Inhibition effects of endostatin on EDA expression of colorectal carcinoma cells to affect formation of human lymph endothelial cells

Abstract:OBJECTIVE: To investigate whether recombinant human endostatin (Endostar) can suppress the tumor lymphangiogenesis through EDA pathway. METHODS: The control group and endostatin experimental groups were set up. The methods of immunofluorescence and tube-like formation assay were used to examine its inhibitive effect on the lymphangiogenesis. The experimental groups were compared with the control group by the method of Western blot to test the different expressions of integrin α9 among the experimental groups and control group. RESULTS: EDA was highest expressed in SW480, and endostatin supressed the expression of EDA dose-dependently. The supernatant of single-cultured SW480 (conditioned medium I) enhanced the tube-like formation ablities of hLECs, these effects were weakened when SW480 was treated with endostatin (conditioned medium II, P<0.01), and as a special receptor of EDA, the level of integrinα9 of hLECs declined accordingly (P<0.05). CONCLUSIONS: EDA is important in tumor lymphangiogenesis and endostatin can restrain the tube-like formation ablities of hLECs through the EDA-integrin α9 pathway.
