Effects of the Exogenous Hormones on Rapid Propagation of Hypocotyl of the Medicinal Plant of Periploca Sepium Bunge

Abstract:OBJECTIVE To study the technique of the rapid propagation of the medicinal plant of Periploca sepium Bunge,to study the effects of the exogenous hormones on regeneration of hypocotyl and to provide scientific basis for the industrialization.METHODS Select the exogenous hormones of different type and concentration to induce the callus and differentiate the adventitious bud,multiply the bud and induce the plantlet to take root.RESULTS Take the hypocotyl as explant.The appropriate hormone combination of inducting callus and differentiating bud is IBA 0.05-0.5 mg·L-1+BA 0.5-2.0 mg·L-1.The best combination is IBA 0.1 mg·L-1+BA 1.0 mg·L-1.The appropriate hormone combination of successive transfer culture is IBA 0.05-0.5 mg·L-1+BA 0.5-1.0 mg·L-1.The best combination is IBA 0.05 mg·L-1+BA 1.0 mg·L-1.The appropriate hormone of rootage is IBA 0.2～0.5 mg·L-1.The best hormone is IBA 0.5 mg·L-1.CONCLUSION By sifting the appropriate hormone combination,the objective of rapid propagation can be realized.
