Expressions of CD40,TNF-α and Caspase-3 in Colonic Tissue of Rats with Experimental Colitis

Abstract:Background: Inflammatory bowel disease (IBD) is a cause-of-complex disease, its potential etiological factors including immunity, genetics, environment and infection, but the exact etiology and pathogenesis remain unclear. Aims: To investigate the changes of expressions of costimulatory molecule CD40, proinflammatory cytokine TNF-α, and apoptotic effector enzyme caspase-3 in colonic tissue of rats with experimental colitis, and to study the roles of these factors in IBD. Methods: Fifty healthy female Sprague-Dawley rats were randomized into two groups, 40 in TNBS-colitis group, and 10 in control group. Rats were sacrificed in batches on day 7, day 14 and day 28 after model construction, respectively, and the expressions of CD40, TNF-α and caspase-3 in colonic tissue were determined by real time-PCR and immunohistochemical method. Results: The expressions of CD40 and TNF-α mRNA and protein in colonic tissue of model rats on day 7, day 14 and day 28 were higher than those of the controls, the differences were statistically significant (P<0.05) except for CD40 expression on day 28; while the expression of caspase-3 mRNA and protein in colonic tissue of model rats were significantly increased on day 7, and decreased on day 14 and day 28 when compared with the controls (P<0.05). Conclusions: Immunologic abnormality, inflammatory mediator and apoptosis might be involved in the pathogenesis of IBD, which provides a potential highlight for the treatment of IBD.
