Clinical significance of VEGF-C expression and LVD detection in predicting the prognosis of stage Ⅱ postoperative colorectal cancer

Abstract:Objective: To evaluate the relationship of vascular endothelial growth factor-C (VEGF-C) and lymphatic vessel density (LVD) with clinicopathological characteristics and prognosis by detecting the expression of VEGF-C and the level of LVD to provide some useful references for comprehensive therapy of stage II colorectal cancer. Methods: We collected and statistically analyzed the clinical data of 56 colorectal cancer patients admitted to our hospital between January 2000 and December 2005. We studied their diagnosis, therapy and survival status, analyzed the factors that may affect the prognosis, and detected the expression of VEGF-C and the level of LVD of 56 stage II postoperative colorectal cancer tissues and 13 normal colorectal tissues by P-V immunohistochemical technique. Results: The expression of VEGF-C and the level of LVD in cancer tissues were obviously higher than those in normal tissues (P<0.05). One-year, three-year and five-year survival rate of the 56 stage II colorectal cancer patients was 98%, 85% and 70% , respectively. The overall survival time and disease-free survival time were not significantly shorter in VEGF-C positive group than in VEGF-C negative one. The prognosis of patients with positive VEGF-C expression was poor, but without significant difference (P>0.05), while that of patients with high level of LVD was significantly poor (P<0.05). The prognosis of patients with both high level of LVD and positive expression of VEGF-C was significantly poorer (P = 0.036). Multifactor analysis revealed that VEGF-C and LVD were independent factors affecting overall survival in stage II colorectal cancer patients. Conclusion: The expressions of VEGF-C and LVD in cancer tissues are obviously high. The combined detection of the expression of VEGF-C and the level of LVD might indicate the prognosis of colorectal cancer.
