Genome variation and genetic diversity of triploid loquats

Abstract:Triploid loquats (Eriobotrya japonica Lindl.) were investigated from both spontaneous triploid seedlings from open pollinated diploids (referred to as natural triploids) and diploid × tetraploid crosses (referred to as synthetic triploids). Genome variation and genetic diversity of these triploids were analyzed by amplified fragment length polymorphism technique. The results indicated that 12 pairs of AFLP primers amplified 2454 bands in 6 natural triploid loquats, 204.5 bands per primer. Triploids, in contrast with diploid maternal parent, had 208 difference bands, a polymorphism rate of 8.5%. There were 2 types: added bands (112) at a rate of 53.9%, and lost bands (96) at a rate of 46.1%. The AFLP analysis of 7 synthetic triploid lines showed that 12 pairs of primers amplified 3122 bands, 260.2 bands per primer, and there were 294 different bands, a polymorphism rate of 9.4%. There were four types of different bands: added, lost, paternal, and maternal. The numbers of sites for these four types were 82, 58, 49 and 105, respectively; the different rates were 27.9, 19.7, 16.7 and 35.7%.
