Study of the levels of IL-8 and GM-CSF secreted by bone marrow stromal cells in psoriasis patients

Abstract:Objective: To reveal the abnormity of bone marrow microenvironment of psoriasis patients by comparing the levels of IL-8 and GM-CSF secreted by bone marrow stromal cells between patients and control. Methods: Bone marrow mononuclear cells of patients and control were separated by density gradient centrifugation. Bone marrow stromal cells were cultured with the method of adherence. When the cells were cultured for three passages and another 72 h, bone marrow stromal cells and supernatant liquid were collected. The phenotypes of cells were identified by flow cytometry and the levels of IL-8 and GM-CSF were assayed by ELISA kits. Results: The purity of bone marrow stromal cells was over 90%. The level of IL-8 secreted by bone marrow stromal cells of patients was lower than that of control (P<0.05), however, the level of GM-CSF was no significant difference between the two groups (P>0.05). Conclusions: The level of IL-8 secreted by psoriatic bone marrow stromal cells is abnormal, indicating that psoriatic bone marrow microenvironment may be abnormal.
