichlorodiphenyltrichloroethane (DDT), dichlorodiphenyldichloroethane (DDD), and dichlorodiphenyldichloroethylene (DDE) (DDTs) as its sole carbon and energy source was isolated and identified as Sphingobacterium sp. The degradation of DDTs by strain D6 in mineral salt medium and in field soil was investigated. The half-lives of the degradation of DDTs increased with increasing concentration ranging from 1 to 50mgL−1. Favorable degradation conditions for DDTs by strain D6 were found to be pH 7.0 and 30°C. The degradation of DDTs by strain D6 was found to be statistically significantly enhanced (p≤0.05) by the addition of glucose. Based on the metabolites detected, a pathway was proposed for DDT degradation in which it undergoes dechlorination, hydrogenation, dioxygenation, decarboxylation, hydroxylation, and phenyl ring-cleavage reactions to complete the mineralization process. The addition of strain D6 into the contaminated soils was found to statistically significantly enhance (p≤0.05) the degradation of DDTs. The results indicate that the isolate D6 can be used successfully for the removal or detoxification of residues of DDTs in contaminated soil.

Abstract:An effective phenol-degrading strain was isolated from activated sludge,and was identified as Candida maltosa based on morphological,biochemical and physiological experiments.The strain could utilize phenol as sole carbon and energy source.The optimum degradation conditions were temperature of 30 ℃,initial pH of 7.0 and shaking speed of 200 r/min.The degradation experiment indicated that the strain could completely degrade 1 800 mg/L phenol within 69 h.The higher the phenol concentration was,the stronger the substrate inhibition was.Growth kinetics of the strain fitted the Haldane equation,which could be used to simulate phenol degradation at different initial concentrations.

Abstract:Biodegradation of phenol and 4-chlorophenol (4-cp) using a pure culture of Candida tropicalis was studied. The results showed that C. tropicalis could degrade 2,000 mg l −1 phenol alone and 350 mg l −1 4-cp alone within 66 and 55 h, respectively. The capacity of the strain to degrade phenol was obviously higher than that to degrade 4-cp. In the dual-substrate system, 4-cp intensely inhibited phenol biodegradation. Phenol beyond 800 mg l −1 could not be degraded in the presence of 350 mg l −1 4-cp. Comparatively, low-concentration phenol from 100 to 600 mg l −1 supplied a sole carbon and energy source for C. tropicalis in the initial phase of biodegradation and accelerated the assimilation of 4-cp, which resulted in the fact that 4-cp biodegradation velocity was higher than that without phenol. And the capacity of C. tropicalis to degrade 4-cp was increased up to 420 mg l −1 with the presence of 100–160 mg l −1 phenol. In addition, the intrinsic kinetics of cell growth and substrate degradation were investigated with phenol and 4-cp as single and mixed substrates in batch cultures. The results illustrated that the models proposed adequately described the dynamic behaviors of biodegradation by C. tropicalis .

Abstract:Aerobic granules effectively degrade phenol at high concentrations. This work cultivated aerobic granules that can degrade phenol at a constant rate of 49 mg-phenol/g x VSS/h up to 1,000 mg/L of phenol. Fluorescent staining and confocal laser scanning microscopy (CLSM) tests demonstrated that an active biomass was accumulated at the granule outer layer. A strain with maximum ability to degrade phenol and a high tolerance to phenol toxicity isolated from the granules was identified as Candida tropicalis via 18S rRNA sequencing. This strain degrades phenol at a maximum rate of 390 mg-phenol/g x VSS/h at pH 6 and 30 degrees C, whereas inhibitory effects existed at concentrations >1,000 mg/L. The Haldane kinetic model elucidates the growth and phenol biodegradation kinetics of the C. tropicalis. The fluorescence in situ hybridization (FISH) and CLSM test suggested that the Candida strain was primarily distributed throughout the surface layer of granule; hence, achieving a near constant reaction rate over a wide range of phenol concentration. The mass transfer barrier provided by granule matrix did not determine the reaction rates for the present phenol-degrading granule.
STUDY ON THE STRAIN OF PHENOL-DEGRADING BACTERIUM C-14-1 AND ITS DEGRADING CAPABILITY

Abstract:An aerobic bacterial strain Nocardia sp.C-14-1 was isolated from an acrylic fiber wastewater.It was identified to have good phenol-degrading capability after acclimating.The bacterium growth and phenol degradation was measured by UV spectrophotometer.Results showed the lag period was extended when increasing the concentration of phenol, but still shorter than that of reported strains.At 35℃ and at log period, the shorter degrading time for 600 mg·L-1 phenol was within 20 hours.The degradation of phenol happened in log period and the optimum conditions were 35℃, pH 7～8 and aeration.The degrading dynamics was well recorded with Andrews equation.
Isolation of Phenol-Degrading Strain and Preliminary Studies on Its Degrading Characteristics

Abstract:In order to investigate the biodegradation mechanisms of phenol by aerobic granular sludge,18 strains were isolated from the sludge using basal medium with phenol as sole carbon source and strain 1# which have high degradation efficiency was studied.This strain could endurance concentration of phenol up to 2000 mg/L when inoculated in medium with phenol as the sole energy and carbon source.The optimization experiments of phenol degradation with strain 1# showed that,with inoculum amount 3%,temperature 30 ℃,pH value 6.85,liquid volume in flask 100 mL and rate of shaking incubator 120 r/min,phenol with initial concentration 1000 mg/L could be degraded up to 95% after 24 h.Moreover,it was revealed that addition of glucose with low concentration(0.5~1.0 g/L)could promote growth of strain,thus increase the phenol removal.The colony morphology of strain 1# was circular,smooth and pinkish,and it was identified as Candida tropicalis according to the 23Sr DNA sequence analysis.
Phenol biodegradation by the yeast Candida tropicalis in the presence of m-cresol

Abstract:Biodegradation of phenol and m-cresol using a pure culture of Candida tropicalis was studied. The results showed that C. tropicalis could degrade 2000mgl−1 phenol alone and 280mgl−1 m-cresol alone within 66 and 52h, respectively. The capacity of the strain to degrade phenol was obviously higher than that to degrade m-cresol. The presence of m-cresol intensely inhibited phenol biodegradation. Only 1000mgl−1 phenol can be completely degraded in the presence of 280mgl−1 m-cresol. Abstract:Brevibacterium aureum DG-12, a new bacterial strain isolated from active sludge, was able to degrade and utilize cyfluthrin as a growth substrate in the mineral medium. Response surface methodology using central composite rotatable design of cultural conditions was successfully employed for optimization resulting in 88.6% degradation of cyfluthrin (50 mg L-1) within 5 days. The bacterium degraded cyfluthrin by cleavage of both the carboxylester linkage and diaryl bond to form 2,2,3,3-tetramethyl-cyclopropane-methanol, 4-fluoro-3-phenexy-benzoic acid, 3,5-dimethoxy phenol, and phenol, and subsequently transformed these compounds with a maximum specific degradation rate, half-saturation constant and inhibition constant of 1.0384 day(-1), 20.4967 mg L-1, and 141.9013 mg L-1, respectively. A novel degradation pathway for cyfluthrin was proposed based on analysis of these metabolites. In addition, this strain was found capable of degrading a wide range of synthetic pyrethroid insecticides. Our results suggest that B. aureum DG-12 may be an ideal microorganism for bioremediation of the pyrethroid-contaminated environments. (C) 2013 Elsevier Ltd. All rights reserved.
Optimization of Phenol Degradation by Candida Tropicalis Z-04 Using Plackett-Burman Design and Response Surface Methodology

Abstract:Statistical experimental designs were used to optimize the process of phenol degradation by Candida tropicalis Z-04, isolated from phenol-degrading aerobic granules. The most important factors influencing phenol degradation (p < 0.05), as identified by a two-level Plackett-Burman design with 11 variables, were yeast extract, phenol, inoculum size, and temperature. Steepest ascent method was undertaken to determine the optimal regions of these four significant factors. Central composite design (CCD) and response surface analysis were adopted to further investigate the mutual interactions between these variables and to identify their optimal values that would generate maximum phenol degradation. The analysis results indicated that interactions between yeast extract and temperature, phenol and temperature, inoculum size and temperature affected the response variable (phenol degradation) significantly. The predicted results showed that the maximum removal efficiency of phenol (99.10%) could be obtained under the optimum conditions of yeast extract 0.41 g/L, phenol 1.03 g/L, inoculum size 1.43% (V/V) and temperature 30.04 degrees C. These predicted values were further verified by validation experiments. The excellent correlation between predicted and experimental values confirmed the validity and practicability of this statistical optimum strategy. This study indicated the excellent ability of C. tropicalis Z-04 in degrading high-strength phenol. Optimal conditions obtained in this experiment laid a solid foundation for further use of this microorganism in the treatment of high-strength phenol effluents.

Abstract:A novel phenol-degrading filamentous fungus, strain JH6, was isolated from activated sludge and identified as a member of Paecilomyces variotii based on standard morphological and phylogenetic analysis. The degradation assays suggested that the strain was able to utilize phenol as the sole source of carbon and energy at concentrations up to 1800 mg/l. The strain exhibited optimum phenol degradation performance with the addition of 100 mg/l glucose at pH 5, 37°C. Haldane's model could be fitted to the growth kinetics data well over a wide range of initial phenol concentrations (100-1800 mg/l), with kinetic values μ(max)=0.312 h(-1), K(s)=130.4 mg/l, and K(i)=200 mg/l. The decay coefficient was found to be 0.0073 h(-1). Complete phenol degradation by strain JH6 could be achieved in the presence of other toxicants, such as m-cresol and quinoline, which were often found in the real phenol-containing wastewater.

Abstract:A phenol-degrading bacterial strain named PN6-15 was isolated from phenol contaminated soil which is Gram positive and can grow on a medium with phenol as the sole carbon and energy sources.The optimum degradation condition is temperature 30 ℃ and pH 7.0,respectively.PN6-15 could degrade phenol at a concentration as high as 500 mg/L completely in 36 h.Heavy metal ions such as Cu2+,Hg2+ and Ag+ had different degrees of inhibition on phenol degradation,while no obvious effect of Zn2+ on phenol degradation was observed.Metabolic pathway research showed that PN6-15 could be induced to synthesize extracellular catechol-2,3-dioxygenase.Homolog comparison analysis of 16S rDNA sequences showed that PN6-15 belongs to the genus clostridium.
